Continuous Dual Glucose-Ketone Monitoring.

Diabetic ketoacidosis (DKA) occurring during pregnancy is an obstetric emergency that can result in significant adverse outcomes for both the pregnant person and the fetus. While DKA generally presents with glucose levels >250 mg/dL, up to 30% of DKA cases involve euglycemic DKA (euDKA) wherein glucose levels <200 mg/dL have been reported. However, detection and prevention of DKA in pregnancy can be challenging for both the pregnant individual and health care providers due to variable clinical presentations of the disease and the limitations of current ketone monitoring technologies. Ketone body metabolism and cardiovascular disease.

Ketone bodies are metabolized through evolutionarily conserved pathways that support bioenergetic homeostasis, particularly in brain, heart, and skeletal muscle when carbohydrates are in short supply. The metabolism of ketone bodies interfaces with the tricarboxylic acid cycle, β-oxidation of fatty acids, de novo lipogenesis, sterol biosynthesis, glucose metabolism, the mitochondrial electron transport chain, hormonal signaling, intracellular signal transduction pathways, and the microbiome. Here we review the mechanisms through which ketone bodies are metabolized and how their signals are tra

Photoreduction and ketone-sensitized reduction of alkaloids.

The photoprocesses of berberine, palmatine, coralyne, sanguinarine, flavopereirine and ellipticine were studied in several solvents. The quantum yields Φ(Δ) of singlet molecular oxygen formation of berberine, palmatine and sanguinarine are moderate in dichloromethane (0.2-0.6) and much smaller in acetonitrile or trifluoroethanol. For the other alkaloids examined, Φ(Δ) is rather independent of solvent polarity. The direct and ketone-sensitized photolysis, using steady-state irradiation at 313 nm or 248/308 nm laser pulses, was studied by absorption and fluorescence spectroscopy. Thereby, radica

Medical aspects of ketone body metabolism.

Ketone bodies are produced in the liver, mainly from the oxidation of fatty acids, and are exported to peripheral tissues for use as an energy source. They are particularly important for the brain, which has no other substantial non-glucose-derived energy source. The 2 main ketone bodies are 3-hydroxybutyrate (3HB) and acetoacetate (AcAc). Biochemically, abnormalities of ketone body metabolism can present in 3 fashions: ketosis, hypoketotic hypoglycemia, and abnormalities of the 3HB/AcAc ratio. Normally, the presence of ketosis implies 2 things: that lipid energy metabolism has been activated

The janus face of ketone bodies in hypertension.

Hypertension is the most important risk factor for the development of terminal cardiovascular diseases, such as heart failure, chronic kidney disease, and atherosclerosis. Lifestyle interventions to lower blood pressure are generally desirable prior to initiating pharmaceutical drug treatments, which may have undesirable side effects. Ketogenic interventions are popular but the scientific literature supporting their efficacy is specific to certain interventions and outcomes in animal models and patient populations. For example, although caloric restriction has its own inherent difficulties (e.

Potential metabolic activities of raspberry ketone.

Novel food and food compounds interventions have attracted a lot of attention nowadays for the prevention and treatment of metabolic diseases. Raspberry ketone (RK) is aromatic compound found within red fruits and berries, has been used as an over-the-counter product for weight loss. However, actually, the effect of RK on weight loss is still controversial, and the mechanism is largely unknown. Besides, in vivo and in vitro studies have demonstrated the beneficial effect of RK on the development of other metabolic diseases. In this review, we comprehensively highlighted the synthesis, bioavail

Ketone Body Metabolism in the Ischemic Heart.

Ketone bodies have been identified as an important, alternative fuel source in heart failure. In addition, the use of ketone bodies as a fuel source has been suggested to be a potential ergogenic aid for endurance exercise performance. These findings have certainly renewed interest in the use of ketogenic diets and exogenous supplementation in an effort to improve overall health and disease. However, given the prevalence of ischemic heart disease and myocardial infarctions, these strategies may not be ideal for individuals with coronary artery disease. Although research studies have clearly de

Ketone bodies as a therapeutic for Alzheimer's disease.

