The Development and Characterization of a Cell Culture System from Indian Mud Crabs Scylla serrata.

Commercially available culture media and supplements were tested for their potential to produce primary cell cultures from tissues of Indian mud crabs Scylla serrata. Eight commercially available culture media from Sigma-Aldrich (Leibovitz's L-15, Medium 199, Grace's Insect Medium, Minimal Essential Medium, Dulbecco's Modified Eagle Medium, TC-100 Insect Medium, IPL-41 Insect Medium, and Roswell Park Memorial Institute) were examined. Three different supplements (amino acid and sugar [AS], crab muscle extract [CME], and natural seawater [NSW]) were also examined. The hemocyte culture appeared

Variation in expression of Biomphalaria glabrata SOD1: a potential controlling factor in susceptibility/resistance to Schistosoma mansoni.

The snail Biomphalaria glabrata kills the blood fluke Schistosoma mansoni by a mechanism involving production of hydrogen peroxide, the enzymatic product of cytosolic Cu/Zn superoxide dismutase (SOD1). This enzyme exhibits higher activity in blood cells (hemocytes) from a predominantly resistant strain of B. glabrata than in hemocytes from a susceptible strain. Additionally, B. glabrata SOD1 polymorphisms have been associated with susceptibility/resistance to the parasite. To address the hypothesis that SOD1 transcription levels differ in accordance with variation at the SOD1 locus, quantitati

Characterization of temporal expression of immune genes in female locust challenged by fungal pathogen, Aspergillus sp.

INTRODUCTION: The innate immune system provides defense against invading pathogens in insects and mammals. METHODS: We conducted transcriptomic analyses of the locust Locusta migratoria under Aspergillus oryzae infection to clarify temporal variation in its molecular immune response. RESULTS: We found that fat body cells and hemocytes play different roles in the immune response of locusts to Aspergillus infection at different time points after inoculation, and melanization was the main process underlying the immune response of female locusts. Most pattern recognition receptors (PRR) genes we

Cellular and molecular complementary immune stress markers for the model species Dreissena polymorpha.

This study aimed to combine cellular and molecular analyses for better detail the effects of various stresses on a sentinel species of freshwater invertebrate. For this purpose, the hemocytes of the zebra mussel, Dreissena polymorpha, were exposed to different stresses at two different intensities, high or low: chemical (cadmium and ionomycin), physical (ultraviolet B), or biological ones (Cryptosporidium parvum and Toxoplasma gondii). After exposure, flow cytometry and droplet digital PCR analyses were performed on the same pools of hemocytes. Several responses related to necrosis, apoptosis,

The molecular mechanism of ferroptosis in the Pacific oyster Crassostrea gigas under Erastin treatment or high temperature stress.

Ferroptosis is an iron- and lipotoxicity-dependent form of programmed cell death, and it is distinct from apoptosis, pyroptosis, and autophagy. In the present study, the hemocytes were found to be shrunken under Erastin treatment or high temperature stress. The mitochondrial atrophy, crest loss and fracture were observed in hemocytes under high temperature stress. In addition, the fluorescence intensity of mitochondrial probe JC-1 monomers increased significantly in hemocytes under high temperature stress. Hemocytes were found to be wrinkled under ultrastructure and the contents of LPO, ROS an

Comparison of virulence between Paracoccidioides brasiliensis and Paracoccidioides lutzii using Galleria mellonella as a host model.

Paracoccidioidomycosis is a systemic mycosis, endemic in Latin America. The etiologic agents of this mycosis are composed of 2 species: Paracoccidioides brasiliensis and P. lutzii. Murine animal models are the gold standard for in vivo studies; however, ethical, economical and logistical considerations limit their use. Galleria mellonella is a suitable model for in vivo studies of fungal infections. In this study, we compared the virulence of P. brasiliensis and P. lutzii in G. mellonella model. The deaths of larvae infected with P. brasiliensis or P. lutzii were similar, and both species were

Polystyrene Microplastics Impair Cellular Immune Responses in Galleria mellonella Following Controlled Internal Exposure.

