Strain level variation in Proteus mirabilis chondroitin sulfate degradation kinetics and regulation by urea.

UNLABELLED: To establish infection, uropathogens must overcome several host defenses including the glycosaminoglycan (GAG) layer coating the apical surface of the bladder urothelium. GAGs are thought to protect against urinary tract infection (UTI) by serving as scaffolding sites for commensals, providing barrier function and preventing uropathogen adherence. However, the ability of uropathogens to degrade and utilize GAGs and the contribution of these activities toward UTI progression is largely unknown. We previously discovered that the uropathogen Proteus mirabilis, a common cause of cathet

Inhibition of Proteus mirabilis growth through gallium-based disruption of iron and heme metabolism.

The increased incidence of multi-drug resistant (MDR) Proteus mirabilis highlights the need for novel antibiotics targeting P. mirabilis. In this study, we examined the potential antibacterial effects of gallium-based antimicrobials on P. mirabilis. We performed in vitro susceptibility testing of gallium compounds against P. mirabilis UTI isolates. Ga(NO3)3 and gallium protoporphyrin IX (GaPP) effectively inhibited P. mirabilis growth, with minimum inhibitory concentrations (MIC) of 64 μg/ml and 2.5 μg/ml, respectively. To further assess the effects of gallium compounds on P. mirabilis growth

Proteus mirabilis in ICU Patients: Prevalence, Risk Factors, and Future Expectations of Bloodstream Infections.

Background Proteus mirabilis is an emerging but under-recognized cause of bloodstream infections (BSIs) in intensive care unit (ICU) settings, where invasive procedures and immunocompromised states increase vulnerability. Despite its clinical importance, the epidemiology and risk factors associated with P. mirabilis BSIs remain poorly understood, especially in low- and middle-income countries. Gap Most existing studies generalize Gram-negative pathogens without isolating the specific impact of P. mirabilis in ICU populations. There is a lack of data on species, specific risk factors, infection

Genomic epidemiology of antimicrobial resistance in Proteus mirabilis: core genome and plasmid-mediated drivers.

Proteus mirabilis has emerged as a clinically significant multidrug-resistant (MDR) pathogen, yet the genomic drivers and dissemination mechanisms of its antimicrobial resistance (AMR) remain poorly characterized. To address this gap, we conducted a pangenome analysis of 2,013 P. mirabilis genomes, including 1,990 publicly available strains and 23 newly sequenced clinical isolates, to delineate the species-wide AMR landscape. Our study identified 197 AMR gene subtypes spanning 12 antibiotic classes, with seven resistance determinants embedded in the core genome. Clinically critical resistance

Septic discitis and septic pulmonary emboli: rare complications of Proteus mirabilis urinary tract infection.

Proteus mirabilis is a gram-negative bacterium frequently considered a pathogen of the urinary tract. Septic discitis and septic pulmonary emboli resulting from P. mirabilis urosepsis is a rare phenomenon. We report a 39-year-old woman who was admitted to our hospital with a complicated urinary tract infection resulting in bacteraemia, septic discitis, paraspinal abscesses and septic emboli. She was treated with a prolonged course of intravenous antibiotics resulting in the clinical resolution of her symptoms. Based on our PubMed search of the English literature, this is only the second report

Tuberculous Otitis with Proteus mirabilis Co-Infection: An Unsuspected Presentation Encountered in Clinical Practice.

Tuberculosis, a contagious bacterial disease which is caused by Mycobacterium tuberculosis, primarily involves the lungs.Though Pulmonary tuberculosis (PTB) is the commonest clinical presentation, there is a need for alertness towards uncommon presentations which involve other organs. Tuberculous otitis media (TOM) is one such rare presentation seen in paediatric practice. It is characterized by painless otorrhoea which fails to respond to the routine antibacterial treatment. TOM usually occurs secondary to PTB. Here is a case of tuberculous otitis media with Proteus mirabilis co-infection, wi

Frequent association with neurosurgical conditions in adult Proteus mirabilis meningitis: report of five cases.

Adult Proteus (P.) mirabilis meningitis is relatively rare and has not been examined individually in the English-language literature. During a period of 15 years (January 1986-December 2000), four adult patients with P. mirabilis meningitis and one adult patient with mixed bacterial meningitis involving P. mirabilis were identified at Chang Gung Memorial Hospital, Kaohsiung. These five patients included one man and four women, aged from 19 to 74 years (mean age=55.4). P. mirabilis infection accounted for 1.7% (4/229) of cases of our culture-proven monomicrobial adult bacterial meningitis and w

Investigation of the effects of ampicillin and ceftazidime on Proteus mirabilis using metabolomic approaches and bioinformatics analyses.

