Targeting ACE2 with luteolin improves imbalance of lung epithelial sodium transport under Omicron infection.

BACKGROUND: As a progressive pathological process, pulmonary edema driven by Omicron infection is closely related to the imbalance of epithelial sodium channel (ENaC)-regulated sodium transport. Luteolin (Lut), a flavonoid compound, exerts therapeutic effects against infectious diseases, yet its detailed involvement in fluid homeostasis during pulmonary edema remains unclear. PURPOSE: This study investigated the effects of Lut against pulmonary edema induced by Omicron infection, and elucidated the associated molecular mechanisms. METHODS: An Omicron S pseudovirus infected mouse model was es

Luteolin use in Integrated Stress Response: insight from a case of EIF2AK2-related dystonia.

This study presents the first reported case of a 3-year-old child with EIF2AK2-related dystonia treated with adjunctive luteolin supplementation. EIF2AK2-related dystonia, characterized by exacerbations during infections, is associated with disruptions in the integrated stress response (ISR). The ISR, a cellular signaling pathway activated in response to stress, culminates in the phosphorylation of eIF2α, which modulates protein synthesis and can induce cell death. Pathogenic variants in EIF2AK2 disrupt this pathway, contributing to the development of dystonia. Luteolin, a flavonoid possessing

Luteolin increases baseline apneas and downregulates brainstem HIF in normoxia and chronic intermittent hypoxia.

Hypoxia-inducible factor (HIF) is a key regulator of cellular adaptation to reduced oxygen availability. Chronic intermittent hypoxia (CIH), characterized by recurrent cycles of hypoxia and reoxygenation, promotes oxidative stress and activates hypoxia-sensitive signaling pathways involved in ventilatory control. Luteolin-7-O-glucoside (luteolin), a flavonoid with antioxidant and anti-inflammatory properties, has been proposed as a potential modulator of HIF activity. This study evaluated the effects of a 7-day luteolin treatment on the ventilatory pattern and brainstem HIF expression in mice

Mechanistic Evaluation of Luteolin in Wound Healing of Diabetic Foot Ulcer via Nrf2 Signaling.

Diabetic foot ulcers (DFUs) are a leading cause of lower-limb amputation, with oxidative stress and ferroptosis playing critical roles in impaired wound healing. This study aimed to evaluate the therapeutic potential of luteolin (LU), a naturally occurring plant-derived flavonoid, in DFU repair and to elucidate its underlying molecular mechanisms. A DFU rat model was established using a high-fat/high-glucose diet combined with streptozotocin injection, followed by full-thickness skin wounding. LU was topically administered at different concentrations. Wound closure was monitored macroscopicall

Luteolin Inhibits NLRP3 Inflammasome Activation to Ameliorate DSS-Induced Colitis by Regulating AMPK Signalling.

Luteolin alleviates DSS-induced ulcerative colitis in mice by targeting the NLRP3 inflammasome, as it shows no effect in NLRP3-/- mice. It inhibits NLRP3 activation and IL-1β secretion in macrophages by reducing ROS, mtROS and calcium levels via AMPK binding and signalling. Metabolomic changes suggest lipid metabolism involvement. Luteolin represents a promising NLRP3-targeted therapeutic candidate for UC.

Application of PVA hydrogel loaded with luteolin nanoparticles in anti EMT treatment after GBM.

BACKGROUND: Glioblastoma multiforme (GBM) is the most common and worst-prognosed primary malignant tumor in the central nervous system. Epithelial mesenchymal transition (EMT) is an important cause of postoperative invasion and recurrence in GBM. Due to the presence of the blood-brain barrier, local therapy may be the preferred route for treatment of GBM after surgery. METHODS: Our objective is to develop an anti- EMT functional hydrogel for post-GBM surgery tamponade. We fabricated self-assembled luteolin nanoparticles (LU NPs) via the solvent evaporation method, and characterized their morp

Targeted Inhibition of CD74+ Macrophages by Luteolin via CEBPB/P65 Signaling Ameliorates Osteoarthritis Progression.

Synovial inflammation represents a hallmark pathological process in osteoarthritis (OA), yet the cellular drivers orchestrating this response remain incompletely defined. Through single-cell transcriptomic profiling of human OA synovial tissues, a distinct subset of CD74⁺ macrophages is identified that displayed robust pro-inflammatory transcriptional signatures, underscoring the pivotal role of macrophages in shaping the inflammatory microenvironment. To explore therapeutic opportunities, computational ligand-target interaction analysis predicted luteolin as a high-affinity binder of CD74. Me

Luteolin improves vasoconstriction function and survival of septic mice via AMPK/NF-κB pathway.

