Recurrent prosthetic joint infections: A review of evaluation, management, and outcomes.

Periprosthetic joint infection (PJI) remains one of the most serious complications of total joint arthroplasty. While two-stage revision arthroplasty remains the most commonly utilized treatment strategy for chronic PJI in North America, recurrent infection following revision represents an increasingly encountered and particularly challenging clinical problem. These patients often have substantial host comorbidity, prior surgical burden, compromised soft tissues, and limited reconstructive options, contributing to high rates of treatment failure and morbidity. This narrative review summarizes

Molecular Regulation of HIV-1 Expression and Persistence Across Diverse Cellular Reservoirs.

Despite the remarkable success of antiretroviral therapy (ART) in suppressing human immunodeficiency virus type 1 (HIV-1) replication, viral persistence remains a major barrier to cure. This persistence is sustained by heterogeneous cellular reservoirs in which viral expression is tightly regulated by host-dependent molecular mechanisms. Beyond the canonical cluster of differentiation 4 (CD4+) T-cell reservoirs, HIV-1 establishes long-lived infection in myeloid cells, glial populations within the central nervous system (CNS), and additional non-canonical cellular niches, each characterized by

Host genetic polymorphisms, immune response, and susceptibility to pleural tuberculosis: A systematic review.

BACKGROUND: Pleural tuberculosis is one of the most frequent extrapulmonary manifestations of Mycobacterium tuberculosis infection and is characterized by a pronounced immune-mediated response with a low bacillary burden. Host genetic factors appear to play a central role in its immunopathogenesis. METHODS: A systematic review was conducted in accordance with PRISMA guidelines and prospectively registered in PROSPERO (CRD420251051395). Searches were performed across MEDLINE/PubMed, Embase, and SciELO databases, covering publications from 1970 to December 2023. The search strategy combined ter

yst gene expression in Yersinia enterocolitica is positively regulated by a chromosomal region that is highly homologous to Escherichia coli host factor 1 gene (hfq).

Yersinia enterocolitica produces heat-stable enterotoxin (Y-ST) as one of its virulence factors. The yst gene, however, frequently and spontaneously becomes inactive (silent) during storage, which is accompanied by concurrent changes in some biological properties such as colony morphology, growth rate, carbon fermentation and ornithine decarboxylase activity. Northern blot analysis revealed that the level of mRNA for yst was repressed. To investigate the regulatory region, we transformed a yst-silent strain with a chromosomal gene library of Y-ST producing an isogenic counterpart. Out of 3604

Human Three Prime Repair Exonuclease 1 Promotes HIV-1 Integration by Preferentially Degrading Unprocessed Viral DNA.

Three prime repair exonuclease 1 (TREX1) is the most abundant 3'→5' exonuclease in mammalian cells. It has been suggested that TREX1 degrades HIV-1 DNA to enable the virus to evade the innate immune system. However, the exact role of TREX1 during early steps of HIV-1 infection is not clearly understood. In this study, we report that HIV-1 infection is associated with upregulation, perinuclear accumulation, and nuclear localization of TREX1. However, TREX1 overexpression did not affect reverse transcription or nuclear entry of the virus. Surprisingly, HIV-1 DNA integration was increased in TREX

Reconstructing the intestinal microbiota ecological network based on graph neural network and contrastive learning: Predicting key microbiota regulatory targets in Crohn's Disease.

Crohn's disease (CD) is a heterogeneous inflammatory bowel disease in which microbial, metabolic, and host factors interact dynamically. Therefore, we aimed to (i) integrate microbiota, metabolite, and host-gene data into a heterogeneous dynamic graph and (ii) use contrastive learning to prioritize microbiota-related regulatory targets in CD under limited-sample conditions. We combined a heterogeneous graph dynamic network (HGDN) with a contrastive learning module (CENet) to model microbiota-metabolite-gene interactions and to improve the robustness of target prioritization. In the current dat

Highly pathogenic avian influenza: a One Health perspective on risks, surveillance, and resilience.

