Glycosyltransferase-Like Toxin of Burkholderia gladioli Strain NGJ1 Is a Potent Antifungal Protein with Potential for Control of Sheath Blight Disease in Rice.

Sheath blight disease caused by the fungal pathogen Rhizoctonia solani poses a significant challenge for sustainable rice cultivation. It is important to develop environmentally friendly measures for its control. Previously, a rice-associated Burkholderia gladioli strain NGJ1 was shown to exhibit mycophagous and antifungal activity on R. solani. Here, we report that a B. gladioli glycosyltransferase-like 1 (BGT1) protein with a canonical D×D (aspartic acid × aspartic acid) motif that is homologous to the glycosyltransferase toxin of different bacteria is encoded in the antibacterial type VI se

Enhancing defense against rice blast disease: Unveiling the role of leaf endophytic firmicutes in antifungal antibiosis and induced systemic resistance.

Rice remains the primary staple for more than half of the world's population, yet its cultivation faces numerous challenges, including both biotic and abiotic stresses. One significant obstacle is the prevalence of rice blast disease, which substantially diminishes productivity and increases cultivation costs due to frequent fungicide applications. Consequently, the presence of fungicide residues in rice raises concerns about compliance with international maximum residue limits (MRLs). While host resistance has proven effective, it often remains vulnerable to new variants of the Magnaporthe or

Disruption of OsSEC3A increases the content of salicylic acid and induces plant defense responses in rice.

The exocyst, an evolutionarily conserved octameric protein complex involved in exocytosis, has been reported to be involved in diverse aspects of morphogenesis in Arabidopsis. However, the molecular functions of such exocytotic molecules in rice are poorly understood. Here, we examined the molecular function of OsSEC3A, an important subunit of the exocyst complex in rice. The OsSEC3A gene is expressed in various organs, and OsSEC3A has the potential ability to participate in the exocyst complex by interacting with several other exocyst subunits. Disruption of OsSEC3A by CRISPR/Cas9 (clustered

Inducible biosynthesis and immune function of the systemic acquired resistance inducer N-hydroxypipecolic acid in monocotyledonous and dicotyledonous plants.

Recent work has provided evidence for the occurrence of N-hydroxypipecolic acid (NHP) in Arabidopsis thaliana, characterized its pathogen-inducible biosynthesis by a three-step metabolic sequence from l-lysine, and established a central role for NHP in the regulation of systemic acquired resistance. Here, we show that NHP is biosynthesized in several other plant species in response to microbial attack, generally together with its direct metabolic precursor pipecolic acid and the phenolic immune signal salicylic acid. For example, NHP accumulates locally in inoculated leaves and systemically in

Cyanide, a coproduct of plant hormone ethylene biosynthesis, contributes to the resistance of rice to blast fungus.

Rice (Oryza sativa) plants carrying the Pi-i resistance gene to blast fungus Magnaporthe oryzae restrict invaded fungus in infected tissue via hypersensitive reaction or response (HR), which is accompanied by rapid ethylene production and formation of small HR lesions. Ethylene biosynthesis has been implicated to be important for blast resistance; however, the individual roles of ethylene and cyanide, which are produced from the precursor 1-aminocyclopropane-1-carboxylic acid, remain unevaluated. In this study, we found that Pi-i-mediated resistance was compromised in transgenic rice lines, in

Plant Spatio-Temporal Integration Network (PSTN): a framework for dynamic single-cell and spatial transcriptomics in fungal-plant interactions.

MOTIVATION: Rice blast caused by Magnaporthe oryzae (M. oryzae) is a dynamic cross-kingdom infection process involving rapid transcriptional reprogramming and spatial tissue remodeling across successive stages. Although single-cell RNA sequencing (scRNA-seq) and spatial transcriptomics (ST) provide complementary information, most integration methods analyze stages independently and ignore temporal continuity. RESULTS: We developed the Plant Spatio-Temporal Integration Network (PSTN), which jointly learns stage-specific cell-to-space mappings through expression reconstruction and bidirectional

A hypermorphic allele of CNGC confers broad-spectrum blast resistance through ectopic calcium influx-induced cell death in rice.

