Arteriovenous shunting and cholestasis in hepatic hemangiomatosis associated with metoclopramide.

Diffuse hemangiomatosis of the liver became apparent in a 22-year-old woman while she was receiving medication with metoclopramide and experiencing the well-known adverse effect of the drug, hyperprolactinemia with secondary amenorrhea and galactorrhea. The hemangiomatosis was demonstrated by ultrasonography, computerized tomography, arteriography, and laparotomy with biopsy. When arteriovenous shunting became life-threatening and severe abdominal pain and cholestasis developed, the patient's name was placed on the waiting list for liver transplantation. However, after stopping the medication

Effect of metoclopramide on gastric ulceration and secretion in albino rats.

The antiulcer and antisecretory effects of metoclopramide were studied in aspirin-induced and pylorus-ligated gastric ulcers. The effects were compared with ranitidine (10 mg/kg, orally). The effect of propantheline alone and in combination with metoclopramide was studied against aspirin-induced gastric ulcers only. Metoclopramide (20 mg and 50 mg/kg) produced an ulcer protective effect both in aspirin-induced and pylorus-ligated experimental models. Its ulcer protective effect against aspirin-induced and pylorus-ligated gastric ulcers was comparable to ranitidine. Metoclopramide reduced the v

The effect of metoclopramide and prochlorperazine on the absorption of effervescent paracetamol in migraine.

Antiemetics modify gastric emptying, a rate-limiting step in drug absorption. The absorption of effervescent paracetamol in water solution was studied in three groups of 10 female patients during acute migraine attacks. Paracetamol was preceded 30 min earlier by a rectal dose of metoclopramide, prochlorperazine maleate, or placebo. Each patient was retested with paracetamol when headache-free. Migraine attacks delayed slightly the absorption of paracetamol solution. Prior administration of rectal prochlorperazine had a minor delaying effect on paracetamol absorption. The peak concentration, th

Clinical use of metoclopramide test in the diagnosis of women with hyperprolactinemia.

UNLABELLED: 14 women with elevated prolactin (PRL) serum levels (greater than 25 ng/ml) were given 2.5 mg of metoclopramide, by bolus intravenous injection, to evaluate its diagnosic potential as a stimulus for PRL release. Following metoclopramide injection there was a prompt increase in serum PRL in normal subjects and in patients with moderate PRL elevations associated with galactorrhea-oligomenorrhea. The women with amenorrhea-galactorrhea regardless of the presence of absence of a pituitary tumor, showed a blunted response. Metoclopramide failed to induce TSH secretion in all cases. IN C

[The effect of domperidone and metoclopramide on antral motility (author's transl)].

The effect of domperidone and metoclopramide on antral motility was studied by measuring the intraluminal pressure in the antrum of 10 normal persons and of 8 patients suffering from duodenal ulcer. Both drugs stimulated the rhythmic activity of the antrum. The antiemetic effect and the therapeutic effect of domperidone on complaints by gastrointestinal retention was the result of the influence on gastrointestinal motility, which is similar to that of metoclopramide.

The effect of metoclopramide on the lower oesophageal sphincter in late pregnancy.

The effects of intravenous metoclopramide (Maxolon) on the lower oesophageal sphincter (LOS) were studied in three groups of patients, one group being normal control and the other two being pregnant females, one without heartburn and the other with. Metoclopramide increases the LOS pressure 20.5, 15.2 and 10.2 cm H2O respectively (p less than 0.005, p less than 0.005 and p less than 0.05). These findings suggest that for patients undergoing elective or emergency obstetrical anaesthesia, intravenous metoclopramide may help reduce the incidence of regurgitation of gastric contents.

Comparison of metoclopramide with other dynamic tests of prolactin secretion.

Metoclopramide, a derivative of procainamide, was compared with thyrotropin releasing hormone (TRH), N3im-methyl-TRH and chlorpromazine as a prolactin (PRL) releaser in 11 euthyroid males. The peak response of serum PRL and the maximum increment of serum PRL (max deltaPRL) were greater (P less than 0.0005) after the administration of metoclopramide (deltaPRL 24.1 +/- 1.5 (SE) ng/ml) than after either TRH (14.9 +/- 1.5 ng/ml) or chlorpromazine (7.0 +/- 2.1 ng/ml). There were no significant differences between the peak and max deltaPRL responses to metoclopramide or N3im-methyl-TRH. Metocloprami

Metoclopramide: a review of its pharmacological properties and clinical use.

Metoclopramide, 4-amino-5-chloro-2-methoxy-N-(2-diethyl-aminoethyl) benzamide, is advocated for use in gastro-intestinal diagnostics, and in treating various types of vomiting and a variety of functional and organic gastro-intestinal disorders. Published data have indicated that metoclopramide assists radiological identification of lesions in the small intestine, facilitates duodenal intubation and small intestine biopsy, and eases emergency endoscopy in upper gastro-intestinal haemorrhage. Metoclopramide reduces post-operative vomiting and radiation sickness, and ameliorates some types of dru

Unrecognised pheochromocytoma in pregnancy discovered through metoclopramide-triggered hypertensive emergency.

PURPOSE: Pheochromocytoma, a catecholamine-secreting tumour leading to neurological and cardiovascular life-threatening conditions through hypertension crisis, occurs in 0.1-0.5% of hypertensive patients, but it is extremely rare in pregnancy (0.0018-0.006%). Some classes of drugs, even commonly used in pregnancy, can trigger catecholamine secretion, precipitating the clinical situation. MATERIALS AND METHODS AND RESULTS: We report a 33-year-old woman, gravida 2 para 1, with previous mild hypertension, was admitted to the emergency room, at 28 2/7 weeks of gestation due to headache, tachycard

Urinary retention--an unusual dystonic reaction to continuous metoclopramide infusion.

Vomiting is one of the most distressing adverse effects of cancer chemotherapy. Metoclopramide by continuous infusion (400 micrograms/kg/h after a loading dose of 2.5 mg/kg) is a novel administration method for optimizing efficacy. A two-year-old boy developed urinary retention on three occasions, once accompanied by priapism and slurred speech, while receiving a continuous infusion. This was reversed by procyclidine, suggesting that it may have been a dystonic reaction.

Gastroparesis following traumatic brain injury and response to metoclopramide therapy.

Most studies on nutrition in patients with traumatic brain injury (TBI) have focused on the hypermetabolic state immediately after trauma, or the swallowing disorders that frequently follow acute TBI. A less well-known sequela, however, is gastroparesis, which generally manifests itself when patients have advanced from nasogastric to oral feeding programs. Case histories of four patients are reported, all of whom responded to therapeutic intervention with metoclopramide. Results of a radionuclide gastric emptying study, before and after metoclopramide therapy, are presented. The etiologic and
