Suppressor-cell activity in coeliac disease induced by alpha-gliadin, a dietary antigen.

The wheat protein antigen alpha-gliadin, a fraction derived from gluten of molecular weight 60 000, activated suppressor cells from patients with coeliac disease but not from normal subjects or patients with Crohn's disease. Two other dietary antigens, casein and beta-lactoglobulin, failed to produce suppressor-cell activation. Since this phenomenon appears to be specific to coeliac disease, it may be of pathogenetic significance.

Effect of gliadin and other food peptides on expression of MHC class II molecules by HT-29 cells.

Expression of major histocompatibility (MHC) class II molecules by enterocytes is known to be enhanced in coeliac disease and other disorders characterised by intestinal inflammation--an effect thought to be mediated via intestinal lymphocytes. To investigate if food peptides can exert direct effects on class II expression, the influence of gliadins, casein, and beta lactoglobulin on an intestinal epithelial cell line (HT-29) was examined in the absence of immune cells. Class II expression was determined by flow cytometry and immunofluorescence microscopy using antibodies against the beta chai

A comparison of secretory antibodies in breast-fed and formula-fed infants over the first six months of life.

In the present study salivary IgA, anti-Escherichia coli, anti-beta-lactoglobulin and anti-poliovirus type 1 IgA and IgM in serum and saliva were evaluated longitudinally in 13 breast-fed and 14 formula-fed infants over the first six months of life. Salivary IgA was quantified by electroimmunodiffusion; specific IgA and IgM antibodies were determined in serum and saliva by ELISA. Salivary IgA was significantly lower at age one month in breast-fed compared with formula-fed infants but in breast-fed infants salivary IgA increased with age and was significantly higher at six months than at one mo

Mixed Multilayers of Protein Fibrils with Sodium or Calcium Alginates.

A layer-by-layer approach is used for the preparation of multilayer films on the basis of biomacromolecules and their aggregates, protein fibrils. In this work, the fibrils were prepared from β-lactoglobulin and oat globulin, while sodium and calcium alginates were used as oppositely charged polyelectrolytes. Mixed layers of fibrils with alginate salts were formed at the air/liquid interface by the sequential replacement of solutions in the sublayer. The use of alginate salts resulted in a high stiffness of the mixed layers, allowing their transfer to a solid substrate without disturbing their

Using the Relationship between Concentrations of Selected Whey Proteins and BHBA to Characterize the Metabolism of Dairy Cows in Early Lactation.

A negative energy balance alters the concentration of various fractions of cows' milk. Therefore, by analyzing the concentration of individual proteins in milk, it is possible to indirectly monitor the health of the animal. The aim of the present study was to identify the relationship between the concentrations of blood β-hydroxybutyric acid (BHBA) and whey proteins in milk as a marker for the diagnosis of metabolic diseases. The analysis included milk and blood samples from 95 Holstein-Friesian cows, which were divided into three groups that were differentiated in terms of serum BHBA levels 5

Interfacial engineering using mixed protein systems: emulsion-based delivery systems for encapsulation and stabilization of β-carotene.

Emulsion-based delivery systems are needed to encapsulate, protect, and deliver lipophilic bioactive components in the food, personal care, and pharmaceutical industries. The functional performance of these systems can be controlled by engineering the composition and structure of the interfacial layer coating the lipid droplets. In this study, interfacial properties were controlled using two globular proteins with widely differing isoelectric points: lactoferrin (LF: pI ≈ 8.5) and β-lactoglobulin (BLG: pI ≈ 5). Oil-in-water emulsions were prepared with different interfacial properties: [LF]-on

Gastric aspiration in sudden unexpected infant death of Prader-Willi syndrome: immunohistochemical detection of feeding components.

Prader-Willi syndrome (PWS) in infants is characterized by hypotonia and poor sucking with feeding difficulties. Two autopsy cases of sudden unexpected death during sleep after tube feeding are described herein. For one, gastric aspiration caused by the possible milk regurgitation was suspected. Immunohistochemical examination of lung sections was performed using three antibodies to human α-lactalbumin, human gross cystic disease fluid protein 15, and cow whey β-lactoglobulin. Five cases of sudden unexpected infant death occurring earlier than at 6 months old were selected as controls. Marked

[The story of Appendix].

