Ileum transcriptional response to prolonged supplementation with phytogenic product containing menthol, carvacrol and carvone.

Pathogen control is a critical issue in the layer industry. Plant-based natural products are firmly replacing the undesirable use of antibiotics in animal production. The poultry industry embraced the opportunity to distance itself from the negative public perception of antibiotic use. In this study, we investigated the effects of a phytogenic product comprising of menthol, carvacrol and carvone on ileum gene expression profile in layers after 16 weeks of continual supplementation. Phytogen supplementation increased endocytosis and autophagy while showing significant predicted cardiovascular p

Menthol Stereoisomers Exhibit Different Effects on α4β2 nAChR Upregulation and Dopamine Neuron Spontaneous Firing.

Menthol contributes to poor cessation rates among smokers, in part because menthol enhances nicotine reward and reinforcement. Mentholated tobacco products contain (-)-menthol and (+)-menthol, in varying proportions. We examined these two menthol stereoisomers for their ability to upregulate α4β2 nAChRs and to alter dopamine neuron firing frequency using long-term, low-dose (≤500 nm) exposure that is pharmacologically relevant to smoking. We found that (-)-menthol upregulates α4β2 nAChRs while (+)-menthol does not. We also found that (-)-menthol decreases dopamine neuron baseline firing and do

Dietary supplementation of menthol-rich bioactive lipid compounds alters circadian eating behaviour of sheep.

BACKGROUND: Plant bioactive lipid compounds (PBLC), commonly known as essential oils, are increasingly evaluated as feed additives in ruminants due to beneficial effects on animal performance and health; however, there is no study evaluating circadian eating behaviour in ruminants. Altered eating behaviour may be implicated in changes of feed intake in ruminants. Therefore, the present study investigated the influence of menthol-rich PBLC on circadian eating behaviour in 24 growing sheep that were equally divided into three treatments, control (without PBLC), a lower dose (80 mg/d) or a higher

Cancer treatment-related neuropathic pain: proof of concept study with menthol--a TRPM8 agonist.

PurposeEffective treatment of neuropathic pain without unacceptable side effects is challenging. Cancer sufferers increasingly live with long-term treatment-related neuropathic pain, resulting from chemotherapy-induced peripheral neuropathy (CIPN) or surgical scars. This proof-of-concept study aimed to determine whether preclinical evidence for TRPM8 ion channels in sensory neurons as a novel analgesic target could be translated to clinical benefit in patients with neuropathic pain, using the TRPM8 activator menthol.Patients and methodsPatients with problematic treatment-related neuropathic pa

The role of Ca2+ in (-)-menthol-induced human promyelocytic leukemia HL-60 cell death.

A human promyelocytic leukemia HL-60 cell line was selected to examine the effect of (-)-Menthol on cell death. Based on the results from morphological changes and the percentage of viable cells in HL-60 cells after treatment with various concentrations of (-)-Menthol, it was shown that (-)-Menthol induced cell death through necrosis, not apoptosis. No cell cycle arrest was found in HL-60 cells examined by flow cytometry analysis. Also, the DNA gel electrophoresis method showed that (-)-Menthol did not induce apoptosis in HL-60 cells. However, it was found that (-)-Menthol induced the producti

Coarse grained molecular dynamics analysis of menthol interactions with ergosterol in a fungal mimetic membrane.

Menthol, a monoterpene from peppermint oil, has antifungal activity against Candida species. However, how menthol interacts directly with fungal membrane sterols is still not well understood. In this study, coarse-grained molecular dynamics simulations were used with the Martini force field to investigate menthol in a fungal-mimetic POPC/ergosterol (8:2) membrane. Radial distribution function analysis revealed preferential spatial association between menthol and ergosterol, including a local maxima at approximately 5.0 Å and a broader peak in the 9.5 Å range. Contact analysis further demonstra

Deep eutectic solvents based on matrine and menthol: A novel and safe transdermal penetration enhancer.

