Hamamelis virginiana L. Leaf Extracts Inhibit the Growth of Antibiotic-Resistant Gram-Positive and Gram-Negative Bacteria.

Virginian witch hazel (WH; Hamamelis virginiana L.; family: Hamamelidaceae) is a North American plant that is used traditionally to treat a variety of ailments, including bacterial infections. Solvents of varying polarity (water, methanol, ethyl acetate, hexane and chloroform) were used to prepare extracts from this plant. Resuspensions of each extract in an aqueous solution were tested for growth-inhibitory activity against a panel of bacteria (including three antibiotic-resistant strains) using agar disc diffusion and broth microdilution assays. The ethyl acetate, hexane and chloroform extra

Genetic and Chemical Screening Reveals Targets and Compounds to Potentiate Gram-Positive Antibiotics against Gram-Negative Bacteria.

Gram-negative bacteria are intrinsically resistant to a plethora of antibiotics that effectively inhibit the growth of Gram-positive bacteria. The intrinsic resistance of Gram-negative bacteria to classes of antibiotics, including rifamycins, aminocoumarins, macrolides, glycopeptides, and oxazolidinones, has largely been attributed to their lack of accumulation within cells due to poor permeability across the outer membrane, susceptibility to efflux pumps, or a combination of these factors. Due to the difficulty in discovering antibiotics that can bypass these barriers, finding targets and com

Butenolide, a Marine-Derived Broad-Spectrum Antibiofilm Agent Against Both Gram-Positive and Gram-Negative Pathogenic Bacteria.

Bacterial biofilm can cause nosocomial recurrent infections and implanted device secondary infections in patients and strongly promotes development of pathogenic drug resistance in clinical treatments. Butenolide is an effective anti-macrofouling compound derived from a marine Streptomyces sp., but its antibiofilm efficacy remains largely unexplored. In the present study, the antibiofilm activities of butenolide were examined using biofilms formed by both Gram-positive and Gram-negative pathogenic model species. Four Escherichia coli strains, Pseudomonas aeruginosa, and methicillin-resistant S

Inactivation dynamics of 222 nm krypton-chlorine excilamp irradiation on Gram-positive and Gram-negative foodborne pathogenic bacteria.

The object of this study was to elucidate the bactericidal mechanism of a 222 nm Krypton Chlorine (KrCl) excilamp compared with that of a 254 nm Low Pressure mercury (LP Hg) lamp. The KrCl excilamp had higher bactericidal capacity against Gram-positive pathogenic bacteria (Staphylococcus aureus and L. monocytogenes) and Gram-negative pathogenic bacteria (S. Typhimurium and E. coli O157:H7) than did the LP Hg lamp when cell suspensions in PBS were irradiated with each type of UV lamp. It was found out that the KrCl excilamp induced cell membrane damage as a form of depolarization. From the stud

Comparative in vitro activity of imipenem against gram-positive and gram-negative aerobic bacteria from clinical isolates.

Imipenem is a member of a new class of beta-lactam antibiotics, carbapenems, with a very broad antibacterial spectrum. In this work we evaluated the in vitro activity of imipenem against a variety of bacterial strains isolated from clinical specimens as well as the activity of other beta-lactam antibiotics. The results obtained with 501 bacterial strains show that imipenem is active on both gram-negative and gram-positive microorganisms isolated from different infections. The in vitro inhibitory activity is greater than that of aztreonam, cefotaxime, ceftriaxone, piperacillin, amikacin, and ne

Vicia ervilia lectin (VEA) has an antibiofilm effect on both Gram-positive and Gram-negative pathogenic bacteria.

