In plant sciences, single-cell and spatial transcriptomics generate large complex datasets which require structured metadata in order to make them interoperable and reproducible. In this work, we demonstrate how the Annotated Research Context (ARC) can be used for the management of such data using a barley single-cell RNA sequencing dataset integrated with spatial transcriptomics data. Experimental metadata was captured in ISA tables using ontology annotations to ensure machine readability and interpretability. The computational analyses were wrapped as Common Workflow Language (CWL) scripts t

Phosphorus (P) deficiency is one of the most widespread constraints on global crop productivity. Root hairs are widely regarded as a pivotal tissue for P acquisition, yet their quantitative contribution to P uptake and subsequent plant performance remains unclear. We examined 12 cultivated and 12 wild barley genotypes, classified as tolerant or sensitive to low P based on relative total biomass, and quantified root hair number and length in relation to plant performance. While P deficiency generally stimulated root hair proliferation, contrasting strategies emerged: tolerant genotypes have mai

Kashin-Beck disease (KBD) is a multifactorial endemic disease that only occurs in specific Asian areas. Mycotoxin contamination, especially from the Fusarium spp., has been considered as one of the environmental risk factors that could provoke chondrocyte and cartilage damage. This study aimed to investigate whether new mycotoxins could be identified in KBD-endemic regions as a potential KBD risk factor. This was investigated on 292 barley samples collected in Tibet during 2009-2016 and 19 wheat samples collected in Inner Mongolia in 2006, as control, from KBD-endemic and non-endemic areas. Th

Various gluten-related diseases (celiac disease, wheat allergy, gluten sensitivity) are known and their incidence is growing. Gluten is a specific type of plant storage protein that can impair the health of gluten-prone persons following consumption, depending on the origin. The most severe effects are induced by wheat, barley, and rye. The only treatment is based on the absolute avoidance of those foods, as even traces might have severe effects on human well-being. With the goal of binding gluten impurities after ingestion, an in vitro setting was created. A special processed kind of zeolite,

Gluten describes a complex mixture of proteins found in wheat, rye, barley and oats that pose a health risk to people affected by conditions such as coeliac disease and non-coeliac gluten sensitivity. Complete digestion of gluten proteins is of critical importance during quantitative analysis. To this end, chymotrypsin was investigated for its ability to efficiently and reproducibly digest specific classes of gluten in barley. Using proteomics a chymotryptic peptide marker panel was elucidated and subjected to relative quantification using LC-MRM-MS. Thorough investigation of peptide markers r

Sugars and polyols were tested at different steps of anther culture in barley (Hordeum vulgare L.) to elucidate their influence on both the overall yield of androgenesis and the structure of plastids in relation to albinism. During the pretreatment period, the osmotic regulation in the medium was beneficial to microspore embryogenesis regardless of the type and concentration of the tested osmoticum. The use of mannitol (300 mOsm/kg), sorbitol (180 mOsm/kg), PEG (240 mOsm/kg) and sucrose (180 mOsm/kg) gave the best results in terms of green plant production, although the influence of each subst

Meiotic recombination is not only a key mechanism for sexual adaptation in eukaryotes but crucial for the accumulation of beneficial alleles in breeding populations. The effective manipulation of recombination requires, however, a better understanding of the mechanisms regulating the rate and distribution of recombination events in genomes. Here, we identified the genomic features that best explain the recombination variation among a diverse set of segregating populations of barley at a resolution of 1 Mbp and investigated how methylation and structural variants (SVs) determine crossover (CO)

Barley flour is valued for its unique texture and resistance to starch digestion. However, its high bran content often compromises food quality. To address this, the effects of incorporating epigallocatechin gallate (EGCG) and ferulic acid (FA) into whole grain barley noodles were investigated. In terms of digestibility, resistant starch content increased from 24% (Control) to 37% (0.5% EGCG) and 26% (0.5% FA), while rapidly digestible starch decreased from 62% to 51% and 59%, respectively. These results indicate that EGCG and FA enhanced the anti-digestive property of noodles. Analyses using

In this study, using a pharmaceutical approach, we analyzed the NO accumulation and emission from the root tips of barley seedlings and the possible mechanisms of NO catabolism. Application of flavohemeprotein inhibitors, such as azide, cyanide, diphenyleneiodonium and dicumarol, an inhibitor of the plasma membrane electron transport chain, increased the NO level in root tissue and stimulated the NO emission from root tip cells. It can be concluded that barley root tips generate and, at the same time, consume a considerable amount of NO, probably by the plasma membrane flavohemeproteins. This

Encasements formed around haustoria and biotrophic hyphae as well as hypersensitive reaction (HR) cell death are essential plant immune responses to filamentous pathogens. In this study we examine the components that may contribute to the absence of these responses in susceptible barley attacked by the powdery mildew fungus. We find that the effector CSEP0162 from this pathogen targets plant MONENSIN SENSITIVITY1 (MON1), which is important for the fusion of multivesicular bodies to their target membranes. Overexpression of CSEP0162 and silencing of barley MON1 both inhibit encasement formation

