[Comparative study of variola virus and monkeypox virus interferon-gamma-binding].

DNA fragments containing genes for coding IFN-gamma-binding proteins (IFNgammaBPs) of variola virus (VARV) and monkeypox virus (MPXV) were obtained from viral genomes using PCR. Isolated genes coding desired proteins were expressed in the insect Sf21 cells using baculovirus expression system. Secreted recombinant IFNgammaBPs were isolated from culture medium of infected Sf21 cells through affinity chromatography procedure. SDS-PAAG and Western blot analysis of culture medium of infected insect cells and preparations of purified recombinant IFNgammaBPs indicated that recombinant viral proteins

OdTx12/GNA, a chimeric variant of a β excitatory toxin from Odontobuthus doriae, reveals oral toxicity towards Locusta migratoria and Tenebrio molitor.

OdTx12, a beta excitatory toxin from Odontobothus doriae had previously been identified and characterized. It had been shown that OdTx12 causes significant lethal effects on insects by injection but does not show any toxicity on mice. Due to the natural ineffectiveness of scorpion toxins to act as oral toxins, OdTx12 was fused to Galanthus nivalis agglutinin (GNA), a protein with the potential to cross the insect gut. The sequence of OdTx12/GNA gene was chemically synthesized, cloned in Escherichia coli, and expressed. The effect of the purified fusion protein (OdTx12/GNA) was assessed on the

Impact of UV-A radiation on the performance of aphids and whiteflies and on the leaf chemistry of their host plants.

Ultraviolet (UV) radiation directly regulates a multitude of herbivore life processes, in addition to indirectly affecting insect success via changes in plant chemistry and morphogenesis. Here we looked at plant and insect (aphid and whitefly) exposure to supplemental UV-A radiation in the glasshouse environment and investigated effects on insect population growth. Glasshouse grown peppers and eggplants were grown from seed inside cages covered by novel plastic filters, one transparent and the other opaque to UV-A radiation. At a 10-true leaf stage for peppers (53 days) and 4-true leaf stage f

Processing insects for use in the food and feed industry.

Although insects are becoming more accepted as potential protein sources for food and feed, the appearance of the insect may be off-putting due to associations of disgust. Edible insects are more likely to be eaten if they are processed into non-recognizable forms. Thus, insects require the use of commercial processing methods that will render the protein suitable for food/feed formulation, while maintaining the safety, nutritional and sensory quality of the final product. Common methods that can be used include lipid extraction, enzymatic proteolysis, commercial thermal processing (e.g. blanc

Immune response profiles from humans experimentally exposed to Phlebotomus duboscqi bites.

INTRODUCTION: Cutaneous leishmaniasis is a neglected vector-borne parasitic disease prevalent in 92 countries with approximately one million new infections annually. Interactions between vector saliva and the human host alter the response to infection and outcome of disease. METHODS: To characterize the human immunological responses developed against saliva of Phlebotomus duboscqi, a Leishmania major (L. major) vector, we repeatedly exposed the arms of 14 healthy U.S volunteers to uninfected P. duboscqi bites. Blood was collected a week after each exposure and used to assess total IgG antibod

Recombinant SpTrf proteins illustrate putative functions of native SpTrf proteins: from immune response to resolution in sea urchins.

The complex and sophisticated sea urchin innate immune system relies on conserved immune receptor and effector molecules plus the euechinoid-specific Transformer family. Native SpTransformer (natSpTrf) proteins in Strongylocentrotus purpuratus respond to immune challenge by augmenting phagocytosis of microbes. These proteins show significant sequence diversity both within and among sea urchins with the predicted benefit of host protection through the recognition of a broad range of pathogens. Prior reports show that 6 recombinant (r)SpTrf proteins from insect cells induce phagocytosis when cro

Identification and Functional Analysis of Candidate Effectors in the Sorghum Aphid, Melanaphis sorghi.

