Principles of treatment for mast cell tumors.

Mast cell tumors (MCT) are the most common malignant cutaneous tumors that occur in dogs. They are most commonly found on the trunk, accounting for approximately 50% to 60% of all sites. MCTs associated with the limbs account for approximately 25% of all sites. Cutaneous MCTs have a wide variety of clinical appearances. Histologic grade is the most consistent prognostic factor available for dogs. MCTs located at 'nail bed' (subungual), inguinal/preputial area, and any mucocutaneous area like perineum or oral cavity carry a guarded prognosis and tend to metastasize. MCTs usually exfoliate well

Circulating microRNA-214 and -126 as potential biomarkers for canine neoplastic disease.

Circulating microRNAs in the blood may provide diagnostic and prognostic information about canine neoplastic diseases, and their profiles may be conserved between human and canine species. We performed RT-qPCR to obtain the profiles of circulating plasma microRNA-214 and -126 in total 181 cases of canine neoplastic diseases and healthy controls. MicroRNA-214 levels were high in 2 epithelial tumours (thyroid and mammary carcinomas) and 4 non-epithelial tumours (osteosarcoma, histiocytic sarcoma, chondrosarcoma, and hemangiosarcoma). In contrast, microRNA-126 levels were high in 6 epithelial tum

The Use of Biologic Therapeutic Regimens in Cutaneous and Systemic Mastocytosis: A Scoping Review.

BACKGROUND: Mastocytoses, a category of diseases characterized by the clonal proliferation of mast cells and the release of their mediators, can be broadly classified as systemic mastocytosis (SM), cutaneous mastocytosis (CM), or mast cell sarcoma (MCS), with treatment options and prognoses varying based on subtype. Advancements in biologics and genetic testing have increased the variety of therapeutic options for mastocytoses. This paper characterizes the therapeutic landscape for mastocytoses with a focus on biologics and other novel medications. METHODS: A comprehensive literature search o

Systemic mastocytosis in a goat.

Systemic mastocytosis was diagnosed in a 4-year-old, female Nubian goat. Clinically, the animal was depressed and had severe macrocytic hypochromic anemia and leukopenia. Postmortem examination revealed neoplastic mast cells invading the heart, lung, liver, spleen, lymph nodes, and bone marrow. Eosinophils were frequently admixed with infiltrating mast cells in all organs. Using routine light microscopy, histochemistry, and transmission electron microscopy, metachromatic and periodic acid-Schiff-positive granules were identified within the cytoplasm of neoplastic mast cells. Erythrophagocytosi

Primary hematological malignancy of the uterine cervix: A case report.

The present case report investigated the clinical characteristics of primary hematological malignancy of the uterine cervixin five patients. Among the five patients, three patients were diagnosed with non-Hodgkin's lymphoma (NHL) and two patients with myeloid sarcoma (MS) of the uterine cervix. Biopsies of the uterine cervices demonstrated the involvement of lymphoid cells or myeloid blasts cells. Immunohistochemical staining demonstrated the expression of B-lymphoid and myeloid lineage markers. The associated lysozyme, myeloperoxidase and mast/stem cell growth factor receptor (CD117) markers

The Spectrum of Aggressive Mastocytosis: A Workshop Report and Literature Review.

Most cases of mastocytosis are indolent, usually cutaneous mastocytosis or indolent systemic mastocytosis (SM). Aggressive mast cell (MC) diseases are very rare and often fatal. They can develop de novo or due to progression of indolent forms and can present in different ways; either as MC sarcoma or as advanced SM which includes aggressive SM, MC leukemia, and SM with an associated hematological neoplasm. This review will describe these different aggressive forms of mastocytosis, illustrated by cases submitted to the workshop of the 18th Meeting of the European Association for Haematopatholog

Breed-predispositions to cancer in pedigree dogs.

Cancer is a common problem in dogs and although all breeds of dog and crossbred dogs may be affected, it is notable that some breeds of pedigree dogs appear to be at increased risk of certain types of cancer suggesting underlying genetic predisposition to cancer susceptibility. Although the aetiology of most cancers is likely to be multifactorial, the limited genetic diversity seen in purebred dogs facilitates genetic linkage or association studies on relatively small populations as compared to humans, and by using newly developed resources, genome-wide association studies in dog breeds are pr

Metastatic cancer of unknown primary in 21 dogs.

The aim of this retrospective study was to describe clinical features, treatment and outcome of 21 dogs with metastatic cancer of unknown primary (MCUP), a biopsy-proven malignancy being diagnosed at a metastatic stage, in which the anatomical origin of the primary tumour cannot be detected. All dogs underwent total-body computed tomography. Signalment, type and duration of clinical signs, metastasis site, pathology results, treatment and outcome were recorded. Carcinoma was the most common diagnosis (57.1%), followed by sarcoma, melanoma and mast cell tumour. The median number of disease site

Selection and isolation of a new variant of DBA/2 mastocytoma P 815 X 2.

