Siblings in Kars, Turkey, with Uner Tan syndrome (quadrupedal locomotion, severe mental retardation, and no speech): a novel theory for the evolution of human bipedalism.

OBJECTIVE: To investigate siblings from Kars (n  =  2), Turkey, with diagonal-sequence quadrupedal locomotion (QL), severe mental retardation, and no speech (Uner Tan syndrome, UTS), in relation to the evolutionary emergence of human bipedal locomotion (BL). METHODS: Video recordings were made to assess gaits. Brain MRI scanning was performed to visualize the cerebro-cerebellar malformations. Genome-wide association analyses were performed in venous blood samples. RESULTS: One of the two men with UTS showed early-onset QL and late-onset BL without infantile hypotonia, the other consistent QL

Turnover of sex chromosomes in the Lake Tanganyika cichlid tribe Tropheini (Teleostei: Cichlidae).

Sex chromosome replacement is frequent in many vertebrate clades, including fish, frogs, and lizards. In order to understand the mechanisms responsible for sex chromosome turnover and the early stages of sex chromosome divergence, it is necessary to study lineages with recently evolved sex chromosomes. Here we examine sex chromosome evolution in a group of African cichlid fishes (tribe Tropheini) which began to diverge from one another less than 4 MYA. We have evidence for a previously unknown sex chromosome system, and preliminary indications of several additional systems not previously repor

DNA barcoding of Iranian radicine freshwater snails begins to untangle the taxonomy and phylogeography of intermediate hosts of schistosomiasis and fasciolosis from the Middle East and across Central Asia.

In the Middle East radicine snails are of considerable medical and veterinary importance acting as vectors of trematodes. In Iran, such snails are responsible for the transmission of the zoonotic trematodes Schistosoma turkestanicum and Fasciola gigantica. Historically, Radix gedrosiana has been incriminated as an important intermediate host for both trematodes, however, controversy remains over the snail's true taxonomic status. This species has been determined using morphological characters that has resulted in erroneous identification of species, affecting understanding of population biolog

Origin of a major infectious disease in vertebrates: The timing of Cryptosporidium evolution and its hosts.

Protozoan parasites of the genus Cryptosporidium infect all vertebrate groups and display some host specificity in their infections. It is therefore possible to assume that Cryptosporidium parasites evolved intimately aside with vertebrate lineages. Here we propose a scenario of Cryptosporidium-Vertebrata coevolution testing the hypothesis that the origin of Cryptosporidium parasites follows that of the origin of modern vertebrates. We use calibrated molecular clocks and cophylogeny analyses to provide and compare age estimates and patterns of association between these clades. Our study provid

The shoulder girdle of early chondrichthyans grew by skeletal remodelling.

A distinct shoulder region, defined by endoskeletal and dermal girdles and associated pectoral musculature, is a major evolutionary adaptation of jawed vertebrates. In teleost model species, the large (macromeric) pectoral dermal bones can be derived from multiple embryonic tissues, identifying the shoulder of osteichthyans as a developmentally complex area at the head-trunk boundary. The absence of bone in living chondrichthyans makes Palaeozoic stem groups capable of dermal ossification key to understanding the underpinnings of skeletal growth in the shoulder of crown gnathostomes (osteichth

On the evolution of the morphology and resilience of molar cusps in fossil hominid teeth.

Teeth play an important role in evolutionary studies due to their good preservation and direct link to diet. The present work makes use of a previously generated database on molar teeth of fossil hominids which consists of cuspal enamel thickness dc, dentin horn angle φ and section width D, all measured on a given histological tooth section. These data are here interpreted with the aid of "fracture stress" QF = PF/D2 and geological age t, where PF is the occlusal force needed to cause cusp failure as determined from dc and φ. QF is virtually a constant in non-hominins ("apes") while monotonica

Genomics and evolution of Pneumocystis species.

The genus Pneumocystis comprises highly diversified fungal species that cause severe pneumonia in individuals with a deficient immune system. These fungi infect exclusively mammals and present a strict host species specificity. These species have co-diverged with their hosts for long periods of time (> 100 MYA). Details of their biology and evolution are fragmentary mainly because of a lack of an established long-term culture system. Recent genomic advances have unlocked new areas of research and allow new hypotheses to be tested. We review here new findings of the genomic studies in relation

A cretaceous fly trap? remarkable abdominal modification in a fossil wasp.