The sorghum aphid (Melanaphis sorghi), a phloem-feeding insect pest, causes severe economic losses in sorghum-producing regions worldwide. During feeding, aphids secrete salivary proteins with potential effector functions that play pivotal roles in plant-insect interactions. However, relatively few secreted proteins and effectors have been characterized in M. sorghi. In this study, proteomic analysis of aphid-infested sorghum leaves identified 69 putative aphid-derived proteins. Bioinformatic analyses were subsequently used to predict signal peptides within these proteins. Among the five candi

Analysis of blood-induced Anopheles gambiae midgut proteins and sexual stage Plasmodium falciparum interaction reveals mosquito genes important for malaria transmission.

Plasmodium invasion of mosquito midguts is a mandatory step for malaria transmission. The roles of mosquito midgut proteins and parasite interaction during malaria transmission are not clear. This study aims to identify mosquito midgut proteins that interact with and affect P. falciparum invasion. Based on gene expression profiles and protein sequences, 76 mosquito secretory proteins that are highly expressed in midguts and up-regulated by blood meals were chosen for analysis. About 61 candidate genes were successfully cloned from Anopheles gambiae and expressed in insect cells. ELISA analysis

Identification and type III-dependent secretion of the Yersinia pestis insecticidal-like proteins.

Plague, or the Black Death, is a zoonotic disease that is spread from mammal to mammal by fleas. This mode of transmission demands that the causative agent of this disease, Yersinia pestis, is able to survive and multiply in both mammals and insects. In recent years the complete genome sequence of a number of Y. pestis strains have been determined. This sequence information indicates that Y. pestis contains a cluster of genes with homology to insecticidal toxin encoding genes of the insect pathogen Photorhabdus luminescens. Here we demonstrate that Y. pestis KIM strains produced the encoded pr

Functional proteomic insights into the lateral oviduct-associated secretions of a bean bug.

The secretions within the lateral oviduct lumen of the insect female reproductive tract (FRT) play crucial roles in reproduction, yet their protein composition and functional mechanisms remain poorly understood. In this study, we revealed that the lateral oviduct of the bean bug, Riptortus pedestris, is composed of an outer muscle layer and an inner epithelium with a microvillar border. Proteomic analysis identified 159 FRT-highly expressed proteins, with functional characterization focusing on 10 putative secretory proteins, including 2 ovary-specific (RpLoap2 and RpLoap4), 7 oviduct-specific

An Epstein-Barr Virus Nuclear Antigen 1 (EBNA1) Serology Test Strip for Nasopharyngeal Carcinoma Risk Screening.

Antibodies to Epstein-Barr virus (EBV) proteins can predict nasopharyngeal carcinoma (NPC) risk. We previously defined a prototype EBNA1 protein panel and multiplex immunoblot assay that distinguishes NPC risk several years pre-diagnosis. Assay throughput and specificity are critical to effectively implement a population-level screening program. Here, we developed a strip test assay-EBNA1 SeroStrip-HT-with an objective to increase throughput and maximize specificity. EBNA1 full-length (FL) and glycine-alanine repeat deletion mutants (dGAr) were purified from insect and mammalian cells to scree

Insects as sources of iron and zinc in human nutrition.

Dietary deficiencies in Fe and Zn are globally widespread, causing serious health problems such as anaemia, poor pregnancy outcomes, increased risk of morbidity and mortality, stunted growth and impaired physical and cognitive development. Edible insects, of which a diversity of over 2000 species is available, are dietary components for about 2 billion individuals and are a valuable source of animal protein. In the present paper, we review the available information on Fe and Zn in edible insects and their potential as a source of these micronutrients for the rapidly growing human population. T

Microbial pathogens of edible insects: a growing problem for the insect farming industry.

Insect farming is widely extolled as a sustainable alternative to the traditional agricultural production of protein for human and animal consumption. However, pathogen contamination endangers insect health, food safety, production yields and market acceptance. Because insect farming is intensive, growth and transmission of pathogens are promoted, elevating the risk of disease outbreaks with severe economic outcomes. Fungal pathogens can invade host insects through their cuticle, reproducing within the nutrientrich haemolymph within the haemocoel until the host's defences are overwhelmed and t
