Case study: three acute 241Am inhalation exposures with DTPA therapy.

Three workers incurred inhalation exposures to Am oxide as a result of waste sorting and compaction activities. The exposure magnitudes were not fully recognized until the following day when an in-vivo lung count identified a significant lung deposition of Am in a male worker, and DTPA chelation therapy was initiated. Two additional workers (one female and one male) were then identified as sufficiently exposed to also warrant therapy. In-vivo bioassay measurements were performed over the ensuing 6 mo to quantify the 241Am activity in the lungs, liver, and skeleton. Urine and fecal samples were

Acute inhalational exposure to chlorodifluoromethane (freon-22): a report of 43 cases.

We report 43 cases of chlorodifluoromethane (Freon-22) intoxication that occurred on August 5, 2003 when a freezer in a seafood factory exploded. In this accident, 80 workers were exposed to Freon-22 gas and 43 workers developed symptoms and were transferred to six hospitals. Neurological symptoms including dizziness, headache, and nausea were most frequently observed (40 of 43 patients). One patient was comatose but recovered within 1 h with oxygen inhalation. Airway and respiratory symptoms including dysesthesia of the tongue, pharyngitis, and shortness of breath were also frequently observe

Ingestion, inhalation, and dermal exposures to chloroform and trichloroethene from tap water.

Individuals are exposed to volatile compounds present in tap water by ingestion, inhalation, and dermal absorption. Traditional risk assessments for water often only consider ingestion exposure to toxic chemicals, even though showering has been shown to increase the body burden of certain chemicals due to inhalation exposure and dermal absorption. We collected and analyzed time-series samples of expired alveolar breath to evaluate changes in concentrations of volatile organic compounds being expired, which reflects the rate of change in the bloodstream due to expiration, metabolism, and absorp

Exposure of rats to inhalational anesthetics alters the hepatobiliary clearance of cholephilic xenobiotics.

Several recent studies indicate that anesthesia-induced alteration of UDP-glucuronic acid concentrations can affect the rate of xenobiotic glucuronidation by UDP-glucuronosyltransferases. Other data demonstrate that the biliary excretion of several cholephilic drugs is depressed in rats anesthetized with diethyl ether. The present study has examined the effect of 2% halothane, 1.5% isoflurane, 2% enflurane and 3% sevoflurane on the clearance and biliary excretion of acetaminophen, digoxin, phenol red and phenol-3,6-dibromphthalein disulfonate. All volatile anesthetics reduced hepatic UDP-glucu

Experimental human exposures to tetrachloroethylene vapor and elimination in breath after inhalation.

Volunteers were exposed in a controlled environment chamber and concentrations of tetrachloroethylene in alveolar air were determined. Results show a proportional relationship between the level of exposure and the alveolar concentration curve. Accordingly, tetrachloroethylene breath decay curves can be used as a method to monitor time-weighted average exposures.

Gastroesophageal disease risk and inhalational exposure a systematic review and meta-analysis.

Environmental exposure-associated diseases, particularly in the context of rising air pollution and inhalant use, are an active area of research. Our group is dedicated to the study of exposure-related inflammation and its downstream adverse health effects. While many studies have focused on the impact of environmental exposures on respiratory sequelae, there is growing evidence of the involvement of other systems including gastrointestinal. This systematic review provides updates on the associations between inhalation exposures and the risk of upper gastrointestinal disease. Primary search id

Difference in Exposure and Tolerability when Comparing Intratracheal Instillation with Inhaled Delivery.

PURPOSE: To investigate the impact of two different routes of administration on the lung and systemic exposure of drugs designed for local delivery to the lung. METHODS: In our comparative studies, similar lung doses of three different drugs were administered to rodents by both intratracheal instillation and inhaled delivery. RESULTS: An obvious but unexpected difference in the exposure was observed. Immediately after the dose, the initial plasma concentration was much higher whereas the initial fraction of the lung dose in the lung was clearly lower for the instillation compared with the in

Estimating cobalt exposure in respirable dust from cobalt in inhalable dust.

Cobalt is a commonly used element in metal industry. Exposure to workers occurs mainly by inhalation of cobalt-containing dust. For the evaluation of cobalt exposure, risk assessment and investigations on occupational diseases, measurements of cobalt in respirable dust are needed. Up to now, often only data for cobalt in inhalable dust are available, which is due to the earlier classification of the limit value in this fraction. Therefore, a possibility to convert cobalt concentrations mathematically from inhalable into respirable concentrations is desirable. In this study, 639 parallel measur

Estimating nickel exposure in respirable dust from nickel in inhalable dust.

The conversion of dust components is of high importance for retrospective evaluations of exposure levels, of occupational diseases or the time trend of occupational dust exposure. For this purpose, possibilities to convert nickel concentrations from inhalable to respirable dust are discussed in this study. Therefore, 551 parallel measurements of nickel concentrations in inhalable and respirable dust fractions were extracted from the exposure database MEGA (maintained at the Institute for Occupational Safety and Health of the German Social Accident Insurance) and investigated by linear regressi

Benchmark dose analysis for Bacillus anthracis inhalation exposures in the nonhuman primate.

