BACKGROUND: Tuberculosis currently represents a serious problem in prison populations.
METHODS: With the aim of studying the predictive factors for, and the prevalence of, Mycobacterium tuberculosis infection and pulmonary tuberculosis in a Spanish prison, all those admitted during 1991 and 1992 were included (N = 1314). The tuberculin skin test, HIV serology, chest X-ray and bacteriological examination of sputum were carried out. Statistical analysis was done by univariant tests, stratified analysis and logistic regression.
RESULTS: The prevalence of M. tuberculosis infection was 55.5% (95%

Mycobacterium tuberculosis (M. tuberculosis) and Mycobacterium abscessus (M. abscessus) are important pathogens that can cause lung diseases. Given the abundance of shared antigens between these two pathogens, evaluating the cross-immunization between Mycobacterium tuberculosis and Mycobacterium abscessus has implications for the assessment of tuberculosis vaccines based on nontuberculous mycobacteria (NTM). The whole-cell proteins of Mycobacterium abscessus were lysed via ultrasonication and then were subcutaneously injected into BALB/c mice either alone or mixed with adjuvant for three times

Tuberculosis is a highly infectious disease and of high incidence in low-income countries that is caused by Mycobacterium tuberculosis (M. tuberculosis). M. tuberculosis can form biofilms in vitro and in vivo, and the cells in the biofilm can survive at high concentrations of antibiotics. CwlM is a peptidoglycan hydrolase (amidase) and can hydrolyze bacterial cell walls, and the effects of CwlM on autolysis and biofilms is worthy of in-depth study. In this study, we successfully constructed an in vitro biofilm model of M. tuberculosis and Mycobacterium smegmatis (M. smegmatis). Reverse transcr

The Mycobacterium bovis P55 gene, located downstream from the gene that encodes the immunogenic lipoprotein P27, has been characterized. The gene was identical to the open reading frame of the Rv1410c gene in the genome of Mycobacterium tuberculosis H37Rv, annotated as a probable drug efflux protein. Genes similar to P55 were present in all species of the M. tuberculosis complex and other mycobacteria such as Mycobacterium leprae and Mycobacterium avium. By Western blotting, P55 was located in the membrane fraction of M. bovis. When transformed into Mycobacterium smegmatis after cloning, P55 c

It is an acceptable medical practice to use second-line antimycobacterial drugs in combination with isoniazid in treatment of isoniazid-resistant tuberculosis. Recent investigations have demonstrated the importance of determining chemotherapeutic interaction in instances of multiple antibiotic use. We studied the inhibitory effect of combinations of isoniazid with ethambutol, rifampin, ethionamide, cycloserine, viomycin, and kanamycin against three isoniazid-resistant strains of Mycobacterium tuberculosis and three strains of M. fortuitum. The isobologram technique with drug concentrations of

Tuberculosis (TB) is an infectious disease that affects humans and animals. The pathogens that cause TB belong to the Mycobacterium tuberculosis complex (MTBC), with M. tuberculosis and Mycobacterium bovis as the main representatives of human- and animal-adapted strains, respectively. One key genetic regulator of the MTBC members is the PhoPR system, which controls many processes, including the stress response, lipid metabolism and pathogenesis, among others. Previous studies identified a key G71I substitution in the M. bovis PhoR orthologue relative to M. tuberculosis PhoR and suggested that

Zoonotic tuberculosis (TB) caused by Mycobacterium bovis (Mbo) is a neglected disease that hinders efforts to eradicate human tuberculosis. Developing a rapid, high-throughput diagnostic test to distinguish Mbo from Mycobacterium tuberculosis (Mtb) isolates could enhance global zoonotic TB diagnostics and surveillance. This study aimed to evaluate the ability of Fourier Transform Infrared Spectroscopy (FT-IRS), using the IR Biotyper® system, to differentiate clinical isolates of Mbo and Mtb. Two bacterial inactivation protocols, paraformaldehyde and heat, were tested using Mtb and Bacillus Cal

BACKGROUND: Mycobacterium tuberculosis complex (MTBC), the causative agent of tuberculosis (TB), is composed of eight subspecies. TB in West Africa, in contrast to other geographical regions, is caused by Mycobacterium africanum (MAF) in addition to M. tuberculosis (MTB), with both infections presenting similar symptoms. Nevertheless, MAF is considered to be hypovirulent in comparison with MTB and less likely to progress to active disease. In this study, we asked whether MAF and MTB infected patients possess distinct intestinal microbiomes and characterized how these microbiota communities are

Mycobacterium kansasii is an uncommon cause of infection of the hand. Other atypical mycobacteria that cause hand infections are M marinium, M avium, M intracellularis, and M chelonei. Such infections usually occur around aquatic areas, though sometimes the source of infection is elusive. Inoculation of the atypical mycobacterium into the host occurs usually from a traumatic break in the skin. Patients commonly report a history of fish tank cleaning, oyster shucking, swinuning, or other aquatic activities. Several drug regimenshave been suggested and used successfully. Surgical intervention is

