Effects of Global Ripk2 Genetic Deficiency in Aged Mice following Experimental Ischemic Stroke.

Besides the loss of blood and oxygen reaching the ischemic tissue, many secondary effects of ischemic stroke can cause additional tissue death, including inflammation, oxidative stress, and proteomic disturbances. Receptor-interacting serine/threonine kinase 2 (RIPK2) is an important mediator in the post-stroke inflammatory cascade that responds to signals and molecular patterns released by dead or dying cells in the ischemic area. We hypothesize that RIPK2 signaling worsens injury and neurological recovery post-stroke and that global deletion of Ripk2 will be protective following ischemic str

FK506 Enhancement of Neuromuscular Junction Recovery After Nerve Injury Is Macrophage-Dependent.

INTRODUCTION: Motor recovery following nerve injury is dependent on time required for muscle reinnervation. This process is imperfect, however, and recovery is often incomplete. At the neuromuscular junction (NMJ), macrophage signaling aids muscle reinnervation. Tacrolimus (FK506) treatment speeds functional recovery through unknown mechanisms. This study investigated whether macrophages were required for FK506 neuroenhancing effects. METHODS: Wildtype (WT) mice and mice with impaired macrophage recruitment to injury sites (Ccr2 -/- ) were injected subcutaneously with either saline or FK506 f

Leakage-controlled machine learning for European Cd, Hg, and Pb emission inventories.

Timely and spatially resolved inventories of cadmium (Cd), mercury (Hg), and lead (Pb) are needed for transboundary pollution assessment and inventory quality control, yet annual grid products may be delayed, incomplete, or internally inconsistent across releases. We developed an auditable, leakage-controlled framework that converts heterogeneous European emission products and contextual covariates into metal-specific, machine-learning-ready datasets. Its central contribution is the integration of data harmonization, target-family auditing, split-before-processing validation, operational basel

Demonstration and Mitigation of Aerosol and Particle Dispersion During Mastoidectomy Relevant to the COVID-19 Era.

BACKGROUND: COVID-19 has become a global pandemic with a dramatic impact on healthcare systems. Concern for viral transmission necessitates the investigation of otologic procedures that use high-speed drilling instruments, including mastoidectomy, which we hypothesized to be an aerosol-generating procedure. METHODS: Mastoidectomy with a high-speed drill was simulated using fresh-frozen cadaveric heads with fluorescein solution injected into the mastoid air cells. Specimens were drilled for 1-minute durations in test conditions with and without a microscope. A barrier drape was fashioned from

Protocol for automated optimization of feature selection and clustering parameters in single-cell RNA-seq using scAutoTune.

Single-cell RNA sequencing (RNA-seq) analysis requires dataset-specific tuning of feature selection and clustering parameters. Here, we present a protocol for automated optimization of feature selection and clustering parameters in single-cell RNA-seq using scAutoTune. We describe steps for installing scAutoTune, preparing Seurat objects from public single-cell RNA-seq datasets, and performing grid-based parameter sweeping with optional Harmony batch correction. We then detail procedures for computing silhouette-based performance metrics, visualizing generalized additive model (GAM)-smoothed l

Measurement-Based Probabilistic Power Flow Using a Basis Constrained Graph Convolutional Network with Few-Shot Node Adaptation.

Probabilistic power flow quantifies voltage and phase angle uncertainty under variable photovoltaic generation, but repeated AC Monte Carlo simulation is costly. A local topology change also modifies the electrical operator and state dimension when only a small target data set is available. We propose a Basis Constrained Graph Convolutional Network (BCGCN) for few-shot adaptation after local bus additions in small-scale grids. BCGCN predicts nonlinear residuals around a first-order solution using graph Laplacian and Proper Orthogonal Decomposition modes. It transfers source coordinates; adapts

A random grid based molecular epidemiological study on EBLV isolates from Germany.

Germany has reported one of the highest levels of EBLV cases in bats in Europe. So far, all isolates originating from Germany have been identified as EBLV-1, using monoclonal antibodies, and a preliminary epidemiological study has indicated that there is a distinct geographical distribution of EBLV-1 in Germany. To further investigate the spatial and temporal distribution of EBLV-1 variants in Germany and their impact on molecular epidemiology, isolates were selected using a random grid sampling procedure based on GIS. Agrid layer 30 km long over the entire area of Germany was applied to 120 g

Electron cryo-tomography of vestibular hair-cell stereocilia.

High-resolution imaging of hair-cell stereocilia of the inner ear has contributed substantially to our understanding of auditory and vestibular function. To provide three-dimensional views of the structure of stereocilia cytoskeleton and membranes, we developed a method for rapidly freezing unfixed stereocilia on electron microscopy grids, which allowed subsequent 3D imaging by electron cryo-tomography. Structures of stereocilia tips, shafts, and tapers were revealed, demonstrating that the actin paracrystal was not perfectly ordered. This sample-preparation and imaging procedure will allow fo

The entorhinal spatial map integrates visual identity information of landmarks.

The location and identity of landmarks provide spatial and nonspatial information of an environment, respectively, to guide spatial navigation. While the medial entorhinal cortex (MEC) is essential for representing spatial information, it is unclear whether it also encodes landmark identity. Here, we conducted two-photon calcium imaging of the MEC when mice navigated in multiple virtual environments, and discovered a general ability of the MEC to encode landmark identity through cue cells, which responded to individual landmarks during virtual navigation. Cue cells represented landmark identit

High-Resolution Spatial Surveillance of Sexually Transmitted Disease Risk in Urban Japan Using Hierarchical Hexagonal Grids (H3) and Quantum Geographic Information System (QGIS).

