Hippocampal synaptic plasticity and cognition.

Discoveries made over the past 20 years have greatly improved our understanding of how the brain functions. This article focuses on the relation between memory and cellular mechanisms of neuronal and synaptic plasticity in the hippocampus. Several studies indicate that the hippocampal formation is a crucial element of the neurobiological bases of higher cognitive function. Severe damage to the hippocampal formation is known to produce seemingly permanent anterograde amnesia. A generally accepted hypothesis in neurobiology has been that long-lasting activity-dependent changes in the efficacy of

Glutamate receptors in preclinical research on Alzheimer's disease: update on recent advances.

The cognitive and related symptoms of Alzheimer's disease are mainly attributable to synaptic failure. Here we review recent research on how the Alzheimer's disease amyloid ß-protein (Aß) affects glutamate receptors and fast excitatory synaptic transmission and plasticity of that transmission. l-glutamate, the main excitatory neurotransmitter in the brain, has long been implicated in causing NMDA receptor-mediated excitotoxicity leading to neurodegeneration in the late stages of the disease. However there is now extensive evidence that soluble Aß oligomers disrupt synaptic transmission and esp

Plastic abnormalities in experimental Huntington's disease.

Huntington's disease (HD) is a late-onset neurodegenerative disorder that follows an autosomal-dominant pattern of inheritance. In human cases of HD and experimental models of the disease, multiple alterations in neurotransmitters and post-receptor machineries have been described. Dopamine, acetylcholine and glutamate signalling, which usually cooperate in the induction of physiological synaptic plasticity, are all disrupted. Impairment of the induction and reversal of the main forms of neuronal synaptic plasticity influences the computational function of complex neural circuits that mediate e

High-frequency priming stimulation does not enhance the effect of low-frequency rTMS in the treatment of tinnitus.

Based on its ability to reduce the excitability of the cortex locally, low-frequency repetitive transcranial magnetic stimulation (rTMS) has been investigated for the treatment of hyperexcitability disorders such as auditory hallucinations and tinnitus. Results are promising, but characterized by only moderate improvement and a high inter-individual variability. Experimental data from motor cortex stimulation in healthy subjects indicates that the depressant effect of low-frequency rTMS can be enhanced by high-frequency priming stimulation. Here we will investigate whether high-frequency primi

Striatal synaptic plasticity: implications for motor learning and Parkinson's disease.

Changing the strength of synaptic connections between neurons is widely assumed to be the mechanism by which memory traces are encoded and stored in the central nervous system. Plastic changes appear to follow a regional specialization and underlie the specific type of memory mediated by the brain area in which plasticity occurs. Thus, long-term changes occurring at excitatory corticostriatal synapses should be critically involved in motor learning. Indeed, repetitive stimulation of the corticostriatal pathway can cause either a long-lasting increase or an enduring decrease in synaptic strengt

The selective noradrenergic reuptake inhibitor reboxetine restores spatial learning deficits, biochemical changes, and hippocampal synaptic plasticity in an animal model of depression.

Depression is a major psychiatric illness that is associated with cognitive dysfunctions. The underlying mechanism of depression-associated memory impairment is unclear. Previously, we showed altered hippocampal synaptic plasticity in an animal model of depression. Although several antidepressants are beneficial in the treatment of depression, very little is known about the effects of these drugs on depression-associated learning and memory deficits. Prolonged antidepressant treatment might contribute to neuroplastic changes required for clinical outcomes. Accordingly, we evaluated the effect

Posttranslational modifications of AMPA receptors in synaptic function and memory.

AMPA receptors, members of the ionotropic glutamate receptor family, are the principal mediators of fast excitatory synaptic transmission in the brain, and their trafficking to and from synapses is critical for neuronal communication. AMPA receptor trafficking is primarily regulated by posttranslational modifications. These receptors undergo diverse site-specific posttranslational modifications, including phosphorylation, palmitoylation, ubiquitination, S-nitrosylation, N-glycosylation, O-GlcNAcylation, and SUMOylation. Crosstalk among these modifications further adds to the complexity of AMPA

Polymodal activation of the endocannabinoid system in the extended amygdala.

The reason why neurons synthesize more than one endocannabinoid (eCB) and how this is involved in the regulation of synaptic plasticity in a single neuron is not known. We found that 2-arachidonoylglycerol (2-AG) and anandamide mediate different forms of plasticity in the extended amygdala of rats. Dendritic L-type Ca(2+) channels and the subsequent release of 2-AG acting on presynaptic CB1 receptors triggered retrograde short-term depression. Long-term depression was mediated by postsynaptic mGluR5-dependent release of anandamide acting on postsynaptic TRPV1 receptors. In contrast, 2-AG/CB1R-

Abnormal synaptic plasticity in the striatum of mice lacking dopamine D2 receptors.

Dopamine D2 receptors (D2Rs) are of crucial importance in the striatal processing of motor information received from the cortex. Disruption of the D2R gene function in mice results in a severe locomotor impairment. This phenotype has analogies with Parkinson's disease symptoms. D2R-null mice were used to investigate the role of this receptor in the generation of striatal synaptic plasticity. Tetanic stimulation of corticostriatal fibers produced long-term depression (LTD) of EPSPs in slices from wild-type (WT) mice. Strikingly, recordings from D2R-null mice showed the converse: long-term poten

A single episode of neonatal seizures permanently alters glutamatergic synapses.

