The pineal gland secretes melatonin principally at night. Regulated by norepinephrine released from sympathetic nerve terminals, adrenergic receptors on pinealocytes activate aralkylamine N-acetyltransferase that converts 5-hydroxytryptamine (5-HT, serotonin) to N-acetylserotonin, the precursor of melatonin. Previous studies from our group and others reveal significant constitutive secretion of 5-HT from pinealocytes. Here, using mass spectrometry, we demonstrated that the 5-HT is secreted primarily via a decynium-22-sensitive equilibrative plasma membrane monoamine transporter instead of by t

O-Acetylation is a common decoration on endotoxins derived from many Gram-negative bacterial species, and it has been shown to be instrumental (e.g. in Salmonella typhimurium) in determining the final tertiary structure of the endotoxin and the immunogenicity of the molecule. Structural heterogeneity of endotoxins produced by mucosal pathogens such as Neisseria meningitidis is determined by decorations on the heptose inner core, including O-acetylation of the terminal N-acetylglucosamine (GlcNAc) attached to HepII. In this report, we show that O-acetylation of the meningococcal lipooligosaccha

Atherosclerosis (AS) is the leading cause of cardiovascular disease-related mortality worldwide and serves as the core pathological basis for cardiovascular events. Protein acetylation, a widespread and highly dynamic post-translational modification, has emerged as a critical link connecting epigenetic regulation, metabolic homeostasis, and inflammatory signaling, thereby playing an important role in both the initiation and progression of AS. This review systematically summarizes the major forms of protein acetylation, including N-terminal acetylation and lysine acetylation, as well as the key

Oral squamous cell carcinoma (OSCC) is the eighth most common malignancy worldwide and the predominant type of head and neck cancer. N-α-acetyltransferase 40 (NAA40), a histone acetyltransferase specifically modifying histones H4 and H2A, has been implicated in oncogenic processes across multiple tumor types. However, its role in OSCC remains unexplored. This study aimed to investigate the functional significance of NAA40 in OSCC and delineate its underlying molecular mechanisms. Bioinformatics analyses were performed to evaluate NAA40 expression and its prognostic value in OSCC tissues. Quant

Bacterial resistance to β-lactam antibiotics mediated by β-lactamase enzymes is widespread worldwide. Chromobacterium violaceum, an environmental Gram-negative bacterial pathogen, is intrinsically resistant to some β-lactam antibiotics. In this work, we found that mutations in an ampD gene, encoding a peptidoglycan-recycling amidase, cause hyperproduction of two chromosomal β-lactamases (AmpC and CmcB), conferring high β-lactam resistance in C. violaceum. Susceptibility tests using ΔampC, ΔcmcB, and ΔcmcBΔampC mutant strains revealed specific susceptibility profiles to penicillin, cephalospori

To map presynaptic cholinergic terminal densities in normal aging (n = 36), Alzheimer's disease (AD) (n = 22), and Parkinson's disease (PD) (n = 15), we performed single-photon emission computed tomography using [123I]iodobenzovesamicol (IBVM), an in vivo marker of the vesicular acetylcholine transporter. We used coregistered positron emission tomography with [18F]fluorodeoxyglucose for metabolic assessment and coregistered magnetic resonance imaging for atrophy assessment. In controls (age, 22-91 years), cortical IBVM binding declined only 3.7% per decade. In AD, cortical binding correlated i

Cysteine is implicated in important biological processes. It is synthesized through two different pathways. Cystathionine β-synthase and cystathionine γ-lyase participate in the reverse transsulfuration pathway, while serine acetyltransferase and cysteine synthase function in the de novo pathway. Two evolutionarily related pyridoxal 5'-phosphate-dependent enzymes, cystathionine β-synthase TtCBS1 (TTHERM_00558300) and cysteine synthase TtCSA1 (TTHERM_00239430), were identified from a freshwater protozoan Tetrahymena thermophila. TtCbs1 contained the N-terminal heme binding domain, catalytic dom

Glioma is the most common primary tumor in the central nervous system, with glioblastoma (GBM) representing one of the most malignant forms, accounting for 46.1% of cases. GBM is characterized by rapid progression, high malignancy, and poor prognosis, with median survival remaining less than 15 months despite combined surgery, radiotherapy, and chemotherapy. NAT10, the only known acetyltransferase mediating N4-acetylcytidine (ac4C) modification in eukaryotes, has been implicated in promoting tumorigenesis and progression in colon cancer, bladder cancer, pancreatic ductal cancer, among others.

Mucopolysaccharidosis (MPS) IIIC is an autosomal recessive lysosomal storage disorder caused by a deficiency in heparan acetyl CoA: alpha-glucosaminide N-acetyltransferase (HGSNAT). The characteristic feature is the deterioration of the central nervous system, but other symptoms may include coarse facies, developmental delay, macrocrania and motor retardation. HGSNAT is localised to the lysosomal membrane and catalyses a transmembrane acetylation in which the terminal glucosamine residue of heparan sulphate acquires an acetyl group, thus forming N-acetylglucosamine. 54 variants of the HGSNAT g

