Identification of the gregarization-associated dark-pigmentotropin in locusts through an albino mutant.

In response to crowding, locusts develop characteristic black patterns that are well discernible in the gregarious phase at outbreaks. We report here a dark-color-inducing neuropeptide (dark-pigmentotropin) from the corpora cardiaca of two plague locusts, Schistocerca gregaria and Locusta migratoria. The chromatographic isolation of this neuropeptide was monitored by using a bioassay with an albino mutant of L. migratoria. The neurohormone, consisting of 11 amino acids, is identical to [His7] corazonin, previously isolated from corpora cardiaca of another acridid without known function. The pr

Plague-Positive Mouse Fleas on Mice Before Plague Induced Die-Offs in Black-Tailed and White-Tailed Prairie Dogs.

Plague is a lethal zoonotic disease associated with rodents worldwide. In the western United States, plague outbreaks can decimate prairie dog (Cynomys spp.) colonies. However, it is unclear where the causative agent, Yersinia pestis, of this flea-borne disease is maintained between outbreaks, and what triggers plague-induced prairie dog die-offs. Less susceptible rodent hosts, such as mice, could serve to maintain the bacterium, transport infectious fleas across a colony, or introduce the pathogen to other colonies, possibly facilitating an outbreak. Here, we assess the potential role of two

An experimental study on the mechanics and control of SMA-actuated bioinspired needle.

Active needles demonstrate improved accuracy and tip deflection compared to their passive needle counterparts, a crucial advantage in percutaneous procedures. However, the ability of these needles to effectively navigate through tissues is governed by needle-tissue interaction, which depends on the tip shape, the cannula surface geometry, and the needle insertion method. In this research, we evaluated the effect of cannula surface modifications and the application of a vibrational insertion technique on the performance of shape memory alloy (SMA)-actuated active needles. These features were in

Aggregation pheromone 4-vinylanisole promotes the synchrony of sexual maturation in female locusts.

Reproductive synchrony generally occurs in various group-living animals. However, the underlying mechanisms remain largely unexplored. The migratory locust, Locusta migratoria, a worldwide agricultural pest species, displays synchronous maturation and oviposition when forms huge swarm. The reproductive synchrony among group members is critical for the maintenance of locust swarms and population density of next generation. Here, we showed that gregarious female locusts displayed more synchronous sexual maturation and oviposition than solitarious females and olfactory deficiency mutants. Only th

Matched-filter coding of sky polarization results in an internal sun compass in the brain of the desert locust.

Many animals use celestial cues for spatial orientation. These include the sun and, in insects, the polarization pattern of the sky, which depends on the position of the sun. The central complex in the insect brain plays a key role in spatial orientation. In desert locusts, the angle of polarized light in the zenith above the animal and the direction of a simulated sun are represented in a compass-like fashion in the central complex, but how both compasses fit together for a unified representation of external space remained unclear. To address this question, we analyzed the sensitivity of intr

Responses of compass neurons in the locust brain to visual motion and leg motor activity.

The central complex, a group of midline neuropils in the insect brain, plays a key role in spatial orientation and navigation. Work in locusts, crickets, dung beetles, bees and butterflies suggests that it harbors a network of neurons which determines the orientation of the insect relative to the pattern of polarized light in the blue sky. In locusts, these 'compass cells' also respond to simulated approaching objects. Here, we investigated in the locust Schistocerca gregaria whether compass cells change their activity when the animal experiences large-field visual motion or when the animal is

Disruption of segmentation in a short germ insect embryo. II. The structure of segmental abnormalities induced by heat shock.

A heat shock (of 15 min at 48 degrees C) given to early embryos of the locust, Schistocerca gregaria, results in localized abnormalities in the segment pattern subsequently formed. Most defects involve two consecutive segments of the thorax or abdomen, and these are analysed in detail. The abdominal defects fall into three main classes each of which involves the absence of a particular region of the segment pair and, in one class, duplication of the region which remains. The thoracic defects similarly involve absence of parts of the segments and the formation of a single limb base from which o

Discrete roles for secreted and transmembrane semaphorins in neuronal growth cone guidance in vivo.

From the initial stages of axon outgrowth to the formation of a functioning synapse, neuronal growth cones continuously integrate and respond to multiple guidance cues. To investigate the role of semaphorins in the establishment of appropriate axon trajectories, we have characterized a novel secreted semaphorin in grasshopper, gSema 2a. Sema 2a is expressed in a gradient in the developing limb bud epithelium during Ti pioneer axon outgrowth. We demonstrate that Sema 2a acts as chemorepulsive guidance molecule critical for axon fasciculation and for determining both the initial direction and su

Behavioural phase change in the Australian plague locust, Chortoicetes terminifera, is triggered by tactile stimulation of the antennae.

Density-dependent phase polyphenism is a defining characteristic of the paraphyletic group of acridid grasshoppers known as locusts. The cues and mechanisms associated with crowding that induce behavioural gregarization are best understood in the desert locust, Schistocerca gregaria, and involve a combination of sensory inputs from the head (visual and olfactory) and mechanostimulation of the hind legs, acting via a transient increase in serotonin in the thoracic ganglia. Since behavioural gregarization has apparently arisen independently multiple times within the Acrididae, the important ques

Life history and host specificity of Mediorhynchus centurorum Nickol 1969 (Acanthocephala: Gigantorhynchidae).

Examination of 1905 arthropods from a swamp and surrounding pasture in sothern Louisiana revealed larval specimens of Mediorhynchus centurorum in 8 of 228 woodroaches, Parcoblatta pensylvanica, but in no other species. The life cycle was confirmed by feeding eggs of M. centurorum to laboratory-reared woodroaches. Cystacanths later recovered from the woodroaches developed into mature worms when pipetted into esophaguses of red-bellied woodpeckers, Centurus carolinius: red-headed woodpeckers, Melanerpes erythrocephalus; yellow-shafted flickers, Colaptes auratus; and a hairy woodpecker, Dendrocop

Dax, a locust Hox gene related to fushi-tarazu but showing no pair-rule expression.