Rice blast, caused by Magnaporthe oryzae, is one of the most destructive diseases threatening global rice production. Race-independent broad-spectrum resistance (BSR) holds the ideal strategy for durable rice blast management, yet its molecular mechanisms remain largely obscure. Here, we identified and characterized elicitor-triggered cell death 1 (etd1), a rice lesion mimic mutant that displays spontaneous cell death and hypersensitive response (HR)-like necrosis upon challenge with M. oryzae elicitors. Genetic analysis reveals that etd1 encodes a hypermorphic haplotype of rice Cyclic Nucleot

Anti-phytopathogenic activity of sporothriolide, a metabolite from endophyte Nodulisporium sp. A21 in Ginkgo biloba.

Phytopathogenic fungi such as Rhizoctonia solani and Sclerotinia sclerotiorum caused multiple plant diseases resulting in severe loss of crop production. Increasing documents endorsed that endophytes are a striking resource pool for numerous metabolites with various bioactivities such as anti-fungal. Here we reported the characterization and anti-phytopathogenic activity of sporothriolide, a metabolite produced by Nodulisporium sp. A21-an endophytic fungus in the leaves of Ginkgo biloba. Among the total twenty-five endophytic fungi isolated from the healthy leaves of G. biloba, the fermentatio

Aspergillus fumigatus arp1 modulates conidial pigmentation and complement deposition.

Aspergillus fumigatus is an important pathogen causing invasive pulmonary aspergillosis in immunocompromised patients. The fungus propagates by conidia, which are the infectious structures inhaled by the human host. Opsonophagocytosis is thought to contribute to clearance of the inhaled conidia, a process that is facilitated by complement deposition on conidial surfaces. We now show that conidial colour mutants exhibit significant increases in C3 binding capacity compared with wild type. A reddish-pink mutation that led to enhanced C3 binding was complemented by a cosmid clone. A 3.3 kb DNA fr

Fungal effector-based strategies for sustainable rice disease control: Transgenic expression and engineered biocontrol approaches deliver broad-spectrum resistance.

Rice is one of the most important food crops in the world and it is prone to attack by many diseases, such as the rice blast, sheath blight, bacterial leaf blight, and so on. These diseases represent the main constraints in rice production, threatening food security and safety. Here, new control strategies against these major rice diseases have been developed either by heterologous expression of a pathogen effector UvScd1 from false smut fungus Ustilaginoidea virens or by spraying the engineered biocontrol agent Bacillus subtilis secreting the effector UvScd1. Compared to the wild-type rice Zh

Phosphorylation of OsTGA5 by casein kinase II compromises its suppression of defense-related gene transcription in rice.

Plants manage the high cost of immunity activation by suppressing the expression of defense genes during normal growth and rapidly switching them on upon pathogen invasion. TGAs are key transcription factors controlling the expression of defense genes. However, how TGAs function, especially in monocot plants like rice with continuously high levels of endogenous salicylic acid (SA) remains elusive. In this study, we characterized the role of OsTGA5 as a negative regulator of rice resistance against blast fungus by transcriptionally repressing the expression of various defense-related genes. Induction of chlorosis, ROs generation and cell death by a toxin isolated from Pyricularia oryzae.

The ethyl acetate extract of the conidia germination fluid from an Avena isolate (Br58) of Pyricularia oryzae had chlorosis-inducing activity on oat leaf segments. The same activity was also present in the acetone extract of an oatmeal agar culture of Br58. Fungal cultures were used for a large-scale preparation. A series of acetone and ethyl acetate extraction monitored by chromatography was used to isolate an active fraction. The active principle was purified by HPLC. We show by NMR and LC/MS that the toxin was an oxidized C18 unsaturated fatty acid named Mag-toxin. Mag-toxin induced chloros

Seed priming with calcium chloride enhances stress tolerance in rice seedlings.

