Scanning and transmission electron microscopy of mercuric chloride-induced acute tubular necrosis in rat kidney.

Scanning (SEM) and transmission (TEM) electron microscopy were used to elucidate morphological changes associated with acute tubular necrosis induced by high doses of mercuric chloride. Marked morphological changes were demonstrated in proximal tubules with TEM at one hour and with both SEM and TEM at six hours. These changes appeared earlier than reported in previous studies using any dose. The scanning microscopic provided a three-dimensional view of proximal cells showing changes in early injury with subsequent separation of the injured cells from the remaining cells. Certain of these resid

Intracellular glutathione (GSH) levels modulate mercuric chloride (MC)- and methylmercuric chloride (MeHgCl)-induced amino acid release from neonatal rat primary astrocytes cultures.

Mercuric chloride (MC) and methylmercury (MeHg) were found to increase amino acid release from astrocytes. This suggests interaction with sulfhydryl (-SH) groups which are controlled by glutathione [GSH] levels. In the present study, we evaluated the effects of alterations in intracellular glutathione concentrations [GSH]i on the outcome of MC and MeHg treatment. [GSH]i were increased in a time-dependent fashion by incubating the astrocytes with 1 mM L-2-oxothiazolidine-4-carboxylic acid (OTC), a cysteine precursor. OTC attenuated the release of [2,3-3H]D-aspartic acid from astrocytes exposed

Histochemical demonstration of mercury in the olfactory system of salmon (Salmo salar L.) following treatments with dietary methylmercuric chloride and dissolved mercuric chloride.

The deposition of organic and inorganic mercury compounds was studied histochemically in the salmon (Salmo salar L.) olfactory system. One group of salmon was given fodder pellets containing methylmercuric chloride (CH3HgCl, 99 micrograms Hg/g) for 4 weeks. Other groups of fish were exposed to dissolved mercuric chloride (HgCl2, 270 micrograms Hg/liter) for 2, 6, and 12 hr, respectively. In both series of experiments, the radioisotope 203Hg was included in order to determine the accumulation of mercury in the olfactory system. Gamma-spectrometry showed that both mercury compounds accumulated i

Differential effects of methylmercuric chloride and mercuric chloride on the histochemistry of rat thyroid peroxidase and the thyroid peroxidase activity of isolated pig thyroid cells.

This study was designed to characterize the interaction of CH3HgCl or HgCl2 with thyroid peroxidase (TPO). Two types of experiments were performed. First, the thyroids from rats that were given 5.6 mg/kg/day of either CH3HgCl or HgCl2 for 2 weeks by intubation were subjected to histochemical treatment and then to electron microscopy. TPO activities in all cell compartments were inhibited by HgCl2 but not by CH3HgCl. Morphological observation showed that taller epithelia were induced by HgCl2, whereas flattened epithelia forming large follicles were induced by CH3HgCl. The serum thyrotropin lev

Improvement of mercuric chloride-induced testis injuries and sperm quality deteriorations by Spirulina platensis in rats.

The present study was undertaken to investigate the protective effect of the filamentous cyanobacterium Spirulina platensis (S. platensis) on mercury (II) chloride (HgCl(2))-induced oxidative damages and histopathological alterations in the testis of Wistar albino rats. The animals were divided into four equal groups, i) control, ii) HgCl(2), iii) S. platensis and iv) combination of HgCl(2)+S. platensis. Oxidative stress, induced by a single dose of HgCl(2) (5 mg/kg, bw; subcutaneously, s.c.), substantially decreased (P<0.01) the activity level of testicular key enzymatic antioxidant biomarker

Phosphorylation of nuclear proteins of peripheral blood T lymphocytes activated by nickel sulfate and mercuric chloride.

The phosphorylation of nuclear proteins of peripheral blood T lymphocytes activated by nickel sulfate or mercuric chloride, and from nickel-allergic subjects, was studied in polyacrylamide gel separations of 32P-labeled proteins. With a preincubation period of the metal salts for 48 h, a marked increase of 32P label into nonhistone proteins, especially the 30- to 40-kilodalton region, was found compared to the control cultures. This increase was most pronounced in mercuric-chloride-treated cultures, which also showed an increase in labeling of histone H4. The increase in nuclear protein phosph

Prolonged exposure to mercuric chloride induces oxidative stress-mediated nephrotoxicity in freshwater food fish Channa punctatus.

Mercury (Hg) is regarded as a serious hazard to aquatic life and is particularly prevalent in aquatic ecosystems. However, there is little evidence available regarding the toxicity of mercury chloride (HgCl2) in vital organs of fish. This study was conducted to assess the effects of HgCl2 (0.039 mg/L and 0.078 mg/L) on oxidative stress-mediated genotoxicity, poikilocytosis, apoptosis, and renal fibrosis after 15, 30, and 45 days of the exposure period. According to the findings, HgCl2 intoxication in fish resulted in a significantly (P < 0.05) elevated lipid peroxidation (LPO), protein carbony

Mercuric chloride induced reproductive toxicity associated with oxidative damage in male Wistar albino rat, Rattus norvegicus.

