Two novel types of dimeric alkaloids were isolated from the marine sediment-derived mutant strain Saccharopolyspora erythraea SCSIO 07745/Δspo11. Among them, sacchaindol A (1) represents a previously unreported class of dimeric aminoquinolinone alkaloids, whereas sacchaindols B (2) and C [(±)-3] comprise a rare fused 6/6/5/5 tetracyclic indole alkaloid framework. Their structures were elucidated using a combination of spectroscopic analyses, single-crystal X-ray diffraction, and computational methods. Hypothetical biosynthetic pathways for 1-3 were proposed. Notably, sacchaindols C [(±)-3] exh

Total syntheses of the antirhine alkaloids are described. The cyanide-catalyzed imino-Stetter reaction of the aldimine derived from ethyl 2-aminocinnamate and 4-bromopyridine-2-carboxaldehyde provided a 2-pyridinyl substituted indole-3-acetate, which was further converted into the corresponding indoloquinolizidinium intermediate through C-ring formation. Subsequent trans-selective installation of the homoallylic alcohol side-chain at C-15 in the resulting indoloquinolizidinium allowed the total syntheses of antirhine and its known epimer.

Rhynchines A-E (1-5), five new indole alkaloids with an unprecedented skeleton, were isolated from Uncaria rhynchophylla. The new skeleton was characterized by an indole moiety and a 2-oxa-8-azatricyclo[6,5,01,5,01,8]tridecane core, forming a unique 6/5/7/5/5 ring system. Their structures were determined by nuclear magnetic resonance, high-resolution electrospray ionization mass spectrometry, infrared spectroscopy, and calculated electronic circular dichroism (ECD) and were confirmed by X-ray crystallography. Interestingly, 1 and 2 showed strong inhibitory activities against the Cav3.1 calcium

In previous studies, we demonstrated the leishmanicide effect of coronaridine, a natural indole alkaloid isolated from stem bark of Peschiera australis (Delorenzi et al., Antimicrob. Agents Chemother. 45:1349-1354, 2001). In this study we show the leishmanicidal effect of the synthetic coronaridine and its racemic 18-methoxylated analog, 18-methoxycoronaridine. Both alkaloids revealed a potent leishmanicide effect against Leishmania amazonensis, a causative agent of cutaneous and diffuse cutaneous leishmaniasis in the New World. Despite their potent leishmanicide effect, both alkaloids were ne

Several reviews have been published on Artemisia's derived natural products, but it is the first attempt to review the chemistry and pharmacology of more than 80 alkaloids and allied nitrogen compounds obtained from various Artemisia species (covering the literature up to June 2018). The pharmacological potential and unique skeleton types of certain Artemisia's alkaloids provoke the importance of analyzing Artemisia species for bioactive alkaloids and allied nitrogen compounds. Among the various types of bioactive Artemisia's alkaloids, the main classes were the derivatives of rupestine (pyrid

Found recently in stignomatales, the Stig cyclases catalyze the Cope rearrangement and intramolecular cyclization to produce complex indole alkaloids. Five crystal structures were solved of subfamily 1 and 2 Stig cyclases, which adopt a β-sandwich fold like the non-catalytic carbohydrate-binding motif. Several complex structures were also determined of indole-based compounds, which are bound to the hydrophobic terminal cavity, where a conserved Asp residue makes an H-bond to the indole N and triggers the acid-catalyzed Cope rearrangement. Through analyzing the enzyme-ligand interactions and mu

Eleven characteristic monoterpenoid indole alkaloids from Uncaria rhynchophylla were isolated, structurally assigned, and evaluated in parallel for inhibitory effects on human Nav1.7, Nav1.5, Cav3.1, and Kv1.5 channels using whole-cell patch-clamp recordings. Nav1.7 and Nav1.5 showed the broadest responses, with active inhibitors displaying micromolar IC50 values against the two sodium channels. Corynanthe-type alkaloids were the main contributors to Cav3.1 inhibition, whereas Kv1.5 was less sensitive and responded mainly to rhynchophylline and isorhynchophylline. The observed channel preferen

2,3-Indole-fused 1,4-diazocines represent a new family of indole alkaloid compounds and are difficult to access by the reported protocols. Herein, we report a copper-catalyzed decarboxylative cyclization of cyclic propargylic carbamates with imidazolidines via sequential C-N/C-N/C-C bond formation to deliver a series of 2,3-indole-fused 1,4-diazocines, with a broad substrate scope and mild conditions.

INTRODUCTION: Tabernaemontana is a genus of plants traditionally used to treat various diseases, such as syphilis. Many cytotoxic indole alkaloids were isolated and characterized from plants of this genus, suggesting their potential applications as chemotherapeutical drugs or drug leads. Nonetheless, the Malaysian native species Tabernaemontanapolyneura was relatively unknown. This study provides an overview of the botany, ethnobotany, phytochemistry, and in silico ADMET analysis of cytotoxic alkaloids isolated from T. polyneura.
METHODS: A comprehensive literature search was conducted using

Tryptophan-derived metabolites are indole-containing compounds with applications in medicine, functional foods, and agriculture. Conventional production via plant extraction or chemical synthesis is often inefficient and unsustainable, prompting the development of microbial biosynthesis. In this review, these metabolites are classified based on reaction sites and the extent of l-tryptophan scaffold remodeling into side-chain-modified compounds, indole ring-functionalized compounds, oxidative coupling products, and complex indole alkaloids, providing a framework to compare biosynthetic logic ac

An efficient synthesis of N-fused polycyclic and 2,3-disubstituted indoles by copper-catalyzed direct annulation/acyl migration reaction of enaminones is reported. This strategy features cheap and low loading of the catalyst/ligand, readily available starting materials, and good functional group compatibilities. Notably, allyl-containing substrates are also tolerated, which allows the downstream derivatization toward indole alkaloids.

We herein report the copper-catalyzed C-S bond coupling reaction of indoles with N-thiosuccinimides, resulting in moderate to excellent yields of mono- and bis-sulfenylated compounds such as arylthioindoles, alkylthioindoles, selenylated indoles, and cysteine-substituted indoles. Thioarylation and thioglycosylation at the C2 position of indole alkaloids in the Radix Isatidis were achieved via structural modification. The first total syntheses of isatindigotindolosides III and IV have been successfully carried out. The electrophilic sulfenyl bromides generated in situ can play an important role

Malaria is one of the major health problems in developing countries. The disease kills a large number of people every year and also affects financial status of many countries. Resistance of the plasmodium parasite, the causative agent, to the existing drugs, including chloroquine, mefloquine, and artemisinin based combination therapy (ACT), is a serious global issue in malaria treatment and control. This warrants an urgent quest for novel compounds, particularly from natural sources such as medicinal plants. Alkaloids have over the years been recognized as important phytoconstituents with inte
