Cardamom extract: An effective weapon in prevention of cardiorenal syndrome induced in rats by cisplatin and high-fat diet.

BACKGROUND: Overnutrition and unhealthy dietary habits are established risk factors for cardiovascular and renal diseases, which may lead to the development of cardiorenal syndrome (CRS). AIM: To evaluate the cardiorenal protective potential of crude ethanol extract (CEE) of green cardamom (Elettaria cardamomum L., Family Zingiberaceae). METHODS: Rats were fed a high-fat diet to induce dyslipidemia and subsequently administered cisplatin (7.5 mg/kg) to induce CRS. CEE was administered orally to CRS rats at low (100 mg/kg) and high (200 mg/kg) doses for one month. Oxidative stress, inflammato

Chlamydia trachomatis induces the transcriptional activity of host YAP in a Hippo-independent fashion.

INTRODUCTION: The obligate intracellular pathogen Chlamydia trachomatis is the causative agent of the most common bacterial sexually transmitted disease worldwide. While the host response to infection by this pathogen has been well characterized, it remains unclear to what extent host gene expression during infection is the product of Chlamydia-directed modulation of host transcription factors. METHODS: To identify transcription factors potentially modulated by Chlamydia during infection, we infected immortalized endocervical epithelial cells (End1/E6E7) with the anogenital C. trachomatis ser

E-Cadherin-Mediated Cell Contact Controls the Epidermal Damage Response in Radiation Dermatitis.

Radiotherapy is a primary oncological treatment modality that also damages normal tissue, including the skin, and causes radiation dermatitis (RD). Here, we explore the mechanism of acute epidermal damage in radiation dermatitis. Two distinctive phases in the damage response were identified: an early destructive phase, where a burst of reactive oxygen species induces loss of E-cadherin-mediated cell contact, followed by a regenerative phase, during which Wnt and Hippo signaling are activated. A blocking peptide, as well as a neutralizing antibody to E-cadherin, works synergistically with ioniz

Xmu-mp-1 attenuates streptozotocin-induced neurotoxicity in SH-SY5Y cells: potential role of Hippo pathway modulation.

BACKGROUND: Alzheimer’s disease (AD) is the most common neurodegenerative dementia, with its primary symptoms appearing only after substantial neuronal death. Streptozotocin (STZ) is a neurotoxic compound that induces neurodegenerative effects similar to those seen in AD. AD is a complex and multifactorial disorder, and the exact etiology of neuronal loss remains elusive. In recent years, hyperactivation of mammalian ste-20-like kinase − 1/2 (MST1/2)-mediated Hippo signaling is critical for neuronal death in AD pathophysiology. Our previous research on a rat model of sporadic AD revealed that

Essential roles of YAP-TEAD complex in adult stem cell development during thyroid hormone-induced intestinal remodeling of Xenopus laevis.

During amphibian metamorphosis which is triggered by thyroid hormone (TH), the small intestine is extensively remodeled from the larval to adult form. In the Xenopus laevis intestine, some of the larval epithelial cells dedifferentiate into adult stem cells, which newly form the adult epithelium similar to the mammalian one. We have previously shown that TH-activated Shh, Wnt and Notch signaling pathways play important roles in adult epithelial development. Here we focus on the Hippo signaling pathway, which is known to interact with these pathways in the mammalian intestine. Our quantitative

Proteomic screening identifies a YAP-driven signaling network linked to tumor cell proliferation in human schwannomas.

BACKGROUND: Inactivation of the NF2 gene predisposes to neurofibromatosis type II and the development of schwannomas. In vitro studies have shown that loss of NF2 leads to the induction of mitogenic signaling mediated by receptor tyrosine kinases (RTKs), MAP kinase, AKT, or Hippo pathways. The goal of our study was to evaluate the expression and activity of these signaling pathways in human schwannomas in order to identify new potential therapeutic targets. METHODS: Large sets of human schwannomas, totaling 68 tumors, were analyzed using complementary proteomic approaches. RTK arrays identifi

Wnt signaling inhibition by monensin results in a period of Hippo pathway activation during intestinal adaptation in zebrafish.

