Cellular membranes of the related fission yeast species differ in their dipole potential.

The dielectric environment and membrane dipole potential are key electrostatic properties of biological membranes, which modulate the function of membrane-associated proteins. If and how these properties are shaped by species-specific cellular lipid landscapes are fundamental questions. Here, we use two related fission yeast species, Schizosaccharomyces pombe (S. pombe) and Schizosaccharomyces japonicus (S. japonicus), which exhibit remarkable differences in membrane lipid composition, to address this problem. S. pombe synthesizes membranes from largely unsaturated glycerophospholipids with 18

The Effects of Ca2+ on Membrane Potential with Altered Function of NALCN and K2P Channels.

Cells exhibit a membrane potential due to the differential distribution of ions and the density of channels, pumps, and exchangers. It is known in larval Drosophila that the membrane potential gravitates towards the equilibrium potential of K+ due to the high density of K2P channels. Higher extracellular Ca2+ ([Ca2+]o) tends to drive the resting membrane potential to a more negative state, while lowering it has an opposite effect. The expression of the K2P channel and NALCN was altered genetically to determine the sensitivity to changes in [Ca2+]o, and computational simulations using the theor

Bacterial Membrane Vesicles as Heteroantigen Reservoirs for Potentiated Colorectal Cancer Immunotherapy.

Cancer immunotherapy, including immune checkpoint blockade (ICB), often has limited efficacy due to inadequate neoantigen exposure. Although neoantigen vaccines can enhance immune sensitization, their clinical application is hindered by high identification costs and tumor heterogeneity. Alternatively, heteroantigen presentation by cancer cells offers a promising strategy to improve immunotherapy. Herein, hyaluronic acid (HA)-modified bacterial membrane vesicles were developed as heteroantigen reservoirs (HLGV) and then further loaded with PD-L1 siRNA (siPD-L1) for immune checkpoint blockade. Resorbable Membranes for Guided Bone Regeneration: Critical Features, Potentials, and Limitations.

Lack of horizontal and vertical bone at the site of an implant can lead to significant clinical problems that need to be addressed before implant treatment can take place. Guided bone regeneration (GBR) is a commonly used surgical procedure that employs a barrier membrane to encourage the growth of new bone tissue in areas where bone has been lost due to injury or disease. It is a promising approach to achieve desired repair in bone tissue and is widely accepted and used in approximately 40% of patients with bone defects. In this Review, we provide a comprehensive examination of recent advance

The Role of Sperm Membrane Potential and Ion Channels in Regulating Sperm Function.

During the last seventy years, studies on mammalian sperm cells have demonstrated the essential role of capacitation, hyperactivation and the acrosome reaction in the acquisition of fertilization ability. These studies revealed the important biochemical and physiological changes that sperm undergo in their travel throughout the female genital tract, including changes in membrane fluidity, the activation of soluble adenylate cyclase, increases in intracellular pH and Ca2+ and the development of motility. Sperm are highly polarized cells, with a resting membrane potential of about -40 mV, which

Membranous nephropathy: Clearer pathology and mechanisms identify potential strategies for treatment.

Primary membranous nephropathy (PMN) is one of the common causes of adult-onset nephrotic syndrome and is characterized by autoantibodies against podocyte antigens causing in situ immune complex deposition. Much of our understanding of the disease mechanisms underpinning this kidney-limited autoimmune disease originally came from studies of Heymann nephritis, a rat model of PMN, where autoantibodies against megalin produced a similar disease phenotype though megalin is not implicated in human disease. In PMN, the major target antigen was identified to be M-type phospholipase A2 receptor 1 (PLA

Coronavirus membrane fusion mechanism offers a potential target for antiviral development.

The coronavirus disease 2019 (COVID-19) pandemic has focused attention on the need to develop effective therapies against the causative agent, SARS-CoV-2, and also against other pathogenic coronaviruses (CoV) that have emerged in the past or might appear in future. Researchers are therefore focusing on steps in the CoV replication cycle that may be vulnerable to inhibition by broad-spectrum or specific antiviral agents. The conserved nature of the fusion domain and mechanism across the CoV family make it a valuable target to elucidate and develop pan-CoV therapeutics. In this article, we revie

Mitochondrial membrane potential and delayed graft function following kidney transplantation.

Delayed graft function (DGF) complicates 20%-40% of deceased-donor kidney transplants and is associated with increased length of stay and subsequent allograft failure. Accurate prediction of DGF risk for a particular allograft could influence organ allocation, patient counseling, and postoperative planning. Mitochondrial dysfunction, a reported surrogate of tissue health in ischemia-perfusion injury, might also be a surrogate for tissue health after organ transplantation. To understand the potential of mitochondrial membrane potential (MMP) in clinical decision-making, we analyzed whether lowe

ELECTROCHEMICAL POTENTIAL OF THE INNER MITOCHONDRIAL MEMBRANE AND Ca2+ HOMEOSTASIS OF MYOMETRIUM CELLS.