Bacterial growing resistance to antibiotics poses a critical threat to global health. This study investigates, for the first time, the antibiofilm properties of Vicia ervilia agglutinin (VEA) from six different V. ervilia accessions against pathogenic bacteria, and the yeast Candida albicans. In the absence of antimicrobial properties, purified VEA significantly inhibited biofilm formation, both in Gram-positive and Gram-negative bacteria, but not in C. albicans. With an inhibitory concentration ranging from 100 to 500 µg/ml, the VEA antibiofilm activity was more relevant against the Gram-posi

The Etiological Bacterial Spectrum of Neck Abscesses of Lymph Node Origin - Gram-Positive and Gram-Negative Bacteria.

According to our medical practice as maxillofacial and oral surgeons, operated patients with purulent infections of the cervical lymph nodes are not many in number. On the other hand, the presence of a purulent infection requires not only the surgical evacuation of the pus but also the application of antimicrobial preparations. This necessitates good knowledge of the spectrum of the bacterial causative agents of the disease, the determination and analysis of which is the purpose of this original article. The bacteria studied in 181 patients with a mean age of 26.25 years, ranging between 29 da

Rapid detection and differentiation of Gram-negative and Gram-positive pathogenic bacteria in urine using TaqMan probe.

Urinary tract infection has been shown to be quite complicated and often difficult to diagnose and treat. For appropriate diagnosis, it is very important to find the correct Gram stain classification as soon as possible, especially in severe cases where there is a possibility of severe sepsis developing. In order to solve this problem, we developed a new method to detect a Gram stain of bacteria obtained from 1 ml of urine from urinary tract infection patients using a consensus real-time PCR protocol with a TaqMan probe that allows detection of spiked bacterial 16S DNA from urine. We extracted

Lipidome plasticity and envelope remodeling under environmental stress in Gram-negative bacteria.

Gram-negative bacterial cell envelopes comprise complex lipid components essential for survival and virulence, functioning as a dynamic barrier against environmental stress. Beyond canonical biosynthetic pathways, Gram-negative bacteria employ multiple adaptive strategies to preserve envelope integrity under nutrient limitation and host-imposed pressures. These include stress-induced lipid remodeling processes such as headgroup substitution, acyl-chain modification, and host lipid-driven membrane incorporation through phosphatidylcholine biosynthesis and the assimilation of host-derived fatty

Random Forest Modeling to Predict Small Molecule Accumulation in Gram-Negative Bacteria.

Despite extensive efforts over the past ∼60 years to discover new classes of Gram-negative-active antibiotics, the development pipeline remains relatively dry. These failures can be largely ascribed to the complexity of the Gram-negative membranes and an inadequate understanding of physicochemical properties associated with compound permeation, efflux evasion, and overall accumulation in Gram-negative pathogens. Recent work using unbiased accumulation assays and advanced chemical descriptors has utilized machine learning algorithms, particularly random forest modeling, to correlate physicochem

Organosulfur scaffolds as quorum sensing and biofilm modulators in Gram-negative bacteria.

Quorum sensing (QS) is a cell-density-dependent communication system that coordinates bacterial virulence, biofilm formation, motility, and stress adaptation, making it a compelling target for antivirulence intervention. Due to the possibility of sulfonyl (-SO₂-) and sulfinyl (-S = O-) functional groups to impart specific electronic and steric properties that can modulate bacterial signaling and receptor interactions, organosulfur compounds are increasingly gaining attention. This review highlights recent advances in the development of sulfonyl- and sulfinyl-containing organosulfur compounds a

A potential benzimidazole-derived compound, NSC348884, combats carbapenem-resistant gram-negative bacteria.

Carbapenem-resistant Acinetobacter baumannii (CRAB) has been classified by WHO as a critical-priority pathogen that poses the most serious threat to public health and urgently requires the development of novel antimicrobial agents. Here, we identified a benzimidazole analogs NSC348884 with additive or synergistic therapeutic effects against carbapenem-resistant gram-negative bacteria (CR-GNB) when combined with imipenem. Further investigation found that NSC348884 exhibited direct antibacterial efficacy without inducing resistance, and comparable anti-biofilm activity to polymyxin B (PMB) again

Colistin Resistance in Gram-Negative Bacteria: Mechanisms, Transmission, and Novel Intervention Strategies.

