Thrombocytopenia secondary to iron deficiency anemia responding to iron therapy.

A broad spectrum of diseases can cause anemia and thrombocytopenia. Some of these diseases are a hematological emergency; others are benign diseases, so early and accurate diagnosis is crucial in managing such patients. Usually, IDA is associated with thrombocytosis or normal platelets; however, in rare cases, IDA can be associated with thrombocytopenia; even though, thrombocytopenia that occurs with IDA responds to iron therapy. Iron therapy rarely causes transient thrombocytopenia per se. We are reporting an African female patient who is found to have thrombocytopenia secondary to iron defic

Higher iron bioavailability of a human-like collagen iron complex.

Iron deficiency remains a public health problem around the world due to low iron intake and/or bioavailability. FeSO4, ferrous succinate, and ferrous glycinate chelate are rich in iron but have poor bioavailability. To solve the problem of iron deficiency, following previous research studies, a thiolated human-like collagen-ironcomplex supplement with a high iron content was prepared in an anaerobic workstation. In addition, cell viability tests were evaluated after conducting an MTT assay, and a quantitative analysis of the thiolated human-like collagen-iron digesta samples was performed usin

Mammalian iron metabolism and its control by iron regulatory proteins.

Cellular iron homeostasis is maintained by iron regulatory proteins 1 and 2 (IRP1 and IRP2). IRPs bind to iron-responsive elements (IREs) located in the untranslated regions of mRNAs encoding protein involved in iron uptake, storage, utilization and export. Over the past decade, significant progress has been made in understanding how IRPs are regulated by iron-dependent and iron-independent mechanisms and the pathological consequences of IRP2 deficiency in mice. The identification of novel IREs involved in diverse cellular pathways has revealed that the IRP-IRE network extends to processes oth

Iron chelation therapy for transfusional iron overload: a swift evolution.

Chronic transfusional iron overload leads to significant morbidity and mortality. While deferoxamine (DFO) is an effective iron chelator with over four decades of experience, it requires tedious subcutaneous infusions that reflect negatively on patient compliance. The novel oral iron chelators deferiprone (L1) and deferasirox (DFRA) opened new horizons for the management of transfusional siderosis. A large body of evidence is now available regarding their efficacy and safety in various populations and settings. Nevertheless, experience with both drugs witnessed some drawbacks, and the search f

Ineffectiveness of oral iron hydroxide polymaltose in iron-deficiency anemia.

Two hundred and forty one patients with iron deficiency anemia (IDA) were identified in a single institution over a 24-year period; of these, 75 individuals were studied as the result of persistent IDA despite the administration of oral iron hydroxide polymatose (IP). The levels of hemoglobin when the patients were referred for study after being given oral IP had a median of 10.3 g/dl; after administration of oral iron fumarate during periods ranging from 1 to 14 months, the levels of hemoglobin rose to a median of 12.5 g/dl (p>0.01). Our data support previous observations made in other countr

Molecular control of vertebrate iron homeostasis by iron regulatory proteins.

Both deficiencies and excesses of iron represent major public health problems throughout the world. Understanding the cellular and organismal processes controlling iron homeostasis is critical for identifying iron-related diseases and in advancing the clinical treatments for such disorders of iron metabolism. Iron regulatory proteins (IRPs) 1 and 2 are key regulators of vertebrate iron metabolism. These RNA binding proteins post-transcriptionally control the stability or translation of mRNAs encoding proteins involved in iron homeostasis thereby controlling the uptake, utilization, storage or

Iron(VI) and iron(V) oxidation of copper(I) cyanide.

Copper(Il) cyanide (Cu(CN)4(3-)) in the gold mine industry presentsthe biggest concern in cyanide management because it is much more stable than free cyanide. Cu(CN)4(3-) is highlytoxic to aquatic life; therefore, environmentally friendly techniques are required for the removal of Cu(CN)4(3-) from gold mine effluent. The oxidation of Cu(CN)4(3-) by iron-(VI) (FeVIO4(2-), Fe(VI)) and iron(V) (FeVO4(3-), Fe(V)) was studied using stopped-flow and premix pulse radiolysis techniques. The stoichiometry with Fe(VI) was determined to be 5HFeO(4-) + Cu(CN)4(3-) + 8H2O - > 5Fe(OH)3 + Cu2+ + 4CNO- +3/202

Intestinal iron absorption studies in mouse models of iron-overload.

