Metabolic stress in the tumor microenvironment (TME) promotes T cell dysfunction and immune checkpoint inhibitor (ICI) resistance. We examined the contribution of activating transcription factor 4 (ATF4), the central node of the integrated stress response (ISR), to T cell dysfunction in tumors. CD8+ tumor-infiltrating lymphocytes (TILs) in patient samples exhibited chronic ATF4 activity, which was reflected across various tumor models. Hypoxia in the TME imposed chronic ATF4 activity via the ISR kinases. ATF4 overexpression in CD8+ T cells induced metabolic polarity, mitochondrial oxidative st

The leukodystrophy Pelizaeus-Merzbacher disease (PMD) is caused by myelin protein proteolipid protein gene (PLP1) mutations. PMD is characterized by oligodendrocyte death and CNS hypomyelination; thus, increasing oligodendrocyte survival and enhancing myelination could provide therapeutic benefit. Here, we use the PMD mouse model Jimpy to determine the impact of the integrated stress response (ISR) on the oligodendrocyte response to mutant PLP expression. Male Jimpy animals in which the ISR-triggering eukaryotic initiation factor (eIF) 2α kinase, protein kinase-like endoplasmic reticulum kinas

Inflammatory responses within the ovarian microenvironment are increasingly recognized as significant disruptors of ovarian function, yet their specific effects on early follicular development, particularly the activation of primordial follicles, remain poorly understood. In this study, we employ a mouse model of transient lipopolysaccharide (LPS)-induced inflammation to mimic the inflammatory conditions associated with chronic pelvic inflammatory disease (PID) and systemic infections. We demonstrate that LPS stimulation triggers the premature activation of primordial follicles, leading to a d

Induction of autophagy and the integrated stress response is important for amino acid homeostasis. It remains unknown whether the autophagy coregulates both mechanistic target of rapamycin complex 1 (mTORC1) signaling and the integrated stress response. In mouse C2C12 myotubes, we found that amino acid limitation induced autophagy and that the subsequent release of amino acid is required to sustain mTORC1 signaling. Inhibition of autophagy by bafilomycin A1 or chloroquine treatment during amino acid scarcity abolished mTORC1 signaling, an effect that could be rescued by inhibiting protein synt

As sessile organisms, plants must continuously balance growth and stress adaptation in response to fluctuating environments. This balance is largely governed by two evolutionarily conserved kinase systems, the Target of Rapamycin Complex (TORC) and Snf1-Related Kinases (SnRKs). Under favorable conditions, plants adopt a "growth mode" in which sufficient nutrients, energy, and growth-promoting hormones activate TORC to drive anabolic metabolism, cell proliferation, and biomass accumulation. By contrast, under adverse conditions such as energy limitation, nutrient deprivation, and other environm

Replication and transcription share the DNA template and must be coordinated to preserve genome integrity. Although temporally organized across the cell cycle, essential transcriptional programs-encoding replication machinery, canonical histones, and DNA repair factors-operate concurrently with DNA synthesis during S phase. Transcription-replication conflicts (TRCs) therefore constitute a recurrent endogenous source of replication stress (RS), particularly under hypertranscriptional or chromatin-constrained conditions. A frequent outcome of TRCs is the formation of R-loops-RNA:DNA hybrids that

The cell envelope fortifies bacterial cells against antibiotics and other insults. Species in the Mycobacteriales order have a complex envelope that includes an outer layer of mycolic acids called the mycomembrane (MM) and a cell wall composed of peptidoglycan and arabinogalactan. This envelope architecture is unique among bacteria and contributes significantly to the virulence of pathogenic Mycobacteriales like Mycobacterium tuberculosis. Characterization of pathways that govern envelope biogenesis in these organisms is therefore critical in understanding their biology and for identifying new

The gaseous plant hormone ethylene (ET) regulates diverse growth responses and adaptive processes to environmental stimuli. Although the role of ET in stress signalling is well established, its effects on stomatal responses remain paradoxical. ET induces stomatal closure in some contexts yet antagonizes abscisic acid-mediated closure in others. While diverse studies have reported that ET regulates stomatal responses, the differential role of ET in stomatal signalling has not been reconciled into a unified framework. Here, we synthesize current evidence into a bidirectional model of ET-dependen

Climate change is intensifying extreme heat events that compromise poultry health, welfare, and productivity. Although RNA-seq has been widely used to investigate heat stress responses, individual studies are constrained by small sample sizes, experimental heterogeneity, and discordant differential expression signatures, limiting the identification of conserved molecular responses across tissues. We conducted an integrative analysis of 11 independent RNA-seq datasets comprising 212 samples (110 liver, 62 breast muscle, and 40 heart) from broiler chickens exposed to heat stress. Differential ex

Listeria monocytogenes is a significant concern in the food industry due to its association with outbreaks of listeriosis, particularly affecting vulnerable populations. High-throughput technologies such as RNA sequencing (RNA-seq) and proteomics offer valuable insights into the molecular responses of L. monocytogenes to stress environments. In this study, a combined transcriptomic and immunoproteomic approach was applied to explore the stress response mechanisms of the L. monocytogenes strain ST7, which was responsible for an outbreak in central Italy. The bacterium was exposed to both optima

