Macrophage-Mimicking Biomimetic Microbubbles for Early Ultrasound Molecular Imaging of Acute Rejection in Heart Transplantation.

Acute rejection (AR) is an important cause of heart graft injury and failure. Accurate diagnosis of AR is essential to achieving early intervention and improving the prognosis of patients. Current clinical monitoring of AR requires repeated invasive endocardial biopsies, and there is an urgent need to develop a noninvasive diagnostic technique. As antigen-presenting cells, macrophages play an important role in adaptive immunity, such as the early activation of T cells in AR, and are considered to be key drivers of AR. Macrophage membrane biomimetic microbubbles (MB-Mφ) inherit a variety of fun

Mucoadhesive Air Microbubbles With Sustained Antibiotic Release for Ultrasound-Guided Intrauterine Infusion Therapy of Endometritis.

Endometritis, a common gynecological disease caused by ascending bacterial infection, is a leading cause of infertility. Currently, the clinical treatment relies primarily on antibiotic therapy, administered via intravenous injection, oral administration, or local delivery (e.g., intrauterine infusion). Nevertheless, intravenous injection aggravates hepatic and renal burdens, whereas oral administration elicits gastrointestinal reactions; intrauterine infusion suffers from drug leakage from the uterus due to gravity. Therefore, we developed mucoadhesive doxycycline hydrochloride-loaded air mic

Magnetic drug-loaded microbubbles for treating lower limb venous thrombosis under controllable rotating magnetic field.

OBJECTIVE: This study aimed to develop a poly (lactic-co-glycolic acid) (PLGA)-based magnetic loaded iron oxide (Fe3O4) and single-chain urokinase-type plasminogen activator (proUK) for enhancing thrombolysis under a controlled rotating magnetic field, specifically targeting acute lower limb venous thrombosis. BACKGROUND: Acute thrombotic disorders are significant health threats, however, the exploration of magnetic actuation as a treatment for acute thrombosis has been limited. METHODS: Magnetic microbubbles were prepared using a double emulsion method, loaded with Fe3O4 nanoparticles and p

Selective Photochemical Conversion of Carbon Dioxide to Formic Acid at Gas-Water Interface of Microbubbles.

We report a selective photochemical conversion of carbon dioxide (CO2) to formic acid (HCOOH, FA) at the gas-water interface (GWI) of microbubbles. The microbubbles with an average diameter of 42 μm are produced by passing CO2 gas through a porous thermoplastic bubbler immersed in an aqueous solution of the copper(II)-phenanthroline complex [Cu(Phen)2]2+. The average FA production rate at room temperature is found to be 47.5 μM h-1 for 5 mM of [Cu(Phen)2]2+. When 5 mM iodide (I-) is added to the system, the FA production rate increases to a maximum value of 63.8 μM h-1. We also demonstrated th

Multi-modality imaging assessment of microbubbles and cerebral emboli in left ventricular pulsed field ablation.

BACKGROUND: Pulsed field ablation (PFA) may have a superior safety profile compared to other technologies, but it has the potential to cause gaseous microbubbles (MB), which may be associated with cerebral emboli. Limited relative safety data has been published regarding PFA in the left ventricle (LV). METHODS: Healthy and chronic myocardial infarction (MI) swine underwent PFA (monopolar, biphasic, 25 Amps) in the LV using an irrigated focal catheter under intra-cardiac echocardiography (ICE) guidance for MB monitoring. Two control swine received air MBs through the lumen of the ablation cath

Accelerating thrombolysis using a precision and clot-penetrating drug delivery strategy by nanoparticle-shelled microbubbles.

Conventional thrombolytic drugs for vascular blockage such as tissue plasminogen activator (tPA) are challenged by the low bioavailability, off-target side effects and limited penetration in thrombi, leading to delayed recanalization. We hypothesize that these challenges can be addressed with the targeted and controlled delivery of thrombolytic drugs or precision drug delivery. A porous and magnetic microbubble platform is developed to formulate tPA. This system can maintain the tPA activity during circulation, be magnetically guided to the thrombi, and then remotely activated for drug release

A non-surgical model of cervical spinal cord injury induced with focused ultrasound and microbubbles.

