Ornithine transcarbamylase and disaccharidase activities in damaged intestinal mucosa of children--diagnosis of hereditary ornithine transcarbamylase deficiency in mucosa.

Ornithine transcarbamylase (OTC) and disaccharidase activities were measured in the intestinal mucosa from 182 children. Sixty-nine had normal mucosa, whereas the others had different degrees of mucosal damage. Brush border disaccharidases are significantly decreased in all degrees of villous atrophy. In contrast, OTC is not affected in moderate atrophy and only slightly decreased in severe atrophy. Consequently, the OTC-to-lactase ratio increases with the degree of atrophy and permits discrimination between normal and damaged mucosa. The assay of OTC activity in intestinal mucosa for the diag

Studies on the histogenesis of the tunica mucosa of the small and large intestines of the camel (Camelus dromedarius). II. Histogenesis of the colonic mucosa.

The colon is at first a simple tube, and its mucosa has a smooth luminal surface, surrounding a previous lumen. At the 150 mm stage the mucosa is thrown into a number of stout projections, each has a vascularized mesenchymal core and an epithelial cloth. These projections are in turn split into finger-like villi by transverse furrows. The villi reach their maximal length at the 260 mm stage and then gradually degenerate and slough into the lumen. The villi were completely disappeared at the 730 mm stage. The colonic glands were firstly observed at the 200 mm stage as downgrowths of the intervi

Compound Chinese medicine (F1) improves spleen deficiency diarrhea by protecting the intestinal mucosa and regulating the intestinal flora.

Compound Chinese medicine (F1) is a traditional prescription in Chinese medicine that is commonly used to treat spleen deficiency diarrhea (SDD). It has demonstrated remarkable effectiveness in clinical practice. However, the precise mechanism by which it exerts its antidiarrheal effect is still unclear. This study aimed at investigating the antidiarrheal efficacy and mechanism of F1 on senna-induced secretory diarrhea (SDD). Senna was utilized to induce the development of a mouse model of senna-induced secretory diarrhea (SDD) in order to observe the rate of diarrhea, diarrhea index, blood bi

Intestinal mucosa permeability following oral insulin delivery using core shell corona nanolipoparticles.

Chitosan nanoparticles (NC) have excellent capacity for protein entrapment, favorable epithelial permeability, and are regarded as promising nanocarriers for oral protein delivery. Herein, we designed and evaluated a class of core shell corona nanolipoparticles (CSC) to further improve the absorption through enhanced intestinal mucus penetration. CSC contains chitosan nanoparticles as a core component and pluronic F127-lipid vesicles as a shell with hydrophilic chain and polyethylene oxide PEO as a corona. These particles were developed by hydration of a dry pluronic F127-lipid film with NC su

The Vibrio cholerae cytolysin promotes chloride secretion from intact human intestinal mucosa.

BACKGROUND: The pathogenicity of the Vibrio cholerae strains belonging to serogroup O1 and O139 is due to the production of virulence factors such as cholera toxin (CT) and the toxin-coregulated pilus (TCP). The remaining serogroups, which mostly lack CT and TCP, are more frequently isolated from aquatic environmental sources than from clinical samples; nevertheless, these strains have been reported to cause human disease, such as sporadic outbreaks of watery diarrhoea and inflammatory enterocolitis. This evidence suggested the possibility that other virulence factor(s) than cholera toxin migh

Combined LDI/SAT test to evaluate intestinal lactose digestion and mucosa permeability.

BACKGROUND: Intestinal mucosal damage causes impaired digestive capacity and increased mucosal permeability. Quantification of damage can be used to improve treatment options. Currently, the Lactose Digestion Index (LDI) and the Sugar Absorption Test (SAT) are used for evaluation. The investigation studied whether both tests could be combined to provide a useful multifunctional test and whether measurements in blood (LDI) could be replaced by measurements in urine. MATERIALS AND METHODS: The LDI (25 g 13C-lactose, 0.5 g 2H-glucose), the SAT (5 g lactulose, 1 g L-rhamnose) and the LDI/SAT comb

Benefits of intraluminal injection of glutamine for intestinal mucosa during ischemia-reperfusion.

BACKGROUND: We investigated the effect of intraluminal glutamine on the intestinal mucosa in the presence of ischemia-reperfusion injury (IRI). METHODS: Six blind sacs were created in the small bowel (3 at the jejunum and 3 at the ileum) in 10 Wistar rats. The lateral sacs of both bowel regions were submitted to IRI (30/30 min), while the medial sacs were left free to receive blood supply. In the lateral sacs, a solution containing either saline plus 4% glutamine or pure saline was injected at the bowel lumen. No fluid was injected in the medial sacs. RESULTS: Both at the jejunum and at the

Distribution of dominant T cell receptor beta chains in human intestinal mucosa.