Stress responses are essential for cellular survival and organismal, developmental progression. In somatic cells and embryonic stem cells (ESC), there exists the sequential transcriptional programs of the integrated stress response (ISR) and senescence-associated secretory phenotype (SASP), which enable stepwise adaptation to adverse conditions. We recently identified a distinct developmentally associated stress response (DASR) in mouse ESC that hypothetically emerges only after successful completion of cellular adaptation (ISR), reflecting a shift toward organismal-level adaptation. In explan

INTRODUCTION: Ankylosing spondylitis (AS) is a chronic inflammatory disorder with poorly defined pathogenic mechanisms. The integrated stress response (ISR), an evolutionarily conserved signaling network, is implicated in AS development. This research endeavored to identify biomarkers for AS, thereby offering novel targets and approaches for therapeutic intervention.
METHODS: Transcriptomic profiling of peripheral blood from AS patients identified differentially expressed genes. Mendelian randomization (MR) was applied to infer causal associations between ISR-related genes and AS susceptibili

BACKGROUND: Chronic obstructive pulmonary disease (COPD) has high morbidity and mortality, and integrated stress response (ISR) participates in its progression. Traditional COPD diagnosis relies mainly on GOLD criteria, while ISR-based auxiliary molecular markers are still lacking. This study aimed to comprehensively analyze and identify ISR-related biomarkers in COPD, and to uncover their underlying molecular mechanisms.
METHODS: COPD data were obtained from the GEO database. Biomarkers were identified and validated through a comprehensive approach, including differential expression analysis

Salinity fluctuations pose a severe threat to the survival and physiological homeostasis of freshwater crustaceans. Here, an integrated transcriptomic and metabolomic approach was utilised to characterise the dynamic regulatory responses in the hepatopancreas of adult Macrobrachium rosenbergii subjected to acute high-salinity stress (20‱). The number of differentially expressed genes (DEGs) and metabolites (DMs) increased from 24 to 48 h, signalling a transition from an initial acute stress reaction to systemic homeostatic remodelling. At the molecular level, M. rosenbergii activated autophagy

BACKGROUND: Heme-regulated inhibitor (HRI/EIF2AK1) is a protein kinase that initiates the integrated stress response (ISR) following mitochondrial damage and/or oxidative stress. Dependent on a context, ISR may promote death or recovery of cells under stress. This study was initiated to determine HRI contributions to brain damage after experimental intracerebral hemorrhage (ICH) where oxidative toxicity of hemoglobin is a major trigger of neural cell loss.
MATERIALS AND METHODS: Publicly available transcriptomic data from a mouse model of ICH were analyzed using standard bioinformatic tools.

BACKGROUND AND AIMS: Drug-induced liver injury (DILI) is one of the most frequent reasons for failure of drugs in clinical trials or market withdrawal. Early assessment of DILI risk remains a major challenge during drug development. Here, we present a mechanism-based weight-of-evidence approach able to identify certain candidate compounds with DILI liabilities due to mitochondrial toxicity.
METHODS: A total of 1587 FDA-approved drugs and 378 kinase inhibitors were screened for cellular stress response activation associated with DILI using an imaging-based HepG2 BAC-GFP reporter platform inclu

For bipolar disorder (BD), the inconsistency of treatment guidelines and the long phases of pharmacological adjustment remain major challenges. BD is known to be comorbid with many medical and psychiatric conditions and they may share inflammatory and stress-related aetiologies, which could give rise to this association. The integrated stress response (ISR) responds to various stress conditions that lead to alterations in cellular homeostasis. However, as a causative mechanism underlying cognitive deficits and neurodegeneration in a broad range of brain disorders, the impact of ISR on BD is un

Transcription factors (TFs) play critical roles in mediating the plant response to various abiotic stresses, particularly heat stress. Plants respond to elevated temperatures by modulating the expression of genes involved in diverse metabolic pathways, a regulatory process primarily governed by multiple TFs in a networked configuration. Many TFs, such as WRKY, MYB, NAC, bZIP, zinc finger protein, AP2/ERF, DREB, ERF, bHLH, and brassinosteroids, are associated with heat shock factor (Hsf) families, and are involved in heat stress tolerance. These TFs hold the potential to control multiple genes,

BACKGROUND: Mitochondria is a key organelle for energy production and cellular adaptive response to intracellular and extracellular stresses. Mitochondrial stress can be evoked by various stimuli such as metabolic stressors or pathogen infection, which may lead to expression of 'mitokines' such as growth differentiation factor 15 (GDF15).
SCOPE OF REVIEW: This review summarizes the mechanism of GDF15 expression in response to organelle stress such as mitochondrial stress, and covers pathophysiological conditions or diseases that are associated with elevated GDF15 level. This review also illus