The etiology of Ménière's disease (MD) is still unknown, but it is likely to be multifactorial, one of the factors being an immunological causation. Antifood allergens as well as anti-baker's yeast antibodies are humoral factors that may be linked with allergenic disorders and other autoimmune conditions. To determine their possible role in MD activity, we investigated 29 MD sera for the presence of antibodies against gliadin, beta-lactoglobulin, albumin, ovalbumin, soya, and Dermatophagoides pteronyssinus and Saccharomyces cerevisiae strains using an ELISA technique. The patients were compare

A simple test for lacrimal gland function: a tear lactoferrin assay by radial immunodiffusion.

A radial immunodiffusion assay for tear lactoferrin is described. Tear samples, collected on filter paper discs, could be applied directly to immunodiffusion plates after blotting. In 58 tear samples from healthy subjects an average lactoferrin concentration of 1.42 g/l was found. A high degree of correlation was found between the lactoferrin radial immunodiffusion and the lysozyme agar diffusion assay in a heterogeneous group, ranging from healthy subjects to patients with severe keratoconjunctivitis sicca. A lower limit of normal (0.78 g/l) could be determined for the tear lactoferrin concen

Perturbation-Modulated Native Mass Spectrometry Excludes a Nonspecific Drug Target Protein Binder Based on Conformation Stability Change.

Native electrospray ionization mass spectrometry (ESI-MS) is a standard technique for drug-protein screening but may be affected by nonspecific binding. To address this, a reference protein and ligand must be selected for each protein investigated. We report a versatile, informative, high-throughput screening approach to differentiate specific and nonspecific interactions based on charge state distribution (CSD) changes caused by ionization perturbations (e.g., methanol or heat) without the need for a surrogate. We show that specific binding stabilizes the protein-ligand complex against pertur

Hyphenation of surface plasmon resonance imaging to matrix-assisted laser desorption ionization mass spectrometry by on-chip mass spectrometry and tandem mass spectrometry analysis.

Most of the recent developments aiming to the coupling between surface plasmon resonance (SPR) and mass spectrometry (MS) are based on the use of a biochip with a limited number of flow cells requiring elution steps for the recovery of the captured biomolecules. In this work, a direct on-chip MALDI-MS detection is presented using a SPRi-sensor biochip in a microarray format that allows a multiplex SPR-MS analysis. The biochip gold surface was functionalized by a self-assembled monolayer (SAM) of short polyoxyethylene (POE) chains carrying a N-hydroxysuccinimide (NHS) group for the immobilizati

Lymphocytes in Peyer patches regulate clinical tolerance in a murine model of food allergy.

BACKGROUND: Food allergy can lead to severe, potentially life-threatening anaphylactic reactions. To generate efficient strategies aimed at an active cure, a better understanding of the pathogenic mechanisms is strongly needed. OBJECTIVE: To investigate T-cell-related mechanisms of food allergy and tolerance to beta-lactoglobulin (BLG) in gut-associated lymphoid structures. METHODS: Beta-lactoglobulin-specific IgG1, IgG2a, and IgE in serum from mice anaphylactic to BLG were analyzed by ELISA and compared with those obtained in mice actively tolerized to BLG. The number of Ab-secreting cells

Secretory antibodies in IgA-deficient and immunosuppressed individuals.

Total levels of IgM and secretory IgM as well as specific antibodies to poliovirus type I antigen, Escherichia coli O antigens, and beta-lactoglobulin were measured in unstimulated and stimulated saliva as well as nasal secretion using an enzyme-linked immunosorbent assay (ELISA). The levels of these antibodies in IgA-deficient adults with and without frequent respiratory infections and children under immunosuppressive therapy for malignant disease were compared to those in normal adults and infants 1-7 months of age. The IgA-deficient adults had significantly higher IgM levels (P less than 0.

A way to probe the microenvironment of free sulfhydryls in intact proteins with a series of monofunctional organic mercurials.