We describe an unusual Antennapedia class homeobox gene from the grasshopper Schistocerca gregaria (Orthoptera, African Plague Locust). Its sequence is not sufficiently similar to that of any other insect Hom-Hox gene to identify it unambiguously, but short conserved elements suggest a relationship to the segmentation gene fushi-tarazu, (ftz). We term it Sg Dax (divergent Antennapedia class homeobox gene). Antibodies raised against the protein encoded by this gene reveal two phases of expression during embryogenesis. In the early embryo, it is a marker for the posterior part of the forming emb

Cell recognition during neuronal development.

Insect embryos, with their relatively simple nervous systems, provide a model system with which to study the cellular and molecular mechanisms underlying cell recognition during neuronal development. Such an approach can take advantage of the accessible cells of the grasshopper embryo and the accessible genes of Drosophila. The growth cones of identified neurons express selective affinities for specific axonal surfaces; such specificities give rise to the stereotyped patterns of selective fasciculation common to both species. These and other results suggest that early in development cell linea

Biomechanical Factors in the Adaptations of Insect Tibia Cuticle.

Insects are among the most diverse groups of animals on Earth. Their cuticle exoskeletons vary greatly in terms of size and shape, and are subjected to different applied forces during daily activities. We investigated the biomechanics of the tibiae of three different insect species: the desert locust (Schistocerca gregaria), American cockroach (Periplaneta americana) and Death's Head cockroach (Blaberus discoidalis). In a previous work, we showed that these tibiae vary not only in geometry (length, radius and thickness) but also in material quality (Young's modulus) and in the applied stress r

Body condition constrains immune function in field populations of female Australian plague locust Chortoicetes terminifera.

The insect innate immune system comprises both humoral and cellular defence responses. In the laboratory, the insect immune system is well characterized. In the field, however, little is known about the role of constitutive insect immune function and how it varies within and between populations. Laboratory studies suggest that host nutrition has significant impact upon insect immune function. Thus, the rationale for this study was to sample natural populations of the Australian Plague Locust Chortoicetes terminifera to establish whether locust body condition (as determined by protein and lipid

[Using of antibodies against Microsporia Hsp70 family proteins for analysis of secretome of intracellular parasites].

Microsporidia is a large group of fungi-related unicellular parasites with obligate intracellular lifestyle. Unlike other protozoan intracellular parasites (Kinetoplastida and Apicomplexa), most microsporidian species develop in direct contact with the host cell cytoplasm. This fact, acquisition of unique transporters to exploit host metabolic system (alongside the strong minimization of own machinery) and predicted repertoire of microsporidia secretome altogether suggest an active role of parasite proteins in the control of infected cell. Lack of information about secretome of microsporidia i

Phytotoxic and antifungal metabolites from Curvularia sp. FH01 isolated from the gut of Atractomorpha sinensis.

Two main phytotoxic and antifungal phthalic acid butyl isobutyl ester (1) and radicinin (2) were isolated from the culture of Curvularia sp. FH01, a fungus residing in the Atractomorpha sinensis gut. The structures of isolated metabolites were established on the basis of spectral analysis. Metabolites 1 and 2 exhibited significant phytotoxic activity against the radical growth of Echinochloa crusgalli with their IC(50) values of 61.9 and 5.9 μg/mL, respectively, which were comparable to that 2,4-dichlorophenoxyacetic acid (2.0 μg/mL) used as a positive control. The antifungal test results show

Aerial treatment of the Australian plague locust, Chortoicetes terminifera (Orthoptera: Acrididae) with Metarhizium anisopliae (Deuteromycotina: Hyphomycetes).

Between October 1999 and April 2000, nearly 4000 ha of nymphal bands and adult swarms of Chortoicetes terminifera (Walker) were aerially treated using a ULV oil formulation of strain FI-985 of Metarhizium anisopliae var. acridum. During the mild weather (maxima 22-30 degrees C) of spring (October), there was little change in nymphal bands during the first week but at all doses between 25-100 g (1-4 x 10(12) conidia) ha(-1), the bands rapidly declined 9-12 days after treatment reaching > 90% mortality by 14 days. Metarhizium persisted for some time as there was 50% mortality of locusts fed vege

Flavonoid-mediated inhibition of ovarian glutathione S-transferase in Schistocerca gregaria: Biochemical characterization and in vitro evaluation.

The desert locust, Schistocerca gregaria, remains one of the most destructive agricultural pests, posing a serious threat to crop production and food security. Current control strategies rely heavily on synthetic insecticides, raising concerns about environmental contamination and the development of insecticide resistance. Glutathione S-transferases (GSTs) are major phase II detoxification enzymes involved in xenobiotic metabolism and insecticide tolerance; however, their physiological role in reproductive tissues remains poorly understood. This study aimed to purify and characterize ovarian G

Linking shape conspicuous asymmetry with shape covariation patterns and performance in the insect head and mandibles.

Bilateral symmetry is widespread across animals, yet, among bilaterians, many cases of conspicuous asymmetries evolved. This means that bilaterally homologous structures on the left and right sides display divergent phenotypes. The evolution of such divergent phenotypes between otherwise similarly shaped structures can be thought to be favored by modularity, but this has rarely been studied in the context of left-right differences. Here, we provide an empirical example, using geometric morphometrics to assess patterns of asymmetry and covariation between landmark partitions in a grasshopper wi
