Subtractive genomics profiling for potential drug targets identification against Moraxella catarrhalis.

Moraxella catarrhalis (M. catarrhalis) is a gram-negative bacterium, responsible for major respiratory tract and middle ear infection in infants and adults. The recent emergence of the antibiotic resistance M. catarrhalis demands the prioritization of an effective drug target as a top priority. Fortunately, the failure of new drugs and host toxicity associated with traditional drug development approaches can be avoided by using an in silico subtractive genomics approach. In the current study, the advanced in silico genome subtraction approach was applied to identify potential and pathogen-spec

Identification of Moraxella lacunata from pulmonary abscesses in three zoo herbivores.

Although Moraxella lacunata causes conjunctivitis, keratitis, endocarditis, and otolaryngitis in humans, its infection is rare in animals. We report three cases of asymptomatic pulmonary abscesses caused by M. lacunata in zoo herbivores, including two elks (Cervus canadensis) and a common eland (Taurotragus oryx). In all cases, macroscopic findings included coalescence of lung lobes and severe pulmonary abscesses filled with cheese-like materials in cysts. Microscopic findings included pneumonia characterized by marked fibrin exudates in alveolar spaces and infiltration of inflammatory cells.

Novel mechanism responsible for high-level macrolide resistance in Moraxella catarrhalis.

BACKGROUND: High-level macrolide-resistant Moraxella catarrhalis strains have been isolated; however, the underlying mechanism has not been well elucidated. We investigated the role of mutations in the 23S rRNA gene and the L4 and L22 ribosomal proteins using spontaneous erythromycin-resistant mutants and transformants. MATERIALS AND METHODS: The erythromycin-susceptible M. catarrhalis ATCC25238 and clinical isolate Mc19 were used as parental strains. To obtain spontaneous erythromycin-resistant mutants, in vitro stepwise selection was performed using brain-heart infusion agar plates containi

Respiratory syncytial virus promotes Moraxella catarrhalis-induced ascending experimental otitis media.

Otitis media (OM) is a polymicrobial disease wherein prior or concurrent infection with an upper respiratory tract virus plays an essential role, predisposing the middle ear to bacterial invasion. In episodes of acute bacterial OM, respiratory syncytial virus (RSV) is the most commonly isolated virus and thus serves as an important co-pathogen. Of the predominant bacterial agents of OM, the pathogenesis of disease due to Moraxella catarrhalis is the least well understood. Rigorous study of M. catarrhalis in the context of OM has been significantly hindered by lack of an animal model. To bridge

Comparative analysis and supragenome modeling of twelve Moraxella catarrhalis clinical isolates.

BACKGROUND: M. catarrhalis is a gram-negative, gamma-proteobacterium and an opportunistic human pathogen associated with otitis media (OM) and exacerbations of chronic obstructive pulmonary disease (COPD). With direct and indirect costs for treating these conditions annually exceeding $33 billion in the United States alone, and nearly ubiquitous resistance to beta-lactam antibiotics among M. catarrhalis clinical isolates, a greater understanding of this pathogen's genome and its variability among isolates is needed. RESULTS: The genomic sequences of ten geographically and phenotypically diver

Serological cross-reactions between Moraxella (Branhamella) catarrhalis and other oropharyngeal bacteria.

Oropharyngeal bacteria belonging to different species were tested for serological cross-reactions with Moraxella catarrhalis using sera from immunized rabbits. Sera were tested using immunofluorescence, an enzyme immunoassay (EIA) and immunoblotting. On immunofluorescence, significant cross-reactions were demonstrated with beta-hemolytic streptococci group A and group G, as well as with streptococci of the viridans group. Some cross-reactions were also noted with Neisseria meningitidis. In the EIA, strong cross-reactions were demonstrated with beta-hemolytic streptococci. No cross-reactions we

Enzyme-linked immunosorbent assay for measurement of anti-Moraxella bovis antibodies.

