Control of bull sperm cell volume during epididymal maturation.

Mature spermatozoa have a mechanism by which they can reduce cellular swelling caused by hypo-osmotic stress. The development of this ability during epididymal maturation in the bull was investigated. Caput and cauda sperm preparations were exposed to various osmotic stresses at 38 degrees C and measurements of cell volume made by electronic cell sizing. (1) Epididymal spermatozoa recovered and incubated in a medium isotonic with caudal epididymal plasma (360 mOsm kg(-1)) showed better viability and better volume regulatory ability than those incubated in a medium isotonic with seminal plasma

[News in paediatrics].

Every pediatrician will be confronted with newborns oryoung infants with skin lesions in proximity of the vertebral column. It is important not to miss a spinal dysraphism because of the risk of meningeal infection or of the possible presence of a tethered cord. A practical algorithm is presented. Non-accidental injury in young infants and toddlers is not rare but difficult to detect. Bruises and fractures are highly suspicious for non-accidental injury and should trigger specific investigations. Emergency departments and hospitals are switching from hypotonic to isotonic solutions as maintena

An electrolyte-based solution that increases corneal glycogen and conjunctival goblet-cell density in a rabbit model for keratoconjunctivitis sicca.

Thirty-two rabbits with monocular surgically induced keratoconjunctivitis sicca (KCS) underwent masked treatment for 12 weeks with 1 of 4 artificial tear solutions. Disease in each group of treated rabbits was compared with disease in untreated KCS controls. One of the solutions tested was a unique electrolyte-based formulation shown previously to preserve normal goblet-cell density after extended exposure in normal rabbits. Only the electrolyte-based solution decreased elevated tear osmolarity and sodium after 9 weeks of treatment (P less than 0.05). At 20 weeks, mean corneal glycogen and con

Further data on the inhibitory enterogastric reflex triggered by intestinal osmotic changes in cats.

The relationship between osmotic pressure in the intestinal lumen and gastric motor activity was studied in anesthetized cats. For this purpose the EMG was recorded in the antrum while the small intestine (duodenum and the first part of jejunum) was perfused with various solutions: tap water (5 mOsm), NaCl, mannitol and glucose having an osmotic pressure of 70, 138, 275, 550 or 1100 mOsm. Hypotonic and hypertonic solutions both induced a decrease in the gastric activity, i.e. an increase in period of electrical control activity. This effect was found to depend both on the osmolarity value (the

Regulatory volume increase in epithelial cells isolated from the mouse fourth ventricle choroid plexus involves Na(+)-H(+) exchange but not Na(+)-K(+)-2Cl(-) cotransport.

The aim of this study was to determine the ability of choroid plexus epithelial cells to volume regulate when exposed to hypertonic solutions, and furthermore to identify the ion transporters involved in any volume regulation. Experiments were performed on cells freshly isolated, using the enzyme dispase, from the mouse fourth ventricle choroid plexus. Cell volume was measured using a video-imaging method. Cells used in this study were all of a similar morphology and had a mean volume of 0.71pl. Cells shrank when superfused with hypertonic solutions to a minimum relative cell volume of 0.84+/-

Effect of osmolarity of artificial tear drops on relief of dry eye symptoms: BJ6 and beyond.

Patients with clinically well defined keratoconjunctivitis sicca (KCS) participated in two trials of hypo-osmolar tear substitutes. The trials were double masked, single crossover studies with computer generated random order allocation and were conducted by postal questionnaire. In the first trial a hypo-osmolar formulation using polyvinylpyrrolidone and hydroxyethyl cellulose was compared with the patients' usual treatment (BJ6 or hypromellose) and found to be inferior, especially in respect of blurring and stickiness caused by higher viscosity. In the second trial hypo-osmolar preparations o

Segmental differences along the crypt axis in the response of cell volume to secretagogues or hypotonic medium in the rat colon.

VIP caused a decrease in the diameter of rat colonic crypts. This decrease was followed by a volume increase at the middle and the upper third of the crypt, which finally led to an increase of crypt diameter above the initial control values. At the fundus only a cell shrinkage was observed. The volume increase at the upper parts of the crypt was suppressed by the inhibitor of the Na(+)-K(+)-Cl(-)-cotransporter, furosemide. When crypts were exposed to a hypertonic medium, cell shrinkage was followed by a regulatory volume increase at the middle and the upper third of the crypt but not at the fu

Low salt mimics effects of dark pulses on circadian pacemaker in cultured chick pineal cells.

Chick pineal cells in static culture display a persistent photosensitive circadian rhythm of melatonin production and release. Pulses of white light or darkness, in otherwise constant red light, induce phase shifts in subsequent cycles whose magnitude and direction depend on the phase at which the pulse is given. Such "phase-dependent phase shifts" are mediated by effects on the underlying pacemaker. We reported previously that inhibiting the Na-K-ATPase with ouabain or salt solutions lacking potassium evokes phase shifts with the same phase dependence as those induced by pulses of darkness. O

Efficacy and safety of isotonic versus hypotonic intravenous maintenance fluids in hospitalized children: an updated systematic review and meta-analysis of randomized controlled trials.

