[Effect of electroacupuncture on electroconvulsive shock--an autoradiographic study for opioid receptors].

Radioreceptor-binding assay combined with autoradiography technique in the brain slices of rats was used to investigate cerebral areas related to electroconvulsive shock (ECS) and to analyze changes of opioid receptors of these areas in electroacupuncture (EA) anticonvulsion. The results from image processing and detection of optic density (O.D.) showed that repeated ECS resulted in an increase of opioid receptor densities in the caudate nucleus, hippocampus, habenulae nucleus and amygdala. In the group of EA plus ECS, however, the opioid receptor densities in the hippocampus, habenula nucleus

Use of halothane to maintain anesthesia induced with etorphine in juvenile African elephants.

Sixteen 3- to 5-year-old African elephants were anesthetized one or more times for a total of 27 diagnostic and surgical procedures. Xylazine (0.1 +/- 0.04 mg/kg of body weight, mean +/- SD) and ketamine (0.6 +/- 0.13 mg/kg) administered IM induced good chemical restraint in standing juvenile elephants during a 45-minute transport period before administration of general anesthesia. After IM or IV administration of etorphine (1.9 +/- 0.56 micrograms/kg), the mean time to lateral recumbency was 20 +/- 6.6 and 3 +/- 0.0 minutes, respectively. The mean heart rate, systolic blood pressure, and resp

New Sinomenine Derivative as a Potential Drug for Cervical Cancer via Inducing Ferroptosis.

Cervical cancer remains a serious threat to women's health due to chemotherapy resistance, poor prognosis, and survival rates. Novel anti-cervical cancer agents are urgently needed. Sinomenine derivatives, which were prepared from sinomenine (a natural morphinane alkaloid), have been previously reported by us to be cytotoxic toward various cancer cell lines. Also, an increase in ROS production, which is associated with ferroptosis, was observed in cancer cells. In this study, we synthesized 18 sinomenine derivatives and evaluated their antitumor activity in vitro against four human tumor cell

Polyglutamic acid and Pluronic F127-based hydrogel for loading sinomenine hydrochloride liposomes in the treatment of rheumatoid arthritis.

Sinomenine hydrochloride (SNH), a key therapeutic agent for rheumatoid arthritis (RA), is limited by its low bioavailability, rapid metabolism, and frequent dosing requirements. This study innovatively developed a composite delivery system (SNH-Lip@PPGels) based on polyglutamic acid-poloxamer F127 thermosensitive hydrogel-encapsulated liposomes. In vitro drug release conformed to the Korsmeyer-Peppas kinetic model, demonstrating sustained-release characteristics. In vivo studies confirmed that SNH-Lip@PPGels precisely targets the joint cavity, with intra-articular retention exceeding 24 h. The

Dendrimer mediated targeted delivery of sinomenine for the treatment of acute neuroinflammation in traumatic brain injury.

Traumatic brain injury (TBI) is a significant medical problem with limited treatment options and is one of the main causes of life-long disability. Neuroinflammation orchestrated by activated microglia/macrophages at the site of injury plays a critical role in the onset of many pathological events following TBI, leading to blood brain barrier (BBB) dysfunction, neuronal damage and long term neuronal and behavioral deficits. Current treatment involves intravenous administration of anti-inflammatory drugs which have limited clinical outcomes only when dosed within the early time window after inj

Sample preparation and separation techniques for bioanalysis of morphine and related substances.

In present time the use or misuse of morphine and its derivatives are monitored by assaying the presence of the drug and its metabolites in biofluids. In the present review, focus is placed on the sample preparation and on the separation techniques used in the current best practices of bioanalysis of morphine and its major metabolites. However, as methods for testing the misuse of heroin, a morphine derivative, often involve bioanalytical methods that cover a number of other illicit drug substances, such methods are also included in the review. Furthermore, the review also includes bioanalysis

[Cumulative pharmacokinetics and clinical effects of nalbuphine in patients in the postoperative phase. An open multicenter study].