Monofunctional organic mercurials (MFOHgs) of different sizes and electronic properties were used to evaluate the accessibility of SH groups in intact proteins. The change in conformation of proteins, such as BLG and OVA, when treated with MFOHgs of increasing size was used as a measure of the accessibility of SH groups contained within these proteins.

An In Vitro and In Vivo Translational Research Approach for the Assessment of Sensitization Capacity and Residual Allergenicity of an Extensive Whey Hydrolysate for Cow's Milk-Allergic Infants.

INTRODUCTION: Hypoallergenic formulas prepared from hydrolyzed cow's milk proteins are often used for the management of cow's milk allergy (CMA) in infants. In this study, both in vitro assays and an in vivo mouse model for CMA were used to assess the sensitizing and allergenic potential of a newly developed, extensive whey hydrolysate (eWH). METHODS: Gel permeation chromatography was used to characterize the molecular weight distribution of the peptides. Residual antigenicity was measured using a beta-lactoglobulin ELISA as well as with immunoblotting using anti-beta-lactoglobulin (BLG) and

Unusual presentation of adverse reactions to cow's milk proteins.

Allergy to cow's milk most commonly give symptoms from the gastrointestinal tract, the skin or the respiratory tract. Here two patients are described, with symptoms not so common. One with cerebral symptoms and the other with enuresis. Infantile colic occurs more or less in about 20% of all infants. We have found that in about 1/3 of breastfed infants with infantile colic the symptoms disappeared when the mothers had a cow's milk free diet. We have also managed to analyse the content of the cow's milk protein beta-lactoglobulin in the human milk.

Investigation of the Structure and Allergic Potential of Whey Protein by Both Heating Sterilization and Simulation with Molecular Dynamics.

As the main allergens in milk, whey proteins are heat-sensitive proteins and are widespread in dairy products and items in which milk proteins are involved as food additives. The present work sought to investigate the effect of heating sterilization on the allergenicity of α-lactalbumin (α-LA) and β-lactoglobulin (β-LG), the main composite and allergen in whey protein isolate (WPI), by combining molecular dynamics with experimental techniques for detecting the spatial structure and IgE binding capacity. The structure of WPI was basically destroyed at heat sterilization conditions of 95 °C for

Model studies on reactions of plant phenols with whey proteins.

Whey proteins were modified by reaction with selected phenolic compounds (ferulic-, chlorogenic-, caffeic- and gallic acid) and related substances (quinic acid and p-quinone) as well as with extracts from coffee, tea, potato and pear at pH 9. The derivatives formed were characterized in terms of their physicochemical and digestion properties. The derivatization was accompanied by a reaction at the lysine and tryptophan side chains, whereby their content was decreased in comparison to that in the control whey proteins. Moreover, the solubility of the derivatives decreased over a broad pH range

Comparison of commercial allergen extract with various milk products of skin prick tests in diagnosing cow's milk protein allergy.

Introduction: In this study, we aimed to solve the incompatibilities between the skin prick test (SPT) result and the clinical picture encountered among patients. Thus, we used various milk products (raw, ultrahigh temperature [UHT], and pasteurized cow's milk) with commercial milk allergen extract in patients we think may have cow's milk protein allergy (CMPA) and made a comparison among the milk types. Methods: This study was conducted retrospectively between February 1, 2019, to June 1, 2019, at a pediatric allergy polyclinic of the university training/research hospital. A pair of 79 indivi

Increased gliadin antibodies in dermatitis herpetiformis and pemphigoid.

The mean titres of serum IgG and IgA gliadin antibodies were significantly increased in thirty-four patients with bullous pemphigoid, and in twenty-three patients with dermatitis herpetiformis, compared with twenty-four healthy controls. The patients with pemphigoid also had increased IgG and IgA gliadin antibodies compared with nine patients with pemphigus. The reason for the high titres of gliadin antibodies in pemphigoid is obscure. These patients may have increased intestinal permeability (suggested by the presence of beta-lactoglobulin antibodies in some patients). Alternatively gliadin m