Intestinal adaptation (IA) is a critical response to increase epithelial surface area after intestinal loss. Short bowel syndrome (SBS) may follow massive intestinal resection in human patients, particularly without adequate IA. We previously validated a model in zebrafish (ZF) that recapitulates key SBS pathophysiological features. Previous RNA sequencing in this model identified upregulation of genes in the Wnt and Hippo pathways. We therefore sought to identify the timeline of increasing cell proliferation and considered the signaling that might underpin the epithelial remodeling of IA in S

TNFR2 overexpression ameliorates allergic airway inflammation and restores immune homeostasis associated with Hippo pathway modulation in an ovalbumin-induced allergic asthma mouse model.

OBJECTIVE: Asthma remains a major global health burden with limited therapeutic options. Tumor necrosis factor receptor 2 (TNFR2) has been implicated in immune regulation, we aimed to investigate its role and underlying mechanisms in asthma pathogenesis. METHODS: An ovalbumin (OVA)-induced murine asthma model was established to investigate TNFR2 function. Recombinant adeno-associated virus serotype 9 vectors were administered three weeks prior to OVA sensitization to achieve prophylactic TNFR2 overexpression. Airway inflammation was assessed by histological staining and inflammation scoring.

Abnormal expression of Hippo-YAP1 signalling pathway and progesterone resistance mechanism in endometrial polyps.

BACKGROUND: Endometrial polyps (EPs) are a localised hyperproliferative disorder of the endometrium, and its pathogenesis remains unclear. Progesterone resistance may influence the recurrence rate of progesterone-treated EPs. Expression of the Hippo-YAP1 signalling pathway has been studied in various diseases. However, few studies have explored their expression in EPs and association with progesterone resistance. Prolactin (PRL), a molecular marker of endometrial mesenchymal cell metaplasia, indirectly reflects progesterone action in vivo. This study aimed to explore the role of Hippo-YAP1 sig

YAP Mediates Hair Cell Regeneration in Balance Organs of Chickens, But LATS Kinases Suppress Its Activity in Mice.

Loss of sensory hair cells causes permanent hearing and balance deficits in humans and other mammals, but for nonmammals such deficits are temporary. Nonmammals recover hearing and balance sensitivity after supporting cells proliferate and differentiate into replacement hair cells. Evidence of mechanical differences between those sensory epithelia and their supporting cells prompted us to investigate whether the capacity to activate YAP, an effector in the mechanosensitive Hippo pathway, correlates with regenerative capacity in acceleration-sensing utricles of chickens and mice of both sexes.

SAV1 in Cooperation With FKBP52 Participates in Implantation.

Endometrial receptivity is restricted to an implantation window, during which progesterone (P4) and estrogen (E2) orchestrate uterine remodeling to support blastocyst attachment. While E2 is indispensable for receptivity, the P4-primed uterus undergoes a transitional phase marked by luminal epithelial remodeling, stromal proliferation, and glandular differentiation, establishing competence for E2 action. Precise coordination of P4-progesterone receptor (PR) signaling with estrogenic cues is essential; upstream regulatory mechanisms remain unclear. We identify Salvador homolog 1 (SAV1), a scaff

Liquiritigenin inhibits hepatic fibrogenesis and TGF-β1/Smad with Hippo/YAP signal.

BACKGROUND: Recent reports highlighted the possibility that Yes-associated protein (YAP) and transforming growth factor-β1 (TGF-β1) can act as critical regulators of hepatic stellate cells (HSCs) activation; therefore, it is natural for compounds targeting Hippo/YAP and TGF-β1/Smad signaling pathways to be identified as potential anti-fibrotic candidates. PURPOSE: Liquiritigenin (LQ) is an aglycone of liquiritin and has been reported to protect the liver from injury. However, its effects on the Hippo/YAP and TGF-β1/Smad pathways have not been identified to date. METHODS: We conducted a serie

The Hippo pathway effector YAP is an essential regulator of ductal progenitor patterning in the mouse submandibular gland.