Microplastics (MPs) are emerging environmental contaminants with increasing evidence of immunotoxic effects in invertebrates; however, their underlying cellular mechanisms remain insufficiently understood. This study investigated whether short-term internal exposure to 1 μm polystyrene microplastics affects immune and oxidative responses in Galleria mellonella. Larvae were injected with 1, 5, 20, 40, and 80 mg/mL PS-MPs (n = 16 per parameter), and total hemocyte count, encapsulation and melanization responses, phenoloxidase (PO) activity, antioxidant enzymes (SOD and CAT), malondialdehyde (MDA

C3-CD18-VATPase-ATG16L1 and C3-ATG16L1 axes restrict the escape of microbes through the autophagy-lysosome pathway.

As the key component of the complement system, C3 plays important roles in complement activation to regulate phagocytosis, lyse cells, mediate inflammation, and clear immune complexes. In the present study, CgC3 in the cell-free hemolymph of the Pacific oyster Crassostrea gigas was found to be able to bind various polysaccharides and microbes and then interacted with membrane receptor CgCD18 to mediate the entry of the CgC3-coated Vibrio into hemocytes. CgATPV1D in hemocytes sensed the CgC3-coated Vibrio vacuole and recruited CgATG16L1. The free CgC3 in hemocytes could also recognize and bind

The Extracellular Matrix Proteins Pericardin and Lonely Heart Modulate the Directionality of Heart Contractions in Infected Mosquitoes.

The cardiac extracellular matrix provides structural support to the primary circulatory organ of an insect: a dorsal vessel that is divided into an aorta in the thorax and a heart in the abdomen. In the fruit fly, Drosophila melanogaster, the extracellular matrix components, Pericardin and Lonely heart, are needed for proper heart contractility, as they mediate adhesion between cardiomyocytes, pericardial cells, hemocytes, and alary muscles. In the mosquito, Anopheles gambiae, Pericardin and Lonely heart are necessary for the infection-induced migration of hemocytes to the heart. In these infe

Effects of fucoidan and quercetin on protecting crayfish (Procambarus clarkii) against infection with white spot syndrome virus.

White spot syndrome virus (WSSV) acts as a dangerous pathogen which threats the farming of crustaceans. This study aims to investigate the effect of diet supplemented with fucoidan and quercetin on the immunity and disease resistance of crayfish by studying the apoptosis of hemocytes, crayfish mortality rate and related immune parameters. The feed additive combination of 30 mg/kg fucoidan and 40 mg/kg quercetin (fucoidan + quercetin) significantly improved by 45 % the survival of crayfish (Procambarus clarkii) after infection with WSSV. The fucoidan + quercetin enhanced the total hemocyte coun

Clearance of bacteria injected into the hemolymph of the ridgeback prawn, Sicyonia ingentis (Crustacea: Decapoda): Role of hematopoietic tissue.

In an earlier investigation, radiolabelled bacteria injected into the hemolymph of the shrimp Sicyonia ingentis were cleared rapidly from circulation. Most of the bacteria were localized in the gills, followed by other organs including the heart, abdominal musculature, and hematopoietic nodules (HPN). We determined the organ-specific effectiveness of bacterial clearance and found the HPN to be the most effective on a per gram basis. Light and electron microscopy were used to determine the mechanism of clearance in the HPN. The HPN are readily permeable to individual bacteria, which then are re

Evidence for an early immunosuppressive role for related Campoletis sonorensis venom and ovarian proteins in Heliothis virescens.

Shared epitopes among venom, ovarian, and viral proteins may indicate that related proteins have similar functional roles during parasitization of Heliothis virescens by Campoletis sonorensis. Venom and ovarian proteins are introduced directly into the hemolymph during parasitization where they may target hemocytes or other components of the immune system. Polydnavirus expression has been detected in hemocytes, fat body, and other tissues but has not been detected earlier than 4 h after parasitization. Therefore, effects on hemocytes at times earlier than 4 h may not be caused by polydnavirus

Differential Protein Expression in the Hemolymph of Bithynia siamensis goniomphalos Infected with Opisthorchis viverrini.

Bithynia siamensis goniomphalos is a freshwater snail that serves as the first intermediate host of the human liver fluke Opisthorchis viverrini. This parasite is a major public health problem in different countries throughout the Greater Mekong sub-region (Thailand, southern Vietnam, Lao PDR and Cambodia). Chronic O. viverrini infection also results in a gradual increase of fibrotic tissues in the biliary tract that are associated with hepatobiliary diseases and contribute to cholangiocarcinoma (a fatal type of bile duct cancer). Infectivity of the parasite in the snail host is strongly corre

A Comprehensive Review on Crustaceans' Immune System With a Focus on Freshwater Crayfish in Relation to Crayfish Plague Disease.