BACKGROUND: The most commonly used animal models of spinal cord injury (SCI) involve surgical exposure of the dorsal spinal cord followed by transection, contusion or compression. This high level of invasiveness often requires significant post-operative care and can limit post-operative imaging, as the surgical incision site can interfere with coil placement for magnetic resonance imaging (MRI) during the acute phase of SCI. While these models are considered to be similar to human SCI, they do not occur in a closed vertebral system as do the majority of human injuries. NEW METHOD: Here we des

The use of cationic microbubbles to improve ultrasound-targeted gene delivery to the ischemic myocardium.

We synthesized a cationic microbubble (CMB) with the aim of enhancing its DNA-carrying capacity to improve targeted gene transfection of the ischemic heart for cardiac regeneration. We previously reported that ultrasound-targeted microbubble destruction (UTMD) employing the commercial Definity microbubble (MB) successfully transfected genes into rodent hearts, but the transfection efficiency was modest. We synthesized a CMB and compared its DNA-carrying capacity and reporter gene transfection efficiency with the Definity MB. The CMB bound 70% more plasmid DNA than the Definity MB. UTMD-mediate

Assessment of hepatic fibrosis by analysis of the dynamic behaviour of microbubbles during contrast ultrasonography.

BACKGROUND/AIMS: Microbubble behaviour from the portal vein to the liver parenchyma may reflect haemodynamic changes because of hepatic fibrosis. The aim of this study was to determine the efficacy of contrast-enhanced ultrasound (US) with Sonazoid™ for the assessment of the grade of hepatic fibrosis. METHODS: This prospective study evaluated 117 patients with chronic liver disease (chronic hepatitis 85; cirrhosis 32) and 34 controls. All subjects received both contrast-enhanced US with Sonazoid™ for 1 min after the agent injection and subsequent liver biopsy. Flow velocity and flow volume in

Microbubble-activated CO2 regeneration in scalable microporous liquids enabled by contact electrocatalysis.

Conventional carbon dioxide (CO2) capture systems are constrained by limited uptake capacity and energy-intensive regeneration. Here, we formulate a microporous carbon capture solution (MCCS) by dispersing a permanent-porosity framework (ZIF-67) in a liquid absorbent, coupling solvent-excluded physisorption with chemical uptake and boosting the CO2 capture capacity by ∼45%. Instead of bulk thermal stripping, we implement interfacial, microbubble-activated regeneration (MAR) with a decreased monoethanolamine (MEA) loss rate by ∼38%. Microbubble activation refreshes gas-liquid-solid contact to l

Ultrasound-driven microbubble motors for targeted myocardial ischemia-reperfusion injury treatment.

Myocardial ischemia-reperfusion injury (MIRI) is a secondary pathological process that occurs after restoration of blood flow during the treatment of acute myocardial infarction. The effective prevention of MIRI is pivotal for optimizing therapeutic efficacy and improving long-term patient outcomes. In this study, we developed an ultrasound-driven, multilevel targeted bionic microbubble motor (CsA@NM-MBs motors) that can break through the vascular endothelial and cell membrane barriers to achieve precise, targeted drug delivery. The CsA@NM-MBs motors were coated with a neutrophil membrane, a h

Microbubble Shell Stiffness Engineering Enhances Ultrasound Imaging, Drug Delivery, and Sonoporation.

Microbubbles (MB) are widely used as contrast agents for ultrasound (US) imaging and US-enhanced drug delivery. While the majority of studies utilize commercial MB formulations, increasing experimental evidence indicates that distinct MB features critically determine their diagnostic and therapeutic performance. Here, it is shown that shell stiffness engineering of poly(alkyl cyanoacrylate) (PACA) MB, via introducing monomers with varying alkyl chain lengths and glass transition temperatures, preserves a narrow size distribution ≈2-3 µm, while enhancing MB drug loading, in vitro sonoporation c

Effect of Lipid Composition and Stirring Dynamics on Oxygen Microbubble Stability and Oxygen Release.