Highly pathogenic avian influenza (HPAI) poses an expanding global threat with direct relevance to immunologists studying host-pathogen dynamics, cross-species transmission, and immune responses to emerging viruses. Driven primarily by evolving H5 and H7 influenza A subtypes, HPAI now affects a broadening host range, including spillovers into mammals such as seals, cattle, and humans, raising concern for pandemic potential. Applying a One Health perspective, this review emphasizes the immunological and ecological factors that underlie HPAI emergence and spread. Migratory bird flyways, shared a

Advances in semiochemicals of the sweet potato weevil, Cylas formicarius, and its application in pest management.

The sweet potato weevil (SPW), Cylas formicarius, is one of the most destructive pests of sweet potato, Ipomoea batatas L., across tropical and subtropical regions worldwide. The concealed feeding habits of larvae and the nocturnal activity of adults render conventional chemical-based control largely ineffective. Semiochemical-based management, especially the use of female-produced sex pheromone, has emerged as a promising strategy for integrated pest management (IPM) for this pest insect. This review provides a comprehensive analysis of the chemical ecology of C. formicarius, including the id

Tea and digestive system health: integrating gut microbiota-involved ADME and personalized nutrition for gastrointestinal disorders intervention.

The global burden of digestive diseases is escalating, marked by rising incidence and severity. Tea, a widely consumed beverage rich in polyphenols, offers a promising dietary intervention due to its demonstrated capacity to modulate gut microbiota, suppress inflammation, and enhance gastrointestinal barrier function. This review provides a systematic integration of the ADME characteristics of key tea compounds with their complex interplay with the gut microbiome, underscoring the central role of microbial biotransformation in mediating the health benefits of tea. We critically examine the spe

Analysis of sequential cytokine release after ACL reconstruction.

PURPOSE: Rupture of the anterior cruciate ligament is common and may necessitate surgical reconstruction. Surgical reconstruction aims to restore normal kinematics and biology within the knee. The acute phase response after surgical reconstruction remains poorly defined but may influence graft integration through modulation of host tissue remodelling. METHODS: The very early host production of key cytokines after surgery was studied. A consecutive series of 14 patients undergoing reconstructive surgery were studied per-operatively, 1 and 6 h after surgery, examining the hypothesis that the ac

Modulation of HIV Replication in Monocyte-Derived Macrophages (MDM) by Host Antiviral Factors Secretory Leukocyte Protease Inhibitor and Serpin Family C Member 1 Induced by Steroid Hormones.

The impact of steroid hormones estrogen and progesterone on human immunodeficiency virus type 1 (HIV-1) replication is well documented. However, the exact mechanism involved in the regulation of HIV-1 replication by estrogen and progesterone is still unclear. In the present study, we wanted to elucidate the molecular mechanisms underlying the modulation of HIV-1 replication by estrogen and progesterone. To achieve this goal, we used real-time quantitative PCR arrays (PCR arrays) to identify differentially expressed host genes in response to hormone treatments that are involved in antiviral res

Identification of integrative and conjugative elements in pathogenic and commensal Neisseriaceae species via genomic distributions of DNA uptake sequence dialects.

Mobile genetic elements (MGEs) are key factors responsible for dissemination of virulence determinants and antimicrobial-resistance genes amongst pathogenic bacteria. Conjugative MGEs are notable for their high gene loads donated per transfer event, broad host ranges and phylogenetic ubiquity amongst prokaryotes, with the subclass of chromosomally inserted integrative and conjugative elements (ICEs) being particularly abundant. The focus on a small number of model systems has biased the study of ICEs towards those conferring readily selectable phenotypes to host cells, whereas the identificati

WSSV355 enhances IDH1 enzymatic activity and promotes efficient WSSV replication.

White spot syndrome virus (WSSV) drives extensive metabolic reprogramming to support viral replication, including activation of reductive carboxylation, a pathway in which host isocitrate dehydrogenase 1 (IDH1) has a central role. However, the mechanisms by which viral factors regulate IDH1 and associated metabolic networks remain unclear. Here, we identified WSSV355 as a key viral regulator that contributed to IDH1 binding and activation during infection. Using yeast two-hybrid screening, WSSV355 was identified as an LvIDH1-interacting protein and further validated by co-immunoprecipitation a

Mce transport systems in Mycobacterium tuberculosis: architecture, regulation, lipid import, and translational perspectives.

