Imaging mass cytometry unveils functional and spatial remodeling of peri-lesional cells in jaw osteonecrosis.

Medication-related osteonecrosis of the jaw (MRONJ) is a severe complication associated with antiresorptive therapy, characterized by compromised bone and soft tissue integrity. However, the underlying tissue-level mechanisms remain poorly understood. To uncover cell functions and spatial organization surrounding ONJ lesions, imaging mass cytometry is used to profile lesions at single-cell resolution across epithelial, stromal, and vascular regions. Widespread immune infiltration is observed, with regulatory T cells, M2-like macrophages, exhausted T cells, and natural killer cells shifting fro

Luminescent Metal-Organic Framework Probes with Metallic and Fluorescent Dual-Properties for Mass Cytometry and Imaging.

Mass cytometry (CyTOF) and imaging mass cytometry (IMC), as cutting-edge technologies in single-cell analysis, are capable of detecting more than 40 biomarkers simultaneously on a single cell. However, their sensitivity and multiparameter detection capabilities have been long constrained by the development of metal labeling materials. Meanwhile, as an imaging technique, IMC has suffered from a rather slow data acquisition rate. Here, we present a luminescent PCN-224-OH material that exhibits both fluorescent and mass dual-functionality and is enriched with Zr-OH-/H2O active sites. Without the

Application of imaging mass cytometry for spatially profiling the microenvironment of salivary glands in primary Sjögren's syndrome.

Primary Sjogren's syndrome (pSS) is a slowly progressive, systemic autoimmune disorder characterized by gradual lymphocytic infiltration of exocrine glands. However, the spatially profiling the immune microenvironment in pSS is largely unclear, limiting the understanding of the complex interplay among cells within the microenvironment. Based on imaging mass cytometry (IMC) analysis of clinical pSS samples, we first revealed that labial salivary gland (LSG) comprised of epithelial, immune cells and stromal cells, and epithelial was the main cell type in LSG. Eight immune cells populations were

Detection of bladder cancer cells using quantitative interferometric label-free imaging flow cytometry.

Bladder cancer is one of the most common cancers with a high recurrence rate. Patients undergo mandatory yearly scrutinies, including cystoscopies, which makes bladder cancer highly distressing and costly. Here, we aim to develop a non-invasive, label-free method for the detection of bladder cancer cells in urine samples, which is based on interferometric imaging flow cytometry. Eight urothelial carcinoma and one normal urothelial cell lines, along with red and white blood cells, imaged quantitatively without staining by an interferometric phase microscopy module while flowing in a microfluidi

Imaging Mass Cytometry in Psoriatic Disease Reveals Immune Profile Heterogeneity in Skin and Synovial Tissue.

Imaging mass cytometry is a technology that enables comprehensive analysis of cellular phenotypes at the tissue level. We performed a multiparameter characterization of structural and immune cell populations in psoriatic skin and synovial tissue samples aimed at characterizing immune cell differences in patients with psoriasis and psoriatic arthritis. A panel of 33 antibodies was used to stain selected immune and structural cell populations. Imaging mass cytometry data were segmented into single cells on the basis of combinations of antibody stains. Single cells were then clustered into cell c

A 40-Marker Panel for High Dimensional Characterization of Cancer Immune Microenvironments by Imaging Mass Cytometry.

Multiplex immunophenotyping technologies are indispensable for a deeper understanding of biological systems. Until recently, high-dimensional cellular analyses implied the loss of tissue context as they were mostly performed in single-cell suspensions. The advent of imaging mass cytometry introduced the possibility to simultaneously detect a multitude of cellular markers in tissue sections. This technique can be applied to various tissue sources including snap-frozen and formalin-fixed, paraffin-embedded (FFPE) tissues. However, a number of methodological challenges must be overcome when devel

Application of imaging flow cytometry for characterization of acute inflammation in non-classical animal model systems.

Phagocytes display marked heterogeneity in their capacity to induce and control acute inflammation. This has a significant impact on the effectiveness of antimicrobial immune responses at different tissue sites as well as their predisposition for inflammation-associated pathology. Imaging flow cytometry provides novel opportunities for characterization of these phagocyte populations through high spatial resolution, statistical robustness, and a broad range of quantitative morphometric cell analysis tools. This study highlights an integrative approach that brings together new tools in imaging f

OpenIMC: an open-source platform for analyzing single-cell and spatial proteomics by imaging mass cytometry.

BACKGROUND: Imaging Mass Cytometry (IMC) enables highly multiplexed, spatially resolved single-cell proteomics, providing simultaneous measurement of dozens of protein markers while preserving tissue architecture. Despite its analytical power, IMC data analysis remains fragmented across multiple software environments, requiring researchers to combine independent tools for visualization, preprocessing, segmentation, feature extraction, phenotyping, batch correction, and spatial analysis. This fragmentation increases technical barriers, complicates reproducibility, and limits accessibility for n

Application of an imaging flow cytometry γ-H2AX assay for biodosimetry using supervised machine learning.

PURPOSE: Phosphorylation of the histone H2AX (γ-H2AX) is a rapid response to radiation-induced DNA double strand breaks (DSBs) and is a good biomarker for exposure to ionizing radiation. The signal has traditionally been detected by microscopy (spot counting) or by flow cytometry (fluorescent intensity). An imaging flow cytometry (IFC) method has been developed, which combines the high resolution of microscopy with the statistical power of flow cytometry methods to measure γ-H2AX in human lymphocytes. MATERIALS AND METHODS: The assay was optimized and validated for both sample acquisition and

Imaging Flow Cytometry of Legionella-Containing Vacuoles in Intact and Homogenized Wild-Type and Mutant Dictyostelium.

