Effects of exercise training on kinin receptors expression in rats with myocardial infarction.

OBJECTIVES: The objective of this study is to determine the role of kinin B1 and B2 receptors in exercise-induced cardiac muscle angiogenesis. METHOD: Thirty Wistar rats were randomly assigned to the control group, the myocardial infarction group and the exercise training group (myocardial infarction model was made and received 30 min exercise training on a treadmill). After 4 weeks of experiment, cardiac muscle was harvested. RESULTS: B1 and B2 receptor mRNA and protein levels in the exercise-training group were significantly higher than those in the myocardial infarction group, which were

Role of the kinin B1 receptor in insulin homeostasis and pancreatic islet function.

Kinins are potent vasoactive peptides generated in blood and tissues by the kallikrein serine proteases. Two distinct kinin receptors have been described, one constitutive (subtype B2) and one inducible (subtype B1), and many physiological functions have been attributed to these receptors, including glucose homeostasis and control of vascular permeability. In this study we show that mice lacking the kinin B1 receptor (B1-/- mice) have lower fasting plasma glucose concentrations but exhibit higher glycemia after feeding when compared to wild-type mice. B1-/- mice also present pancreas abnormali

Comparison of tissue kallikrein and kinin receptor expression in gastric ulcers and neoplasms.

Recent studies have suggested the possible involvement of the kallikrein-kinin cascade in gastric inflammatory diseases and malignant transformation in peptic ulcers. Germ-line mutations of genes responsible for repairing DNA mismatch may cause the transformation of chronic peptic ulceration to malignancy. The aim of the study was to compare the expression of the serine protease, tissue kallikrein (TK) and kinin peptide receptors, B1 and B2, in human gastric carcinoma and ulcers. Furthermore, experiments were designed to ask the question whether holistic changes known to occur in carcinoma wou

Effect of Chronic Blockade of the Kallikrein-Kinin System on the Development of Hypertension in Rats.

-The kallikrein-kininogen-kinin system is an important vasodilator and vasodepressor component of the cardiovascular system. Acting mainly through B(2) receptors, kinins may counterbalance the pressor effect of angiotensin II, salt, and mineralocorticoids plus salt. Using rats lacking the bradykinin precursors low- and high-molecular-weight kininogen or a B(2) kinin receptor antagonist (icatibant), we investigated whether absence or blockade of the kallikrein-kinin system alters blood pressure (BP) in rats given (1) chronic infusion of Ang II, (2) a normal or high salt diet, or (3) chronic adm

[The kallikrein-kinin system in acute peritonitis and methods of correcting its disorders].

In experimental fecal peritonitis in 140 albino rats the kallikrein-kinin system was shown to undergo the following changes: its inhibition during the first day was followed by an activation on the second day and inanition on the third day. Under conditions of hyperbaric oxygenation the normalizing effect on the kinin system was observed during two days. The application of salicylates against the background of HBO was shown to enhance the normalizing effect which lasted for three days.

Coagulation, fibrinolysis, platelet and kinin-forming systems during toxemia of pregnancy.

The coagulation, fibrinolysis, and platelet and kinin-forming systems were studied during toxemia of pregnancy by multifactorial examinations. In severe toxemia of pregnancy, decreases in extrinsic coagulation factors, platelet, antithrombin III, plasminogen, alpha 2-plasmin inhibitor, and increases in soluble fibrin-monomer complex, fibrin degradation products, beta-thromboglobulin were observed. This suggests that intravascular coagulation, induced probably through the extrinsic pathway and secondary fibrinolysis, is associated with toxemia of pregnancy. Prekallikrein increased during pregna

[Renal kallikrein-kinin system and control of blood pressure (author's transl)].

Kallikrein excreted with the urine appears to be formed in the kidney. The kallikrein-kinin system in the kidney is localized in the distal nephron from the juxtaglomerular apparatus to the collecting duct. It has been shown that intrarenal infusion of kinins produces an increase in renal blood flow as well as diuresis and natriuresis. Part of the effect of kinins appears to be mediated by the release of prostaglandins. However, the precise role of the renal kallikrein-kinin system in sodium and volume homeostasis and in blood pressure regulation still remains to be determined. Mineralocortico

The significance of kinin-destroying enzymes activity in Plasmodium knowlesi malarial infection.

Experiments were performed on healthy (non-infected) and Plasmodium knowlesi-infected rhesus monkeys (Macaca mulatta) to determine the activity of the kinin-destroying enzymes (kininases) present in the circulation. Methods for the studies and estimation of kininases in the circulating blood of control and P. knowlesi-infected animals are described. The activity of the enzymes, present in the blood in all animals investigated, was evaluated from both methods employed by incubation with synthetic bradykinin. The isolated rat uterus served as the test organ. Studies were made on protein peak fra

Effect of ultraviolet radiation and kinins on oxygen uptake by tissue slices and on basal metabolism in rabbits.

