All extant jawless vertebrates (lampreys and hagfishes) possess a unique adaptive immune system characterized by highly variable lymphocyte receptors (VLR) that are assembled in developing lymphocytes using leucine-rich-repeat donor cassettes. Five VLR types have been identified in lampreys: VLRA, VLRB, VLRC, VLRD, and VLRE. VLRB-expressing lymphocytes are functional analogs to B cells, whereas VLRA, VLRC, VLRD, and VLRE-expressing lymphocytes are more akin to T cells of jawed vertebrates. Here we define an additional VLR, designated VLRF. VLRF is phylogenetically closest to VLRA, with which i

The hypothalamic-pituitary (HP) system, which is specific to vertebrates, is considered to be an evolutionary innovation that emerged prior to or during the differentiation of the ancestral jawless vertebrates (agnathans) leading to the neuroendocrine control of many complex functions. Along with hagfish, lampreys represent the oldest lineage of vertebrates, agnathans (jawless fish). This review will highlight our discoveries of the major components of the lamprey HP axis. Generally, gnathostomes (jawed vertebrates) have one or two hypothalamic gonadotropin-releasing hormones (GnRH) while lamp

Sea lampreys (Petromyzon marinus) are members of the ancient class Agnatha and undergo a metamorphosis that transforms blind, sedentary, filter-feeding larvae into free-swimming, parasitic juveniles. Thyroid hormones (THs) appear to be important for lamprey metamorphosis, however, serum TH concentrations are elevated in the larval phase, decline rapidly during early metamorphosis and remain low until metamorphosis is complete; these TH fluctuations are contrary to those of other metamorphosing vertebrates. Moreover, thyroid hormone synthesis inhibitors (goitrogens) induce precocious metamorpho

In this study we sought to demonstrate that Comet assay can be applied to sea lamprey sperm DNA fragmentation and used to describe the relationship between sperm DNA damage and sperm fertilizing ability. We show that the assay can be used reliably and accurately, and unlike in the case of mammals, there is no need for additional steps related to improvement of efficacy of lysis and DNA decondensation. This agrees with the presence of histone proteins in lamprey sperm. An increase in DNA fragmentation was noted during short-term storage of milt on ice (0-4 days). We demonstrated genotoxic effec

BACKGROUND: The genus Caspiomyzon is comprised of three species, two of which are found in Greece (Caspiomyzon hellenicus and Caspiomyzon graecus). Both species are endemic with very restricted distribution and are classified as Critically Endangered. Caspiomyzon hellenicus is restricted to Tenagi, Philippi and C. graecus to Louros River. No studies have characterized their mitochondrial genomes.
METHODS AND RESULTS: The complete mitochondrial genome of C. hellenicus and C. graecus was generated with Oxford Nanopore long-read technology, and it was processed using various bioinformatics appro

The occurrence and localization of molecular components of pro-opiocortin in the pituitary and brain of two species of adult lamprey, Petromyzon marinus and Entosphenus tridentatus, were studied immunocytochemically using antisera generated against human pro-gamma-MSH (N-terminal fragment 1Trp to 71Gly of pro-opiocortin), porcine ACTH, alpha-MSH, human beta-endorphin, gamma-endorphin, and methionine enkephalin. (1) In both species of lamprey most cells of the rostral pars distalis and some cells of the caudal pars distalis contained Met-enkephalin-like immunoreactivity. Some of these cells als

Invasive sea lamprey (Petromyzon marinus) populations in the Great Lakes are controlled by applying the piscicide, 3-trifluoromethyl-4-nitrophenol (TFM), to infested streams with larval sea lamprey (ammocoetes). While treatment mortality is >90%, surviving lamprey, called residuals, can undermine control efforts. A key determinant of TFM effectiveness is water pH, which can fluctuate daily and seasonally in surface waters. The objectives of this research were to evaluate the influence of pH on the uptake, elimination, and accumulation of TFM by larval sea lamprey using radio-labeled TFM (14C-T

The dopaminergic system influences motor behavior, signals reward and novelty, and is an essential component of the basal ganglia in all vertebrates including the lamprey, one of the phylogenetically oldest vertebrates. The intrinsic organization and function of the lamprey basal ganglia is highly conserved. For instance, the direct and indirect pathways are modulated through dopamine D1 and D2 receptors in lamprey and in mammals. The nucleus of the tuberculum posterior, a homologue of the substantia nigra pars compacta (SNc)/ventral tegmental area (VTA) is present in lamprey, but only scarce

Within the RFamide peptide family, PQRFamide peptides that include neuropeptide FF and AF possess a C-terminal Pro-Gln-Arg-Phe-NH(2) motif. We previously identified PQRFamide peptides, lamprey PQRFa, PQRFa-related peptide (RP)-1 and -RP-2 by immunoaffinity purification in the brain of lamprey, one of the most ancient vertebrate species [13]. Lamprey PQRFamide peptide precursor mRNA was expressed in regions predicted to be involved in neuroendocrine regulation in the hypothalamus. However, the putative function(s) of lamprey PQRFamide peptides (PQRFa, PQRFa-RP-1 and PQRFa-RP-2) were not examine

The distribution of the central neural connections of the pineal organ of the larval sea lamprey was investigated by means of anterograde and retrograde tracing with the fluorescent lipophilic dye, DiI (1,1'-dioctadecyl 3,3,3',3'-tetramethylindocarbocyanine perchlorate). Pinealofugal projections are well developed in larvae, extending from the posterior commissure into the diencephalon and mesencephalon. Small numbers of neurons were retrogradely labelled in the transition zone between the diencephalon and the mesencephalic tegmentum. These cells may constitute the first pinealopetal system de