Mammalian cell entry (Mce) systems are now recognized as central determinants of lipid uptake, cell-envelope homeostasis, and host adaptation in Mycobacterium tuberculosis (Mtb). Although the term "mammalian cell entry" originated from early studies linking the Mce loci to host-cell invasion phenotypes, subsequent genetic, biochemical, and structural work has substantially reframed their biological significance. Current evidence supports a model in which Mce1 and Mce4 function as multiprotein lipid-import systems specialized primarily for fatty-acid and cholesterol uptake, respectively, wherea

A graph neural network-based approach for predicting SARS-CoV-2-human protein interactions from multiview data.

The COVID-19 pandemic has demanded urgent and accelerated action toward developing effective therapeutic strategies. Drug repurposing models (in silico) are in high demand and require accurate and reliable molecular interaction data. While experimentally verified viral-host interaction data (SARS-CoV-2-human interactions published on April 30, 2020) provide an invaluable resource, these datasets include only a limited number of high-confidence interactions. Here, we extend these resources using a deep learning-based multiview graph neural network approach, coupled with optimal transport-based

Host-Microbiota Metabolic Interactions in Atherosclerosis: Oral, gut, and Blood Perspectives.

Atherosclerosis is a chronic inflammatory disease influenced by host-microbiota interactions beyond traditional risk factors. Microbial communities in the oral cavity, gut, and blood contribute to vascular dysfunction through metabolic and immune mechanisms, yet an integrated perspective across these compartments remains lacking. This narrative review synthesizes current evidence on the distinct and interconnected roles of oral, gut, and blood microbiotas in atherosclerosis pathogenesis. We critically evaluate key microbial metabolites, trimethylamine N-oxide (TMAO), short-chain fatty acids (S

Influence of Staphylococcus aureus Strain Background on Sa3int Phage Life Cycle Switches.

Staphylococcus aureus asymptomatically colonizes the nasal cavity of mammals, but it is also a leading cause of life-threatening infections. Most human nasal isolates carry Sa3 phages, which integrate into the bacterial hlb gene encoding a sphingomyelinase. The virulence factor-encoding genes carried by the Sa3-phages are highly human-specific, and most animal strains are Sa3 negative. Thus, both insertion and excision of the prophage could potentially confer a fitness advantage to S. aureus. Here, we analyzed the phage life cycle of two Sa3 phages, Φ13 and ΦN315, in different phage-cured S. a

Host factors in parainfluenza virus replication: from entry to innate immunity evasion.

Human parainfluenza viruses (HPIVs)are significant pathogens responsible for acute respiratory infections in infants, the elderly, and immunocompromised individuals. Among children under five, HPIVs are the second most common viral cause of acute lower respiratory tract infections (ALRIs), surpassed only by respiratory syncytial virus (RSV). Despite their significant health impact, there are currently no licensed vaccines or specific antiviral treatments for HPIV infection. Recent advances in functional genomics, including CRISPR-Cas9 screening, cDNA library screening, and multi-omics approach

Knowledge graph and machine learning-guided virtual screening prioritizes putative blockers of the PSMD11/Rpn6-EBOV glycoprotein interface.

INTRODUCTION: Host-directed antiviral discovery requires a traceable route from host-factor identification to testable molecular hypotheses. This study developed such a workflow around a host factor associated with Ebola virus glycoprotein (EBOV GP). METHODS: GFP-GP pulldown coupled with co-immunoprecipitation/mass spectrometry was used to identify GP-associated host candidates. A PSMD11/Rpn6-centred workflow integrated knowledge-graph inference, ligand-only machine learning, ADMETlab profiling and interface-guided docking. Candidate prioritization was assessed through score-weight sensitivit

Hiding in Plain Sight: HIV-1 Membraneless Organelles as Nuclear Hubs-Host Hijacking, Replication, Immune Evasion, and Drug-Access Implications.

Theories on the early steps of the HIV-1 life cycle have been radically revised over the past five years. The long-held assumption that the capsid fully disassembles in the cytoplasm has given way to a more nuanced view: Cytoplasmic disassembly does occur and, in several myeloid systems, is increasingly linked to cytosolic cDNA sensing and to abortive infection. However, a substantial fraction of intact or nearly intact capsid cores instead traverse the nuclear pore complex (NPC) and, upon interacting with the host factor CPSF6, induce liquid-liquid phase separation. This leads to the formatio
