Human Metapneumovirus Induces Formation of Inclusion Bodies for Efficient Genome Replication and Transcription.

Human metapneumovirus (HMPV) causes significant upper and lower respiratory disease in all age groups worldwide. The virus possesses a negative-sense single-stranded RNA genome of approximately 13.3 kb encapsidated by multiple copies of the nucleoprotein (N), giving rise to helical nucleocapsids. In addition, copies of the phosphoprotein (P) and the large RNA polymerase (L) decorate the viral nucleocapsids. After viral attachment, endocytosis, and fusion mediated by the viral glycoproteins, HMPV nucleocapsids are released into the cell cytoplasm. To visualize the subsequent steps of genome tra

Regulatory role of noncoding RNAs in infections caused by Rhabdoviridae family viruses.

The Rhabdoviridae family comprises negative-sense single-stranded RNA viruses that cause severe infections in a wide range of hosts. Increasing evidence indicates that host noncoding RNAs (ncRNAs), including microRNAs (miRNAs), long noncoding RNAs (lncRNAs), and circular RNAs (circRNAs), play central roles in regulating rhabdovirus replication, pathogenesis, and host immune responses. These ncRNAs exert both antiviral and proviral effects through modulation of innate immune signaling pathways, regulation of gene expression, and control of inflammatory responses. Antiviral ncRNAs suppress viral

Nucleocapsid assembly drives Ebola viral factory maturation and dispersion.

Replication and genome encapsidation of many negative-sense RNA viruses take place in virus-induced membraneless organelles termed viral factories (VFs). Although liquid properties of VFs are believed to control the transition from genome replication to nucleocapsid (NC) assembly, VF maturation and interactions with the cellular environment remain elusive. Here, we apply in situ cryo-correlative light and electron tomography to follow NC assembly and changes in VF morphology and their liquid properties during Ebola virus infection. We show that viral NCs transition from loosely packed helical

Molecular biology, pathogenesis and pathology of mumps virus.

Mumps is caused by the mumps virus (MuV), a member of the Paramyxoviridae family of enveloped, non-segmented, negative-sense RNA viruses. Mumps is characterized by painful inflammatory symptoms, such as parotitis and orchitis. The virus is highly neurotropic, with laboratory evidence of central nervous system (CNS) infection in approximately half of cases. Symptomatic CNS infection occurs less frequently; nonetheless, prior to the introduction of routine vaccination, MuV was a leading cause of aseptic meningitis and viral encephalitis in many developed countries. Despite being one of the oldes

N-phenethylphenazine-1-carboxamide, a novel antiviral compound against infectious bronchitis virus.

Avian infectious bronchitis virus (IBV) is a globally prevalent and highly contagious avian pathogen that imposes significant disease burden and economic losses on the poultry industry. Given the challenges associated with current vaccine-based immunization strategies, there is an urgent need to develop novel therapeutic approaches for the prevention and treatment of IBV infection. In this study, a newly synthesized compound, N-phenethylphenazine-1-carboxamide (SQXA-12), was evaluated for its potent inhibitory effects on IBV replication. The cytotoxicity assay revealed CC50 values of 151.2 µM

Phylogenetic analysis of Bunyamwera and Ngari viruses (family Bunyaviridae, genus Orthobunyavirus) isolated in Kenya.

Orthobunyaviruses, tri-segmented, negative-sense RNA viruses, have long been associated with mild to severe human disease in Africa, but not haemorrhagic fever. However, during a Rift Valley fever outbreak in East Africa in 1997-1998, Ngari virus was isolated from two patients and antibody detected in several others with haemorrhagic fever. The isolates were used to identify Ngari virus as a natural Orthobunyavirus reassortant. Despite their potential to reassort and cause severe human disease, characterization of orthobunyaviruses is hampered by paucity of genetic sequences. Our objective was

Real-Time RT-PCR for the Detection of Lyssavirus Species.

The causative agents of rabies are single-stranded, negative-sense RNA viruses in the genus Lyssavirus of Rhabdoviridae, consisting of twelve classified and three as yet unclassified species including classical rabies virus (RABV). Highly neurotropic RABV causes rapidly progressive encephalomyelitis with nearly invariable fatal outcome. Rapid and reliable diagnosis of rabies is highly relevant for public and veterinary health. Due to growing variety of the genus Lyssavirus observed, the development of suitable molecular assays for diagnosis and differentiation is challenging. This work focused

Development of high-content imaging assays for lethal viral pathogens.