The causative agent of a severe pneumonia termed "Legionnaires' disease", Legionella pneumophila, replicates within protozoan and mammalian phagocytes in a specialized intracellular compartment called the Legionella-containing vacuole (LCV). This compartment does not fuse with bactericidal lysosomes but communicates extensively with several cellular vesicle trafficking pathways and eventually associates tightly with the endoplasmic reticulum. In order to comprehend in detail the complex process of LCV formation, the identification and kinetic analysis of cellular trafficking pathway markers on

Imaging flow cytometry data analysis using convolutional neural network for quantitative investigation of phagocytosis.

Macrophages play an important role in the adaptive immune system. Their ability to neutralize cellular targets through Fc receptor-mediated phagocytosis has relied upon immunotherapy that has become of particular interest for the treatment of cancer and autoimmune diseases. A detailed investigation of phagocytosis is the key to the improvement of the therapeutic efficiency of existing medications and the creation of new ones. A promising method for studying the process is imaging flow cytometry (IFC) that acquires thousands of cell images per second in up to 12 optical channels and allows mult

Semi-automated background removal limits data loss and normalizes imaging mass cytometry data.

Imaging mass cytometry (IMC) allows the detection of multiple antigens (approximately 40 markers) combined with spatial information, making it a unique tool for the evaluation of complex biological systems. Due to its widespread availability and retained tissue morphology, formalin-fixed, paraffin-embedded (FFPE) tissues are often a material of choice for IMC studies. However, antibody performance and signal to noise ratios can differ considerably between FFPE tissues as a consequence of variations in tissue processing, including fixation. In contrast to batch effects caused by differences in

Characterization of functional variables in epididymal alpaca (Vicugna pacos) sperm using imaging flow cytometry.

Epididymal alpaca sperm represent an alternative model for the study of alpaca semen. The objective of this study was to characterize the normal values of some functional variables in epididymal alpaca sperm using imaging flow cytometry. Alpaca testicles (n=150) were processed and sperm were recovered from the cauda epididymides. Only 76 samples with acceptable motility and sperm count were considered for assessment by imaging flow cytometry. Acrosome integrity and integrity/viability were assessed by FITC-PSA/PI and FITC-PNA/PI. Mitochondrial membrane potential (MMP) was assessed by MitoTrack

Image cytometry accurately detects DNA ploidy abnormalities and predicts late relapse to high-grade dysplasia and adenocarcinoma in Barrett's oesophagus following photodynamic therapy.

BACKGROUND AND AIMS: DNA ploidy abnormalities (aneuploidy/tetraploidy) measured by flow cytometry (FC) are strong predictors of future cancer development in untreated Barrett's oesophagus, independent of histology grade. Image cytometric DNA analysis (ICDA) is an optical technique allowing visualisation of abnormal nuclei that may be undertaken on archival tissue. Our aim was to determine the accuracy of ICDA vs FC, and evaluate DNA ploidy as a prognostic biomarker after histologically successful treatment with photodynamic therapy (PDT). METHODS: Nuclei were extracted from 40 mum sections of

Imaging Flow Cytometry Detection of Cytogenetic Abnormalities in Circulating CD34+ Cells Predicts Leukemic Transformation in Myelofibrosis.

Myelofibrosis is a myeloproliferative neoplasm with potential to transform to acute myeloid leukemia. This evolution is unpredictable and current assays lack the sensitivity and applicability needed to predict this transformation. While population-level data utilizing comprehensive genomic profiling can identify subgroups at higher risk of progression, they do not provide individualized information on the likelihood or timing of leukemia. Cytogenetic alterations are typically present in secondary leukemia. We aimed to determine whether these changes could be detected at an early stage. To achi

Unveiling the immune landscape and microenvironmental architecture of IgA nephropathy using single-cell imaging mass cytometry.

OBJECTIVE: The heterogeneity in the pathology of IgA nephropathy (IgAN) represents a significant gap in current research, emphasising the need for individualised treatment strategies. The role of the glomerular microenvironment in driving IgAN heterogeneity remains largely unknown. METHODS: This study employed Imaging Mass Cytometry (IMC) to analyse 34 primary IgAN renal biopsies. A panel of 35 protein markers was used to identify and characterise 17 cell clusters. RESULTS: Immune cells composed of 9.64% of the glomerular cell population, with macrophages being the predominant subset. While

Associating the morphology of helper T cells with cytokine activity through imaging flow cytometry and deep learning.

The immune system protects the body from foreign substances, such as viruses and bacteria. Among its key orchestrators are helper T cells, which coordinate immune responses by activating other immune cells to eliminate these threats. Helper T cell activation has been widely studied, often by stimulating cells in vitro and measuring changes in receptor expression. However, systematic assessment of activation at scale remains challenging because it requires both large single-cell imaging datasets and advanced computational methods to analyse complex morphological features. Here, using imaging fl

A novel capillary-driven dual-mode imaging flow cytometry system for malaria parasite detection and quantification.

Malaria remains a significant global health burden, particularly in tropical and subtropical regions with limited resources. Accurate quantification of parasitemia is critical for effective treatment and disease management. While microscopy is the gold standard in diagnostics, it is labour-intensive and highly dependent on operator expertise. Here, we present a capillary-driven image flow cytometer with a cost comparable to microscopy-based diagnostics. This system integrates brightfield and fluorescence imaging into a single image, encoding morphological information in the red channel and flu
