[Glutamine metabolic reprogramming in regulating the occurrence and development of osteosarcoma].

Excessive cellular proliferation and metabolic reprogramming are important characteristics of cancer cells. Cancer cells promote excessive proliferation and growth by altering coordinated metabolic pathways. In terms of glucose metabolism, most cancer cells exhibit increased glucose uptake and lactate production even under aerobic conditions, known as the Warburg effect. Increased glucose uptake not only provides energy but also supplies essential carbon sources for the biosynthesis of nucleotides, lipids, and proteins. During this process, decreased pyruvate dehydrogenase activity leads to di

Metabolism and Nutrition of L-Glutamate and L-Glutamine in Ruminants.

Although both L-glutamate (Glu) and L-glutamine (Gln) have long been considered nutritionally nonessential in ruminants, these two amino acids have enormous nutritional and physiological importance. Results of recent studies revealed that extracellular Gln is extensively degraded by ruminal microbes, but extracellular Glu undergoes little catabolism by these cells due to the near absence of its uptake. Ruminal bacteria hydrolyze Gln to Glu plus ammonia and, intracellularly, use both amino acids for protein synthesis. Microbial proteins and dietary Glu enter the small intestine in ruminants. Bo

The essential role of glutamine metabolism in diabetic cardiomyopathy: A review.

Diabetic cardiomyopathy (DCM) is a pathophysiological condition caused by diabetes mellitus and is the leading cause of diabetes mellitus-related mortality. The pathophysiology of DCM involves various processes, such as oxidative stress, inflammation, ferroptosis, and abnormal protein modification. New evidence indicates that dysfunction of glutamine (Gln) metabolism contributes to the pathogenesis of DCM by regulating these pathophysiological mechanisms. Gln is a conditionally essential amino acid in the human body, playing a vital role in maintaining cell function. Although the precise molec

Evaluation of Glutamine Utilization in Patients With Sickle Cell Disease.

Glutamine (Gln) was FDA-approved in 2017 to reduce acute sickle cell disease (SCD) pain and acute chest syndrome. However, typical pediatric patients with SCD exhibit moderate adherence, measured by a medication possession ratio <80%. This study examined Gln utilization in a "real-world" clinical setting to determine factors influencing medication adherence and to characterize the impact of an interdisciplinary team approach at an institution with specialty pharmacy services. A retrospective chart review identified 40 patients prescribed Gln by sickle cell specialists over a 2-year period and

Glutamine transporters in mammalian cells and their functions in physiology and cancer.

The SLC (solute carrier)-type transporters (~400 in number) in mammalian cells consist of 52 distinct gene families, grouped solely based on the amino acid sequence (primary structure) of the transporter proteins and not on their transport function. Among them are the transporters for amino acids. Fourteen of them, capable of transporting glutamine across the plasma membrane, are found in four families: SLC1, SLC6, SLC7, and SLC38. However, it is generally thought that the members of the SLC38 family are the principal transporters for glutamine. Some of the glutamine transporters are obligator

A tracer bolus method for investigating glutamine kinetics in humans.

Glutamine transport between tissues is important for the outcome of critically ill patients. Investigation of glutamine kinetics is, therefore, necessary to understand glutamine metabolism in these patients in order to improve future intervention studies. Endogenous glutamine production can be measured by continuous infusion of a glutamine tracer, which necessitates a minimum measurement time period. In order to reduce this problem, we used and validated a tracer bolus injection method. Furthermore, this method was used to measure the glutamine production in healthy volunteers in the post-abso

Glutamine as indispensable nutrient in oncology: experimental and clinical evidence.

BACKGROUND: In hypermetabolic situations, glutamine is intensively used by rapidly dividing cells such as enterocytes, lymphocytes, and fibroblasts as nitrogen source and/or alternative energy fuel. It is hypothesized that in cancer patients the increased glutamine demands of the host increase the capacity of endogenous production resulting in a strong glutamine deprivation with detrimental effects on organ functions. In long-term periods of cancer cachexia, an adequate nutrition support including glutamine can essentially contribute to cover glutamine needs and, thus, to spare energy reserves

[Protective effect of glutamine on microcirculation of the intestine in experimental colitis].

Parenteral glutamine application can stabilize intestinal permeability and mucosal integrity. It is not known whether glutamine influences the microcirculation in the large intestine. This study thus employs intravital microscopy to investigate mucosal microcirculation in the ascending and descending Colon of Sprague-Dawley rats with TNBS colitis. The animals were randomized and treated with either saline solution (placebo) or glutamine (verum). In the severely inflamed descending colon, TNBS colitis involves a significant capillary blood flow reduction that is not improved by glutamine applic

Role of glutamine in bacterial transcytosis and epithelial cell injury.