The Shorthead lamprey Mordacia mordax (Mordaciidae, Agnatha) represents one of the earliest stages of vertebrate evolution. This study investigates the ultrastructural anatomy of the cornea, iris and anterior chamber in the eyes of this species in both the downstream and upstream migrant phases of its protracted lifecycle to assess the morphological and quantitative changes associated with growth, corneal function and vision. Using light and both scanning and transmission electron microscopy, the cornea is found to be divided into dermal and scleral components separated by a mucoid layer. A ra

The nucleotide oligomerization domain (NOD)-like receptor (NLR) is a relatively conserved receptor family involved in natural immunity that plays a key role in the resistance to pathogen invasion and regulation of the innate immune response. Lethenteron reissneri (lamprey) is a representative species of existing ancient jawless vertebrates. Studies of the evolutionary relationship of immune system-related molecules in lampreys can provide an important reference for the origin and evolution of innate immunity. However, the characterization and evolutionary patterns of the NLR family remain uncl

A Ca(2+)-activated, non-selective cation current (I(CAN)) has been suggested to contribute to plateau potentials in lamprey reticulospinal neurons, providing the drive for locomotor initiation. Flufenamic acid (FFA) is commonly used as a blocker of I(CAN). To explore the effects of FFA on spinal locomotor pattern generation, we induced fictive locomotion in the isolated lamprey spinal cord. Bath-applied FFA (100-200microM) caused a marked reduction of amplitude and regularity of the locomotor burst activity. We next analyzed the NMDA-induced membrane potential oscillations in single spinal neu

Niclosamide (2',5-dichloro-4'-nitrosalicylanalide) is a piscicide used to control invasive sea lamprey (Petromyzon marinus) in the Laurentian Great Lakes. It is also a molluscide used in tropical and sub-tropical freshwaters to control snail populations that are intermediate hosts to the blood flukes that causes schistosomiasis in humans. While the mechanism of niclosamide toxicity is known, its corresponding physiological effects on non-target fishes are not well-established. To better understand how niclosamide could adversely affect non-target fishes, rainbow trout (Oncorhynchus mykiss) wer

The innate immune system is the body's first line of defense against pathogens and involves antibody and complement system-mediated antigen removal. Immune-response-related complement molecules have been identified in lamprey, and the occurrence of innate immune response via the mannose-binding lectin-associated serine proteases of the lectin cascade has been reported. We have previously shown that lamprey (Lampetra japonica) serum can efficiently and specifically eliminate foreign pathogens. Therefore, we aimed to understand the immune mechanism of lamprey serum in this study. We identified a

The results of this study were found to support the hypothesis put forth by Boenig (Z. Mikrosk-Anat. Forsch., 17:125-184, 1929) that the caudal pancreas of adult lamprey develops from the epithelium of the extrahepatic common bile duct in that the bile duct cells were found to undergo a great proliferation during the early stages of metamorphosis, with a large number of the cells incorporating 3H-thymidine. If the bile duct degenerated as suggested by Barrington (The Biology of Lampreys. Academic Press, London, pp. 135-169, 1972), this uptake would not be expected. The cranial pancreas was det

Fresh lamprey (F-La) or sardine (F-Sa) oil is known to contain a large amount of n-3 polyunsaturated fatty acids such as eicosapentaenoic (EPA) and docosahexaenoic (DHA) acids. When F-La or F-Sa was deodorized with steam at 280 degrees C under 1 mmHg for 1 h (H-La and H-Sa, respectively), the contents of EPA and DHA were reduced and unidentified peaks were newly detected by gas-liquid chromatography. To know the biological influences of these high-temperature deodorized oils, the sterilizing function of the macrophage against Listeria monocytogenes was investigated in male ddY mice fed H-La or

1. Somatostatin concentrations were measured in homogenates of the pancreas-intestinal tissues from each period of the life cycle of Petromyzon marinus using radioimmunoassay. 2. Levels were very low in larva (4.0 pg/mg wet weight) and in the first three stages of metamorphosis, but increased from stage 4 onwards and reached a high in upstream-migrating adults (210.0 ng/mg). 3. These data correlate well with our previous morphological and immunohistochemical observations on the morphogenesis of somatostatin-containing D-cells during the life cycle and indicate that the increased concentration

Synaptic aggregation of α-synuclein often occurs in Parkinson's disease (PD), dementia with Lewy bodies (DLB) and other synucleinopathies and is associated with cognitive deficits and dementia. Thus, it is important to understand how accumulation of α-synuclein affects synapse structure and function. Native, physiological α-synuclein comprises a mixture of tetramers and related physiological oligomers (60-100 kDa) in equilibrium with monomeric α-synuclein. We previously demonstrated that acutely increasing the levels of physiological α-synuclein impaired intracellular synaptic vesicle traffick

Two orthologues of the gene encoding the Na+-Cl- cotransporter (NCC), termed ncca and nccb, were found in the sea lamprey genome. No gene encoding the Na+-K+-2Cl- cotransporter 2 (nkcc2) was identified. In a phylogenetic comparison among other vertebrate NCC and NKCC sequences, the sea lamprey NCCs occupied basal positions within the NCC clades. In freshwater, ncca mRNA was found only in the gill and nccb only in the intestine, whereas both were found in the kidney. Intestinal nccb mRNA levels increased during late metamorphosis coincident with salinity tolerance. Acclimation to seawater incre
