Analgesic-induced asthma caused by 2.0% ketoprofen adhesive agents, but not by 0.3% agents.

A 74-year-old woman was admitted with an asthma attack. She had a 40-year history of sinusitis, nasal polyp and analgesic-induced asthma; however, asthma had never occurred when she used a 0.3% ketoprofen adhesive patch (Mohrus) for stiff shoulder or lumbago. In the hospital, a life-threatening asthma attack suddenly occurred two and a half hours after application of a 2.0% ketoprofen adhesive tape (Mohrus tape) to her shoulder. She was treated with bronchodilator and glucocorticoid and extubated after 20 hours. A drug lymphocyte stimulating test (DLST) was strongly positive for ketoprofen. We

Interaction of ultraviolet-B-rich and ultraviolet-A-rich radiation in ketoprofen-induced photohemolysis.

Interactions between different wavelengths can cause inhibition or enhancement of various biological reactions. We evaluated in vitro the effect of UVB-rich irradiation on UVA-induced phototoxicity. Suspensions of human erythrocytes were incubated with ketoprofen, a phototoxic nonsteroidal anti-inflammatory drug. These samples were exposed to 0, 20, 40, 80, 160 or 320 mJ/cm2 UVB followed by irradiation with 0 or 6 J/cm2 UVA. Photo-induced hemolysis was calculated as a percentage of complete hemolysis. Ketoprofen-dependent hemolysis due to UVA alone was 63% (median). UVB did not induce ketoprof

A novel Ag-BiOBr-rGO photocatalyst for enhanced ketoprofen degradation: Kinetics and mechanisms.

Ag-BiOBr-reduced graphene oxide (rGO) was synthesized for the first time and used to promote photocatalytic activity under visible-light irradiation. The Ag-BiOBr-rGO showed an excellent photocatalytic activity to degrade ketoprofen compared with other photocatalysts. The composites were comprehensively characterized to explore the mechanisms of the enhancement. Electron Paramagnetic Resonance and scavenger experiments demonstrated that the superoxide radical was the active species. Ketoprofen was completely removed in 120 min. The high photocatalytic activity and stability of the catalyst ind

Stereoselective Differences in the Percutaneous Permeation of Ketoprofen Enantiomers and the Underlying Chiral Recognition Mechanisms.

This research aims to investigate the stereoselective permeation differences and molecular recognition mechanisms of ketoprofen enantiomers in the chiral microenvironment of the stratum corneum (SC). This also aims to provide theoretical support for developing highly efficient and precise single-enantiomer transdermal drug delivery systems. The permeation behaviors of dexketoprofen and its racemate through excised full-thickness rat skin in three different solvent systems were studied using Franz diffusion cells. The solubility, apparent partition coefficients, and differential scanning calori

Ketoprofen Associated with Hyaluronic Acid Hydrogel for Temporomandibular Disorder Treatment: An In Vitro Study.

Temporomandibular disorders (TMD) are a public health problem that affects around 12% of the global population. The treatment is based on analgesics, non-steroidal anti-inflammatory, corticosteroids, anticonvulsants, or arthrocentesis associated with hyaluronic acid-based viscosupplementation. However, the use of hyaluronic acid alone in viscosupplementation does not seem to be enough to regulate the intra-articular inflammatory process. So, we propose to develop and evaluate the physicochemical and biological properties in vitro of hyaluronic acid hydrogels (HA) associated with ketoprofen (KE

Surprising results of patch testing with the baseline series in patients with photocontact allergy to ketoprofen.

OBJECTIVE: Photoallergic reactions due to topical ketoprofen are common. As some simultaneous contact allergies have been described in the literature, we aimed to get an overview of the pattern of reactivity towards common allergens in the baseline series in ketoprofen-photoallergic individuals. METHODS: Using our database, we found 94 patients with photocontact allergy to ketoprofen diagnosed during 1999-2018. Approximately 12 800 patients patch tested with the baseline series during the same time frame served as controls. Data on patch testing with the baseline series of 518 individuals bel

Ketoprofen extended-release capsules: a new formulation for the treatment of osteoarthritis and rheumatoid arthritis.

Ketoprofen is a nonsteroidal anti-inflammatory drug (NSAID) of the propionic acid family with demonstrated efficacy in the treatment of rheumatoid arthritis and osteoarthritis; a distinguished safety profile; and good tolerability. This report describes a new extended-release formulation that maintains therapeutic plasma ketoprofen concentrations for up to 24 hours. A single 200-mg capsule thus provides daytime and nighttime symptom control. Small pellets, enclosed in a gelatin capsule, are released in the stomach but release their contained ketoprofen only after reaching the nonacidic environ

A double-blind, parallel comparison of ketoprofen, aspirin, and placebo in patients with postpartum pain.

