Vertebrate photoreceptor cells operate under very dim and bright illumination regimes. A protein machinery in rod and cone cells underlying the light response mediates photoexcitation, the return to the dark state, and adaptation processes. The machinery controls the homeostasis and mutual dependence of two cytoplasmic messengers, cGMP and Ca2+. The signaling pathway starts with light absorption by visual pigments (rhodopsin in rods or cone opsin in cones), which triggers an amplified signaling cascade, leading to the hydrolysis of cGMP and closure of cyclic nucleotide-gated channels in the ph

Abnormalities in T-lymphocyte populations and function are observed in autism. Soluble amyloid precursor protein α (sAPP-α) is elevated in some patients with autism and is known to be produced by immune cells. In light of the well-established role of sAPP-α in proliferation, growth, and survival of neurons, we hypothesized an analogous role in the immune system. Thus, we explored whether sAPP-α could modulate immune development and function, especially aspects of the pinnacle cell of the adaptive arm of the immune system: the T cell. To do this, we generated mice overexpressing human sAPP-α an

Evolutionary forces have designed a large family of rod and cone photoreceptors, each member of which suits the lifestyle requirements and circadian patterns of a particular species. The three-segment architecture of signal transduction is conspicuous in the biochemistry of photoreceptors and supports their demonstrated properties of extreme sensitivity, low noise levels, extended dynamic range, and light adaptation. The designs elaborated by evolution reflect a gradual process of modification, with the sequential elaboration of layers of control and refinements in control. The end results of

The retina is one of the highest oxygen-consuming tissues because visual transduction and light signaling processes require large amounts of ATP. Thus, because of the high energy demand, oxygen-rich environment, and tissue transparency, the eye is susceptible to excess production of reactive oxygen species (ROS) resulting in oxidative stress. Oxidative stress in the eye is associated with the development and progression of ocular diseases including cataracts, glaucoma, age-related macular degeneration, and diabetic retinopathy. ROS can modify and damage cellular proteins, but can also be invol

Photoreceptors in the retina are highly specialized neurons with photosensitive molecules in the outer segment that transform light into chemical and electrical signals, and these signals are ultimately relayed to the visual cortex in the brain to form vision. Photoreceptors are composed of rods and cones. Rods are responsible for dim light vision, whereas cones are responsible for bright light, color vision, and visual acuity. Photoreceptors undergo progressive degeneration over time in many hereditary and age-related retinal diseases. Despite the remarkable heterogeneity of disease-causing g

A distance-dependent electrochemiluminescence resonance energy transfer (ERET) system based on CdTe nanocrystals and Au nanoclusters (Au NCs) was designed with the aid of ligase for highly selective detection of microRNA (miRNA). First, Au NCs functionalized hairpin DNA was synthesized via Au-S chemistry, and characterized with transmission electron microscopy and dynamic light scattering. The resulting hairpin DNA-Au NCs composite can be bound to the carboxylated CdTe nanocrystals via amide reaction on glass carbon electrode. The strong interaction between CdTe nanocrystals and AuNCs led to t

BACKGROUND: Low-level red light (LLRL) irradiation may inhibit myopia occurrence and progression. Understanding how LLRL inhibits fibrosis in human retinal pigment epithelial (hRPE) cells is critical to inhibiting myopia progression and developing novel therapeutic strategies. Here, we explored the effects of LLRL on hRPE cells in a myopia-simulating hypoxic microenvironment and elucidated the mechanisms through which it inhibits scleral remodeling.
METHODS: We first used the MTT assay to analyze hRPE cell proliferation under hypoxic conditions after LLRL irradiation at varying frequencies ov

Adequate photoprotection is essential to control UV-related disorders, including sunburn, photoaging and photocarcinogenisis. Sun avoidance, protection of skin with clothing, and sunscreens are presently the best way of photoprotection, assuming that they are used properly. However, new strategies, which are based on or make use of the endogenous protective response to UV light, may further improve currently used photoprotective means. The addition of repair enzymes and/or antioxidants has a positive effect on skin's recovery from UV-induced DNA-damage. Several botanical agents, mainly vitamin

Male Sprague-Dawley rats injected (i.p.) at 1500h with L-acetyl-carnitine in doses of 10, 30 or 90 mg/kg exhibited a notable increase in their pineal and serum melatonin content 1 hr later. Likewise, L-acetyl-carnitine administered in the same dose range induced a significant increase of pineal and serum melatonin content in rats treated at 0100h, following exposure of 30 min to bright white light to suppress endogenous melatonin. Under in vitro experimental conditions, however, 60 min of coincubation of isolated rat pineal glands with L-acetyl-carnitine (10(-5) M) did not result in an elevati

Fish body color changes play vital roles in adapting to ecological light environment and influencing market value. However, the initial mechanisms governing the changes remain unknown. Here, we scrutinized the impact of light spectrum on turbot (Scophthalmus maximus) body coloration, exposing them to red, blue, and full light spectra from embryo to 90 days post hatch. Transcriptome and quantitative real-time PCR (qRT-PCR) analyses were employed to elucidate underlying biological processes. The results showed that red light induced dimorphism in turbot juvenile skin pigmentation: some exhibited