BACKGROUND: Iatrogenic hyponatremia is a common complication following intravenous maintenance fluid therapy (IV-MFT) in hospitalized children. Despite the American Academy of Pediatrics' 2018 recommendations, IV-MFT prescribing practices still vary considerably. OBJECTIVES: This meta-analysis aimed to compare the safety and efficacy of isotonic versus hypotonic IV-MFT in hospitalized children. DATA SOURCES: We searched PubMed, Scopus, Web of Science, and Cochrane Central from inception to October 1, 2022. STUDY ELIGIBILITY CRITERIA: We included randomized controlled trials (RCTs) comparing

Hypotonic polyuria: at the cross-roads of copeptin.

The aetiology of hypotonic polyuria, after excluding solute diuresis, is one of primary polydipsia, central, or nephrogenic diabetes insipidus. Theoretically, these disorders should be relatively easily distinguished based on history and the results of an indirect water deprivation test. Practically, however, there is a significant overlap in diagnostic evaluation, potentially leading to an erroneous diagnosis and deleterious management plan. The ability to measure a stimulated copeptin level, either with hypertonic saline or arginine infusion, has led to greater diagnostic accuracy.

[THE ROLE OF HYDROGEN SULFIDE IN VOLUME-DEPENDENT MECHANISMS OF REGULATION OF VASCULAR SMOOTH MUSCLE CELLS CONTRACTILE ACTIVITY].

The hydrogen sulfide (H2S) influence on the contractile activity of vascular smooth muscle cells (SMC) was studied on endothelium-denuded aortic ring segments of male Wistar rats with method of mechanography. Contractions of SMS were induced by incubation in high potassium solution as well as in hyper-, hypo- and isosmotic solutions. 5-100 LM of H2S donor--sodium hydrosulfide (NaHS) increased mechanical tension of SMC precontracted with high potassium solution that was abolished by bumetanide--the inhibitor of Na+, K+, 2Cl(-) -cotransporter (NKCC), but 100-1000 microM of NaHS relaxed SMS. Improvement of the ocular surface using hypotonic 0.4% hyaluronic acid drops in keratoconjunctivitis sicca.

BACKGROUND: The ocular surface changes of keratoconjunctivitis sicca (KCS) could be the result of the effect of an altered tear film on the epithelial environment. PURPOSE: To evaluate the possibility of improving the environmental conditions of the ocular surface by lowering tear osmolarity, increasing tear film volume and stabilising the tear film. Also, to study the effect of such an improvement on the epithelial cells of the ocular surface. METHODS: One hundred and thirty-five patients with a diagnosis of KCS were treated on a randomised basis with either unpreserved hypotonic 0.4% hyalu

Reduction of water permeability by anisotonic solutions in frog corneal epithelium.

PURPOSE: To study the effects of bathing solution osmolarity and Cl- secretagogues on the diffusional water permeability (Pdw) of the isolated frog corneal epithelium. METHODS: Isolated frog corneas, with the endothelium scraped off, were mounted as a partition between Ussing-type hemichambers. Unidirectional diffusional water fluxes (Jdw) were measured by adding 3H2O to one hemichamber and sampling from the other. Electrical parameters were measured simultaneously. Jdw was determined in control isosmotic conditions and after either changes in osmolarity of the bathing solutions or the additi

Choosing fluids to reduce the risks of acute electrolyte disturbances in children after a kidney transplant.

Intravenous (i.v.) fluid therapy is critically important in pediatric kidney transplantation. Because of the high volumes given perioperatively, transplant recipients can develop significant electrolyte abnormalities depending on the types of fluids used. Current practices in pediatric transplantation aim to balance risks of hyponatremia from traditionally used hypotonic fluids, such as 0.45% sodium chloride, against hyperchloremia and acidosis associated with isotonic 0.9% sodium chloride. Using the balanced solution Plasma-Lyte 148 as an alternative might mitigate these risks.

Hyponatraemia-treatment standard 2024.

Hyponatraemia is the most common electrolyte disorder in hospital patients associated with increased morbidity, mortality, hospital stay and financial burden. The speed of a correction with 3% sodium chloride as a 100- to 150-ml intravenous bolus or continuous infusion depends on the severity and persistence of the symptoms and needs frequent biochemical monitoring. The rapid intermittent administration of hypertonic saline is preferred for treatment of symptomatic hyponatraemia. In asymptomatic mild hyponatraemia, an adequate solute intake with an initial fluid restriction (FR) of 500 ml/day

The effects of tonicity, glucose concentration and temperature of an oral rehydration solution on its absorption and elimination.

Effects of different tonicities, glucose concentrations and temperatures of an oral rehydration solution (ORS) on its uptake and elimination in resting horses were studied. Fluid and electrolyte deficits similar to those occurring during prolonged exercise were induced by the administration of 1 mg/kg bwt of frusemide i.m., 3 h prior to the ORS. Fluid was administered via nasogastric tube at a volume equivalent to 4% bodyweight, which approximated diuretic induced losses. The uptake of fluid was evaluated by changes in haematocrit (PCV) and plasma total protein concentration (TP). Changes in e
