Equine Models of Temporomandibular Joint Osteoarthritis: A Review of Feasibility, Biomarkers, and Molecular Signaling.

Osteoarthritis (OA) of the temporomandibular joint (TMJ) occurs spontaneously in humans and various animal species, including horses. In humans, obtaining tissue samples is challenging and clinical symptoms appear late in the disease progression. Therefore, genetically modified, induced, and naturally occurring animal models play a crucial role in understanding the pathogenesis and evaluating potential therapeutic interventions for TMJ OA. Among the naturally occurring models, the equine TMJ OA model is characterized by slow, age-related progression, a wide range of clinical examinations, and

Molecular Mechanisms of ZIKV-Induced Teratogenesis: A Systematic Review of Studies in Animal Models.

Zika virus (ZIKV) is a teratogen that causes congenital anomalies, being linked to microcephaly in children exposed during pregnancy. Animal studies have been conducted to investigate the molecular mechanisms related to ZIKV teratogenesis. Although animal models can mimic the effects of ZIKV in human embryo development, few in vivo studies have addressed molecular changes following ZIKV infection in embryos. Moreover, few literature reviews have been conducted with these studies. The aim of this systematic review is to evaluate the molecular mechanisms of ZIKV teratogenesis determined from stu

Interaction of the antimicrobial peptide ∆M3 with the Staphylococcus aureus membrane and molecular models.

Staphylococcus aureus is one of the most pathogenic bacteria; infections with it are associated with significant morbidity and mortality in health care facilities. Antimicrobial peptides are a promising next generation antibiotic with great potential against bacterial infections. In this study, evidence is presented of the biological and biophysical properties of the novel synthetic peptide ΔM3. Its antimicrobial activity against the ATCC 25923 and methicillin-resistant S. aureus strains was evaluated. The results showed that ΔM3 has activity in the same μM range as vancomycin. Calpastatin ablation aggravates the molecular phenotype in cell and animal models of Huntington disease.

Deciphering the molecular pathology of Huntington disease is of particular importance, not only for a better understanding of this neurodegenerative disease, but also to identify potential therapeutic targets. The polyglutamine-expanded disease protein huntingtin was shown to undergo proteolysis, which results in the accumulation of toxic and aggregation-prone fragments. Amongst several classes of proteolytic enzymes responsible for huntingtin processing, the group of calcium-activated calpains has been found to be a significant mediator of the disease protein toxicity. To confirm the impact o

Molecular Pathology and Pharmacological Treatment of Autism Spectrum Disorder-Like Phenotypes Using Rodent Models.

Autism spectrum disorder (ASD) is a heterogeneous neurodevelopmental disorder with a high prevalence rate. The core symptoms of ASD patients are impaired social communication and repetitive behavior. Genetic and environmental factors contribute to pathophysiology of ASD. Regarding environmental risk factors, it is known that valproic acid (VPA) exposure during pregnancy increases the chance of ASD among offspring. Over a decade of animal model studies have shown that maternal treatment with VPA in rodents recapitulates ASD-like pathophysiology at a molecular, cellular and behavioral level. Her

Molecular MR imaging of liver fibrosis: a feasibility study using rat and mouse models.

BACKGROUND & AIMS: Liver biopsy, the current clinical gold standard for fibrosis assessment, is invasive and has sampling errors, and is not optimal for screening, monitoring, or clinical decision-making. Fibrosis is characterized by excessive accumulation of extracellular matrix proteins including type I collagen. We hypothesize that molecular magnetic resonance imaging (MRI) with a probe targeted to type I collagen could provide a direct and non-invasive method of fibrosis assessment. METHODS: Liver fibrosis was induced in rats with diethylnitrosamine and in mice with carbon tetrachloride.

Molecular mechanics and dynamics simulations of various dispersant models on the water surface (001).

Five dispersant-molecule models of succinimide, acrylate, imide, phenylsulfonic and salicyl were used to study their interactions with the water surface (001). The interaction energy, molecular configuration, charge distribution and radial distribution function (RDF) curve for each of the dispersant molecules were analyzed from the molecular mechanics (MM) and molecular dynamics (MD) simulation results. It can be seen that the system energies, mostly electrostatic and hydrogen bond energies, were reduced significantly when the dispersant molecules interacted with the water surface. The hydroph

Cyclic ester insertion into a bismuth-amide bond: isolable molecular models for ROP initiation.

Reaction of a bismuth amide with L-lactide, ε-caprolactone and γ-butyrolactone gives isolable ring-opened products which are also observed during polymerisation studies, including rare crystallographically authenticated amidolactate and γ-butyrolactone-derived complexes. Monomer-dependent divergence between post-insertion transesterification, delayed propagation and arrested insertion provides insight into ROP initiation.

Transforming Molecular Science With Large Language Models: From Molecule Understanding to Autonomous Scientific Discovery.