This paper suggests to rigorously test the hypothesis that there are causal links between visible light and the development of leukaemia and other cancers in children. Light can be considered as a candidate risk factor because it suppresses melatonin biosynthesis which may play a role in a series of anticancer defences. Indeed, melatonin may offer some protection against all "hallmarks of cancer" [i.e., self-sufficiency in growth signals; insensitivity to growth-inhibitory signals; evasion of programmed cell death (apoptosis); limitless replicative potential; sustained angiogenesis; tissue inv

Self-powered wearable photonic sensors enable energy-autonomous operation by integrating optical sensing, energy harvesting, and signal transduction within a unified platform. These systems exploit photophysical mechanisms such as photovoltaic, photogating, and triboelectric effects to convert ambient or body-derived energy into electrical signals while simultaneously performing sensing. Advances in hybrid semiconductors, two-dimensional materials, and nanocomposites have significantly improved light absorption, carrier transport, and mechanical compliance, enabling integration into flexible a

Ultraviolet (UV) light is emerging as an important tool for biosensing, biomedical signal readout, and dose monitoring because of its strong and selective interactions with nucleic acids, proteins, and other biological components. This review summarizes recent progress in UV sensing-guided biomedical systems, with emphasis on three interconnected directions: label-free and surface-weighted imaging, wearable and embedded UV dosimetry, and sensor-assisted therapeutic guidance. Representative examples include ultraviolet photoacoustic microscopy (UV-PAM) for label-free nuclear imaging, microscopy

Dendrobium officinale Kimura et Migo (D. officinale) has promising lung moisturizing, detoxifying, and immune boosting properties. Light is an important factor influencing functional metabolite synthesis in D. officinale. The mechanisms by which lasers affect plants are different from those of ordinary light sources; lasers can effectively address the shortcomings of ordinary light sources and have significant interactions with plants. Different light treatments (white, blue, blue laser) were applied, and the number of red leaves under blue laser was greater than that under blue and white ligh

The multipotent mesenchymal stem/stromal cells (MSCs), initially discovered from bone marrow in 1976, have been identified in nearly all tissues of human body now. The multipotency of MSCs allows them to give rise to osteocytes, chondrocytes, adipocytes, and other lineages. Moreover, armed with the immunomodulation capacity and tumor-homing property, MSCs are of special relevance for cell-based therapies in the treatment of cancer. However, hampered by lack of knowledge about the controversial roles that MSC plays in the crosstalk with tumors, limited progress has been made with regard to tran

Understanding the complex interplay among gasotransmitters is of great significance but remains technically challenging. In this study, we present the design and synthesis of a dually responsive BOD-NH-SC reporter for probing the dynamic and alternating existence of NO and H2 S in living cells. This designed reporter can repeatedly cycle S-nitrosation and transnitrosation reactions when successively treated with NO and H2 S, thus affording the interchange of NIR fluorescence at 645 nm (NO) and NIR II fluorescence at 936 nm (H2 S). In light of this unique fluorescence alternation between two co

We report a dual-readout, AuNP-based sandwich immunoassay for the device-free colorimetric and sensitive scanometric detection of disease biomarkers. An AuNP-antibody conjugate serves as a signal transduction and amplification agent by promoting the reduction and deposition of either platinum or gold onto its surface, generating corresponding colorimetric or light scattering (scanometric) signals, respectively. We apply the Pt-based colorimetric readout of this assay to the discovery of a novel monoclonal antibody (mAb) sandwich pair for the detection of an anthrax protective antigen (PA83). T

The action of the promoting agent 12-0-tetradecanoyl phorbol-13-acetate (TPA), an active component of croton oil, on the cell membrane, is described. TPA primarily acts on Protein Kinase C (PKC), which is the prime target for this agent. PKC activation and calcium mobilization are the basic pathways for signal transduction and the regulation of differentiation, explaining how TPA affects cell growth and proliferation in some cell types. The effects of TPA on leukaemic cells in-vitro, is reviewed and the changes in cell surface features, membrane phenotype, regulation of growth and differentiat

Circadian rhythms in physiological, endocrine and metabolic functioning are controlled by a neural clock located in the suprachiasmatic nucleus (SCN). This structure is endogenously rhythmic and the phase of this rhythm can be reset by light information from the eye. A key feature of the SCN is that while it is a small structure containing on the order of about 20,000 cells, it is amazingly heterogeneous. It is likely that anatomical heterogeneity reflects an underlying functional heterogeneity. In this review, we examine the physiological responses of cells in the SCN to light stimuli that re

This review explores contemporary ideas about the relationship between light exposure and vitamin biology. Nutritional biochemistry has long recognized the relationship between vitamins A and D and light exposure, but in recent years other vitamins have also been implicated in photoresponsive biological mechanisms that influence health, well-being, and even evolutionary processes. Interactions between light and vitamins can modify genotype-phenotype relationships across the life cycle, providing a basis for interesting new explanations relevant to wide aspects of human biology. This review exa