The expression of O-acetylated sialic acids in human colonic mucins is developmentally regulated, and a reduction of O-acetylation has been found to be associated with the early stages of colorectal cancer. Despite this, however, little is known about the enzymatic process of sialic acid O-acetylation in human colonic mucosa. Recently, we have reported on a human colon sialate-7(9)-O-acetyltransferase capable of incorporating acetyl groups into sialic acids at the nucleotide-sugar level [Shen et al., Biol. Chem. 383 (2002), 307-317]. In this report, we show that the CMP-N-acetyl-neuraminic aci

N-terminal acetyltransferase D (NatD) acetylates histones H4 and H2A at serine 1 and has been implicated in oncogenesis, yet its role in glioblastoma (GBM) remains unexplored. Here, we show that NatD expression is significantly elevated in GBM at both transcript and protein levels. NatD knockdown markedly suppressed proliferation, colony formation, migration, invasion and tumoursphere formation in GBM cell lines and patient-derived models, indicating a critical role in maintaining cancer stem-like properties. These inhibitory effects were rescued by reintroduction of wild-type NatD but not by

As the primary metabolite of alcohol and the most abundant carcinogen in tobacco smoke, acetaldehyde is linked to a number of human diseases associated with chronic alcohol consumption and smoking including cancers. In addition to direct DNA damage as a result of the formation of acetaldehyde-DNA adducts, acetaldehyde may also indirectly impact proper genome function through the formation of protein adducts. Histone proteins are the major component of the chromatin. Post-translational histone modifications (PTMs) are critically important for the maintenance of genetic and epigenetic stability.

Arginine metabolism in plants is targeted by phytopathogens such as Pseudomonas syringae strains, which produce toxins that affect enzymes of plant arginine metabolism, causing devastating infections in crops. In this work, the structural and functional properties of plant N-acetylornithine glutamate acetyltransferase (NAOGAT), the key deacetylating enzyme of the cyclic ornithine pathway and the target of mangotoxin, were investigated. We determined the structures of rice and maize NAOGATs to near-atomic resolution. Plant NAOGATs demonstrate a characteristic ArgJ-like N-terminal nucleophile fo

The NAA10 gene encodes the catalytic subunit of the N-terminal acetyltransferase protein complex A (NatA), which is supposed to acetylate approximately 40% of the human proteins. After the advent of next-generation sequencing, more variants in the NAA10 gene and Ogden syndrome (OMIM# 300855) have been reported. Individuals with NAA10-related syndrome have a wide spectrum of clinical manifestations and the genotype-phenotype correlation is still far from being confirmed. Here, we report a three years old Chinese girl carrying a heterozygous de novo NAA10 [NM_003491: c. 247C > T, p. (Arg83Cys)]

Mucopolysaccharidosis (MPS) type IIIC or Sanfilippo syndrome type C is a rare autosomal recessive disorder caused by the deficiency of the lysosomal membrane enzyme, heparan sulfate acetyl-CoA (AcCoA): alpha-glucosaminide N-acetyltransferase (HGSNAT; EC 2.3.1.78), which catalyzes transmembrane acetylation of the terminal glucosamine residues of heparan sulfate prior to their hydrolysis by alpha-N-acetylglucosaminidase. Lysosomal storage of undegraded heparan sulfate in the cells of affected patients leads to neuronal death, causing neurodegeneration and severely impaired development accompanie

1. Interferon-gamma (IFN-gamma) has been shown to suppress N-acetyltransferase (NAT) activity in cultured rat pineal glands when stimulated with isoproterenol (ISO). 2. Conversely, IFN-gamma has also been shown to increase the melatonin content of the rat pineal gland in organ culture. 3. Circumstantial evidence leads to a hypothesis that the NAT suppressive effect may be due to the action of IFN-gamma on the sympathetic nerve terminals. 4. To test this hypothesis, pineal glands from intact (INT) and superior cervical ganglionectomized (SCGX) rats, which had been operated 5 days earlier, were

Secretion of melatonin by the mammalian pineal gland is primarily regulated by the release of norepinephrine (NE) from sympathetic nerve terminals that originate from the superior cervical ganglia. Peptidergic nerves that originate in the perikarya located in the sensory trigeminal ganglia also innervate the pineal gland. Some of these peptidergic nerve fibers contain substance P. Previously, we have characterized neurokinin 1 type substance P receptors in the pineal gland. However, the function of this receptor in the pineal gland remains unclear. Here, we examined the modulatory effect of su

Epstein-Barr virus (EBV), the causative agent of infectious mononucleosis, persistently infects over 90% of the human adult population and is associated with several human cancers. To establish life-long infection, EBV tampers with the induction of type I interferon (IFN I)-dependent antiviral immunity in the host. How various EBV genes help orchestrate this crucial strategy is incompletely defined. Here, we reveal a mechanism by which the EBV nuclear antigen 3A (EBNA3A) may inhibit IFNβ induction. Using proximity biotinylation we identify the histone acetyltransferase P300, a member of the IF

Cohesin-mediated loop extrusion is central to IgH V(D)J recombination in early developing B cells, driving D-to-JH rearrangements and VH-to-DJH rearrangements, with the latter step regulated by the cohesin release factor WAPL. Here, we investigate the distinct contributions of two cohesin subunits, STAG1 and STAG2, which are incorporated in the somatic cohesin variants, cohesin-STAG1 and cohesin-STAG2, respectively. Using WAPL-sufficient mouse v-Abl pro-B cells to model the D-to-JH recombination stage, we show that STAG2 promotes balanced D-to-JH joining and restrains STAG1 expression. Loss of