Lipid-coated oxygen microbubbles (OMBs) are being investigated for biomedical applications to alleviate hypoxia such as systemic oxygenation and image-guided radiosensitization therapy. Additionally, they hold potential for boarder application as oxygen carriers beyond the biomedical filed. Understanding the stability and oxygen release properties of OMBs in dynamic aqueous environments is critical for these applications. In this study, we found that OMBs composed of longer acyl chain phospholipids (DSPC and DBPC) were stable in storage for at least 1 week, unlike the shorter acyl chain phosph

Focused ultrasound-microbubble treatment arrests the growth and formation of cerebral cavernous malformations.

Cerebral cavernous malformations (CCMs) are vascular lesions within the central nervous system that cause debilitating neurological symptoms. Currently, surgical excision and stereotactic radiosurgery, the primary treatment options, pose risks to some patients. Here we tested whether pulsed, low intensity, focused ultrasound-microbubble (FUS-MB) treatments control CCM growth and formation in a clinically representative Krit1 null murine model. FUS-MB under magnetic resonance imaging (MRI) guidance opened the blood-brain barrier, with gadolinium contrast agent deposition most evident at periles

An investigation into the cytotoxic effects of microbubbles and their constituents on osteosarcoma and bone marrow stromal cells.

BACKGROUND: Ultrasound-responsive microbubbles offer a means of achieving minimally invasive, localised drug delivery in applications including regenerative medicine. To facilitate their use, however, it is important to determine any cytotoxic effects they or their constituents may have. The aim of this study was to test the hypothesis that phospholipid-shelled microbubbles are non-toxic to human bone-derived cells at biologically-relevant concentrations. METHODS: Microbubbles were fabricated using combinations of 1,2-distearoyl-sn-glycero-3-phosphocholine (DSPC), 1,2-dibehenoyl-sn-glycero-3-

Ultrasound-Targeted Microbubble Destruction: Modulation in the Tumor Microenvironment and Application in Tumor Immunotherapy.

Tumor immunotherapy has shown strong therapeutic potential for stimulating or reconstructing the immune system to control and kill tumor cells. It is a promising and effective anti-cancer treatment besides surgery, radiotherapy and chemotherapy. Presently, some immunotherapy methods have been approved for clinical application, and numerous others have demonstrated promising in vitro results and have entered clinical trial stages. Although immunotherapy has exhibited encouraging results in various cancer types, however, a large proportion of patients are limited from these benefits due to speci

Ultrasound-targeted microbubble destruction remodels tumour microenvironment to improve immunotherapeutic effect.

Cancer immunotherapy (CIT) has gained increasing attention and made promising progress in recent years, especially immune checkpoint inhibitors such as antibodies blocking programmed cell death 1/programmed cell death ligand 1 (PD-1/PD-L1) and cytotoxic T lymphocyte-associated protein 4 (CTLA-4). However, its therapeutic efficacy is only 10-30% in solid tumours and treatment sensitivity needs to be improved. The complex tissue environment in which cancers originate is known as the tumour microenvironment (TME) and the complicated and dynamic TME is correlated with the efficacy of immunotherapy

Development of an ultrasound triggered nanomedicine-microbubble complex for chemo-photodynamic-gene therapy.

Recently, combination therapy has received much attention because of its highly therapeutic effect in various types of cancers. In particular, chemo-photodynamic combination therapy has been considered as an outstanding strategy. However, an abnormal increase in tumor angiogenesis caused by reactive oxygen species (ROS) generated during photodynamic therapy (PDT) has been reported. In this study, the complex of doxorubicin (DOX)-encapsulating anti-angiogenic small interfering RNA (siRNA) nanoparticle and chlorin e6 (Ce6)-encapsulating microbubble has been developed to suppress tumor angiogenes
