Modeling, molecular docking, probing catalytic binding mode of acetyl-CoA malate synthase G in Brucella melitensis 16M.

There are enormous evidences and previous reports standpoint that the enzyme of glyoxylate pathway malate synthase G (MSG) is a potential virulence factor in several pathogenic organisms, including Brucella melitensis 16M. Where the lack of crystal structures for best candidate proteins like MSG of B. melitensis 16M creates big lacuna to understand the molecular pathogenesis of brucellosis. In the present study, we have constructed a 3-D structure of MSG of Brucella melitensis 16M in MODELLER with the help of crystal structure of Mycobacterium tuberculosis malate synthase (PDB ID: 2GQ3) as tem

Compression-induced metabolic adaptation drives confined tumor cell migration and distant metastasis via malate-dependent microtubule reinforcement.

Metastasis, responsible for > 90% of cancer-related mortality, represents the most lethal yet least mechanistically understood phase of cancer progression. A critical bottleneck is tumor cell migration through physically confined environments, including dense extracellular matrix, narrow capillaries and endothelial gaps. Although tumor cells reprogram their metabolism to facilitate cancer progression, it remains unclear how specific metabolic adaptations enable them to overcome the unique physical challenges posed by these confined spaces, thereby promoting distant metastasis. We conducted a C

Malate reduced kidney injury molecule (KIM-1) expression and selectively upregulated the renal nitric oxide production in obstructive nephropathy.

BACKGROUND: Malate, the tricarboxylic acid (TCA) cycle intermediary, upregulates renal nitric oxide (NO) signaling, and NO is renoprotective in nephropathy. OBJECTIVES: This study explored the hypothesis that malate could increase renal NO and decrease renal injury and fibrotic markers in obstructive nephropathy. METHODS: Kidney injury was induced in rats via unilateral surgical ligation of the ureter, there after, rats were treated with malate (600 mg/kg, p.o.) for ten days. Urine was collected on days 0, 4, 7 and 10. Urinary sodium excretion was also determined. Western blot and biochemica

Effect of geranylated dihydrochalcone from Artocarpus altilis leaves extract on Plasmodium falciparum ultrastructural changes and mitochondrial malate: Quinone oxidoreductase.

Nearly half of the world's population is at risk of being infected by Plasmodium falciparum, the pathogen of malaria. Increasing resistance to common antimalarial drugs has encouraged investigations to find compounds with different scaffolds. Extracts of Artocarpus altilis leaves have previously been reported to exhibit in vitro antimalarial activity against P. falciparum and in vivo activity against P. berghei. Despite these initial promising results, the active compound from A. altilis is yet to be identified. Here, we have identified 2-geranyl-2', 4', 3, 4-tetrahydroxy-dihydrochalcone (1) f

A targeted proteomic multiplex CSF assay identifies increased malate dehydrogenase and other neurodegenerative biomarkers in individuals with Alzheimer's disease pathology.

Alzheimer's disease (AD) is the most common cause of dementia. Biomarkers are required to identify individuals in the preclinical phase, explain phenotypic diversity, measure progression and estimate prognosis. The development of assays to validate candidate biomarkers is costly and time-consuming. Targeted proteomics is an attractive means of quantifying novel proteins in cerebrospinal and other fluids, and has potential to help overcome this bottleneck in biomarker development. We used a previously validated multiplexed 10-min, targeted proteomic assay to assess 54 candidate cerebrospinal fl

Human MALAT-1 long non-coding RNA is overexpressed in cervical cancer metastasis and promotes cell proliferation, invasion and migration.

Источник пометил публикацию как отозванную

PURPOSE: Long non-coding RNAs (lncRNAs) have been shown to play a crucial role in the development as well as the prognosis of various human cancers, particularly in human metastasis associated lung adenocarcinoma transcript-1 (MALAT-1), which has been shown to be upregulated in some tumors. However, the role of MALAT-1 in cervical cancer (CC) is yet to be elucidated. This study aimed to establish a correlation between MALAT-1 and its role in CC progression and prognosis. METHODS: The expression of MALAT-1 was examined in tissue samples of 30 CC patients and was compared with the adjacent non-

Integrated Multi-Omics and Molecular Docking Analysis of Malate Degradation by Lactiplantibacillus plantarum S4 During Sea Buckthorn Juice Fermentation.

