Expression patterns and proliferative effect of OTUD6A on Sertoli cells and Leydig cells in cattleyak.

Cattleyak, a hybrid between cattle and yak, can adapt to the harsh environment of the highlands and also exhibit high production performance. However, its male infertility problem has not yet been solved, which limits the application of its hybrid vigor. In this work, we cloned the OTUD6A gene of cattleyak and analyzed its expression patterns and proliferative effect on Sertoli cells and Leydig cells. The expression level of OTUD6A in testicular tissues, Sertoli cells and Leydig cells were significantly lower than those in yak. Over-expression of OTUD6A in Sertoli cells and Leydig cells of cat

Effects of Midazolam on the Development of Adult Leydig Cells From Stem Cells In Vitro.

BACKGROUND: Midazolam is a neurological drug with diverse functions, including sedation, hypnosis, decreased anxiety, anterograde amnesia, brain-mediated muscle relaxation, and anticonvulsant activity. Since it is frequently used in children and adolescents for extended periods of time, there is a risk that it may affect their pubertal development. Here, we report a potential effect of the drug on the development of Leydig cells (LCs), the testosterone (T)-producing cells in the testis. METHODS: Stem LCs (SLCs), isolated from adult rat testes by a magnetic-activated cell sorting technique, we

Chlorocholine chloride disrupts differentiation of rat stem/progenitor Leydig cells: A combined in vivo and in vitro study by Leydig cell regeneration model.

Chlorocholine chloride (CCC), a widely-used plant growth regulator, impairs testosterone biosynthesis, but the mechanism remains elusive. The Ethane dimethane sulfonate (EDS)-induced rat Leydig cell regeneration model was adopted to investigate the effect of CCC on Leydig cell development and testosterone levels. Rats received 75, 137.5, or 200 mg/kg bw CCC from post EDS d7 to d28 or d56. At d28, CCC exposure inhibited stem/progenitor Leydig cell differentiation. At d56, CCC exposure affected the maturation status of adult Leydig cells, reduced expression of StAR, HSD3B1, and serum and testicu

Leydig cell ovarian tumour: a rare cause of hyperandrogenism.

We present a case of a postmenopausal woman with androgenic alopecia and very high testosterone levels of 16 nmol/L (normal range 0.4-1.2). CT showed an enlarged right ovary with a high-density nodule and an incidental adrenal nodule. She underwent bilateral salpingo-oophorectomy and pathology confirmed a benign Leydig cell tumour of the right ovary and Leydig cell hyperplasia of the left. Testosterone levels improved to near normal postoperatively. Adrenal investigations favoured a benign adenoma, although adrenal androgen production could not be ruled out. This case highlights the challenges

Human benign Leydig cell tumor - Biochemical evaluation.

Recently we reported expressional alterations in 219 genes and their transcripts in Leydig cell tumors but nowadays there is still a lack of full basic biochemical characteristics of these tumors. The discovery of potential biochemical markers for tumor management from early detection, treatments, and control of therapy results may markedly supplement genetic data. Leydig cell micronodules were obtained from patients with azoospermia who were qualified for testicular biopsy. The biochemistry of Leydig cell tumors was analyzed using histological staining and spectrophotometric measurements of t

Management of testicular Leydig cell tumor: A case report.

RATIONALE: Testicular Leydig cell tumor (LCT) is a rare neoplasm. It commonly presents as a painless testicular mass with or without endocrine changes. Histological and immunohistochemical examination play important roles in differentiating LCT from testicular germ cell tumors. PATIENT CONCERNS: We highlight the imaging phenotype, as well as the pathological findings of a case of LCT in a 62-year-old male. DIAGNOSES: Preoperative noncontrast CT scan of the abdomen revealed a 7.0 × 6.4 × 5.3 cm oval mass with heterogeneous density, located in the right testis. Pelvic noncontrast MRI showed a

Two Years of Gynecomastia Caused by Leydig Cell Tumor.

Gynecomastia is a common incidental finding in males that can be caused by various benign or malignant diseases. In rare cases, it results from Leydig cell tumors, a rare entity accounting for 3% of all testicular neoplasms. Some of them are hormonally active but seldom cause symptomatic endocrine disturbance. Here we report on a 32-year-old male presenting with gynecomastia which he had already been suffering from for two years. Although he had been seen by three other specialists, including a urologist, none of them found the small mass in the upper pole of his right testis. We decided to pe

Testis sparing surgery for Leydig cell pathologies in children.