Antibiotic resistance is the ability of microorganisms to survive and proliferate despite exposure to antibiotics that would normally inhibit or kill susceptible strains. This resistance can make antibiotics ineffective or diminish their ability to combat microorganisms, complicating the treatment of infections and potentially leading to serious complications. To combat antibiotic resistance, a comprehensive analysis of changes in the metabolic activities of microorganisms is crucial for understanding the underlying mechanisms. Proteus mirabilis is a critical pathogen, particularly as a common

The effectiveness and risk comparison of EDTA with EGTA in enhancing Cd phytoextraction by Mirabilis jalapa L.

In the previous study, Mirabilis jalapa L. had revealed the basic Cd hyperaccumulator characteristics, but the accumulation ability was not as strong as that of other known Cd hyperaccumulators. In order to improve the accumulation ability of this ornamental plant, the chelants were used to activate the Cd in soil. As a substitute, ethylene glycol bis(2-aminoethyl) tetraacetic acid (EGTA) was selected to testify whether it has better effectiveness and can bring lesser metal leaching risk than EDTA. The data showed that the growth of M. jalapa was inhibited, while the Cd concentration of the pl

ZapA, a virulence factor in a rat model of Proteus mirabilis-induced acute and chronic prostatitis.

Our knowledge of pathogenesis has benefited from a better understanding of the roles of specific virulence factors in disease. To determine the role of the virulence factor ZapA, a 54-kDa metalloproteinase of Proteus mirabilis, in prostatitis, rats were infected with either wild-type (WT) P. mirabilis or its isogenic ZapA(-) mutant KW360. The WT produced both acute and chronic prostatitis showing the typical histological progressions that are the hallmarks of these diseases. Infection with the ZapA(-) mutant, however, resulted in reduced levels of acute prostatitis, as determined from lower le

The detection of raised levels of IgM to Proteus mirabilis in sera from patients with rheumatoid arthritis.

An analysis by ELISA of 100 rheumatoid factor (RF)-positive sera selected at random from a collection of sera from patients with various auto-immune diseases and joint pains, and 100 RF-negative sera from the same collection matched by patient age and gender, showed that the RF-positive sera had highly significantly (p < 0.0001) raised levels of IgM antibody, but not IgG antibody, to Proteus mirabilis over those of the RF-negative sera. This response was subsequently found to be associated with sera from patients who clinically had rheumatoid arthritis (RA). Sera from the RA patients had signi

The impact of EmrA deficiency on antibiotic-susceptibility and virulence- and stone-associated phenotypes in Proteus mirabilis.

BACKGROUND: Proteus mirabilis is a common uropathogen responsible for urinary tract infections (UTIs). The emrA gene encodes the EmrA protein, which is a critical component of the EmrAB-TolC multidrug efflux pump. In addition to its established role in antibiotic resistance, emerging evidence indicates that EmrA is also implicated in bacterial stress adaptation and virulence-associated processes. Nevertheless, its specific functions in the pathogenesis of P. mirabilis remain unclear. RESULTS: To define the function of EmrA in pathogenicity of P. mirabilis, this study combined phenotypic assay

Outbreak Caused by VIM-1- and VIM-4-Positive Proteus mirabilis in a Hospital in Zagreb.

BACKGROUND/OBJECTIVES: Proteus mirabilis is a frequent causative agent of urinary and wound infections in both community and hospital settings. It develops resistance to expanded-spectrum cephalosporins (ESCs) due to the production of extended-spectrum β-lactamases (ESBLs) or plasmid-mediated AmpC β-lactamases (p-AmpCs). Recently, carbapenem-resistant isolates of P. mirabilis emerged due to the production of carbapenemases, mostly belonging to Ambler classes B and D. Here, we report an outbreak of infections due to carbapenem-resistant P. mirabilis that were observed in a psychiatric hospital

Study of the molecular characteristics and homology of carbapenem-resistant Proteus mirabilis by whole genome sequencing.

Introduction. Proteus mirabilis is part of the family Enterobacteriaceae, and is naturally resistant to various antimicrobial drugs. In recent years, outbreaks of severe nosocomial infections caused by carbapenem-resistant P. mirabilis (CR-PMI) have been frequently reported. Few studies exist on the whole-genome molecular characteristics of this bacterium in China and elsewhere, which stimulated the implementation of this study.Hypothesis. CR-PMI strains contained the multiple drug resistance genes and exhibited a high resistance rate to commonly used antimicrobial drugs.Aim. Our goals here we