Multidrug resistance (MDR) in Gram-negative bacteria is a global issue and needs to be addressed urgently. MDR can emerge through genetic mutations and horizontal gene transfer and deteriorate under antibiotic selective pressure. The emergence of resistance to last-resort antibiotics, which are used to treat MDR bacteria, is of particular concern. Colistin has been recognized as a last-line antibiotic for the treatment of MDR Gram-negative bacterial infections caused by Escherichia coli, Acinetobacter baumannii, Klebsiella pneumoniae, and Pseudomonas aeruginosa. Recently, the increasing report

Molecular Epidemiological Surveillance of Carbapenem-Resistant Gram-Negative Bacteria in Southern Lebanon.

Introduction: Carbapenem-resistant Gram-negative bacteria (CR-GNB) are rapidly spreading pathogens that increase morbidity and mortality in hospital settings and significantly restrict available treatment options worldwide. The lack of molecular epidemiological data and the limited use of next-generation sequencing (NGS) in South Lebanon have hindered comprehensive surveillance efforts. This study represents the first molecular characterization of CR-GNB in this region. Methods: A total of 477 clinical Gram-negative bacterial isolates were collected from intensive care unit (ICU) patients admi

Management of Severe Infections: Multidrug-Resistant and Carbapenem-Resistant Gram-Negative Bacteria.

This article provides an overview of the mechanisms behind carbapenem resistance and the antibiotic management for severe infections caused by key carbapenem-resistant gram-negative bacteria, specifically Enterobacterales, Acinetobacter baumannii, and Pseudomonas aeruginosa. For Enterobacterales, it highlights the relative advantages of meropenem-vaborbactam and imipenem-relebactam in treating Klebsiella pneumoniae carbapenemase (KPC)-producing strains with resistance to ceftazidime-avibactam, the preference for ceftazidime-avibactam in addressing oxacillin-hydrolyzing carpapenemase (OXA)-48-l

Fluorescent probe-based detection of outer membrane damage of Gram-negative bacteria.

Infections caused by drug-resistant bacteria, particularly Gram-negative species, represent one of the most significant global public health challenges. The outer membrane (OM) is a crucial target for the development of drugs against Gram-negative bacteria. For the confirmation of the mechanism of OM-targeted drugs, the evaluation of OM damage is necessary. In this study, we optimized the method for detecting OM damage employing N-phenyl-1-naphthylamine (NPN) and ethidium bromide (EtBr) as probes in Escherichia coli (E. coli), including the bacterial loading, probe concentration, and incubatio

Noncytotoxic polymyxin derivatives enhance antibiotic action against multidrug-resistant Gram-negative bacteria.

The widespread emergence of multidrug-resistant (MDR) Gram-negative bacteria prompted the reintroduction of polymyxins in the clinic despite their adverse effects. Ongoing research is primarily focused on the development of non-nephrotoxic and -neurotoxic polymyxins as not only standalone agents but also as potentiators that enhance the activity of a partner antibiotic. Safer derivatives of polymyxin B3, a minor component of polymyxin B, were synthesized and utilized as a potentiator of multiple antibiotics. Compound 1, consisting of Dap residues, was nontoxic to kidney cells and is a promisin

MCR-1-dependent lipid remodelling compromises the viability of Gram-negative bacteria.

The global dissemination of the mobilized colistin resistance gene, mcr-1, threatens human health. Recent studies by our group and others have shown that the withdrawal of colistin as a feed additive dramatically reduced the prevalence of mcr-1. Although it is accepted that the rapid reduction in mcr-1 prevalence may have resulted, to some extent, from the toxic effects of MCR-1, the detailed mechanism remains unclear. Here, we found that MCR-1 damaged the outer membrane (OM) permeability in Escherichia coli and Klebsiella pneumonia and that this event was associated with MCR-1-mediated cell s