Introduction Urgent action is required to combat sexually transmitted diseases (STDs), such as syphilis. This study aimed to examine whether the combination of hierarchical hexagonal grids (H3) (Uber Technologies, Inc., San Francisco, CA, USA) and Quantum Geographic Information System (QGIS) (OSGeo, Grut, Switzerland) can capture localized clustering and short-term fluctuations of estimated STD-related risk behaviors with high spatial resolution while preserving anonymity, and to evaluate its potential utility as a public infectious disease surveillance method. Methods A field survey was condu

First cell-patterning of primary, patient-derived mesenchymal glioblastoma brain cancer cells on parylene-C/SiO2 substrates.

The mesenchymal subtype of the WHO grade IV Glioblastoma (GBM) is the most common and lethal form of adult brain cancer, characterised by its rapid growth, invasiveness and colonisation. Current standard of care includes surgical resection, chemotherapy and radiotherapy. Despite this, mortality remains dismal with median patient survival of less than 15 months after treatment. For these reasons, the World Health Organisation (WHO) classifies GBM as an incurable disease. To address this challenge, the GBM scientific community are investigating new approaches to gain insights into GBM function a

Nascent flagellar basal bodies are immobilized by rod assembly in Bacillus subtilis.

UNLABELLED: Flagella are complex, trans-envelope nanomachines that localize in species-specific patterns on the cell surface. Here, we study the localization dynamics of the earliest stage of basal body formation in Bacillus subtilis using a fluorescent fusion to the C-ring protein FliM. We find that B. subtilis basal bodies do not exhibit dynamic subunit exchange and are largely stationary at steady state, consistent with flagellar assembly through the peptidoglycan (PG). However, rare mobile basal bodies were observed, and the prevalence of mobile basal bodies is elevated both early in basal

Correlative Light and Scanning Electron Microscopy to Study Interactions of Salmonella enterica with Polarized Epithelial Cell Monolayers.

Live cell fluorescence imaging is the method of choice to visualize dynamic cellular processes in time and space, such as adhesion to and invasion of polarized epithelial cells by Salmonella enterica sv. Typhimurium. Scanning electron microscopy provides highest resolution of surface structures of infected cells, providing ultrastructure of the apical side of host cells and infecting Salmonella. Combining both methods toward correlative light and scanning electron microscopy (CLSEM) enables new insights in adhesion and invasion mechanisms regarding dynamics over time, and high spatial resoluti

Curcumin prevents neuronal loss and structural changes in the superior cervical (sympathetic) ganglion induced by chronic sleep deprivation, in the rat model.

BACKGROUND: In modern societies, sleep deprivation is a serious health problem. This problem could be induced by a variety of reasons, including lifestyle habits or neurological disorders. Chronic sleep deprivation (CSD) could have complex biological consequences, such as changes in neural autonomic control, increased oxidative stress, and inflammatory responses. The superior cervical ganglion (SCG) is an important sympathetic component of the autonomic nervous system. CSD can lead to a wide range of neurological consequences in SCG, which mainly supply innervations to circadian system and oth

Real-time confocal imaging for evaluation of dose-dependent effects of gamma knife radiosurgery on U87 glioblastoma cell line.

PURPOSE: To characterize, in real time, how single-fraction low-dose Gamma Knife stereotactic radiosurgery (GKRS) affects proliferation and cell-cycle dynamics in glioblastoma (GBM) cells across a range of doses. We hypothesized that a 2.5 Gy dose would induce mitotic failure and cell death, whereas low doses (<1 Gy) might elicit adaptive or hormetic responses. METHODS: Human U-87 MG glioblastoma cells were cultured on coated glass-bottom dishes, stained with Hoechst 33,342, and maintained at 37°C/5% CO₂. A custom agarose phantom with an embedded dish and metal grid enabled precise delivery o

Cerebellar histogenesis and synaptic maturation following pre- and postnatal alcohol administration. An electron-microscopic investigation of the rat cerebellar cortex.

Fetal alcohol syndrome was produced in rats by feeding an ethanol-enriched, vitamine -supplemented liquid diet over a period of 4 weeks before mating and throughout pregnancy. The amount of ethanol was equivalent to 38-40% of the daily calories; control animals were fed an isocaloric amount of sucrose. The offspring of experimental animals displayed a diminished birth weight. In contrast to previous experiments, the ethanol diet was also administered during the lactating period. On postnatal day 7, experimental animals and controls were killed by perfusion and the cerebella prepared for electr

Achieving High Spatial Resolution Transmission Electron Microscopy Studies in Closed Liquid Cells.

Liquid cell transmission electron microscopy (LCTEM) is a rising technique for probing dynamic processes and structural transformations in liquid environments at the nanoscale. However, its broader application is largely constrained by limited spatial resolution, which arises from electron scattering by the encapsulating membranes and the liquid layer. Recent advances in instrument design, micro-electro-mechanical system (MEMS)-based cells, and ultrathin window materials have substantially improved imaging quality, yet achieving atomic-scale resolution in liquids remains a formidable challenge