Transdermal drug delivery has attracted extensive attention due to its ability to bypass the first-pass metabolism, maintain plasma drug concentrations, and improve patient compliance. However, the transdermal delivery of hydrophilic drugs is still quite challenging. This study aims to develop a novel deep eutectic solvent (DES) composed of matrine (MT) from sophora flavescens Aiton and menthol (Men) from Mentha canadensis to enhance the transdermal delivery of hydrophilic drugs. Geniposide (GS) was selected as the model drug. Combining multiple experimental techniques and theoretical calculat

Calcium channel blocking activity as a mechanism of the spasmolytic effect of menthol on isolated bovine ileum.

Menthol is an organic compound commonly derived from mint oils, known for its cooling and soothing properties, often used in various pharmaceutical, cosmetic and therapeutic applications. It exerts beneficial effects on the digestive system by relaxing smooth muscles, reducing spasms and improving gastrointestinal motility. The present study aimed to evaluate the effects of menthol on bovine ileal smooth muscle contractions in vitro. Ileal tissue segments were collected from adult cattle at an abattoir and subsequently immersed in 25.00 mL organ baths containing Tyrode's solution. The baths we

Menthol's disruptive effects on kanamycin-resistant Escherichia coli energy metabolism and ion fluxes.

The presented study aimed to investigate the antibacterial activity of menthol-the main component of one of the widespread plants of the Lamiaceae family-Mentha arvensis. To investigate the mode of action of menthol, we studied its influence on kanamycin-resistant E. coli pARG-25 and wild-type E. coli BW25113 strains. For this, the effect of menthol on ATPase activity, proton and potassium fluxes, and intracellular pH was investigated under aerobic and anaerobic conditions. The results showed that menthol influences these parameters in a concentration- and condition-dependent way. It likely in

Molecular insights into the inhibition of MexCD-OprJ efflux pumps by menthol and thymol in Pseudomonas aeruginosa.

MexCD-OprJ is one of the major efflux systems which confers significant drug resistance to Pseudomonas aeruginosa. This study aimed to investigate the effects of two herbal compounds, menthol and thymol, on the MexCD-OprJ efflux pump in P. aeruginosa isolates. Ethidium bromide (EtBr) agar cartwheel, disk diffusion, and antibiogram tests were carried out for the isolates, with and without exposure to sub-MIC values of menthol and thymol. The expression level of the oprJ gene was evaluated through reverse transcription quantitative PCR (RT-qPCR). In order to check the interactions between the Op

Evaluating a Real World Ban on Menthol Cigarettes: An Interrupted Time-Series Analysis of Sales.

BACKGROUND: Menthol in cigarettes has been shown to increase regular cigarette smoking and nicotine dependence, and decrease success in smoking cessation. Owing to these reasons, in May 2015, the province of Ontario introduced a menthol ban on tobacco products that came into effect in January 2017 prior to a Federal Canadian Ban in October 2017. The objective of this article was to assess the effect of a provincial menthol ban on cigarette wholesale sales in Ontario. METHODS: Wholesale data submitted by tobacco manufacturers to Health Canada pursuant to the federal Tobacco Reporting Regulatio

Inhibition by menthol and its related chemicals of compound action potentials in frog sciatic nerves.

AIMS: Transient receptor potential (TRP) vanilloid-1 (TRPV1) and melastatin-8 (TRPM8) channels play a role in transmitting sensory information in primary-afferent neurons. TRPV1 agonists at high concentrations inhibit action potential conduction in the neurons and thus have a local anesthetic effect. The purpose of the present study was to know whether TRPM8 agonist menthol at high concentrations has a similar action and if so whether there is a structure-activity relationship among menthol-related chemicals. MAIN METHODS: Compound action potentials (CAPs) were recorded from the frog sciatic

The effect of menthol application to the skin on sweating rate response during exercise in swimmers and controls.

We tested the hypothesis that menthol application would reduce the magnitude and initiation of sweating via excitation of cold-sensitive afferent pathways and concurrently via a cross-inhibition of heat loss pathways in acclimatized (swimmers, SW) and non acclimatized (control, CON) subjects in cool water. It was expected this effect to be exaggerated in SW subjects. Eight SW and eight CON subjects cycled at 60% of their VO(2)max, as long as to reach 38 degrees C in rectal temperature (Tre), without or with (4.6 g per 100 ml of water) all-body application of menthol sediment. Heart rate (HR),