An early feature of Alzheimer's disease (AD) is region-specific declines in brain glucose metabolism. Unlike other tissues in the body, the brain does not efficiently metabolize fats; hence the adult human brain relies almost exclusively on glucose as an energy substrate. Therefore, inhibition of glucose metabolism can have profound effects on brain function. The hypometabolism seen in AD has recently attracted attention as a possible target for intervention in the disease process. One promising approach is to supplement the normal glucose supply of the brain with ketone bodies (KB), which inc

Ketone body metabolism during development.

This paper briefly reviews the role of ketone bodies during development in the rat. Regulation of ketogenesis is in part dependent on the supply to the liver of medium- and long-chain fatty acids derived from mother's milk. The partitioning of long-chain fatty acids between the hepatic esterification and oxidation pathways is controlled by the concentration of malonyl-CoA, a key intermediate in the conversion of carbohydrate to lipid. As hepatic lipogenesis is depressed during the suckling period, [malonyl-CoA] is low and entry of long-chain acyl-CoA into the mitochondria for partial oxidation

Metal-Free Three-Component Coupling Reaction of Ketones with Electron-rich Arenes and Selenium Dioxide for the Synthesis of α-Arylselanyl Ketones.

A metal-free three-component coupling reaction of aryl alkyl/alkyl ketones, SeO2, and phenols/anisoles is described. This multicomponent reaction provides a straightforward and facile pathway for the synthesis of α-((4-hydroxy/methoxyphenyl)selanyl)-aryl alkyl/alkyl ketones in the presence of p-toluenesulfonic acid for the C-Se bond formation process. The method offers an attractive and simple procedure using commonly available shelf reagents to deliver organoselenides that, to our knowledge, are being reported here for the first time.

Visible-Light-Mediated Additive-Free Decarboxylative Ketonization Reaction of Acrylic Acids: An Access to α-Thiocyanate Ketones.

Visible-light-mediated additive-free decarboxylative functionalization of acrylic acids has been developed. The reaction uses inexpensive organic dye 9,10-dicyanoanthracene as a photocatalyst and uses the ubiquitous dioxygen as both an oxygen source and an oxidant. Through this mild and environmentally friendly method, a series of important α-thiocyanate ketones can be generated from easily available acrylic acids and ammonium thiocyanate. In addition, the facile transformation of product α-thiocyanate ketones makes this method have great potential for application in organic and pharmaceutical

Concise trichlorination of methyl ketones using sodium hypochlorite pentahydrate.

We report a practical and efficient method for the synthesis of trichloromethyl ketones from the corresponding methyl ketones via a two-step sequence involving trifluoroacetylation followed by selective chlorination. The key chlorination step proceeds under mild, weakly acidic conditions using sodium hypochlorite pentahydrate, thus avoiding the use of hazardous chlorine gas or strong bases. A wide range of aromatic and aliphatic methyl ketones was successfully converted into the corresponding trichloromethyl ketones in high yields. Mechanistic studies suggest that the reaction involves the for

Oxidant-Free α-Amination of Ketones with Nitrogen Nucleophiles.

α-Amino carbonyl compounds are valuable building blocks for diverse pharmaceuticals and natural products. However, current synthetic methods often rely on specialized nitrogen reagents, stoichiometric oxidants, or metal catalysts, highlighting the need for powerful and green synthetic methodologies. Herein, we report an unprecedented method for the synthesis of α-amino ketones from α-chloroketones using available nucleophilic amines. The process features a ring-opening rearrangement of cyclopropanones to generate para-quinone methide intermediates, enabling subsequent 1,6-additions with N-nucl

Manganese-Catalyzed Ortho-C-H Alkenylation of Aromatic Ketones.

Ortho-C-H alkenylation of aromatic ketones is a key transformation for bioactive molecules and functional materials, but all existing methods rely on scarce noble metals, hindering industrial applications. Herein, we report the first manganese-catalyzed ortho-C-H alkenylation of aromatic ketones with commercial acrylates. Using MnBr-(CO)5 as the precatalyst, the reaction proceeds under mild aqueous conditions with broad substrate scope, scalable to 10 mmol, and allows tunable mono/bis-alkenylation via acrylate stoichiometry. Mechanistic studies support an electrophilic C-H metalation pathway,