In the field of toxicology, male reproductive hazards attributed to metal exposure is a fast-developing issue. Mercury has been identified as an environmental pollutant that causes potential adverse impacts on organisms. This study aimed to assess the reprotoxic consequences of mercuric chloride (HgCl2). Five groups of sexually mature albino rats were given oral mercuric chloride (HgCl2) treatment. (G1) control group received saline treatment; (G2) (5.25 mg/kg of HgCl2 for 30 days); (G3) (5.25 mg/kg of HgCl2 for 60 days); (G4) (10.5 mg/kg of HgCl2 for 30 days); (G5) (10.5 mg/kg of HgCl2 for 60

Bone marrow cells contribute to tubular epithelium regeneration following acute kidney injury induced by mercuric chloride.

BACKGROUND & OBJECTIVES: Acute tubular necrosis (ATN) caused by renal ischaemia, renal hypo-perfusion, or nephrotoxic substances is the most common form of acute kidney injury (AKI). There are a few treatment options for this life-threatening disease and the mortality rate exceeds 50 per cent. In critical cases of AKI the only option is renal transplantation. In the present study we evaluated whether bone marrow cells (BMCs) are involved in regeneration of kidney tubules following acute tubular necrosis in the mouse. METHODS: Six to eight week old C57BL6/J and congenic enhanced green fluoresc

A protein from Cajanus indicus Spreng protects liver and kidney against mercuric chloride-induced oxidative stress.

Mercuric chloride (HgCl(2)) is a widespread environmental toxin that affects mainly liver and kidney. The present study has been carried out to investigate the protective action of a protein (the CI protein) isolated from the herb, Cajanus indicus Spreng against HgCl(2) induced renal and hepatic toxicities in mice. Intraperitoneal administration of HgCl(2) at a dose of 5 mg/kg body weight for 1 d significantly reduced the activities of antioxidant enzymes like superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx). Moreover, it also depleted the glutathione to oxidized glu

Methyl mercuric chloride toxicokinetics in mice. I: Effects of strain, sex, route of administration and dose.

The toxicokinetics of methyl mercury is studied most intensively in the rat. However, the toxicokinetics of methyl mercury in man is closer to the toxicokinetics in the mouse. This study describes the effects of dose, route of administration, and strain and sex on the toxicokinetics of methyl mercuric chloride in mice. Half-time values, fractional whole-body retentions and relative organ distributions of mercury were compared after a single oral or intraperitoneal administration of methyl mercuric chloride. The intestinal absorption was almost complete in accordance with earlier published resu

Elevation of rat erythrocyte nucleotide levels following acute renal failure induced by glycerol or mercuric chloride.

Biochemical changes in the blood following induction of renal failure by glycerol or mercuric chloride have been studied in 16 rats. Plasma creatinine, urea and Pi levels indicated that renal impairment followed the same time course in both renal failure models, with the severest effects on day 3 and returning to normal by day 7. Erythrocyte ATP and guanine triphosphate (GTP) levels were significantly elevated above contorl values on day 1 and remained elevated in both models. ATP/ADP and GTP/GDP ratios also increased in both models. In renal failure the increased purine 'salvage' in the eryth

Mercury Toxicity and Contamination of Households from the Use of Skin Creams Adulterated with Mercurous Chloride (Calomel).

Inorganic mercury, in the form of mercurous chloride, or calomel, is intentionally added to some cosmetic products sold through informal channels in Mexico and the US for skin lightening and acne treatment. These products have led to multiple cases of mercury poisoning but few investigations have addressed the contamination of cream users' homes. We report on several cases of mercury poisoning among three Mexican-American families in California from use of mercury-containing skin creams. Each case resulted in widespread household contamination and secondary contamination of family members. Uri

The effects of chronic ingestion of mercuric chloride on fertility and testosterone levels in male Sprague Dawley rats.

Although male infertility is well researched, the effects of inorganic mercury on male reproduction and fertility are less well known. Studies pertaining to mercury and male fertility identified reduced concentration of testosterone in the serum of male workers, a toxic influence on fertility of organic mercury compounds within concentrations at the workplace, and increased days to pregnancy. We evaluated the effect of chronic mercuric chloride (HgCl(2)) exposure in male rats on reproductive endpoints. Thirty-day old male Sprague Dawley rats (n = 31) were exposed to 0.0, 1.0, or 2.0 mg/kg/day