Salivary glands, such as submandibular glands (SMGs), are composed of branched epithelial ductal networks that terminate in acini that together produce, transport and secrete saliva. Here, we show that the transcriptional regulator Yap, a key effector of the Hippo pathway, is required for the proper patterning and morphogenesis of SMG epithelium. Epithelial deletion of Yap in developing SMGs results in the loss of ductal structures, arising from reduced expression of the EGF family member Epiregulin, which we show is required for the expansion of Krt5/Krt14-positive ductal progenitors. We furt

Human cytomegalovirus (CMV) dysregulates neurodevelopmental pathways in cerebral organoids.

Human cytomegalovirus (CMV) infection is the leading non-genetic aetiology of congenital malformation in developed countries, causing significant fetal neurological injury. This study investigated potential CMV pathogenetic mechanisms of fetal neural malformation using in vitro human cerebral organoids. Cerebral organoids were permissive to CMV replication, and infection dysregulated cellular pluripotency and differentiation pathways. Aberrant expression of dual-specificity tyrosine phosphorylation-regulated kinases (DYRK), sonic hedgehog (SHH), pluripotency, neurodegeneration, axon guidance,

Yes-associated protein 1 translocation through actin cytoskeleton organization in trophectoderm cells.

A mammalian embryo experiences the first cell segregation at the blastocyst stage, in which cells giving form to the embryo are sorted into two lineages; trophectoderm (TE) and inner cell mass (ICM). This first cell segregation process is governed by cell position-dependent Hippo signaling, which is a phosphorylation cascade determining whether Yes-associated protein 1 (YAP1), one of the key components of the Hippo signaling pathway, localizes within the nucleus or cytoplasm. YAP1 localization determines the transcriptional on/off switch of a key gene, Cdx2, required for TE differentiation. Ho

The mammalian Ste20-like kinase 2 (Mst2) modulates stress-induced cardiac hypertrophy.

The Hippo signaling pathway has recently moved to center stage in cardiac research because of its key role in cardiomyocyte proliferation and regeneration of the embryonic and newborn heart. However, its role in the adult heart is incompletely understood. We investigate here the role of mammalian Ste20-like kinase 2 (Mst2), one of the central regulators of this pathway. Mst2(-/-) mice showed no alteration in cardiomyocyte proliferation. However, Mst2(-/-) mice exhibited a significant reduction of hypertrophy and fibrosis in response to pressure overload. Consistently, overexpression of MST2 in

Activation of the Legionella pneumophila LegK7 Effector Kinase by the Host MOB1 Protein.

Legionella pneumophila infects alveolar macrophages and can cause life-threatening pneumonia in humans. Upon internalization into the host cell, L. pneumophila injects numerous effector proteins into the host cytoplasm as a part of its pathogenesis. LegK7 is an effector kinase of L. pneumophila that functionally mimics the eukaryotic Mst kinase and phosphorylates the host MOB1 protein to exploit the Hippo pathway. To elucidate the LegK7 activation mechanism, we determined the apo structure of LegK7 in an inactive form and performed a comparative analysis of LegK7 structures. LegK7 is a non-RD

Research progress on Hippo signaling pathway effector molecules in rheumatic immune system diseases.

The core components of the Hippo signaling pathway encompass upstream regulatory molecules, core kinase cascade complexes, and downstream transcriptional regulation complexes. This pathway modulates cellular behaviors by influencing the effector molecules of its core components and plays a pivotal role in immune regulation. Effector molecules, such as Yes-associated protein (YAP), transcriptional coactivator with PDZ-binding motif (TAZ), transcriptional enhanced associate domain transcriptional factor (TEAD), monopolar spindle-one binder (MOB1), large tumor suppressor (LATS), can stimulate fibr

Targeting the PTCHD1-hippo axis by rhPTH(1-34) restores contact inhibition and suppresses hyperproliferation in psoriatic keratinocytes.

Psoriasis is a chronic, immune-mediated inflammatory skin disease induced by genetic and environmental factors, with dysfunctional contact inhibition of keratinocytes being a core pathological feature leading to hyperproliferation. Our previous gene chip screening identified a significant downregulation of PTCHD1 following treatment with recombinant human parathyroid hormone (1-34) (rhPTH(1-34)), which inhibited keratinocyte proliferation. PTCHD1 (Patched domain-containing protein 1) is a transmembrane protein that regulates intercellular communication and proliferative signaling. This study a
