Mutagenic activity of vinyl compounds and derived epoxides.

Many vinyl compounds, such as vinyl chloride and some inhalational anesthetics, are known to be mutagens. In the present study, 10 vinyl compounds or derived epoxides, widely used in industry, were assayed in the Salmonella typhimurium/mammalian microsome system. 3 strains of histidine-dependent S. typhimurium, TA1535, TA98 and TA100 were used. Of the 10 compounds, 4 were mutagens. They were 9-vinylanthracene, vinylcarbazole, 3-vinyl-7-oxabicyclo[4.1.0]heptane and 3-epoxyethyl-7-oxabicyclo[4.1.0]-heptane. The study confirmed the overall genotoxicity of vinyl compounds and epoxides and the need

Mutagenic and carcinogenic effects of pesticides.

A discussion of the mutagenic and carcinogenic effects of various classes of commonly used pesticides. Classes discussed include halogenated hydrocarbons organophosphorus insecticides, and certain herbicides and fungicides. Halogenated hydrocarbons were found generally hazardous, causing sterility and displaying marginal or definite carcinogenic activity. Several organophosphorus insecticides have been shown to be mutagens; some have been shown to be carcinogens. Both the herbicide impurity tetrachlorodibenzodioxin, TCDD, and maleic hydrazide have been shown to be carcinogenic in mice and rats

On the pathogenesis of erythroleukaemia (H0493).

In erythroleukaemia megaloblastic changes can co-exist with leukaemic changes in the marrow. The cause of the disease must therefore be such as can cause megaloblastosis and at the same time be mutagenic. Failure of the thymidylate synthelase reaction, the commonest cause of megaloblastic anaemia, can be eliminated in erythroleukaemia because (a) the dU suppression test is normal in the disease and (b) failure of the thymidylate synthelase reaction is not mutagenic. The deamination of both cytosine and adenine is mutagenic but the deamination of cytosine alone is apparent and the nucleotide of

Use of in silico and in vitro methods as a potential new approach methodologies (NAMs) for (photo)mutagenicity and phototoxicity risk assessment of agrochemicals.

The increased use of agrochemicals raises concerns about environmental, animal, and mainly human toxicology. The development of New Approach Methodologies (NAMs) for toxicological risk assessment including new in vitro tests and in silico protocols is encouraged. Although agrochemical mutagenicity testing is well established, a complementary alternative approach may contribute to increasing reliability, with the consequent reduction of false-positive results that lead to unnecessary use of animals in follow-up in vivo testing. Additionally, it is unreasonable to underestimate the phototoxic ef

Formation of mutagenic/carcinogenic heterocyclic amines under moderate conditions.

More than 10 kinds of heterocyclic amines (HCAs), showing mutagenic and carcinogenic potency, have been isolated from cooked fish and meat. But many researchers say that the contribution ratio of HCAs to human cancer is very low. Our purpose in this experiment was to investigate the possibility of the formation of HCAs under moderate conditions, including in vivo. A mixture of d-glucose, creatinine, and amino acid such as glycine, methionine, threonine, and proline was dissolved in phosphate-buffered solution (pH7.4) and incubated at 37 degrees C, 50 degrees C, 128 degrees C. At an appropriate

Cytotoxic, genotoxic/antigenotoxic and mutagenic/antimutagenic effects of the venom of the wasp Polybia paulista.

Hymenoptera venoms are constituted by a complex mixture of chemically or pharmacologically bioactive agents, such as phospholipases, hyaluronidases and mastoparans. Venoms can also contain substances that are able to inhibit and/or diminish the genotoxic or mutagenic action of other compounds that are capable of promoting damages in the genetic material. Thus, the present study aimed to assess the effect of the venom of Polybia paulista, a neotropical wasp, by assays with HepG2 cells maintained in culture. The cytotoxic potential of the wasp venom, assessed by the methyl thiazolyl tetrazolium

Ecotoxicity and Mutagenicity Assessment of Novel Antifungal Agents VT-1161 and T-2307.

Antifungal substances are essential for managing fungal infections in humans, animals, and plants, and their usage has significantly increased due to the global rise in fungal infections. However, the extensive application of antifungal agents in pharmaceuticals, personal care products, and agriculture has led to their widespread environmental dissemination through various pathways, such as excretion, improper disposal, and agricultural runoff. Despite advances in wastewater treatment, many antifungal compounds persist in the environment, affecting non-target organisms and contributing to resi

A study of the mutagenicity of anesthetics and their metabolites.

The commonly used volatile anesthetics, several of their metabolites, and drugs frequently employed by the anesthesiologist were screened for mutagenicity in the Salmonella/rat-liver microsomal assay system developed by Dr. B. Ames and his colleagues. Chloral hydrate, both a sedative and metabolite of trichloroethylene, was found to be weakly mutagenic. Other compounds testing including halothane, isoflurane, methoxyflurane, diazepam and chlordiazepoxide were not mutagenic. Non-volatile compounds were tested for their ability to inhibit growth of bacterial strains with decreased capacity to re

Oxidative mutagens specific for A-T base pairs induce forward mutations to L-arabinose resistance in Salmonella typhimurium.