Calcium is a crucial second messenger in plant cells and contributes to plant resistance against biotic and abiotic stress. Plant defense priming with natural or synthetic compounds leads to quicker and stronger resistance responses. However, whether pretreatment of plant seeds with calcium could improve their resistance to stress remains poorly understood. In this study, we showed that rice seedlings grown from calcium chloride (CaCl2)-pretreated seeds displayed enhanced resistance to the rice blast fungus Magnaporthe oryzae and the rice bacterial pathogen Xanthomonas oryzae pv. Oryzae (Xoo).

Identification and characterization of antifungal compounds using a Saccharomyces cerevisiae reporter bioassay.

New antifungal drugs are urgently needed due to the currently limited selection, the emergence of drug resistance, and the toxicity of several commonly used drugs. To identify drug leads, we screened small molecules using a Saccharomyces cerevisiae reporter bioassay in which S. cerevisiae heterologously expresses Hik1, a group III hybrid histidine kinase (HHK) from Magnaporthe grisea. Group III HHKs are integral in fungal cell physiology, and highly conserved throughout this kingdom; they are absent in mammals, making them an attractive drug target. Our screen identified compounds 13 and 33, w

Assessment of inhibitory potentiality of natural compounds against worrisome rice blast fungus.

Rice blast, a severe fungal disease, is a substantial threat to global food security, particularly in rice-oriented areas. The Magnaporthe oryzae fungus is increasingly resistant and fast developing in nature. However, chemical fungicides are not only detrimental to the environment but eventually also lose their efficiency. To Tackle this issue, we used an in silico based strategy to identify plant metabolites as bio-fungicides to combat rice blast. Therefore, we screened a total of 56 antifungal natural compounds for the ability to inhibit fungal development through the targeted inhibition of

The tandem EH domains of End3 cooperate to interact with dual XPF motifs of Sla1 for the connection of early and late stages in fungal endocytosis.

Clathrin-mediated endocytosis (CME) is imperative for physiological processes in eukaryotic cells. In fungi, the Pan1/End3/Sla1 complex controls the transition between early and late stages of CME. Although it is acknowledged that End3 uses its N-terminal to interact with the C-terminal of Sla1, detailed mechanism remains obscure. Magnaporthe oryzae, the pathogenic fungus of rice, cause blast disease that threatens rice production worldwide. Here we report the detailed interaction mechanism between End3 and Sla1 of M. oryzae, i.e. MoEnd3 and MoSla1. The two EH domains of MoEnd3 (MoEnd3-EH1 and

Rice OsVAMP714, a membrane-trafficking protein localized to the chloroplast and vacuolar membrane, is involved in resistance to rice blast disease.

Membrane trafficking plays pivotal roles in many cellular processes including plant immunity. Here, we report the characterization of OsVAMP714, an intracellular SNARE protein, focusing on its role in resistance to rice blast disease caused by the fungal pathogen Magnaporthe oryzae. Disease resistance tests using OsVAMP714 knockdown and overexpressing rice plants demonstrated the involvement of OsVAMP714 in blast resistance. The overexpression of OsVAMP7111, whose product is highly homologous to OsVAMP714, did not enhance blast resistance to rice, implying a potential specificity of OsVAMP714

FvBck1, a component of cell wall integrity MAP kinase pathway, is required for virulence and oxidative stress response in sugarcane Pokkah Boeng pathogen.

Fusarium verticillioides (formerly F. moniliforme) is suggested as one of the causal agents of Pokkah Boeng, a serious disease of sugarcane worldwide. Currently, detailed molecular and physiological mechanism of pathogenesis is unknown. In this study, we focused on cell wall integrity MAPK pathway as one of the potential signaling mechanisms associated with Pokkah Boeng pathogenesis. We identified FvBCK1 gene that encodes a MAP kinase kinase kinase homolog and determined that it is not only required for growth, micro- and macro-conidia production, and cell wall integrity but also for response