We demonstrated using Ca(2+)-sensitive fluorescent probe, mitochondria binding dyes, and confocal laser scanning microscopy, that elimination of electrochemical potential of uterus myocytes' inner mitochondrial membrane by aprotonophore carbonyl cyanide m-chlorophenyl hydrazone (10 μM), and by a respiratory chain complex IV inhibitor sodium azide (1 mM) is associated with substantial increase of Ca2+ concentration in myoplasm in the case of the protonophore effect only, but not in the case of the azide effect. In particular, with the use of nonyl acridine orange, a mitochondria-specific dye, a

Chlamydial polymorphic membrane proteins: regulation, function and potential vaccine candidates.

Pmps (Polymorphic Membrane Proteins) are a group of membrane bound surface exposed chlamydial proteins that have been characterized as autotransporter adhesins and are important in the initial phase of chlamydial infection. These proteins all contain conserved GGA (I, L, V) and FxxN tetrapeptide motifs in the N-terminal portion of each protein. All chlamydial species express Pmps. Even in the chlamydia-related bacteria Waddlia chondrophila, a Pmp-like adhesin has been identified, demonstrating the importance of Pmps in Chlamydiales biology. Chlamydial species vary in the number of pmp genes an

Pathogenic, diagnostic and vaccine potential of leptospiral outer membrane proteins (OMPs).

Pathogenic Leptospira species are important human and animal pathogen that causes leptospirosis, with more than half a million cases reported annually but little is known regarding the true incidence of leptospirosis due to the limitations in diagnosis. Proteins embedded in the outer membrane are found to be prime drug targets due to its key role as receptors for cellular communication and gatekeepers for iron and substrate transport across cell membranes. The major key issues to be addressed to overcome the disease burden of leptospirosis are: need to identify the genes that turn on in vivo;

Membrane-damaging potential of natural L-(-)-usnic acid in Staphylococcus aureus.

The purpose of this investigation was to try to understand the antibacterial mechanism of L-(-)-usnic acid isolated for the first time from fruticose lichen Usnea subfloridana using clinical isolates of methicillin-resistant Staphylococcus aureus (MRSA). The minimum inhibitory concentration (MIC) of L-(-)-usnic acid against the clinical isolates of MRSA and reference strain S. aureus MTCC-96 (SA-96) was in the range 25-50 μg/ml. Treatment of both reference and clinical strains (MRSA-ST 2071) with four-fold MIC concentrations (100-200 μg/ml) of L-(-)-usnic acid reduced the viability of cells wi

Study of membrane potential in T lymphocytes subpopulations using flow cytometry.

BACKGROUND: Ion channels are involved in the control of membrane potential (psi) in a variety of cells. The maintenance of psi in human T lymphocytes is essential for T-cell activation and was suggested to depend mostly on the voltage-gated Kv1.3 channel. Blockage of Kv1.3 inhibits cytokine production and lymphocyte proliferation in vitro and suppresses immune response in vivo. T lymphocytes are a heterogeneous cell population and the expression of Kv1.3 varies among cell subsets. Oxonol diBA-C4-(3) was used to determine psi by flow cytometry. The presence of distinct T cell subsets was evalua

Calculating event-triggered average synaptic conductances from the membrane potential.

The optimal patterns of synaptic conductances for spike generation in central neurons is a subject of considerable interest. Ideally such conductance time courses should be extracted from membrane potential (V(m)) activity, but this is difficult because the nonlinear contribution of conductances to the V(m) renders their estimation from the membrane equation extremely sensitive. We outline here a solution to this problem based on a discretization of the time axis. This procedure can extract the time course of excitatory and inhibitory conductances solely from the analysis of V(m) activity. We

Indications for acceleration-dependent changes of membrane potential in the flagellate Euglena gracilis.

The effects of the calcium sequester EGTA on gravitactic orientation and membrane potential changes in the unicellular flagellate Euglena gracilis were investigated during a recent parabolic-flight experiment aboard of an Airbus A300. In the course of a flight parabola, an acceleration profile is achieved which yields subsequently about 20 s of hypergravity (1.8 g(n)), about 20 s of microgravity, and another 20 s of hypergravity phases. The movement behavior of the cells was investigated with real-time, computer-based image analysis. Membrane potential changes were detected with a newly develo

Membrane potential stabilization in amphibian skeletal muscle fibres in hypertonic solutions.

This study investigated membrane transport mechanisms influencing relative changes in cell volume (V) and resting membrane potential (E(m)) following osmotic challenge in amphibian skeletal muscle fibres. It demonstrated a stabilization of E(m) despite cell shrinkage, which was attributable to elevation of intracellular [Cl(-)] above electrochemical equilibrium through Na(+)-Cl(-) and Na(+)-K(+)-2Cl(-) cotransporter action following exposures to extracellular hypertonicity. Fibre volumes (V) determined by confocal microscope x z - scanning of cutaneous pectoris muscle fibres varied linearly wi
