Investigating insulin's role in regulating local aromatase in growth plate development.

INTRODUCTION: Obesity-associated hyperinsulinemia is hypothesized to disrupt linear growth trajectories, though the mechanisms remain unclear. This study investigates these mechanisms using a high-fat diet-induced rat model. METHODS: Twelve healthy 1-week-old Sprague-Dawley rats (60-80 g) were randomized into two groups: a control group fed a standard diet and an obese group fed a high-fat diet. After 6 weeks of dietary intervention, weekly body weight and naso-anal length were recorded. At the end of the study, all rats were euthanized, and blood samples were collected for further analysis.

Compressive stress induces spinal vertebral growth plate chondrocytes apoptosis via Piezo1.

Many clinical studies have indicated an association between biomechanical factors and the incidence and pathological progression of adolescent idiopathic scoliosis (AIS). However, at present, the research on AIS is mainly focused on the etiology, and there are few studies reporting the causes of progressive aggravation of AIS. In the present study, we aim to investigate the role of Piezo1 in compressive stress-induced mouse spinal vertebral growth plate chondrocytes apoptosis. First, a scoliosis mouse model was established, and the expression of Piezo1 as well as the degree of apoptosis were i

Vitamin D deficiency and anatomical region alters porcine growth plate properties.

Ossification of growth plate cartilage mediates longitudinal extension of long bones. Biomechanical and biochemical disruptions of growth plate function may lead to abnormal bone growth. In humans and animals, severe dietary vitamin D deficiency can lead to rickets which features growth plate widening, resulting in abnormalities in growth. However, effects of marginal vitamin D deficiencies on growth plates are not well understood. The purpose of this study was to examine the effects of a vitamin D deficient diet in the 26-day nursery phase on mechanical properties (ultimate normal stress, ult

AKAP2 overexpression modulates growth plate chondrocyte functions through ERK1/2 signaling.

In our previous study, the mutation c.2645A > C (p. E882A) was found in the A-Kinase Anchoring Protein 2 (AKAP2) gene, which plays an important role in regulating the development of the skeletal system; however, the specific effect of AKAP2 on chondrocyte proliferation and differentiation and the potential mechanism are still not clear. In the present study, we investigated the effect of AKAP2 in vitro. We successfully isolated human growth plate chondrocytes (GPCs) from growth plate cartilage tissues and identified GPCs by aggrecan expression and flow cytometric analysis. AKAP2 overexpression

Growth factor signaling alters the morphology of the zebrafish ethmoid plate.

Craniofacial development relies on coordinated tissue interactions that allow for patterning and growth of the face. We know a priori that the Wingless, fibroblast growth factor, Hedgehog and transforming growth factor-beta growth factor signaling pathways are required for the development of the face, but how they contribute to the shape of the face is largely untested. Here, we test how each signaling pathway contributes to the overall morphology of the zebrafish anterior neurocranium. We tested the contribution of each signaling pathway to the development of the ethmoid plate during three di

Insulin exerts direct, IGF-1 independent actions in growth plate chondrocytes.

Insufficient insulin production or action in diabetic states is associated with growth retardation and impaired bone healing, while the underling mechanisms are unknown. In this study, we sought to define the role of insulin signaling in the growth plate. Insulin treatment of embryonic metatarsal bones from wild-type mice increased chondrocyte proliferation. Mice lacking insulin receptor (IR) selectively in chondrocytes (CartIR (-/-)) had no discernable differences in total femoral length compared to control littermates. However, CartIR (-/-) mice exhibited an increase in chondrocyte numbers i

Treatment of growth plate injury using IGF-I-loaded PLGA scaffolds.

Growth plate fracture can lead to retarded growth and unequal limb length, which may have a lifelong effect on a person's physical stature. The goal of this research was to develop an in vivo tissue-engineering approach for the treatment of growth plate injury via localized delivery of insulin-like growth factor I (IGF-I) from cell-free poly(lactic-co-glycolic acid) (PLGA) scaffolds. Mass loss and drug release studies were conducted to study the scaffold degradation and IGF-I release patterns. In vitro cell studies showed that rat bone marrow stromal cells seeded on the porous scaffolds coloni

Multiple exostosis: a short study of abnormalities near the growth plate.

The pathogenesis of multiple exostosis has been controversial with many theories put forward including the structural/mechanical theory, which emphasizes that the osteochondroma arises in the displaced growth plate cartilage penetrating a defective periosteum. Recently, molecular genetics has offered the neoplastic model with tumor suppressor genes implicated in the development and pathogenesis of exostosis. In this study, we demonstrated the spectrum of histological abnormalities in the developing exostosis present on the surface of the bone at the physis. Seven skeletally immature patients w

Expression profiling of human fetal growth plate cartilage by EST sequencing.

The differentiation of mesenchymal stem cells into hypertrophic chondrocytes is an integral and multistep process important in pattern formation, endochondral ossification, and postnatal growth of the skeleton. In recent years, novel genes involved in these processes have been identified, but still only little is known about the large-scale gene expression profile during skeletal development. We initiated an expressed sequence tag (EST) project aiming at the identification of genes and pathways involved in this complex process. Candidate genes are expected to be of value for diagnosis and trea

Annexin-mediated Ca2+ influx regulates growth plate chondrocyte maturation and apoptosis.

Maturation of epiphyseal growth plate chondrocytes plays an important role in endochondral bone formation. Previously, we demonstrated that retinoic acid (RA) treatment stimulated annexin-mediated Ca(2+) influx into growth plate chondrocytes leading to a significant increase in cytosolic Ca(2+), whereas K-201, a specific annexin Ca(2+) channel blocker, inhibited this increase markedly. The present study addressed the hypothesis that annexin-mediated Ca(2+) influx into growth plate chondrocytes is a major regulator of terminal differentiation, mineralization, and apoptosis of these cells. We fo

An ultrastructural analysis of osteochondritic growth plate cartilage in growing swine.