OBJECTIVE: The aim was to analyze testis-sparing surgical procedures in boys with Leydig cell pathologies. STUDY DESIGN: The hospital records of four boys with Leydig cell hyperplasia who underwent testis-sparing surgery for testicular masses between 2000 and 2012 were analyzed retrospectively. Tumor markers were evaluated and all boys underwent scrotal ultrasonography preoperatively. The hormonal profile was also analyzed for symptoms of precocious puberty. The testis was delivered through a high transverse inguinal incision and the tumor was excised by enucleation. After confirming the beni

Clinicopathologic features of ovarian Sertoli-Leydig cell tumors.

BACKGROUND: Ovarian Stertoli-Ledig cell tumor (SLCT) is a rare type of sex cord-stromal tumor of the ovary. The present study was to evaluate clinicalopahologic features and prognosis of patients with Sertoli-Leydig cell tumor treated by surgery and adjuvant chemotherapy during short term follow-up. METHODS: A total of sixteen patients with ovarian Sertoli-Leydig cell tumor treated at the Obstetrics and Gynecology Hospital, Shanghai, China, between Jan 2001 and Dec 2011 were reviewed. The clinical data, treatment and prognosis were obtained from medical records. RESULTS: The median age of th

[Bilateral Leydig cell tumor of the test: a case report].

Testicular Leydig cell tumours are uncommon. Bilateral synchronous lesions are exceptional. They cause isosexual pseudo precocious puberty in childhood. The histological diagnosis of malignancy is sometimes difficult to establish and it can be made retrospectively when lymph nodes involvement or visceral metastasis appear in the follow-up. We report a case of a 9 year-old boy presenting bilateral Leydig cell tumour of the testis treated by bilateral radical orchiectomy who developed 2 years after the intervention a pulmonary metastasis.

[Precocious puberty caused by a testicular Leydig cell tumor].

Testicular tumors are very rare in boys, approximately 1.5% of these are Leydig cell tumors. The authors present a 4.5 year-old boy with Leydig cell tumor of left testis, which was associated with increased sex steroid production that caused precocious puberty. These tumors are benign processes in prepubertal children. Beyond the rarity of this case the authors would like to report about its diagnostic difficulties and testis-sparing remove of it. The plasma 17-hydroxyprogesterone levels provide the distinction between congenital adrenal hyperplasia and Leydig cell tumor in patient with precoc

Leydig cell tumour in a man with human immunodeficiency virus.

We report a case of Leydig-cell testicular tumour in a 32-year-old intravenous drug abuser bearing HIV-infection and pulmonary tuberculosis. The testicular mass was initially interpreted as genital tuberculosis and the diagnosis was made by aspiration cytology. Radical orchiectomy was performed and pathology revealed a Leydig-cell tumour. Gynecomastia was found as a possible sign of endocrine activity. Although testicular malignancies in men with human immunodeficiency virus are increasingly reported, this is the first case of Leydig-cell neoplasia associated to HIV-infection.

Bilateral virilizing hilus (Leydig) cell tumors of the ovary.

A 72-year-old postmenopausal woman with high plasma testosterone levels and virilization, as demonstrated by hirsutism and balding, is presented. Urinary 17-ketosteroids and 17-hydroxycorticosteroids, as well as computed axial tomography scan of the adrenal glands were normal. Although no pelvic mass was detected by ultrasonography or pelvis examination, the patient was found to have small bilateral hilus cell tumors of the ovary. Following bilateral salpingo-oophorectomy the plasma testosterone dropped to normal level. This is the third case of bilateral hilus cell tumors of the ovary to be r

Interstitial cell (Leydig) tumor in an eland (Taurotragus oryx).

We observed an interstitial cell tumor in an 18-mo-old captive eland bull (Taurotragus oryx) in Tel Aviv, Israel. The histological description of the tumor in the eland was similar to that described in cattle; however, the appearance of a moderate amount of lipid vacuoles in the cytoplasm of the neoplastic cells was uncharacteristic for bovine interstitial cell tumors. The eland also had clinical signs of gynecomastia.

[Bilateral testicular Leydig cell tumor. Procedure to follow].

The authors report one case of a bilateral testicular Leydig cell tumor in a man of 29 years old. There are few cases of such tumors reported in the literature. Gynecomastia force the patient to consult his doctor. His hormonal profile is practically normal; however, the serum estradiol is at the limit superior of normal range, and serum testosterone at the limit inferior. Testicular palpation is normal. It is the scrotal ultrasonography which confirms the diagnosis of testicular tumor. The scrotal ultrasonography has to be performed in every patient with unexplained gynecomastia. There is no

A 32-year-old patient with a history of surgery for left gynecomastia four years previously presented with right gynecomastia; a tumor in the left testis proved to be a Leydig cell tumor. Preoperative investigations showed elevated but variable levels of plasma estradiol (E2) and estrone (E1), and reduced serum LH and FSH and plasma testosterone (T). After hCG stimulation, E2 response was increased and abnormally prolonged; T reached normal values, which has predictive value for a return to normal of post-operative T level.
