Interferons in the pathogenesis and treatment of human immunodeficiency virus infection.

There still remains several unanswered questions concerning the pathogenesis of human immunodeficiency virus (HIV) infection. Interferons (IFNs), as well as other cytokines, are both dysregulated in HIV infection and serve as effector molecules that modulate the replicative capacity of HIV. Acid-labile IFN-alpha, an aberrant form of interferon earlier described in certain autoimmune diseases, has been detected in HIV-infected individuals. Conversely, a deficient expression of IFN-alpha may occur usually associated with HIV disease. Although conflicting findings have been reported on whether IF

Localization of interferons and interleukin 2 in polymyositis and muscular dystrophy.

Muscle biopsies from nine patients with polymyositis, six with muscular dystrophy, six with other muscle diseases and three controls have been studied with a panel of 10 monoclonal antibodies (MoAb) identifying T lymphocytes, HLA-class I antigens, alpha, beta and gamma interferons and interleukin 2 (IL-2). The result confirm that the staining of the sarcolemma with anti-HLA class I antibody is weak or negative, except in areas adjacent to infiltrating leucocytes or where muscle fibre damage is evident. The very similar tissue distribution of alpha, beta and gamma interferons in the polymyositi

Natural and recombinant interferons inhibit epithelial cell invasion by Shigella spp.

The effect of natural and recombinant interferons (IFNs) on the abilities of Shigella flexneri, S. sonnei, and Salmonella typhimurium to invade different human and murine cells was examined. Pretreatment of cell monolayers with natural and recombinant IFNs reduced the number of Shigella-infected cells in a dose-dependent manner. Establishment of an anti-invasive cellular state was time dependent, requiring 10 h for 50% inhibition of bacterial invasion. The inhibitory effect of IFN was species specific, with human or murine IFN effective against homologous but not heterologous cells. Gamma IFN

[Interferon].

More than 50 years have passed since interferon (IFN) was discovered. Until now, the signaling pathway and function of IFN were basically elucidated. IFN has been clinically used for patients with hepatitis B virus (HBV) or hepatitis C virus (HCV). In Japan, pegylated IFN (PEG-IFN) is just approved for patients with HBV related chronic hepatitis. Also, the adding of direct-acting antiviral agents to the combination therapy with PEG-IFN plus ribavirin will improve anti-HCV effect. Another important effect of IFN is a suppression of hepatocarcinogenesis in patients with viral related chronic liv

Interferon.

Interferons are proving useful in the management of recalcitrant condylomata acuminata. Dosage schedules have been established that balance efficacy and systemic toxicity. Alone, however, interferons are not entirely effective in every patient. In combination with conventional medical and surgical modalities, they offer hope for the control of an often frustrating and potentially serious virus infection.

Programming innate immune memory: The context-dependent role of type I interferons.

Type I interferons (IFNs) are best known for driving antiviral defense; however, a growing body of evidence identifies a pivotal role for type I IFNs in the establishment of innate immune memory. Depending on the nature of the priming stimulus, the identity of the responding cell, and the tissue microenvironment, type I IFN signaling may contribute to both trained immunity and tolerance. This review summarizes recent advances in identifying the molecular mechanisms by which the two best studied type I IFN family members, IFNα and IFNβ, contribute to innate immune memory. Stimuli triggering typ

Type I interferons induced upon respiratory viral infection impair lung metastatic initiation.

Metastatic breast cancer accounts for 7% of cancer-related deaths, with the lungs being a common site of cancer spread. In parallel, lower respiratory tract infections, including those caused by respiratory syncytial virus (RSV), remain a common cause of morbidity and mortality worldwide. Acute viral respiratory infections induce marked changes in the lung. However, how these changes influence metastasis initiation and cancer progression remains unclear. Using breast cancer and other cancer cell types in an experimental lung metastasis model, we show that RSV infection impairs tumor cell seedi

Oropouche virus causes acute hepatitis in mice controlled by type I interferons.

Oropouche virus (OROV), a member of the Peribunyaviridae family endemic to South America, is a current public health threat. The recent OROV outbreak driven by a novel reassortant strain has caused a dramatic increase in cases in 2024 (13,785 in Brazil, versus only 261 from 2015 to 2022) with sustained levels of transmission in 2025. Previously underreported outcomes have been recognized, including miscarriage, microcephaly, encephalitis, and death. OROV lethality in humans has been attributed to severe coagulopathy with liver involvement, and epidemiological data suggest that acute hepatitis

Autoantibodies neutralizing type 1 interferons in two cohorts of people with HIV.

Autoantibodies (autoAbs) neutralizing type I interferons (IFN-I) markedly increase the risk of severe manifestations of some viral diseases. The prevalence of autoAbs neutralizing at least one IFN-I, specifically IFN-α2, IFN-β, and IFN-ω, in individuals from the Swiss human immunodeficiency virus 1 (HIV-1) cohort is largely similar to French age-matched healthy controls, with around 1% in adults by age 65 having neutralizing IFN-I autoAbs and rising thereafter. We analyzed samples from Danish individuals with HIV, including antiretroviral therapy (ART) treated (n = 260) and ART-naive (n = 58)

Sterile production of interferons in the thymus affects T cell repertoire selection.