Three mouse strains have been evaluated as suitable models for investigations into the pathogenesis of iron-overload syndromes. Mice with hereditary heterozygous alpha-thalassaemia had moderately raised reticulocyte counts, but were not anaemic and showed little, if any, iron loading. In contrast, mice with homozygous beta-thalassaemia showed microcytic anaemia, reticulocytosis and splenomegaly. Iron-loading was marked, progressive with age and mainly confined to the spleen. Liver iron-loading increased until the age of 7-8 weeks, with no further increase over successive weeks. Although intest

Iron deposition in the corneal graft. Another corneal iron line.

Linear iron deposits in the corneal epithelium are well-documented phenomena in a variety of circumstances, both normal and pathologic. A characteristic annular pigmented line, appearing clinically as an "iron" line, occurred in the donor epithelium of approximately 13% of corneal grafts with retained sutures. Eight cases of normally functioning grafts had iron lines located just central to the sutures. These lines appeared at various intervals postoperatively and did not correlate with age or race. They were frequently found in association with other surface-related epithelial and subepitheli

Serum iron and total iron binding capacity in leprosy patients.

Serum Iron and Total Iron Binding capacity were estimated on the sera collected from 45 leprosy patients attending the out-patient department of the Central JALMA Institute for Leprosy, Agra. The Sera from 15 healthy subjects were included in the study as controls. Hypoferraemia was observed in lepromatous leprosy and was particularly marked during the reactive phase. Further investigations to elucidate the pathogenesis of anaemia in leprosy are being planned.

Ferritin and intestinal iron absorption: pancreatic enzymes and free iron.

Rat intestinal mucosa gave low yields of ferritin purified by standard procedures. The resulting ferritin had less protein relative to iron and migrated faster electrophoretically than ferritin from other rat tissues. Pancreatic duct ligation reduced these differences, suggesting digestive enzyme attack during ferritin isolation. Even in ligated rats, ferritin accounted for only 5-10% of mucosal iron. However, shortly after giving 59FeCl3 orally, 50% of mucosal radioactivity occurred in cell sap, about equally distributed between ferritin and low-molecular-weight (chelated?) iron. No other cel

Iron and Pregnancy.

Iron is essential for nearly all living organisms, but iron requirements vary throughout one's lifetime. Pregnancy is a period notable for vastly increased iron requirements. Iron is needed to support maternal adaptations throughout pregnancy and to enable the growth and development of both the placenta and fetus. Insufficient iron has long been linked to adverse pregnancy outcomes; thus, universal iron supplementation is common practice before and during pregnancy. In high-income countries, however, where foods are often iron-fortified and red meat consumption is common, too much iron supplem

Asbestos and Iron.

Theories of disease pathogenesis following asbestos exposure have focused on the participation of iron. After exposure, an open network of negatively charged functional groups on the fiber surface complexes host metals with a preference for iron. Competition for iron between the host and the asbestos results in a functional metal deficiency. The homeostasis of iron in the host is modified by the cell response, including increased import to correct the loss of the metal to the fiber surface. The biological effects of asbestos develop in response to and are associated with the disruption of iron

Iron and the liver.

Humans have evolved to retain iron in the body and are exposed to a high risk of iron overload and iron-related toxicity. Excess iron in the blood, in the absence of increased erythropoietic needs, can saturate the buffering capacity of serum transferrin and result in non-transferrin-bound highly reactive forms of iron that can cause damage, as well as promote fibrogenesis and carcinogenesis in the parenchymatous organs. A number of hereditary or acquired diseases are associated with systemic or local iron deposition or iron misdistribution in organs or cells. Two of these, the HFE- and non-HF

Iron and the liver.

Iron is an important bio-catalyst of oxidation-reduction reactions in the cell and is essential for life. Paradoxically, it may also be lethal when the fraction of redox-active metal ions exceeds that sequestered in specialized proteins or cellular compartments, and uncontrolled production of free radical species may arise. The liver is the main body site for iron stores and central in the regulation of iron homeostasis. Important iron-proteins, such as hepcidin, the iron regulatory hormone, are specifically produced by the liver. Pathogenic mutations in hepatic iron transporters and regulator

Iron as a hepatotoxin.

Although essential for life, iron in excessive amounts may be toxic. The liver is particularly subject to the toxic effects of iron, since it is the major site of iron storage. Several inherited and acquired human disorders may result in hepatic iron overload, the most common of which are genetic hemochromatosis (GH) and transfusional iron overload. GH is an inherited disorder of iron metabolism, and in patients with GH excess iron absorbed from the gut is transported through the portal vein to the liver. The mechanisms by which excess iron exerts its cytotoxic effects include enhanced formati