Pharmacokinetic data and analgetic potency of nalbuphine hydrochloride (Nubain) under conditions of postoperative respiratory treatment and repeated dosage was investigated in a multicenter study. Patients received 20 mg nalbuphine intravenously at intervals of 3 h. Blood samples for analysis of nalbuphine plasma concentration were taken after the first injection, immediately before further medication and after the last one. Data were fitted to 2-compartment model using non-linear fit procedures. The half-life time in plasma was slightly shorter than in other studies, while the peak plasma con

Dextrorphan and dextromethorphan attenuate hypoxic injury in neuronal culture.

The dextrorotatory opioid derivatives, dextrorphan and dextromethorphan, can attenuate hypoxic injury in cortical cell cultures. This effect is concentration-dependent in the micromolar range, and not strongly stereospecific, as it can also be demonstrated with the levorotatory enantiomer of dextrorphan, levorphanol. The possibility that these clinically available compounds may have therapeutic utility in hypoxic or ischemic encephalopathy warrants further investigation.

Forty-eight newly captured free-ranging feral stallions (Equus caballus) from two different locations and six captive stallions were immobilized using combinations of etorphine hydrochloride, xylazine hydrochloride and atropine sulfate with or without acepromazine. Six animals were immobilized twice, 1 mo apart. The drugs were administered either intramuscularly (n = 13) or intravenously (n = 44). Mean immobilization time (+/- SE) after intravenous (i.v.) injection of etorphine, xylazine and atropine was 55 +/- 4 sec (range 20 to 185 sec) compared to 708 +/- 131 sec (range 390 to 1,140 sec) fo

Time-dependent influence of some sedating agents on basic haematological values in various artio- and perissodactylids.

Basic haematological values in 32 animals of five species were estimated after administration of sedating agents. In all species under investigation, a time-dependent decrease of erythrocyte counts, haematocrit values and haemoglobin content was noted during the first 30 min after sedation, for the following 30 min the lowered values remained essentially without any change in zebras. Derived parameters, mean corpuscular haemoglobin, mean corpuscular Hb concentration and mean corpuscular volume did not change during the period of observation. Only insignificant changes in leukocyte count and in

Etorphine pharmacokinetics in the rat: experimental data and mathematical model.

Rats were injected i.v. with 3H-etorphine (200 ng/Kg). At least 85% of the dose was cleared from the blood in the first 2 min. Blood levels continued to fall slowly from about 5% of the administered dose after 15 min to less than 2% after 3 hours. Although more than 15% of the dose was found in the liver and kidney after 15 min, labeled material did not further accumulate in these organs, but decreased to about 3% after 3 hours. The concentration of labeled material (dpm/mg tissue) in cerebellum was less than half that attained in other brain regions at early time points, probably reflecting t

Natural product fragment combination to performance-diverse pseudo-natural products.

Natural product structure and fragment-based compound development inspire pseudo-natural product design through different combinations of a given natural product fragment set to compound classes expected to be chemically and biologically diverse. We describe the synthetic combination of the fragment-sized natural products quinine, quinidine, sinomenine, and griseofulvin with chromanone or indole-containing fragments to provide a 244-member pseudo-natural product collection. Cheminformatic analyses reveal that the resulting eight pseudo-natural product classes are chemically diverse and share b

Associated mood changes with naloxegol therapy for opioid-induced constipation in a patient with psychiatric disease.

The objective of this clinical case report is to highlight unusual adverse effects brought on by naloxegol therapy in a patient with underlying psychiatric illness. The patient is a 68-year-old female, with a psychiatric history of bipolar disorder, who presented for chronic pain management and opioid-induced constipation. After failing other therapies, she was trialed on naloxegol on three separate occasions. She experienced mood lability with symptoms including agitation, confusion, irritability, hysteria and unprompted crying spells on each occasion. Notably, the drug manufacturer does not

Urinary excretion of morphine and biosynthetic precursors in mice.

It has been firmly established that humans excrete a small but steady amount of the isoquinoline alkaloid morphine in their urine. It is unclear whether it is of dietary or endogenous origin. There is no doubt that a simple isoquinoline alkaloid, tetrahydropapaveroline (THP), is found in human and rodent brain as well as in human urine. This suggests a potential biogenetic relationship between both alkaloids. Unlabeled THP or [1,3,4-D(3)]-THP was injected intraperitoneally into mice and the urine was analyzed. This potential precursor was extensively metabolized (96%). Among the metabolites fo