BACKGROUND: Carnivorous insects have evolved a range of prey and host capture mechanisms. However, insect predation strategies in the fossil record remain poorly understood. RESULTS: Here, we describe †Sirenobethylus charybdis n. gen. & sp., based on sixteen adult female wasps in Kachin amber from the mid-Cretaceous, 99 Mya (million years ago), and place it in Chrysidoidea: †Sirenobethylidae n. fam. The fossils display unique morphological modifications on the tip of the abdomen consisting of three flaps from the modified abdominal sternum 6 and tergum and sternum 7; the lower flap formed fro

Analysis of Amino Acid Mutations of the Foot-and-Mouth Disease Virus Serotype O Using both Heparan Sulfate and JMJD6 Receptors.

Foot-and-mouth disease (FMD) is an economically devastating animal disease. Adapting the field virus to cells is critical to the vaccine production of FMD viruses (FMDV), and heparan sulfate (HS) and Jumonji C-domain-containing protein 6 (JMJD6) are alternative receptors of cell-adapted FMDV. We performed serial passages of FMDV O/SKR/Andong/2010, classified as the O/Mya-98 topotype/lineage and known as a highly virulent strain, to develop a vaccine seed virus. We traced changes in the amino acid sequences of the P1 region, plaque phenotypes, and the receptor usage of the viruses, and then str

Three Blind Moles: Molecular Evolutionary Insights on the Tempo and Mode of Convergent Eye Degeneration in Notoryctes typhlops (Southern Marsupial Mole) and Two Chrysochlorids (Golden Moles).

Golden moles (Chrysochloridae) and marsupial moles (Notoryctidae) are textbook examples of convergent evolution. Both taxa are highly adapted to subterranean lifestyles and have powerful limbs for digging through the soil/sand, ears that are adapted for low-frequency hearing, vestigial eyes that are covered by skin and fur, and the absence of optic nerve connections between the eyes and the brain. The eyes of marsupial moles also lack a lens as well as retinal rods and cones. Two hypotheses have been proposed to account for the greater degeneracy of the eyes of marsupial moles than golden mole

Tracing the origins of Antarctic periwinkles: an integrative morphological-molecular perspective on Laevilacunaria evolution.

In biogeography, related lineages often share similar range sizes and species richness due to inherited traits and shared geographic history. This expectation is based on phylogenetic niche conservatism and shared dispersal constraints. However, within the subfamily Laevilitorininae, this pattern is not observed: Laevilitorina shows broader distributions and higher species richness than Laevilacunaria despite similar developmental mode and habitat preferences. Combining multilocus genetic data, radular morphology and model-based historical biogeography, we aim to understand the origins of thes

Polyploidy meets the Cape: Evolutionary dynamics of Heliophila (Brassicaceae).

The Cape Floristic Region (CFR), a global biodiversity hotspot, provides an exceptional setting for studying plant diversification. The South African genus Heliophila (Brassicaceae), notable for its species richness and morphological diversity, offers a unique system for examining the evolutionary role of polyploidy. Using an integrative framework combining phylogenomics, cytogenetics, and floral pigment biochemistry, we reconstructed the genus's evolutionary history based on ITS and 48 nuclear markers from 392 accessions representing over 120 taxa. Our analyses resolved four major clades orig

Sodium new houttuyfonate inhibits the growth of Trichophyton spp. by potentially inducing oxidative stress and metabolic disruption.

INTRODUCTION: Dermatophytes are a group of common pathogenic fungi that infect the skin, nails, and hair of humans and other animals, affecting approximately 25% of the global human population. Their antifungal resistance has intensified, underscoring the urgent need for novel antifungal agents. Sodium new houttuyfonate (SNH), a derivative of the Houttuynia cordata Thunb., a plant native to Southeast Asian, shows broad-spectrum antimicrobial activity, but its effects on dermatophytes are unclear. METHODS: The in vitro activity of SNH against Trichophyton mentagrophytes standard strain ATCC MY

Candida dubliniensis fungemia in a patient with severe COVID-19: A case report.