Strain TA102 of S. typhimurium is a new histidine-requiring mutant, particularly suited to the detection of oxidative mutagens acting at A.T base pairs. 10 oxidizing chemicals, previously tested in strain TA102, were used to evaluate the mutagenic sensitivity of the L-arabinose forward mutation assay of S. typhimurium with respect to those types of mutagens. The mutagenicity of each compound was determined by liquid test, measuring both the frequency of mutants among the survivors and the absolute number of mutants growing in selective plates with traces of D-glucose. Strain BA13 with a wild-t

The importance of glutathione and glutathione transferase for somatic mutations in Drosophila melanogaster induced in vivo by 1,2-dichloroethane.

Two principal pathways of metabolism of the carcinogenic compound 1,2-dichloroethane (DCE) have been proposed. One is a mixed function oxidase dependent pathway requiring oxygen and NADPH. The other pathway depends on the presence of glutathione (GSH) and glutathione transferase (GST). The aim of this study was to investigate the role of the latter pathway for the in vivo mutagenicity of DCE in the somatic wing spot test in Drosophila melanogaster. DCE caused a dose-dependent increase of wing spots. In order to investigate the role of cellular GSH for the mutagenicity, the level of GSH was dec

Modulation of the potency of promutagens and direct acting mutagens in bacteria by inhibitors of the multidrug resistance mechanism.

Multiple drug resistance (MDR) mechanisms are known to limit the effectiveness of some cancer chemotherapies, probably through enhancing P-glycoprotein-mediated drug efflux from mammalian cells. Similar mechanisms appear to act in other organisms, including bacteria, and may affect not only the toxicity but also the mutagenicity of certain chemicals. At least in some experimental situations, MDR can be overcome through concomitant treatment of the cells with various types of inhibitors. Two MDR inhibitors, verapamil, a calcium channel blocker, and trifluoperazine, a calmodulin inhibitor, were

Mutagenicity of fine (less than 2.5 microns) airborne particles: diurnal variation in community air determined by a Salmonella micro preincubation (microsuspension) procedure.

A simple modification of the Salmonella liquid incubation assay previously developed for detecting mutagens in urine was used to determine mutagenic activity of airborne particulate matter. The modification consists of adding ten times more bacteria (approximately 10(9) per incubation tube) and five to ten times less metabolic enzymes compared to the plate incorporation method. The mixture volume is approximately 0.2 ml, and the mixture is incubated for 90 min before pouring it according to the standard protocol. The modified procedure (micro preincubation or microsuspension) was approximately

Mutagenicity of inhalation anaesthetics: trichloroethylene, divinyl ether, nitrous oxide and cyclopropane.

The mutagenic potential of trichloroethylene, divinyl ether, nitrous oxide and cyclopropane was assessed in vitro by microbial assay employing two histidine-dependent strains of Salmonella typhimurium, TA1535 and TA100. Anaesthetic agents in various concentrations were incubated with bacteria in the presence or absence of an enzyme system prepared from enzyme-induced rat liver. Nitrous oxide and cyclopropane were not mutagenic, whereas divinyl ether gave a strongly positive response. Results for trichloroethylene were equivocal. These and previous studies with the salmonella system, together w

Strong mutagenic effects of diesel engine emissions using vegetable oil as fuel.

Diesel engine emissions (DEE) are classified as probably carcinogenic to humans. In recent years every effort was made to reduce DEE and their content of carcinogenic and mutagenic polycyclic aromatic compounds. Since 1995 we observed an appreciable reduction of mutagenicity of DEE driven by reformulated or newly designed fuels in several studies. Recently, the use of rapeseed oil as fuel for diesel engines is rapidly growing among German transportation businesses and agriculture due to economic reasons. We compared the mutagenic effects of DEE from two different batches of rapeseed oil (RSO)

Comparison of chromosome aberration and micronucleus induction in human lymphocytes after occupational exposure to vinyl chloride monomer and microwave radiation.

Differences and similarities between the mutagenicity of microwave radiation and VCM were studied on the lymphocytes of persons occupationally exposed to either of these two clastogens. The results of the micronucleus test and the chromosome aberration assay were compared. Our findings provided evidence that the mutagenicity of both VCM and microwave radiation can be detected with both of these tests. However, we detected both quantitative and qualitative differences between VCM and microwave radiation in the induction of aberrations and micronuclei. VCM causes quantitatively more chromosome d

Contact allergy as a therapeutic tool for alopecia areata: application of squaric acid dibutylester.

Squaric acid dibutylester (SADBE), a potent contact allergen, was tested for mutagenicity in the bacterial plate incorporation assay (Ames test), in the presence and absence of mammalian microsomes. In contrast to dinitrochlorobenzene which is mutagenic in this test, SADBE was found not to be mutagenic. In 53 patients with extensive or total alopecia areata, SADBE dissolved in acetone was applied weekly to one side of the head, the other side serving as control. In 46 patients (87%), hair regrew either exclusively on the treated side, or considerably faster and denser on this side. In some pat

UV and ethyl methanesulfonate effects in hyperthermophilic archaea and isolation of auxotrophic mutants of Pyrococcus strains.

The lethal and mutagenic effects of ethyl methanesulfonate (EMS) and UV on nine archaeal strains belonging to each of the two described genera of Thermococcales, Pyrococcus and Thermococcus, were investigated. To test the efficiency of the EMS and UV mutagenesis under a variety of experimental conditions, we chose Pyrococcus abyssi strain GE5 as a model strain. We observed a strong induced mutagenicity in both cases, since the spontaneous mutation frequency (expressed as the frequency of resistance to 5-fluoroorotic acid) increased up to 150-fold with EMS and 400-fold with UV, after mutagen ex