Sea buckthorn juice (SBJ) is nutritionally valuable, but its high malic acid content contributes to excessive sourness that limits consumer acceptance. In this study, the deacidification phenotypes of two lactic acid bacterial isolates, Lactiplantibacillus plantarum S4 and Lacticaseibacillus paracasei YJ2, were compared using fermentation phenotyping, sensory evaluation, enzyme assays, untargeted metabolomics, transcriptomics, and molecular docking. After 96 h of fermentation, S4 degraded 91.98% of malic acid, compared with 69.50% for YJ2, and exhibited higher viable-cell counts and overall se

TCA cycle-related lactate and malate dehydrogenases differentially modulate fungal development, immune evasion and pathogenicity in the entomopathogen Beauveria bassiana.

The tricarboxylic acid (TCA) cycle plays a pivotal role in fungal physiological processes. In this study, two TCA-cycle related enzyme were functionally analyzed in a model entomopathogenic fungus Beauveria bassiana, including NAD-dependent lactate dehydrogenase (BbLdh) and malate dehydrogenase (BbMdh1 and 2). Domain annotation indicated all these enzymes contained an Ldh_1 domain. Functional analyses indicated that BbLdh and BbMdh2 had differential contributions to fungal growth, development, stress tolerance, virulence, and mycosis. Notably, BbLdh played a more important role in fungal inter

Effects of 3 days of citrulline malate supplementation on short-duration repeated sprint running performance in male team sport athletes.

Citrulline malate (CM) is purported to be an ergogenic aid during various types of exercise performance. However, the effects of CM on repeated sprint performance (RSP) are under-explored. In a placebo-controlled, double-blind, counterbalanced cross-over design, male university-level team sport athletes (n = 13) performed two familiarization trials, after which CM or placebo (PLA) (8 × 1 g tablets each day) were taken on the 2 days prior to, and with breakfast on the morning of, each main experimental trial. The main experimental trials employed a RSP protocol consisting of 10 repetitions of 4

Magnesium malate-modified calcium phosphate bone cement promotes the repair of vertebral bone defects in minipigs via regulating CGRP.

Active artificial bone substitutes are crucial in bone repair and reconstruction. Calcium phosphate bone cement (CPC) is known for its biocompatibility, degradability, and ability to fill various shaped bone defects. However, its low osteoinductive capacity limits bone regeneration applications. Effectively integrating osteoinductive magnesium ions with CPC remains a challenge. Herein, we developed magnesium malate-modified CPC (MCPC). Incorporating 5% magnesium malate significantly enhances the compressive strength of CPC to (6.18 ± 0.49) MPa, reduces setting time and improves disintegration

LncRNA AC020978 facilitates non-small cell lung cancer progression by interacting with malate dehydrogenase 2 and activating the AKT pathway.

Long non-coding RNA AC020978 (lncRNA AC020978) is an oncogenic regulator of non-small cell lung cancer (NSCLC). However, the function of AC020978 in regulating NSCLC metastasis and the potential molecular mechanism remains largely unknown. In this study, we evaluated the expression levels of AC020978 in a series of NSCLC tissues using FISH assays and found that higher AC020978 expression levels were closely associated with metastasis and unfavorable prognosis. Functional studies showed that AC020978 promoted NSCLC migration and invasion both in vitro and in vivo. Further investigation demonstr

Citrulline malate supplementation does not improve German Volume Training performance or reduce muscle soreness in moderately trained males and females.

BACKGROUND: Use of supplements to aid performance is common practice amongst recreationally active individuals, including those without a sufficient evidence base. This investigation sought to assess whether acute supplementation with 8 g of citrulline malate (CM) (1.11: 1 ratio) would improve anaerobic performance. METHODS: A randomised double blind placebo control trial was employed, using a counterbalanced design. We recruited recreationally active men and women to take part in an isokinetic chair protocol, based on German Volume Training (GVT) whereby participants attempted to perform 10

Malate-aspartate shuttle inactivation drives pulmonary hypertension through glucose carbon flux rewiring and α-ketoglutarate-dependent epigenetic remodeling.

Pulmonary hypertension (PH) is characterized by progressive pulmonary vascular remodeling, primarily driven by the hyperproliferation of pulmonary artery smooth muscle cells (PASMCs) and their resistance to apoptosis. Metabolic reprogramming, particularly enhanced glycolysis, has been proposed to be involved in PASMCs activation. However, the implicated mechanisms remain incompletely understood. Here, using in-vitro cultured PASMCs and in-vivo PH models, we showed that malate-aspartate shuttle (MAS), a critical system connecting glycolysis to oxidative phosphorylation, was significantly inhibi
