Glycosides as Potential Medicinal Components for Ulcerative Colitis: A Review.

Ulcerative colitis (UC) is a chronic, non-specific disease of unknown etiology. The disease develops mainly in the rectum or colon, and the main clinical symptoms include abdominal pain, diarrhea, and purulent bloody stools, with a wide variation in severity. The specific causative factors and pathogenesis of the disease are not yet clear, but most scholars believe that the disease is caused by the interaction of genetic, environmental, infectious, immune, and intestinal flora factors. As for the treatment of UC, medications are commonly used in clinical practice, mainly including aminosalicyl

Carbon tetrachloride-free allylic halogenation-mediated glycosylations of allyl glycosides.

The allylic bromination of allyl glycosides is conducted using NBS/AIBN reagents in (EtO)2CO and PhCF3 solutions, without using CCl4 as a solvent. The activated mixed halo-allyl glycosides led to glycosylations, mediated by a triflate, in a latent-active manner, with the allyl glycosides acting as donors and acceptors. Systematic glycosylation studies are performed with different triflate promoters, non-glycosyl acceptors and various allyl glycosyl donors. One-pot allylic halogenations and subsequent glycosylations are developed in PhCF3 solutions. This newer glycosylation method is utilized t

Cyanogenic glycosides in plant-based foods available in New Zealand.

Cyanogenic glycosides occur in a wide range of plant species. The potential toxicity of cyanogenic glycosides arises from enzymatic degradation to produce hydrogen cyanide, which may result in acute cyanide poisoning and has also been implicated in the aetiology of several chronic diseases. One hundred retail foods were sampled and analysed for the presence of total hydrocyanic acid using an acid hydrolysis-isonicotinic/barbituric acid colourimetric method. Food samples included cassava, bamboo shoots, almonds and almond products, pome fruit products, flaxseed/linseed, stone fruit products, li

Electrochemical detection of cyanogenic glycosides after enzymatic post-column cleavage.

Crude and partly purified extracts from Helix pomatia and linamarase from cassava were immobilized on columns packed with porous glass or silica and used as post-column reactors in the high-performance liquid chromatography of cyanogenic glycosides. Sodium hydroxide (2 M) was added to the flowstream after the enzyme-reactor resulting in the formation of cyanide, which was then detected at a silver electrode by an amperometric measurement at 0 V with reference to a silver-silver chloride electrode. The selective detection of cyanide allows measurements in a complex matrix. The response is linea

Urinary amino acid glycosides and oligosaccharides in health and disease.

Due to heterogeneity and lack of suitable fractionation techniques it is only recently that more detailed information about urinary low molecular weight carbohydrates has been obtained. Ingemar Berggård was one of the pioneers who opened up this field and outlined the directions towards fruitful progress. Today more than one hundred well characterized oligosaccharides and amino acid glycosides have been described. This review covers the latest development and discusses the mechanisms behind the urinary excretion.

Phylogeny constrains and environment modulates glycoside hydrolase architecture.

Glycoside hydrolases (GHs) are central to microbial carbohydrate degradation and frequently occur as multidomain proteins containing accessory domains involved in substrate binding, localization, and retention. However, the extent to which GH multidomain architectures are shaped by phylogeny versus environmental adaptation remains poorly understood. Here, we analyzed 158,829 GH-containing proteins from 17,108 high-quality metagenome-assembled genomes (MAGs) in the GEM catalog, focusing on major GH families involved in polysaccharide and oligosaccharide degradation. We developed domain-domain a

Functional, structural, and taxonomical diversity of glycoside transporters in bacteria.

Glycoside metabolization is a crucial function in bacteria and a key feature in fermentation and synthetic biology. To feed on various glycosides with complex structures, bacteria have developed very diverse mechanisms to recognize, transport, and degrade them. Despite their crucial role, transporters are hard to identify and study due to challenges in expressing membrane proteins in recombinant form and the fact that most bacteria are uncultivable. In this review, we present the available methods and technologies for identifying and characterizing bacterial glycoside transporters. We consolid

Allosteric indicator displacement enzyme assay for a cyanogenic glycoside.

Indicator displacement assays (IDAs) represent an elegant approach in supramolecular analytical chemistry. Herein, we report a chemical biosensor for the selective detection of the cyanogenic glycoside amygdalin in aqueous solution. The hybrid sensor consists of the enzyme β-glucosidase and a boronic acid appended viologen together with a fluorescent reporter dye. β-Glucosidase degrades the cyanogenic glycoside amygdalin into hydrogen cyanide, glucose, and benzaldehyde. Only the released cyanide binds at the allosteric site of the receptor (boronic acid) thereby inducing changes in the affinit

An efficient fermentation method for the degradation of cyanogenic glycosides in flaxseed.

Recently, flaxseed has become increasingly popular in the health food market because it contains a considerable amount of specific beneficial nutrients such as lignans and omega-3 fatty acids. However, the presence of cyanogenic glycosides (CGs) in flaxseed severely limits the exploitation of its health benefits and nutritive value. We, therefore, developed an effective fermentation method, optimised by response surface methodology (RSM), for degrading CGs with an enzymatic preparation that includes 12.5% β-glucosidase and 8.9% cyanide hydratase. These optimised conditions resulted in a maximu

Rate of hydrolysis and degradation of the cyanogenic glycoside - dhurrin - in soil.

Cyanogenic glycosides are common plant toxins. Toxic hydrogen cyanide originating from cyanogenic glycosides may affect soil processes and water quality. In this study, hydrolysis, degradation and sorption of dhurrin (4-hydroxymandelonitrile-beta-d-glucoside) produced by sorghum has been studied in order to assess its fate in soil. The log K(ow) of dhurrin was -1.18+/-0.08 (22 degrees C). Hydrolysis was a first-order reaction with respect to dhurrin and hydroxyl ion concentrations. Half lives ranged from 1.2h (pH 8.6; 25 degrees C) to 530d (pH 4; 25 degrees C). The activation energy of hydroly

The neurotoxic effects of prenatal cardiac glycoside exposure: a hypothesis.

Cardiac glycosides (CGs) are beneficial in treating cardiac conditions; depending on time and dosage, they can also be toxic as they regularly cross the blood brain barrier and the placenta and may affect the unborn baby. This paper therefore focuses on the effects of CGs administered to the mother on normal cellular physiology of the foetus with specific reference to neural tissue. CGs act by binding to the Na+/K+-ATPase and decrease or inhibit Na+-K+ pump activity. In the foetus, CGs may disrupt ion homeostasis. An over-dosage of CGs or when it is taken during pregnancy, can also affect the