In growing swine, ossification failure due to osteochondrosis has an incidence of nearly 100% in the distal ulna of animals at six months of age, yet the etiology of the disease is understood poorly. In this study, the ultrastructure of the chondrocyte and its pericellular matrix is analyzed in normal growth plates and in growth plates with lesions characteristic of osteochondrosis using aldehyde primary fixatives and osmium-ferrocyanide as the secondary fixative. Chondrocytes in lesion areas fail to undergo normal hypertrophic cell maturation, and they have an accumulation of rough endoplasmi

In vitro culture of rabbit growth plate chondrocytes. 2. Chondrodystrophic mutants.

The epiphyseal plates of newborn normal (Cd/?), dwarf (Dw/Dw) and achondroplastic (ac/ac) rabbits were grown in organ culture for 48 hours. The 3H-thymidine labeling index of proliferating zone chondrocytes from both mutants was below that of normals. The generation time of Dw/Dw animals was longer than that of normals (greater than 24 vs. 17 hours respectively). Inasmuch as the medium was supplemented with 10% fetal calf serum, which presumably contains somatomedin, these data suggest that the Dw/Dw mutant cannot be considered simply a model for pituitary dwarfism. Pituitary fibroblast growth

Crystal Growth Engineering for Dendrite-Free Zinc Metal Plating.

The practical implementation of aqueous zinc-ion batteries (AZIBs) is limited by uncontrolled zinc (Zn) dendrite growth during anode plating, compromising both safety and cycle life. Typically, Zn plating proceeds via 2D growth along the six equivalent prismatic [ 10 1 ¯ 0 ] directions of the hexagonal close-packed (HCP) Zn lattice, forming hexagonal platelets that promote dendrite formation. Here, an effective electrolyte engineering strategy is presented using rare-earth ions to regulate Zn plating. Combined multiscale experimental analyses and computational modeling reveal that these ions

Primary cilia in growth plates orchestrate long bone development.

The growth plate plays a crucial role in long bone development and elongation, housing stem cells that contribute to bone formation. This study investigates the role of primary cilia, specialized organelles that regulate stem cell fate, in the development and repair of long bones. Here, we report the presence of primary cilia in all zones of the growth plate, particularly during embryonic development. Spatial transcriptomics and analysis of conditional knockout (CKO) mice identified that primary cilia mediate critical developmental signaling pathways within the growth plate. Disruption of prim

[Stress fractures of the growth plates in the fingers of adolescent rock climbers].

BACKGROUND: Primary periphyseal stress injuries (PPSI) of the hand and fingers are a rare condition overall but are most commonly seen in adolescent rock climbers and is the most common sport-specific injury in young climbers. Early diagnosis and treatment are crucial for a good treatment outcome and to avoid chronic sport-related injury. OBJECTIVE: The aim of the study is to introduce the injury to a wider audience. Based on an analysis of the current literature, the pathophysiology is demonstrated and the diagnostic and treatment standards are analyzed. Prophylactic measures are also report

Roles of Local Soluble Factors in Maintaining the Growth Plate: An Update.

The growth plate is a cartilaginous tissue found at the ends of growing long bones, which contributes to the lengthening of bones during development. This unique structure contains at least three distinctive layers, including resting, proliferative, and hypertrophic chondrocyte zones, maintained by a complex regulatory network. Due to its soft tissue nature, the growth plate is the most susceptible tissue of the growing skeleton to injury in childhood. Although most growth plate damage in fractures can heal, some damage can result in growth arrest or disorder, impairing leg length and resultin

DDRGK1 is required for the proper development and maintenance of the growth plate cartilage.

Loss-of-function mutations in DDRGK1 have been shown to cause Shohat type spondyloepimetaphyseal dysplasia (SEMD). In zebrafish, loss of function of ddrgk1 leads to defects in early cartilage development. Ddrgk1-/- mice show delayed mesenchymal condensation in the limb buds and early embryonic lethality. Mechanistically, Ddrgk1 interacts with Sox9 and reduces ubiquitin-mediated proteasomal degradation of Sox9 protein. To investigate the cartilage-specific role of DDRGK1, conditional knockout mice were generated by intercrossing Prx1-Cre transgenic mice with Ddrgkfl/fl mice to delete its expres

[Effect of Flax lignans on apoptosis of growth plate chondrocytes in rats].

OBJECTIVE: To compare the effects of different concentrations of linolenin on inhibiting apoptosis of chondrocytes in the growth plate, and to screen the optimal concentration of linolenin, so as to provide theoretical support for delaying epiphyseal closure and promoting long bone growth in rats. METHODS: Two 4-week-old male SD rats (SPF grade) with a body mass of 80 g were selected. The growth plate cartilage of rat tibia and femur was dissected and isolated in vitro to obtain growth plate chondrocytes for culture. The chondrocytes were observed and identified by inverted phase contrast mic

Vascularization of primary and secondary ossification centres in the human growth plate.

BACKGROUND: The switch from cartilage template to bone during endochondral ossification of the growth plate requires a dynamic and close interaction between cartilage and the developing vasculature. Vascular invasion of the primarily avascular hypertrophic chondrocyte zone brings chondroclasts, osteoblast- and endothelial precursor cells into future centres of ossification.Vascularization of human growth plates of polydactylic digits was studied by immunohistochemistry, confocal-laser-scanning-microscopy and RT-qPCR using markers specific for endothelial cells CD34 and CD31, smooth muscle cell