Our purpose was to evaluate the analgesic efficacy of single oral doses of ketoprofen 25, 50, and 100 mg compared with aspirin 650 mg and placebo in the relief of moderate to severe postepisiotomy, uterine cramping, or cesarean section pain. One hundred and fifty-six patients participated in a randomized, double-blind, stratified, parallel-group study. They were observed over a six-hour period by one nurse-observer. Several of the standard summary measures of analgesia were derived from the interview data, including the sum of pain intensity differences (SPID) and the sum of the hourly relief

Mechanistic Insights into Membrane- and Release-Controlled Drug Permeation from Ketoprofen-Lidocaine Eutectic Fluids.

PURPOSE: Eutectic fluids (EFs) are liquid pharmaceutical systems formed through intermolecular interactions between their components. This study aims to clarify the mechanistic basis of membrane- and release-controlled drug permeation from the ketoprofen-lidocaine (KP-LD) EFs by characterizing their physicochemical properties and molecular interactions. METHODS: Two types of liquid EFs were prepared by mixing and heating KP with LD. The prepared EFs were characterized using thermodynamic, rheological, and NMR-based analyses to obtain parameters related to phase behavior, viscosity, molecular

Spectroscopic Study of R- and S-Ketoprofen Inclusion Complexes With 2-Hydroxypropyl-β-cyclodextrin.

Ketoprofen, also known as (RS)-2-(3-benzoylphenyl)-propionic acid, has the molecular formula C16H14O3 and is classified as a non-steroidal anti-inflammatory medication. R-ketoprofen has stronger analgesic effects than S-K, and as a result, there is a growing interest in enantio-recognition research, which may be accomplished through supramolecular interactions, particularly host-guest reactions. A 1:1 molar ratio was used to produce the combination of separate RK and SK and a 0.01 M stock solution of HP-β-CD to get the final concentration of 6 × 10-4 M. Ethanol was used to make stock solutions

Ozonolysis of ketoprofen in polluted water: Reaction pathways, kinetics, removal efficiency, and health effects.

Ketoprofen (KET), as a non-steroidal anti-inflammatory drug frequently detected in aqueous environments, is a threat to human health due to its accumulation and low biodegradability, which requires the transformation and degradation of KET in aqueous environments. In this paper, the reaction process of ozone-initiated KET degradation in water was investigated using density functional theory (DFT) method at the M06-2X/6-311++g(3df,2p)//M06-2X/6-31+g(d,p) level. The detailed reaction path of KET ozonation is proposed. The thermodynamic results show that ozone-initiated KET degradation is feasibl

Formation of low melting point binary systems comprising ketoprofen and an amide local anaesthetic.

Liquid forms of active pharmaceutical ingredients, ionic liquids (ILs) and deep eutectic mixtures (DEMs), offer several potential benefits in respect to advancing pharmaceutical formulations. The aim of this study was to develop and characterise ILs/DEMs composed of two active molecules: ketoprofen (KET), as the acidic component, and a local anaesthetics (LA), lidocaine (LID), mepivacaine (MEP) or bupivacaine (BUP), which constituted the basic component. A mechanosynthetic approach was successfully applied to obtain LA-KET low melting systems. Composition/temperature phase diagrams were determ

Comparative mutagenic and genotoxic effects of three propionic acid derivatives ibuprofen, ketoprofen and naproxen.

The mutagenicity of three propionic acid derivatives, namely ibuprofen, ketoprofen and naproxen, was tested in the Ames mutagenicity assay (in strains TA97a, TA100 and TA102) and in vivo genotoxicity was tested by sister chromatid exchange (SCE) in bone marrow cells of mice. These are the anti-inflammatory drugs frequently used in different parts of the world. Mutagenicity results showed no mutagenic effects in strains TA97a, TA100 and TA102 for all three drugs. Results of in vivo SCE assays indicate that these three drugs are weakly genoxic in bone marrow cells of mice. This is the first repo

Double Salts and Racemate in Mefloquine-Ibuprofen and Mefloquine-Ketoprofen Systems. A Structural and Thermochemical Analysis.

The formation of salts remains a highly sought-after yet unpredictable strategy for resolving chiral drugs. Using a drug-drug approach, we have investigated the crystallization behavior of Mefloquine (Mf), a racemic antimalarial drug, in combination with enantiopure (S)-ibuprofen and (S)-ketoprofen, as well as their racemic forms. Double salts and racemates have been obtained and characterized by single-crystal and powder X-ray diffraction, Hirshfeld surface analysis, thermal analysis (DSC and TGA), and solubility measurements. Structural analysis reveals that the formation of double salts oft