Septic shock, the leading cause of death in sepsis, is related to vasoconstriction dysfunction. To investigate the effects of Luteolin (LTL), a flavonoid polyphenol compound, on vasoconstriction dysfunction in septic mice and the underlying mechanism, cecal ligation and puncture (CLP) surgery was performed on wild-type C57BL/6 mice to induce septic shock. Mice were intraperitoneally injected with 0.2 mg/kg LTL within 10 min after CLP surgery with or without 20 mg/kg Compound C (AMPK inhibitor) (CC) 1 h before CLP surgery, and re-administrated every 12 h. Luteolin Attenuates Cardiac Ischemia/Reperfusion Injury in Diabetic Rats by Modulating Nrf2 Antioxidative Function.

Luteolin has been reported to attenuate ischemia/reperfusion (I/R) injury in the diabetic heart through endothelial nitric oxide synthase- (eNOS-) related antioxidative response. Though the nuclear factor erythroid 2-related factor 2 (Nrf2) is regarded as a key endogenous factor to reduce diabetic oxidative stress, whether luteolin reduces cardiac I/R injury in the diabetic heart via enhancing Nrf2 function needs to be clarified. We hypothesized that pretreatment with luteolin could alleviate cardiac I/R injury in the diabetic heart by affecting the eNOS/Nrf2 signaling pathway. The diabetic ra

Luteolin-Mediated Inhibition of Hepatic Stellate Cell Activation via Suppression of the STAT3 Pathway.

Hepatic stellate cell (HSC) activation is responsible for hepatic fibrogenesis and is associated with an overexpression of transcription 3 (STAT3). Luteolin, a common dietary flavonoid with potent anti-inflammatory properties, has previously demonstrated antifibrogenic properties in HSCs but the mechanism has not been fully elucidated. Activated human and rat hepatic stellate cell lines LX-2 and HSC-T6 were used to study the effects of luteolin on HSCs. Cellular proteins were determined by western blot and immunofluorescence. Cell proliferation was assessed with Alamar Blue assay. Luteolin sig

Protective role of luteolin against cognitive dysfunction induced by chronic cerebral hypoperfusion in rats.

Chronic cerebral hypoperfusion, a mild ischemic condition, is associated with the cognitive deficits of Alzheimer's disease (AD). Luteolin, a polyphenolic compound found in foods of plant origin, belonging to the flavone subclass of flavonoids, has been shown to possess antioxidant, anti-inflammatory and antitumorigenic properties. In the present study, the effects of luteolin on chronic cerebral hypoperfusion-associated neurocognitive pathologies were investigated by using rats with permanent bilateral common carotid artery occlusion, a rat model of chronic cerebral hypoperfusion. As expected

Luteolin-Loaded WPI/Pueraria lobata Amylopectin Composite Gel Improves Adipocyte Thermogenesis and Insulin Sensitivity.

Adipose tissue metabolic dysfunction plays a central role in the development of obesity and related metabolic disease. Luteolin is a natural flavonoid with antioxidant, anti-inflammatory, antidiabetic, and antiobesity activities, while it is limited by poor solubility, bioavailability, and stability. This study developed a whey protein isolate (WPI)-Pueraria lobata amylopectin (PLA) composite gel, characterized by a dense 3D network and enhanced mechanical strength. The WPI-PLA gel efficiently encapsulated luteolin in a stable amorphous form and exhibited uniform particle size and excellent co

Neuroprotective effects of luteolin against aluminum-induced brain injury via chelation and antioxidant pathways.

Aluminum (Al) is a widespread environmental neurotoxin linked to several neurodegenerative conditions through oxidative stress, inflammation, and apoptosis. Because it is present in food, water, pharmaceuticals, and consumer products, chronic exposure is nearly unavoidable and poses a public health concern. Luteolin (LUT), a natural flavone with antioxidant, anti-inflammatory, and metal-chelating properties, has emerged as a potential neuroprotective agent. This study investigated the protective effects of LUT against aluminum chloride (AlCl3)-induced neurotoxicity in rats. Sprague-Dawley rats