The majority of human intestinal intraepithelial lymphocytes (iIELs) are CD8+ T cells that use the T cell receptor (TCR)-alpha/beta. Previous studies have shown that iIELs isolated from segments of small intestine or colon contain one or several dominant alpha/beta T cell clones. It is not known whether these clones expand only locally in response to a particular antigen or whether they are widely distributed throughout the intestine. To address this question, iIELs were purified from near the proximal and distal margins in a series of intestinal resections for noninflammatory diseases. TCR-be

Digestion of gliadin peptides by intestinal mucosa from control or coeliac children.

Gliadin, subsequently treated with pepsin, trypsin and pancreatic extract was further digested by small-intestinal mucosal homogenates from 10 control or 8 coeliac children. The amino acids liberated in the incubation mixture were measured and corrected for mucosal damage. In accordance with the data from the literature on adults, the total amount of amino acids released from gliadin peptides by the intestinal mucosa from children with active coeliac disease is significantly lower than that by the mucosa from control subjects. Qualitatively, however, no significant differences for the individu

Dynamics of Intestinal Mucosa Microbiota in Juvenile Sika Deer During Early Growth.

The establishment of gut microbiota in young ruminants has a profound impact on their productive performance in adulthood. The microbial communities of ruminants differ significantly across the different regions of the digestive tract, as well as between the mucosa and lumen. In this study, we analyzed the characteristics of the microbiota of the small intestine (jejunum and ileum) and large intestine (cecum and colon) of sika deer on day 1 (birth), day 42 (transition period) and day 70 (rumination period) using 16S rRNA gene sequencing. The results showed that the microbial diversity of the m

Binary Bacillus subtilis protects the intestinal mucosa barrier and alleviates nonalcoholic steatohepatitis.

BACKGROUND: Nonalcoholic steatohepatitis (NASH) is characterized by liver steatosis, inflammation, and even fibrosis. NASH is likely to develop into cirrhosis and liver cancer, the major causes of liver related deaths. We aimed to study the effect of probiotics on NASH via the gut-liver axis. METHODS: Thirty male Sprague-Dawley rats were divided into three groups. A control group of 10 rats was fed on a standard chow for 16 weeks. Twenty rats fed on a high-fat diet for 8 weeks were separated to two groups: a model group (10 rats) fed on vehicle for 8 weeks and a treatment group (10 rats) supp

Chlorogenic Acid Attenuates Oxidative Stress-Induced Intestinal Mucosa Disruption in Weaned Pigs.

Chlorogenic acid (CGA) is a natural polyphenol that possesses potent antioxidant activity. However, little is known about its exact role in regulating the intestinal health under oxidative stress. This study was conducted to explore the effect of dietary CGA supplementation on intestinal barrier functions in weaned pigs upon oxidative stress. Twenty-four weaned pigs were allocated to three treatments and were given a basal diet (control) or basal diet containing CGA (1,000 mg/kg) for 21 days. Pigs were challenged by sterile saline (control) or diquat [10 mg/kg body weight (BW)] on the 15th day

Screening for intestinal microflora influencing superoxide dismutase activity in mouse cecal mucosa.

We have suggested that intestinal microflora reduces the activity of the antioxidant enzyme superoxide dismutase (SOD) in the mouse cecal mucosa. In this study, gnotobiotic mice were used to examine the species of intestinal microflora influencing SOD activity in the cecal mucosa. The total SOD activity in the cecal mucosa of each germ-free (GF), gnotobiotic mouse with Escherichia coli, Lactobacillus and Bacteroides was significantly higher than that in the cecal mucosa of gnotobiotic mice with chloroform-treated feces (CHF), conventionalized (CVz) mice and conventional (CV) mice (P<0.05). Effect of cholera toxin on ATPase activities in rabbit small intestinal mucosa.

Cholera toxin induced the appearance of ATPase activity in rabbit small intestinal mucosa. This enzyme significantly differed from other ATPases, including Na(+),K(+)-ATPase and HCO3(-)-ATPase in the small intestinal epithelium of rabbits, by some properties, in particular, by relation to divalent and monovalent cations and anions, pH optimum, substrate specificity, and inhibitory analysis.

Innate immunity modulation by the IL-33/ST2 system in intestinal mucosa.

Innate immunity prevents pathogens from entering and spreading within the body. This function is especially important in the gastrointestinal tract and skin, as these organs have a large surface contact area with the outside environment. In the intestine, luminal commensal bacteria are necessary for adequate food digestion and play a crucial role in tolerance to benign antigens. Immune system damage can create an intestinal inflammatory response, leading to chronic disease including inflammatory bowel diseases (IBD). Ulcerative colitis (UC) is an IBD of unknown etiology with increasing worldwi

Histological diagnosis of follicular lymphoma by biopsy of small intestinal normal mucosa.

Due to advances in double-balloon endoscopy, various types of malignant lymphoma of the small intestine have been observed at very early stages. We report here that after remission of diffuse large B-cell lymphoma, follicular lymphoma recurred in the mucosa of the small intestine. Furthermore, these lesions were diagnosed by biopsy from normal mucosa as diminutive erosions and reddish looking. If a small intestinal lesion is suspected of being a malignant lymphoma, a double-balloon endoscopy should be carried out, and a biopsy should be taken and inspected.