Large language models (LLMs) are driving a paradigm shift in molecular science, transitioning from molecule understanding to autonomous scientific discovery. By learning from multimodal molecular representations through advanced training techniques, LLMs address traditional limitations in molecular research, including expert dependency and limited experimental scalability. We analyze strategies for cross-modal alignment and domain adaptation that enable LLMs to interpret complex molecular semantics. This deep semantic understanding translates into broad versatility across key downstream applic

Molecular Pathways, Target Landscape, and Translational Models in Heart Failure with Preserved Ejection Fraction.

Heart failure with preserved ejection fraction (HFpEF) is a substantial global health burden and the greatest unmet medical need for cardiovascular diseases. It is marked by pronounced clinical heterogeneity and complex multi-system pathophysiology with limited therapeutic options. Progress in developing effective therapeutics is constrained by the inadequacy of experimental models to fully recapitulate the multifactorial nature of the disease. Recent evidence underscores the significant involvement of inflammatory, oxidative, and mitochondrial pathways in the pathogenesis of HFpEF, with non-c

Variable Selection via Fused Sparse-Group Lasso Penalized Multi-state Models Incorporating Molecular Data.

In multi-state models based on high-dimensional data, effective modeling strategies are required to determine an optimal, ideally parsimonious model. In particular, linking covariate effects across transitions is needed to conduct joint variable selection. A useful technique to reduce model complexity is to address homogeneous covariate effects for distinct transitions. We integrate this approach to data-driven variable selection by extended regularization methods within multi-state model building. We propose the fused sparse-group lasso (FSGL) penalized Cox-type regression in the framework of

Deciphering Alzheimer's disease: Molecular mechanisms, preclinical models and strategies to overcome Blood-Brain-Barrier.

Neurodegenerative diseases involve a spectrum of diseases, with Alzheimer's being the most prevalent one. Alzheimer's disease (AD) is a progressive neurodegenerative disorder marked by cognitive decline, memory loss, and synaptic dysfunction. Despite extensive research, effective therapeutic interventions remain limited, largely due to the complex molecular underpinnings of the disease and the challenge posed by the blood-brain barrier (BBB). This review explores the critical molecular mechanisms implicated in AD pathogenesis, i.e. disruption of cholinergic neurotransmission, aggregation of β-

Comparative analysis of molecular targeted radiosensitizers in 2D and 3D cancer cell line models.

BACKGROUND AND PURPOSE: Despite being a critical treatment modality, radiotherapy effectiveness is often limited by tumor resistance. Therefore, there exists a need to identify molecular targeted drugs that enhance the therapeutic response to radiation. We hypothesize that a systematic comparison of targeted radiosensitizers across two-dimensional (2D) and three-dimensional (3D) spheroid cultures will reveal context-specific differences in radiosensitivity to guide preclinical prioritization of candidate radiosensitizers. MATERIAL AND METHODS: Radiosensitizing effects of DNA-PKcs (M3814), ATR

Single-Cell Transcriptomic Analysis of Different Liver Fibrosis Models: Elucidating Molecular Distinctions and Commonalities.

Background: Liver fibrosis, a consequence of various chronic liver diseases, is characterized by excessive accumulation of extracellular matrix (ECM), leading to impaired liver function and potentially progressing to cirrhosis or hepatocellular carcinoma. The molecular mechanisms underlying liver fibrosis are complex and not fully understood. In vivo experiments are essential for studying the molecular mechanisms of the disease. However, the diverse principles behind mouse modeling techniques for liver fibrosis can complicate the elucidation of specific fibrotic mechanisms. Methods: Five disti

Redox-regulated bone loss in spaceflight and terrestrial models: Molecular mechanisms and therapeutic strategies.

Exposure to microgravity induces a rapid and profound loss of bone mass, particularly in weight-bearing skeletal regions, closely resembling accelerated osteoporosis on Earth. Traditionally attributed to mechanical unloading, bone loss in space is now recognized as being strongly regulated by oxidative stress. Excessive production of reactive oxygen species (ROS)-driven by mitochondrial dysfunction, cosmic radiation, altered circadian rhythms, and fluid shifts-disrupts osteoblast differentiation, enhances osteoclastogenesis, and compromises osteocyte viability. These effects are mediated throu

Molecular Pathways and Animal Models of Tetralogy of Fallot and Double Outlet Right Ventricle.

Tetralogy of Fallot and double-outlet right ventricle are outflow tract (OFT) alignment defects situated on a continuous disease spectrum. A myriad of upstream causes can impact on ventriculoarterial alignment that can be summarized as defects in either i) OFT elongation during looping morphogenesis or ii) OFT remodeling during cardiac septation. Embryological processes underlying these two developmental steps include deployment of second heart field cardiac progenitor cells, establishment and transmission of embryonic left/right information driving OFT rotation and OFT cushion and valve morph