BACKGROUND: L-Glutamine is the principal energy source for small intestinal enterocytes. Diminution of intestinal function, mucosal atrophy, and increased bacterial translocation have been noted during total parenteral nutrition (TPN). In a rat model of glutamine starvation, we previously showed that luminal glutamine is essential for optimal intestinal function. In this study, we examined the effect of apical vs basolateral glutamine on bacterial translocation in a Caco-2 cell culture system and bacteria-induced tissue injury in a weanling rabbit ileal loop model. METHODS: Caco-2 cells were

Glutamine synthetase in liver of the American alligator, Alligator mississippiensis.

Glutamine synthetase was shown to be localized in liver mitochondria of the American alligator, Alligator mississippiensis, by immunofluorescent staining of frozen liver sections and by the detection of enzymatic activity and immunoreactive protein in the mitochondrial fraction following subcellular fractionation of liver tissue by differential centrifugation. The primary translation product of alligator liver glutamine synthetase mRNA was shown to have an Mr = 45,000 which is similar if not identical in size to that of the mature subunit. This mRNA was found to be heterogeneous in size with a

Regulation of glutamine metabolism in vitro by bicarbonate ion and pH.

The effect of variations of medium pH and bicarbonate concentration on glutamine oxidation was studied in slices and mitochondria from dog renal cortex. Decreasing pH and bicarbonate concentration increased the rate of oxidation of glutamine-U-(14)C to (14)CO(2) in both slices and mitochondria, an effect comparable to the acute stimulation of glutamine utilization produced by metabolic acidosis. Decreases in the concentration of glutamate and alpha-ketoglutarate, which accompany metabolic acidosis in the intact animal, also occurred in tissue slices when pH and [HCO(3) (-)] were lowered; decre

Growth hormone together with glutamine-containing total parenteral nutrition maintains muscle glutamine levels and results in a less negative nitrogen balance after surgical trauma.

BACKGROUND: Muscle protein catabolism, reflected by a decrease in glutamine (GLN), a decrease in muscle protein synthesis, and a negative nitrogen balance can be reduced by either administration of GLN or growth hormone (GH). In this study, the effects of a combination of GH and GLH were studied. METHODS: Patients (n = 16) undergoing abdominal operation were given total parenteral nutrition (TPN) containing either GLN alone or GLN together with GH (GH/GLN) during 3 postoperative days. The amino acid concentration and protein synthesis in muscle tissue and the nitrogen balance were measured.

Rare-Earth-Coordinated Glutamine Nanostructure-Induced "Pseudo-Overnutrition" Metabolic Therapy.

Tumor cells display a strong dependency on nutrients, yet nutrient-deprivation strategies often fail due to their metabolic plasticity and compensatory mechanisms, and can even promote invasion and metastasis. Here, we propose a metabolic therapeutic strategy that leverages rather than suppresses tumor nutrient addiction, termed "pseudo-overnutrition". This approach induces tumor cells to continuously uptake nutrient analogs that cannot be effectively utilized, thereby decoupling nutrient input from nutrient utilization and triggering metabolic collapse. To validate this concept, we designed a

Glutamine Metabolism-associated Molecular Heterogeneity and Prognosis in Pancreatic Adenocarcinoma.

BACKGROUND: Pancreatic Adenocarcinoma (PAAD) is characterized by pronounced molecular heterogeneity and profound metabolic reprogramming. Glutamine metabolism plays a critical role in sustaining tumor growth and shaping the tumor microenvironment; however, its contribution to molecular heterogeneity, immune characteristics, and clinical outcomes in PAAD remains incompletely understood. METHODS: We conducted an integrative transcriptomic analysis utilizing transcriptomic and clinical data from TCGA, GTEx, and an independent GEO cohort to focus on genes involved in glutamine metabolism. Glutamine alleviates Staphylococcus aureus-induced mastitis by modulating macrophage polarisation.

Mastitis, a major dairy industry challenge, is often caused by Staphylococcus aureus (S. aureus). Previous research linked gut microbiota disruption-induced by vancomycin in mice-to reduced microbial glutamine and increased mastitis susceptibility. This study hypothesizes that oral glutamine may regulate macrophage function and mitigate S. aureus mastitis. Our results showed that glutamine attenuated S. aureus-induced mastitis, protected the blood-milk barrier function, increased M2-type macrophages in the mammary gland, and increased the concentration of its metabolite α-Ketoglutaric acid (αK