Freshwater crayfish immunity has received great attention due to the need for urgent conservation. This concern has increased the understanding of the cellular and humoral defense systems, although the regulatory mechanisms involved in these processes need updating. There are, however, aspects of the immune response that require clarification and integration. The particular issues addressed in this review include an overall description of the oomycete Aphanomyces astaci, the causative agent of the pandemic plague disease, which affects freshwater crayfish, and an overview of crustaceans' immun

The effect of infection with the entomopathogenic fungus Conidiobolus coronatus (Entomopthorales) on eighteen cytokine-like proteins in Galleria mellonella (Lepidoptera) larvae.

BACKGROUND: In response to the replace mammal research models with insects in preliminary immunological studies, interest has grown in invertebrate defense systems. The immunological response is regulated by cytokines; however, while their role in mammals is well understood, little is known of their function in insects. A suitable target for studies into insect immunology is Galleria mellonella (Lepidoptera), the wax moth: a common host for human fungal and bacterial pathogens. G. mellonella is also a perfect subject for studies into the presence of cytokine-like proteins. SPECIFIC OBJECTIVES

Antiviral Activity of Myticin C Peptide from Mussel: an Ancient Defense against Herpesviruses.

UNLABELLED: Little is known about the antiviral response in mollusks. As in other invertebrates, the interferon signaling pathways have not been identified, and in fact, there is a debate about whether invertebrates possess antiviral immunity similar to that of vertebrates. In marine bivalves, due to their filtering activity, interaction with putative pathogens, including viruses, is very high, suggesting that they should have mechanisms to address these infections. In this study, we confirmed that constitutively expressed molecules in naive mussels confer resistance in oysters to ostreid herp

Wash functions downstream of Rho1 GTPase in a subset of Drosophila immune cell developmental migrations.

Drosophila immune cells, the hemocytes, undergo four stereotypical developmental migrations to populate the embryo, where they provide immune reconnoitering, as well as a number of non-immune-related functions necessary for proper embryogenesis. Here, we describe a role for Rho1 in one of these developmental migrations in which posteriorly located hemocytes migrate toward the head. This migration requires the interaction of Rho1 with its downstream effector Wash, a Wiskott-Aldrich syndrome family protein. Both Wash knockdown and a Rho1 transgene harboring a mutation that prevents Wash binding

Effects of beauvericin on the blood cells of Bombyx mori.

In this study, silkworms were treated by injection of the bioactive depsipeptide beauvericin (BEA) to explore its effect on the cellular immunity of larvae of the silkworm Bombyx mori. The results showed that: The LC50 of BEA for silkworms on the 3rd day of the 4th instar was 362.36 µM. The total count of circulating hemocytes in the silkworms decreased at 12 h after injection with 350 µM BEA, and reached the minimum value at 72 h post-treatment; at 48 h post-treatment, a large number of nodules formed by the aggregation of blood cells of the silkworms were observed under the light microscope.

Multiplication and distribution of type 2 dengue and Japanese encephalitis viruses in Toxorhynchites splendens after intrathoracic inoculation.

The nonhematophagous mosquito Toxorhynchites (Tx.) splendens was found to be the most susceptible to type 2 dengue (D-2) and Japanese encephalitis (JEV) viruses among three hosts examined by virus titration and replication assays. After inoculation with D-2, the number of viral antigen positive cells in the head, thorax and abdomen increased up to day 15 and D-2 reached the maximum titer of 8.4 log10 PFU/g in the head on day 15. Hemocytes were the earliest cell type that could be detected as D-2 antigen positive on day 2. Multiplication of JEV was faster than that of D-2 in the mosquito. The n

Phenotypic switching in Candida tropicalis alters host-pathogen interactions in a Galleria mellonella infection model.

Candida tropicalis is a human pathogen associated with high mortality rates. We have reported a switching system in C. tropicalis consisting of five morphotypes - the parental, switch variant (crepe and rough), and revertant (crepe and rough) strains, which exhibited altered virulence in a Galleria mellonella model. Here, we evaluate whether switching events may alter host-pathogen interactions by comparing the attributes of the innate responses to the various states. All switched strains induced higher melanization in G. mellonella larvae than that induced by the parental strain. The galiomic
