BACKGROUND: Pine wilt disease, caused by Bursaphelenchus xylophilus, devastates pine ecosystems. As a well-known biocontrol fungus, Trichoderma harzianum has been shown to exhibit broad-spectrum antagonistic effects against multiple pathogens and nematodes. In this study, we employed morphological, gas chromatography-mass spectrometry (GC-MS), and RNA-sequencing analyses to systematically evaluate its efficacy against Bursaphelenchus xylophilus and to elucidate the underlying molecular mechanisms. These findings clarify the biocontrol potential of T. harzianum and support developing new agents

Chitinases are promising enzymes for a multitude of applications, including chitooligosaccharide (COS) synthesis for food and pharmaceutical uses and marine waste management. Owing to fungal diversity, fungal chitinases may offer alternatives for chitin degradation and industrial applications. The rapid reproduction cycle, inexpensive growth media, and ease of handling of fungi may also contribute to reducing enzyme production costs. Thus, this study aimed to identify fungal species with chitinolytic potential and optimize chitinase production by submerged culture and enzyme characterization u

Three new antimalarial compounds, clonocoprogens A, B, and C, were isolated from the static culture of an Okinawan fungus, Clonostachys compactiuscula FKR-0021. These compounds were new analogs of N14-palmitoylcoprogen, reported as a siderophore. They showed moderate antimalarial activity against chloroquine-sensitive and chloroquine-resistant Plasmodium falciparum strains, with IC50 values ranging from 1.7 to 9.9 µM.

Protein tyrosine phosphatase 1B (PTP1B) is implicated as a negative regulator of insulin receptor (IR) signaling and a potential drug target for the treatment of type II diabetes and other associated metabolic syndromes. Thus, small molecule inhibitors of PTP1B can be considered as an attractive approach for the design of new therapeutic agents of type II diabetes and cancer diseases. In a continuing search for new PTP1B inhibitors, a new tetramic acid possessing a rare pyrrolidinedione skeleton named fumosorinone A (1), together with five known ones 2-6 were isolated from the entomogenous fun

Some herbivore-induced-plant volatiles (HIPVs) compounds are vital for the functioning of an ecosystem, by triggering multi-trophic interactions for natural enemies, plants and herbivores. However, the effect of these chemicals, which play a crucial role in regulating the multi-trophic interactions between plant-herbivore-entomopathogenic fungi, is still unknown. To fill this scientific gap, we therefore investigated how these chemicals influence the entomopathogenic fungi growth and efficacy. In this study, Lipaphis erysimi induced Arabidopsis thaliana HIPVs were collected using headspace sys

Fungal biocontrol agents (BCAs) are increasingly used in plant protection, but information about their establishment and fate as well as non-target effects under field conditions is yet limited, hampering the development of efficient and safe field application strategies. In field and semi-field experiments with strawberry, the fate of the fungal BCAs Trichoderma harzianum (strain T22), Clonostachys rosea (strain IK726) and Ulocladium atrum (strain 302), and their non-target effects on the native cultivable strawberry mycobiota on ripe berries was investigated. In two strawberry fields, T. har

This study aimed to produce secondary metabolites (SM) through the co-cultivation of Trichotomospora caesia AC-1134 and Streptoverticillium sp. AC-1375 and to evaluate their antioxidant properties. The fermentation was carried out on GYM, ISP, LB, and YGGS agar media. The radical scavenging activity of metabolites was studied using the ABTS and DPPH assays. The metabolites produced by the bacteria were identified with gas chromatography coupled with mass spectrometry (GC-MS). The GC-MS analysis revealed 39 and 28 compounds for the ethyl acetate and chloroform extracts, respectively. The compou

BACKGROUND: Purpureocillium lilacinum is a type of fungus that is found all over the world and is known for its ability to help control pests, especially those that harm plants. Generally, this fungus does not cause serious problems for healthy people, but it can lead to severe infections in individuals with weakened immune systems. In this report, we discuss a rare case where a healthy person developed an infection after being exposed to a biopesticide containing this fungus.

Terminalia arjuna, an important medicinal plant in traditional Indian systems, has been extensively studied for its cardioprotective bark. However, limited attention has been given to its fruit, which contains several biologically active phytochemicals with potential antioxidant, anti-inflammatory, and immunomodulatory properties. This study aimed to isolate and partially purify phytoactive compounds from the fruit of T. arjuna using chromatographic techniques, characterize them, and assess their effect on the growth of industrially important fungal strains. Dried and powdered T. arjuna fruit

The filamentous ascomycete Trichoderma reesei, known for its prolific cellulolytic enzyme production, recently also gained attention for its secondary metabolite synthesis. Both processes are intricately influenced by environmental factors like carbon source availability and light exposure. Here, we explore the role of the transcription factor STE12 in regulating metabolic pathways in T. reesei in terms of gene regulation, carbon source utilization and biosynthesis of secondary metabolites. We show that STE12 is involved in regulating cellulase gene expression and growth on carbon sources asso

BACKGROUND: Adaptation to complex, rapidly changing environments is crucial for evolutionary success of fungi. The heterotrimeric G-protein pathway belongs to the most important signaling cascades applied for this task. In Trichoderma reesei, enzyme production, growth and secondary metabolism are among the physiological traits influenced by the G-protein pathway in a light dependent manner.
RESULTS: Here, we investigated the function of the SNX/H-type regulator of G-protein signaling (RGS) protein RGS4 of T. reesei. We show that RGS4 is involved in regulation of cellulase production, growth,

Chronic granulomatous disease (CGD) is an inherited immunodeficiency due to defective leukocyte NADPH responsible for recurrent infections and aberrant inflammation. Mutations in the CYBB gene are responsible for the X-linked CGD and account for approximately 70% of the cases. CGD is diagnosed during childhood in males. Female carriers may have biased X-inactivation and may present with clinical manifestations depending on the level of residual NADPH oxidase activity. We report the case of a previously asymptomatic female carrier who was diagnosed at age 67 with a skin infection with the rare

Iron (Fe) deficiency is one of the major limiting factors affecting crop yields. Trichoderma asperellum Q1, a biocontrol and plant growth promoting fungus, can produce the siderophore which has a high affinity to Fe3+ in the absence of iron. In this study, Trichoderma asperellum Q1 was found to be able to promote growth of Arabidopsis thaliana in an iron-deficient or insoluble iron-containing (Fe2O3) medium. It also can produce more siderophore and indole-3-acetic acid (IAA) as the concentration of iron ions decreased. However, it is unclear that the relationship between siderophore and IAA in

The objective of this work was to evaluate the effectiveness of the putative biocontrol agents (PBA) Bacillus paralicheniformis and Trichoderma asperelloides in vitro and in vivo to control two of the most important tomato plant diseases: vascular wilt (Fusarium oxysporum) and early blight (Alternaria alternata). The assessment of the in vitro interactions between the PBA and the phytopathogenic fungi was performed by dual confrontation assays. The biocontrol effectiveness of the individual and combined PBA treatments towards individual phytopathogen inoculations was evaluated in tomato plants