Candida dubliniensis phenotypically mimics Candida albicans in its microbiological features; thus, its clinical characteristics have yet to be fully elucidated. Here we report the case of a 68-year-old Japanese man who developed C. dubliniensis fungemia during treatment for severe coronavirus disease 2019 (COVID-19). The patient was intubated and received a combination of immunosuppressants, including high-dose methylprednisolone and two doses of tocilizumab, as well as remdesivir, intravenous heparin, and ceftriaxone. A blood culture on admission day 11 revealed Candida species, which was con

Genome and population evolution and environmental adaptation of Glyptosternon maculatum on the Qinghai-Tibet Plateau.

Persistent uplift means the Qinghai-Tibet Plateau (QTP) is an ideal natural laboratory to investigate genome evolution and adaptation within highland environments. However, how paleogeographic and paleoclimatic events influence the genome and population of endemic fish species remains unclear. Glyptosternon maculatum is an ancient endemic fish found on the QTP and the only critically endangered species in the Sisoridae family. Here, we found that major transposons in the G. maculatum genome showed episodic bursts, consistent with contemporaneous geological and climatic events during the QTP fo

Impact of Chromosomal Rearrangements on the Interpretation of Lupin Karyotype Evolution.

Plant genome evolution can be very complex and challenging to describe, even within a genus. Mechanisms that underlie genome variation are complex and can include whole-genome duplications, gene duplication and/or loss, and, importantly, multiple chromosomal rearrangements. Lupins (Lupinus) diverged from other legumes approximately 60 mya. In contrast to New World lupins, Old World lupins show high variability not only for chromosome numbers (2n = 32⁻52), but also for the basic chromosome number (x = 5⁻9, 13) and genome size. The evolutionary basis that underlies the karyotype evolution in lup

In vitro antifungal synergy between amphiphilic aminoglycoside K20 and azoles against Candida species and Cryptococcus neoformans.

Several azoles are widely used to treat human fungal infections. Increasing resistance to these azoles has prompted exploration of their synergistic antifungal activities when combined with other agents. The amphiphilic aminoglycoside, K20, was recently shown to inhibit filamentous fungi, yeasts and heterokonts, but not bacteria. In this study, in vitro synergistic growth inhibition by combinations of K20 and azoles (fluconazole, itraconazole, voriconazole, clotrimazole, or posaconazole) were examined against Candida species and Cryptococcus neoformans. Evidence for a population expansion in the West Nile virus vector Culex tarsalis.

Population genetic structure of the West Nile Virus vector Culex tarsalis was investigated in 5 states in the western United States using 5 microsatellite loci and a fragment of the mitochondrial reduced form of nicotinamide adenine dinucleotide dehydrogenase 4 (ND4) gene. ND4 sequence analysis revealed a lack of isolation by distance, panmixia across all populations, an excess of rare haplotypes, and a star-like phylogeny. Microsatellites revealed moderate genetic differentiation and isolation by distance, with the largest genetic distance occurring between populations in southern California

A Young ahsg/fetuin-a Inactive Retrocopy Reflects Recent Retrotransposon Activity in the Xenopus laevis Lineage.

The vertebrate ahsg (alpha 2-HS glycoprotein, also coined fetuin-a) homologs are highly expressed in the liver, and their secreted protein products exert complex systemic effects, including the regulation of biomineralization of soft and skeletal tissues. Here, we report a previously uncharacterized ahsg retrocopy in the allotetraploid frog species Xenopus laevis. We show that this young retrocopy was born from the ahsg.L homeologue less than 10 Mya, and landed in the S subgenome in a locus located between asic2.S and smarcd2.S. The ahsg.L-retrocopy ends with a poly(A) tail, is intronless, and

Does Amyotrophic Lateral Sclerosis (ALS) Have Metabolic Causes from Human Evolution?

As so many drugs have failed in ALS a new approach is needed. The author proposes that recent human genetic variants may play major roles in the disease, changing metabolism. Evolution of hominins was accelerated 3-2.5 Mya, by cytidine monophospho-N-acetylneuraminic acid hydroxylase (CMAH) becoming a unitary pseudogene after a pathogenic infection, changing the sialome, and hence metabolism, brain development and neuromuscular junctions (NMJs). This was when hominins evolved to run in Africa and develop bigger brains. Deletion of CMAH in mice allows them to run for longer (~50%). The enzyme CM