Type I and III interferons (IFNs) are robustly induced during infections and protect cells against viral infection. Both type I and III IFNs are also produced at low levels in the thymus at steady state; however, their role in T cell development and immune tolerance is unclear. Here, we found that both type I and III IFNs were constitutively produced by a very small number of AIRE+ murine thymic epithelial cells, independent of microbial stimulation. Antigen-presenting cells were highly responsive to thymic IFNs, and IFNs were required for the activation and maturation of thymic type 1 convent

Type 1 interferons promote Staphylococcus aureus nasal colonization by inducing phagocyte apoptosis.

Staphylococcus aureus is an important human commensal which persistently colonizes up to 30% of the human population, predominantly within the nasal cavity. The commensal lifestyle of S. aureus is complex, and the mechanisms underpinning colonization are not fully understood. S. aureus can induce an immunosuppressive environment in the nasal tissue (NT) by driving IL-10 and IL-27 to facilitate nasal colonization, indicating that S. aureus has the capacity to modulate the local immune environment for its commensal habitation. Mounting evidence suggests commensal bacteria drive type 1 interferon

Innate Lymphoid Cells and Interferons Limit Neurologic and Articular Complications of Brucellosis.

Brucellosis is a globally significant zoonotic disease. Human patients with brucellosis develop recurrent fever and focal complications, including arthritis and neurobrucellosis. The current study investigated the role of innate lymphoid cells (ILCs) in the pathogenesis of focal brucellosis caused by Brucella melitensis. After footpad infection, natural killer cells and ILC1 cells both limited joint colonization by Brucella. Mice lacking natural killer cells, and in particular mice lacking all ILCs, also developed marked arthritis after footpad infection. Following pulmonary infection, mice la

Evasion mechanisms of the type I interferons responses by influenza A virus.

The type I interferons (IFNs) represent the first line of host defense against influenza virus infection, and the precisely control of the type I IFNs responses is a central event of the immune defense against influenza viral infection. Influenza viruses are one of the leading causes of respiratory tract infections in human and are responsible for seasonal epidemics and occasional pandemics, leading to a serious threat to global human health due to their antigenic variation and interspecies transmission. Although the host cells have evolved sophisticated antiviral mechanisms based on sensing i

The Use of Interferons in the Treatment of Cutaneous T-Cell Lymphoma.

Interferons are polypeptides that naturally occur in the human body as a part of the innate immune response. By harnessing these immunomodulatory functions, synthetic interferons have shown efficacy in combating various diseases including cutaneous T-cell lymphoma. This article closely examines the qualities of interferon alfa and interferon gamma and the evidence behind their use in the 2 most common types of cutaneous T-cell lymphomas, namely, mycosis fungoides and Sézary syndrome.

Type I interferons: key players in normal skin and select cutaneous malignancies.

Interferons (IFNs) are a family of naturally existing glycoproteins known for their antiviral activity and their ability to influence the behavior of normal and transformed cell types. Type I Interferons include IFN- α and IFN- β . Currently, IFN- α has numerous approved antitumor applications, including malignant melanoma, in which IFN- α has been shown to increase relapse free survival. Moreover, IFN- α has been successfully used in the intralesional treatment of cutaneous squamous cell carcinoma (SCC) and basal cell carcinoma (BCC). In spite of these promising clinical results; however, the

Interferons in enteroviral heart disease: modulation of cytokine expression and antiviral activity.

Interferon (IFN)-beta has a more than 120-fold higher antiviral activity than the closely related IFN-alpha in human myocardial fibroblasts infected with the cardiotropic enterovirus coxsackievirus B3 (CVB3). CVB3 replication induces interleukin (IL)-6 and IL-8 expression in myocardial fibroblasts, and suppresses the expression of monocyte chemoattractant protein-1 (MCP-1). We investigated whether the higher antiviral activity of IFN-beta compared to IFN-alpha was a result of a suppression of IL-8 expression by IFN-beta since previous studies had indicated that IL-8 stimulates enterovirus repl

Autoantibodies neutralizing type I interferons in COVID-19 patients from southern Brazil.

Inborn errors of type I IFN (IFN-I) immunity and neutralizing autoantibodies (auto-Abs) against IFN-I (AAN-I-IFNs) impair antiviral immunity and underlie critical COVID-19. Their impact on disease severity in different populations, such as Latin American populations, remains to be better understood. This study assessed the presence of AAN-I-IFNs (anti-IFN-α2, anti-IFN-ω, and anti-IFN-β) in a cohort of 458 COVID-19 patients from southern Brazil, stratified as mild (n = 224) versus non-mild (n = 234), or survivors (n = 369) versus deceased (n = 89). AAN-I-IFNs were detected in 17 patients (3.7%)

The Uncovered Role of Interferons in Sepsis: From Pathogenesis to Precision Immunotherapy.

Sepsis is a life-threatening syndrome characterized by a dysregulated host response to infection resulting in organ dysfunction. While traditional paradigms have emphasized proinflammatory cytokines, such as tumor necrosis factor-α and interleukin-1β, interferons (IFNs) are increasingly recognized as critical yet underexplored mediators of sepsis pathobiology. IFNs are central regulators of innate and adaptive immunity and have emerged as key mediators of the immune alterations observed during sepsis. Type I IFNs (IFN-α/β), type II IFN (IFN-γ), and type III IFNs exert pleiotropic effects on im