The experiments were carried out on 85 rabbits in 15 groups of 5-8 animals in each. The different groups received: 1) ultraviolet radiation once during 45 minutes or daily during 10 minutes for 6 weeks, 2) bradykinin intravenously 10 microgram/kg, 3) kallikrein 5 B.U./kg intramuscularly during 3 weeks. The levels of kinins and kininogen, and the activity of kallikrein and kininases were determined in the blood several times during 24 hours after radiation exposure, and at intervals of two weeks during long-term exposure to radiation. In the same time periods after exposure the basal metabolism

Kallikrein-8 contributes to kallikrein-kinin system dysregulation in Alzheimer's disease.

Neuroinflammation is a central feature of Alzheimer's disease (AD), yet the mechanisms linking inflammatory protease systems to disease pathology remain incompletely understood. The kallikrein-kinin system (KKS), a major source of bradykinin, has been associated with AD mainly at the peripheral level, but its regulation within the human brain remains largely unexplored. Here, we investigated KKS activity in human AD tissue and a transgenic mouse model, with a particular focus on serine protease kallikrein-8 (KLK8). We observed that both bradykinin and bradykinin B1 receptor levels are increase

Heme-induced kallikrein-kinin system activation in hemolytic uremic syndrome.

INTRODUCTION: Heme is released from red blood cells during hemolytic uremic syndrome (HUS). Here, we investigated hemolysis-induced kallikrein-kinin system activation and the interaction between heme and clotting factor XII (FXII). METHODS: Pediatric patients (19 male and 15 female, median age three years, range three months to 14 years) with HUS induced by enterohemorrhagic Escherichia coli (EHEC) and mice infected with EHEC were assessed for activation of the kallikrein-kinin system by degradation of high-molecular-weight kininogen ((HK) by immunoblotting) and levels of residual pre-kallikr

Kinin receptors in adipose tissue: drivers of inflammation and metabolic dysfunction in obesity.

BACKGROUND: Kinins, bioactive peptides produced through the proteolytic activity of kallikrein1, are members of the kallikrein-kinin system (KKS) and play crucial roles in regulating physiological processes such as inflammation, blood pressure, vascular permeability, and cell function and growth. In adipose tissue, bradykinin (BK) and des-Arg9-BK (DBK), produced by plasma kallikrein (KLKB1), act via their receptors B2 (B2R) and B1 (B1R), respectively. B1R predominates in preadipocytes, while B2R is expressed during adipogenesis, likely driving adipose tissue expansion and sustaining chronic lo

Targeting Kinin B1R Attenuates Hypertension Through AT1R-Dependent Mechanisms.

BACKGROUND: Neurogenic hypertension is chronically high blood pressure that is initiated and maintained through excessive sympathetic nervous system activity and has been associated with increased B1R (kinin B1 receptor) activation. We previously reported a central role for B1R in mediating inflammatory pathways in the development of deoxycorticosterone acetate salt hypertension. Additionally, we identified a causal relationship between B1R expression after Ang II (angiotensin II) stimulation, and that B1R can mediate the bidirectional interaction between neuroinflammation and oxidative stress

Kallikrein-kinin system as a potential target for the treatment of intervertebral disc degeneration.

Intervertebral disc degeneration (IVDD) is one of the main causes of chronic low back pain (LBP) and related spinal diseases, severely affecting patients' quality of life and imposing a considerable burden on healthcare systems. Current treatment strategies for IVDD primarily include conservative management and surgical intervention, but the therapeutic outcomes remain limited. Consequently, there is an urgent need to identify novel therapeutic targets. Recent research highlights the kallikrein-kinin system (KKS), a complex polypeptide network, as a key player in the pathological processes of

Kinin-B2 Receptor Activity in Skeletal Muscle Regeneration and Myoblast Differentiation.

The bioactive peptide bradykinin obtained from cleavage of precursor kininogens activates the kinin-B2 receptor functioning in induction of inflammation and vasodilatation. In addition, bradykinin participates in kidney and cardiovascular development and neuronal and muscle differentiation. Here we show that kinin-B2 receptors are expressed throughout differentiation of murine C2C12 myoblasts into myotubes. An autocrine loop between receptor activation and bradykinin secretion is suggested, since bradykinin secretion is significantly reduced in the presence of the kinin-B2 receptor antagonist