Mutant-based research is a cornerstone for elucidating gene function in barley (Hordeum vulgare) and other cereals. In this study, we exploited a unique collection of barley tweaky spike (tw) mutants to identify the locus controlling floret and spike development. We combined genetic mapping with structural variant and gene-expression analyses across a tw allelic series, supported by functional validation, and quantified endogenous auxin levels alongside auxin-application assays to test phytohormone involvement. Analysis of independent tw allelic mutants revealed that the phenotype results from

Genome-wide association studies (GWASs) have become a central tool in plant genomics, providing high-resolution access to the genetic architecture of complex traits in cereal crops. This review observes the application of GWASs to biotic and abiotic stress responses and nutritional quality in rice (Oryza sativa), wheat (Triticum aestivum), maize (Zea mays), and barley (Hordeum vulgare). However, conventional SNP-based GWASs may overlook structural and presence-absence variation, and significant associations do not by themselves identify the causal gene or variant. Recent methodological develop

Fungal infection of grasses, including rice (Oryza sativa), sorghum (Sorghum bicolor), and barley (Hordeum vulgare), induces the formation and accumulation of flavonoid phytoalexins. In maize (Zea mays), however, investigators have emphasized benzoxazinoid and terpenoid phytoalexins, and comparatively little is known about flavonoid induction in response to pathogens. Here, we examined fungus-elicited flavonoid metabolism in maize and identified key biosynthetic enzymes involved in the formation of O-methylflavonoids. The predominant end products were identified as two tautomers of a 2-hydroxy

Hemoglobin gene expression is upregulated during hypoxia. To determine whether the induction occurs via similar mechanisms that have been proposed for other hypoxically induced proteins, barley (Hordeum vulgare L.) aleurone layers were treated with various agents that interfere with known components of signal transduction. Ruthenium red, an organelle calcium channel blocker, inhibited anoxia-induced hemoglobin (Hb) and alcohol dehydrogenase (EC 1.1.1.1) (Adh) gene expression in a dose-dependent manner. The divalent ionophore, A23187, combined with EGTA also dramatically reduced anoxia-induced

Mitochondria isolated from the roots of barley (Hordeum vulgare L.) and rice (Oryza sativa L.) seedlings were capable of oxidizing external NADH and NADPH anaerobically in the presence of nitrite. The reaction was linked to ATP synthesis and nitric oxide (NO) was a measurable product. The rates of NADH and NADPH oxidation were in the range of 12-16 nmol min(-1) mg(-1) protein for both species. The anaerobic ATP synthesis rate was 7-9 nmol min(-1) mg(-1) protein for barley and 15-17 nmol min(-1) mg(-1) protein for rice. The rates are of the same order of magnitude as glycolytic ATP production d

The roles of endogenous brassinosteroids (BRs) in the modulation of reaction to drought and genetic regulation of this process are still obscure. In this study, a multidirectional analysis was performed on semi-dwarf barley (Hordeum vulgare) Near-Isogenic Lines (NILs) and the reference cultivar "Bowman" to get insights into various aspects of metabolic reaction to drought. The NILs are defective in BR biosynthesis or signaling and displayed an enhanced tolerance to drought. The BR metabolism perturbations affected the glucose and fructose accumulation under the control and stress conditions. T

Foods with partially hydrolyzed gluten may still trigger immune responses in celiac disease (CeD) patients due to residual CeD-active peptides. We applied nanoliquid chromatography-tandem mass spectrometry (nanoLC-MS/MS) to identify 54 CeD-active peptides in 15 barley malt extracts (BME), seven barley malt vinegars (BMV), and 15 soy sauces. Eight common CeD-active peptides were quantitated with targeted nanoLC-MS/MS and stable isotope dilution assay. Total peptide concentrations ranged from 481 mg/kg to 1305 mg/kg in BMEs, whereas three samples showed peptide concentrations near LOQ. Six BMVs

Efficient foliar delivery of manganese (Mn) remains a major challenge in crop nutrition due to the low phloem mobility of Mn2 + ions and the risk of leaf scorching from conventional Mn salts. This study investigates the uptake and assimilation of foliar-applied ∼25 nm polyacrylic acid-coated manganese dioxide nanoparticles (nPAA-MnO2) in 4-week-old Mn-deficient barley (Hordeum vulgare) and compares them to ionic Mn. Complementary imaging methods showed rapid (2 h) leaf penetration of nPAA-MnO2 through stomata, facilitated by the formulation´s low surface tension and the addition of 3% glycerol

The unfavorable bioaccessibility of polyphenols in cereal-based food limits their physiological functions as most polyphenols bind spontaneously to the cell-wall polysaccharides. Effects of β-glucan and various thermal processing methods including flaking and roasting, stir-frying, steam-flash explosion, and popping expansion on the bioaccessibility and antioxidant properties of polyphenols from hulless barley in vitro and in vivo were investigated in this study. The bioaccessibility and antioxidant capacity (via DPPH, ·OH, and ·O2- free radical scavenging, TAC, and FRAP assays) of polyphenol

Peroxidase enzymes have been found in soluble, ionically bound, and covalently bound forms and have been implicated in several physiological processes in plants. This paper investigates the effect of aphid infestation on soluble and bound-cell wall peroxidase activity and bound-cell wall isoform changes of barley plants. Peroxidase activity was measured in control plants and plants infested with the aphid Schizaphis graminum (Rondani). The activity of soluble peroxidases increased with time of infestation, older plants being more affected than younger ones. The increase in bound-cell wall pero