There is considerable variability in the published lethality values for inhalation exposures of Bacillus anthracis. The lack of consensus on an acceptable dose-response relationship poses a significant challenge in the development of risk-based management approaches for use following a terrorist release of B. anthracis spores. This article reviewed available B. anthracis dose-response modeling and literature for the nonhuman primate, evaluated the use of the U.S. Environmental Protection Agency's Benchmark Dose Software (BMDS) to fit mathematical dose-response models to these data, and reporte

Development of an inhalational Bacillus anthracis exposure therapeutic model in cynomolgus macaques.

Appropriate animal models are required to test medical countermeasures to bioterrorist threats. To that end, we characterized a nonhuman primate (NHP) inhalational anthrax therapeutic model for use in testing anthrax therapeutic medical countermeasures according to the U.S. Food and Drug Administration Animal Rule. A clinical profile was recorded for each NHP exposed to a lethal dose of Bacillus anthracis Ames spores. Specific diagnostic parameters were detected relatively early in disease progression, i.e., by blood culture (∼37 h postchallenge) and the presence of circulating protective anti

Airway response of asthmatic subjects to inhaled allergen after exposure to pollutants.

BACKGROUND: Recent studies have suggested that air pollutants resulting from vehicle exhaust emissions and burning of fossil fuels, either in combination or individually, may enhance the airway response of asthmatic subjects to inhaled allergen. It was hypothesised that the airway response to inhaled allergen after exposure to a combination of 400 ppb nitrogen dioxide (NO2) and 200 ppb sulphur dioxide (SO2) is increased 24-48 hours after exposure. METHODS: Thirteen mild atopic asthmatic volunteers were exposed for six hours to a single exposure of air and three exposures of the combination of

Nasal lavage technique reveals regular inhalation exposure of microplastics, not associated from face mask use.

During the COVID-19 pandemic, the use of face masks has been a worldwide primary protection measure to contain the spread of the virus. However, very little information is known about the possible inhalation of microplastics (MP) from wearing masks. This pilot study evaluates the presence of MP accumulated in nasal cavities through the nasal lavages technique. Six different commercial face masks were tested in 18 participants during five working days (8 h use/day). Eight different polymers (polystyrene, polyamide, poly(ethylene - propylene) diene monomer, polyester, polyethylene, polyvinyliden

Numerical and experimental analysis of pollen inhalation exposure in nasal airways following various middle turbinectomy.

BACKGROUND: Middle turbinectomy (MT) has always been controversial. MT significantly alters the anatomy and redistributes the inhaled air. The current study is designed to quantify the effect of MT with varying resection volumes on airflow and associated pollen inhalation exposure characteristics in the nasal airways. METHODS: Six realistic models following bilateral comprehensive Functional Endoscopic Sinus Surgery (FESS) deriving from CT images were constructed and their corresponding post-MT models with four types of MT procedures were virtually conducted. Chemical exposure and alveolar macrophages responses: 'the role of pulmonary defense mechanism in inhalation injuries'.

Epidemiological and clinical studies have indicated an association between particulate matter (PM) exposure and acute and chronic pulmonary inflammation, which may be registered as increased mortality and morbidity. Despite the increasing evidence, the pathophysiology mechanism of these PMs is still not fully characterised. Pulmonary alveolar macrophages (PAMs), as a predominant cell in the lung, play a critically important role in these pathological mechanisms. Toxin exposure triggers events associated with macrophage activation, including oxidative stress, acute damage, tissue disruption, re

Evaluation of inhalation toxicology after a 90-day xylitol aerosol exposure in Sprague-Dawley rats.

Xylitol is a hygroscopic compound known to protect nasal cavity against bacteria. It has also been developed into nasal spray and evaluated as a potential candidate drug for respiratory diseases. Consequently, it is necessary to study its inhalation toxicity. Based on our previous study on its subacute inhalation toxicity, this study aimed to investigate the safety of xylitol inhalation for long-term use. According to the OECD Test Guideline 413, Sprague-Dawley rats were randomly divided into six groups and exposed with different concentrations of xylitol aerosol or air. After exposure for 90-

Risk assessment of population inhalation exposure to volatile organic compounds and carbonyls in urban China.

Over the past three decades, China has experienced rapid urbanization. The risks to its urban population posed by inhalation exposure to hazardous air pollutants (HAPs) have not been well characterized. Here, we summarize recent measurements of 16 highly prevalent HAPs in urban China and compile their distribution inputs. Based on activity patterns of urban Chinese working adults, we derive personal exposures. Using a probabilistic risk assessment method, we determine cancer and non-cancer risks for working females and males. We also assess the uncertainty associated with risk estimates using