Antibodies of the immunoglobulin (Ig) G and IgA classes specific for Moraxella bovis in serum or lacrimal secretions of cattle were detected with an enzyme-linked immunosorbent assay, using soluble extracts of M bovis or whole organisms. The amount of detectable antibodies was influenced by the type of antigenic preparation used in the enzyme-linked immunosorbent assay. The test indicated that 2 subcutaneous inoculations of M bovis cells induce significantly (P less than 0.0001) increased anti-M bovis serum antibodies. The infection of calves with M bovis induced specific IgG antibodies in ser

Multifactorial inhibition of lactobacilli against the respiratory tract pathogen Moraxella catarrhalis.

Probiotics, mainly lactic acid bacteria (LAB), are widely focused on gastrointestinal applications. However, recent microbiome studies indicate that LAB can be endogenous members of other human body sites such as the upper respiratory tract (URT). Interestingly, DNA-based microbiome research suggests an inverse correlation between the presence of LAB and the occurrence of potential pathogens, such as Moraxella catarrhalis, an important URT pathogen linked to otitis media, sinusitis and chronic obstructive pulmonary disease. However, a direct interaction between these microbes has not been expl

Outer membrane porin M35 of Moraxella catarrhalis mediates susceptibility to aminopenicillins.

BACKGROUND: The outer membrane protein M35 is a conserved porin of type 1 strains of the respiratory pathogen Moraxella catarrhalis. It was previously shown that M35 is involved in the uptake of essential nutrients required for bacterial growth and for nasal colonization in mice. The aim of this study was (i) to characterize the potential roles of M35 in the host-pathogen interactions considering the known multifunctionality of porins and (ii) to characterize the degree of conservation in the phylogenetic older subpopulation (type 2) of M. catarrhalis. RESULTS: Isogenic m35 mutants of the typ

Moraxella bovoculi sp. nov., isolated from calves with infectious bovine keratoconjunctivitis.

Eighteen isolates of a Gram-negative coccus (strain 237(T)) were cultured from the eyes of dairy and beef calves affected with infectious bovine keratoconjunctivitis (IBK; 'pinkeye') in northern California, USA, during summer 2002. These isolates had near full-length (1397 bp) 16S rRNA gene sequences that clustered into three groups with 99.9 % sequence similarity. On the basis of 16S rRNA gene sequence, the isolates were most closely associated with Moraxella bovis and Moraxella ovis in clade I of the classical moraxellae. Biochemically, the novel isolates could be distinguished from the othe

Correlation between susceptibility and BRO type enzyme of Moraxella catarrhalis strains.

Clinical isolates of Moraxella catarrhalis (76 isolates) were screened for beta-lactamase production and antibiotic susceptibility. beta-Lactamases (detected in 90.8% of isolates) were typed using isoelectric focusing to BRO-1 (87%) and BRO-2 (13%). Minor variations in electrofocusing patterns between the two types were seen. Isolates expressing BRO type enzymes showed solid resistance to penicillin, ampicillin and cephalothin, in particular BRO-1 producers. BRO-1 isolates were less susceptible to cephems and to beta-lactamase inhibitors than BRO-2 isolates. Isolates harbouring BRO-1 enzymes h

Goblet cell density in acute otitis media caused by Moraxella catarrhalis.

HYPOTHESIS AND BACKGROUND: Secretory otitis media is associated with a highly increased goblet cell density, confirming the secretory pathogenesis of this disease. Previous studies have shown that the middle ear goblet cell density, and thus the secretory capacity, are massively increased during experimental acute otitis media and at least 6 months thereafter, conceivably predisposing to the subsequent development of secretory otitis media. These studies used middle ear inoculation of either Streptococcus pneumoniae, nontypeable Haemophilus influenzae, or H. influenzae type b. The present stud

First report of Moraxella oculi in Brazil in an infectious bovine keratoconjunctivitis outbreak.

Infectious Bovine Keratoconjunctivitis (IBK) is a widespread ocular disease that affects dairy and beef cattle worldwide, caused by Gram-negative bacteria from the genus Moraxella. It is the most common eye disease in cattle, with symptoms including tearing, ocular pain, corneal opacity, photophobia, ulceration, and, in severe cases, permanent blindness. This study focused on characterizing Moraxella species in a 2022 IBK outbreak in Minas Gerais, Brazil. Ocular swabs from 18 symptomatic Holstein cattle were analyzed through colony isolation, physiological tests, and molecular techniques, incl