OBJECTIVE: The contribution of seizures to cognitive changes remains controversial. We tested the hypothesis that a single episode of neonatal seizures (sNS) on rat postnatal day (P) 7 permanently impairs hippocampal-dependent function in mature (P60) rats because of long-lasting changes at the synaptic level. METHODS: sNS was induced with subcutaneously injected kainate on P7. Learning, memory, mossy fiber sprouting, spine density, hippocampal synaptic plasticity, and glutamate receptor expression and subcellular distribution were measured at P60. RESULTS: sNS selectively impaired working m

Cellular mechanisms of burst firing-mediated long-term depression in rat neocortical pyramidal cells.

During wakefulness and sleep, neurons in the neocortex emit action potentials tonically or in rhythmic bursts, respectively. However, the role of synchronized discharge patterns is largely unknown. We have recently shown that pairings of excitatory postsynaptic potentials (EPSPs) and action potential bursts or single spikes lead to long-term depression (burst-LTD) or long-term potentiation, respectively. In this study, we elucidate the cellular mechanisms of burst-LTD and characterize its functional properties. Whole-cell patch-clamp recordings were obtained from layer V pyramidal cells in som

Synapses between parallel fibres and stellate cells express long-term changes in synaptic efficacy in rat cerebellum.

Various forms of synaptic plasticity underlying motor learning have already been well characterized at cerebellar parallel fibre (PF)-Purkinje cell (PC) synapses. Inhibitory interneurones play an important role in controlling the excitability and synchronization of PCs. We have therefore tested the possibility that excitatory synapses between PFs and stellate cells (SCs) are also able to exhibit long-term changes in synaptic efficacy. In the present study, we show that long-term potentiation (LTP) and long-term depression (LTD) were induced at these synapses by a low frequency stimulation prot

Which comes first in fragile X syndrome, dendritic spine dysgenesis or defects in circuit plasticity?

The salient neuropathological defect in fragile X syndrome is the overabundance of immature dendritic spines in cortical pyramidal neurons. This review examines this anatomical synaptic defect in the context of other alterations in synaptic and circuit plasticity in fragile X mice. In theory, abnormal spines could lead to dysfunctional circuits and vice versa, so it is still not clear which problem comes first. Because of the tight structure-function relationships at the synapse, and given the significant overlap between signaling pathways that regulate spine shape/dynamics and long-term synap

Hardware-Algorithm Co-Optimization of Weight-Update Protocols in Oxide-Based Synaptic Transistor Arrays for Neuromorphic Systems.

The transition from single-device characterization to array-level simulation remains a critical challenge in the development of three-terminal synaptic transistors for neuromorphic computing, as most existing simulation studies either extract parameters from a single representative device and apply them uniformly, or rely on weight-update strategies originally designed for two-terminal memristors. Here, we establish an experimentally calibrated behavioral simulation framework based on differential conductance-pair mapping (W = G+ - G-, where G+ and G- denote the conductances of the positive an

Sex-specific interaction effects of Syntaxin 1A coexpression network and childhood trauma on adult depressive symptoms.

BACKGROUND: Disruptions in synaptic function and exposure to early life trauma are both implicated in the development of depression, though the underlying mechanisms remain poorly understood. METHODS: To investigate this relationship, we developed an expression-based polygenic score (ePRS) that captures individual variations in the expression of genes co-expressed with the synaptic protein Syntaxin 1A (STX1A) in the prefrontal cortex (PFC). This polygenic score allows us to explore the impact of variations on synaptic function and childhood trauma on the susceptibility to depressive symptoms

Ecdysterone and High-Intensity Interval Training Mitigate Alzheimer's Pathology in Rats: Impacts on Depression, Synaptic Plasticity, and Neuroinflammation.

The study focuses on the role of neuroinflammation and impaired synaptic plasticity in the progression of Alzheimer's disease caused by amyloid-beta accumulation. It examines the potential therapeutic effects of Ecdysterone, known for its anti-inflammatory and antioxidant properties, and high-intensity interval training, which may also support brain health. The primary goal is to assess how Ecdy and HIIT, alone or combined, influence depressive-like behavior, synaptic function, inflammation, and Aβ buildup in a rat model of AD. Ten days after Aβ administration, treatments began with Ecdy (10 m

Age-Dependent Contributions of NMDA Receptors and L-Type Calcium Channels to Long-Term Depression in the Piriform Cortex.

In the hippocampus, the contributions of N-methyl-D-aspartate receptors (NMDARs) and L-type calcium channels (LTCCs) to neuronal transmission and synaptic plasticity change with aging, underlying calcium dysregulation and cognitive dysfunction. However, the relative contributions of NMDARs and LTCCs in other learning encoding structures during aging are not known. The piriform cortex (PC) plays a significant role in odor associative memories, and like the hippocampus, exhibits forms of long-term synaptic plasticity. Here, we investigated the expression and contribution of NMDARs and LTCCs in l

Learning as a model for neural plasticity in major depression.

BACKGROUND: The neuroplasticity hypothesis of depression proposes that a dysfunction of neural plasticity-the basic ability of living organisms to adapt their neural function and structure to external and internal cues-might represent a final common pathway underlying the biological and clinical characteristics of the disorder. This study examined learning and memory as correlates of long-term synaptic plasticity in humans to further test the neuroplasticity hypothesis of depression. METHODS: Learning in three tasks, for which memory consolidation has been shown to depend on local synaptic re