BACKGROUND: Multi-drug resistance (MDR) TB is defined as tuberculosis (TB) disease caused by a strain of Mycobacterium tuberculosis (MTB) that was resistant to at least isoniazid and rifampicin (RIF). Emerging Multidrug-Resistant TB is one of the major concerns of health policy and rapid detection of M. tuberculosis and detection of RIF resistance in infected patients are essential for disease management. The aim of this study was to evaluate patterns of RIF resistance in cases of sputum positive pulmonary TB by using GeneXpert MTB/RIF and comparing between phenotypic and genotypic testing of

BACKGROUND: Recent Mycobacterium tuberculosis infection (MTB) carries a higher risk of progression to active tuberculosis (TB) than MTB infection, but the immune features that distinguish these stages remain unclear.
METHODS: In this multicenter observational study in Jiangsu Province, China, we enrolled 228 participants with MTB infection (n = 88), recent infection (n = 44), or active TB (n = 96). Twelve plasma cytokines and chemokines were quantified using a multiplex Luminex platform. Group differences were assessed using non-parametric tests with a two-stage false discovery rate (FDR) cor

Sarcoidosis is a granulomatous systemic inflammatory disease predominantly affecting the lungs. It shares histopathological, clinical, and immune features with tuberculosis (TB). There are currently no diagnostic tests to formally identify sarcoidosis; instead, there is a need first to rule out the presence of other diseases, including TB. We hypothesized that Mycobacterium tuberculosis (Mtb)-specific immune signatures differ between sarcoidosis and TB. We characterized T-cell and monocyte signatures after Mtb antigen in vitro stimulation in the blood of patients with sarcoidosis compared to p

Tuberculosis (TB) is an infectious disease induced by Mycobacterium tuberculosis (Mtb). TB continues to pose a significant global health concern owing to limitation in vaccination effectiveness, diagnostic accuracy, and extended treatment regimen that foster drug resistance. Mtb dormancy is a non-replicative, metabolically modified condition that enables the bacterium to survive inside host macrophages, resulting in latent TB infection for years. This survival strategy is activated by stressors like hypoxia and low pH, resulting in phenotypic drug resistance and the capacity to reactivate when

Tuberculosis (TB) is a health concern resulting in 10.8 million cases every year. In 2024, TB had the highest fatalities second only to COVID-19. Global TB management has become difficult due to rise of multi-drug resistant (MDR) and extensively drug-resistant (XDR) TB. Therefore, novel approaches must be developed to fight with M.tb infection. Studies on many strategies like pathogen-centric, host-directed therapeutics, antimicrobial peptides, nano-based drug delivery, and more are still being explored to develop novel therapeutics. One such important target i.e., pathogen associated Methyltr

The heterogeneity of outcomes after Mycobacterium tuberculosis (Mtb) exposure is a conundrum associated with millennia of host-pathogen co-evolution. We hypothesized that human myeloid cells contain genetically encoded, Mtb-specific responses that regulate critical steps in tuberculosis (TB) pathogenesis. We mapped genome-wide expression quantitative trait loci (eQTLs) in Mtb-infected monocytes with RNAseq from 80 Ugandan household contacts of pulmonary TB cases to identify monocyte-specific, Mtb-dependent eQTLs and their association with cytokine expression and clinical resistance to tubercul

BACKGROUND: Tuberculosis (TB), an infectious disease caused by Mycobacterium tuberculosis, has plagued humans since the early middle-ages. More than one million deaths are recorded annually due to TB, even in present times. These deaths are primarily attributed to the constant appearance of resistant TB strains. Even with the advent of new therapeutics and diagnostics techniques, tuberculosis remains challenging to control due to resistant M. tuberculosis strains. Aided by various molecular changes, these strains adapt to stress created by anti-tuberculosis drugs.

OBJECTIVES: IFNγ-release assays (IGRAs) used for diagnosis of Mycobacterium (M.) tuberculosis infection have limited sensitivity. Alternative cytokines and M. tuberculosis latency-associated antigens may improve immune-based tests.
METHODS: Multiplex cytokine analyses was done in culture supernatants after 6-day in vitro restimulation with M. tuberculosis IGRA and latency-associated antigens (i.e. Rv2628, Rv1733) in tuberculosis patients (n = 22) and asymptomatic contacts (AC)s (n = 20) from Ghana.
RESULTS: Four cytokines (i.e. IFNγ, IP-10, IL-22 and IL-6) were significantly increased after

Alveolar macrophages (AMs) are the first immune cells to encounter Mycobacterium tuberculosis (Mtb) in the lungs, but they frequently fail to eliminate this causative agent of tuberculosis (TB), allowing Mtb to persist or replicate. Interstitial macrophages (IMs) are recruited to restrict Mtb growth and limit immune evasion. While IMs have been implicated in the control of acute Mtb infection, their role during latent tuberculosis infection (LTBI) has not yet been explored. We hypothesized that IMs contribute to maintaining latency and that their depletion during LTBI would promote Mtb reactiv

We report on a female patient suffering from tuberculosis cutis colliquativa. The skin lesions were caused by hematogenic dissemination of mycobacteria tuberculosis spreading from the affected urogenital tract. The diagnosis was confirmed by culturing. Combined treatment with tuberculostatic agents for 5 months resulted in complete remission of the tuberculosis.