A tumor model was developed in DBA/2 mice for studying the progression of the mastocytoma P 815 X 2 tumor. Tumor cells obtained from ascites were grown in vitro. The number of cells derived from a single clone was increased by in vitro culture. Cells were then injected either i.v. or i.p. into DBA/2 mice. Large volumes of tumor ascites were observed after i.p. but not i.v. injection. The latter led to tumor growth at multiple sites, especially in the liver. Mastocytoma cells were released from liver tissue and then injected i.p. into other recipients. For cloning of liver invading tumor cells,

[Sequence complexity of transcribed unique DNA sequences in genome of mouse P815 mastocytoma cells (author's transl)].

The sequence complexity of nuclear RNA from mouse liver, mouse spleen and highly malignant P815 mastocytoma was measured by nRNA driven hybridization to unique DNA sequences of P815 cells. The unique DNA sequences represent 63% of the total nuclear DNA of P815 cells and their availibility in hybridization experiments was found to be 76%. Of these sequences 7.8% formed hybrids with nuclear RNA of this cell, about 11.5% with mouse spleen and about 14.5% with mouse liver nuclear RNA. Assuming an asymmetrical transcription, the complexities of these transcripts are 2.8 X 10(8) nucleotides for mous

Involvement of the larynx by hemopoietic neoplasms. An investigation of autopsy cases and review of the literature.

Involvement of the larynx by hemopoietic tumors is generally considered a rare event and little is known about the associated clinicopathologic features. Laryngeal tissue removed at autopsy from 14 patients with known disseminated hematologic malignancies and at operation from one patient with multicentric malignant lymphoma of low-grade malignancy (MALToma) of the head and neck region was investigated. A systematic survey of the main clinicopathologic features of the published cases of hemopoietic tumors with laryngeal involvement was also performed. Primary involvement of the larynx by hemop

The morphology, immunohistochemistry, and incidence of hematopoietic neoplasms in mice and rats.

Hematopoietic neoplasms in the rodent may be classified into lymphoid or nonlymphoid neoplasms. Lymphoid neoplasms include the following morphologic types: follicular center cell, lymphoblast (lymphocytic), immunoblast, plasma cell, and large granular lymphocyte (LGL). Nonlymphoid hematopoietic neoplasms include histiocytic sarcoma, granulocytic leukemia, erythroid leukemia, and mast cell tumors. Most types of hematopoietic neoplasms, exclusive of LGL lymphoma (leukemia), are more common in mice than in rats. Specific strains of mice have a hematopoietic tumor incidence of more than 50% in age

R634W KIT Mutation in an Adult With Systemic Mastocytosis.

Mastocytosis is a clonal neoplasm with the potential to affect various organs within the body. It can range in clinical severity from benign to extremely aggressive. Mastocytosis can be separated into cutaneous, systemic, and leukemic forms, as well as mast-cell sarcoma and extracutaneous mastocytoma. It is most often an acquired condition but can be inherited; the most commonly identified genetic aberrations leading to mastocytosis are activating mutations involving codon 816 of the KIT gene. Herein, we present the case of a 30-year-old Caucasian man with systemic mastocytosis discovered to h

Immunohistochemical characterization of mast cell disease in paraffin sections using tryptase, CD68, myeloperoxidase, lysozyme, and CD20 antibodies.

To date, the diagnosis of mast cell disease (MCD) relied on routine plus histochemical stains. Its differential diagnosis, however, includes a variety of other hematopoietic and particularly B-cell lymphoid neoplasms that are best identified in paraffin sections using immunostains. To determine the paraffin-section immunoreactivity of MCD, 20 specimens from 14 patients with MCD and 1 bone marrow sample (from a patient with probable MCD) that showed equivocal metachromasia, were stained with antitryptase, CD68 (KP-1), CD20 (L26), antilysozyme, and antimyeloperoxidase antibodies. Unexplained Syncope in Mastocytosis: The Role of Early Dermatological Recognition.

Mastocytosis is a rare disorder characterized by the accumulation of abnormal mast cells in various tissues. The World Health Organization recognizes three disease types: cutaneous mastocytosis, systemic mastocytosis and mast cell sarcoma. (1) Urticaria pigmentosa, a type of cutaneous mastocytosis is the most common form of mastocytosis and manifests as red-brown macules or papules on the skin. While typically benign, systemic involvement may occur, necessitating further evaluation. Clinical manifestations primarily arise from mast cell degranulation and include diarrhea, nausea, vomiting, syn

Ureteral mast cell tumor in a dog.

A 6-year-old, castrated male, mixed-breed dog was diagnosed with partial unilateral ureteral obstruction secondary to a ureteral mass. The ureteral mass was surgically resected, and an ureteroneocystostomy was performed. Histopathology of the ureteral mass was consistent with a poorly differentiated mast cell tumor (MCT). The patient recovered well but was euthanized 5 months postoperatively for central nervous system signs. A choroid plexus tumor was diagnosed during necropsy examination. There was no evidence of recurrence or dissemination of the ureteral MCT. Extracutaneous MCTs are rare in