Filoviruses such as Ebola (EBOV) and Marburg (MARV) are single-stranded negative sense RNA viruses that cause acute hemorrhagic fever with high mortality rates. Currently, there are no licensed vaccines or therapeutics to counter filovirus infections in humans. The development of higher throughput/high-content primary screening assays followed by validation using the low-throughput traditional plaque or real-time PCR assays will greatly aid efforts toward the discovery of novel antiviral therapeutics. Specifically, high-content imaging technology is increasingly being applied for primary drug

SNX17 facilitates entry and infection of Rift Valley fever virus and related bunyaviruses.

Bunyaviruses are a group of multisegmented, negative-sense RNA viruses. Rift Valley fever virus (RVFV) is a representative member that can cause sporadic human outbreaks. We previously identified low-density lipoprotein receptor-related protein 1 (LRP1) as a host entry factor for RVFV and several bunyaviruses. This screen also identified additional factors as proviral host proteins, including sorting nexin 17 (SNX17). Here, we describe efforts to characterize SNX17 in bunyaviral infection. Our findings reveal that the removal of SNX17 results in a substantial reduction of RVFV infection in SNX

PARP14 is an interferon (IFN)-induced host factor that promotes IFN production and affects the replication of multiple viruses.

PARP14 is a 203 kDa multi-domain protein that is primarily known as an ADP-ribosyltransferase, and is involved in a variety of cellular functions including DNA damage, microglial activation, inflammation, and cancer progression. In addition, PARP14 is upregulated by interferon (IFN), indicating a role in the antiviral response. Furthermore, PARP14 has evolved under positive selection, again indicating that it is involved in host-pathogen conflict. We found that PARP14 is required for increased IFN-I production in response to coronavirus infection lacking ADP-ribosylhydrolase (ARH) activity and

Recoding of the vesicular stomatitis virus L gene by computer-aided design provides a live, attenuated vaccine candidate.

UNLABELLED: Codon pair bias (CPB), which has been observed in all organisms, is a neglected genomic phenomenon that affects gene expression. CPB results from synonymous codons that are paired more or less frequently in ORFeomes regardless of codon bias. The effect of an individual codon pair change is usually small, but when it is amplified by large-scale genome recoding, strikingly altered biological phenotypes are observed. The utility of codon pair bias in the development of live attenuated vaccines was recently demonstrated by recodings of poliovirus (a positive-strand RNA virus) and influ

Inhibitory effects of molnupiravir on Crimean-Congo hemorrhagic fever virus polymerase.

The order Bunyavirales encompasses a diverse group of segmented negative-sense RNA viruses, many of which can cause severe human disease. Their global distribution paired with multiple routes of transmission by arthropods and rodents cause risks to public health. However, effective antiviral drugs are not yet approved. Molnupiravir is an orally available prodrug of β-D-N4-hydroxycytidine (NHC) and shows a broad spectrum of antiviral activity. This includes severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and also several members of the Bunyavirales, such as Crimean-Congo hemorrhagi

GSH-ZnS Nanoparticles Exhibit High-Efficiency and Broad-Spectrum Antiviral Activities via Multistep Inhibition Mechanisms.

Despite the good biocompatibility and antibacterial activity of zinc sulfide nanoparticles (ZnS NPs), whether they possess antiviral activity is still unclear. Here, GSH-modified ZnS NPs (GSH-ZnS NPs) were synthesized and their significant antiviral activity was demonstrated using the Arteriviridae family RNA virus, porcine reproductive and respiratory syndrome virus (PRRSV), as a model. Mechanistically, GSH-ZnS NPs were shown to reduce PRRSV-induced ROS production to prevent PRRSV multiplication, with no activating effect on the interferon (IFN) signal pathway, the first defense line against

A region of mumps virus nucleoprotein affects defective interfering particle production.

Mumps virus (MuV) is a negative-sense, single-stranded RNA virus belonging to the family Paramyxoviridae. MuV causes acute infection of the parotid glands, and the infection can result in severe cases of encephalitis, meningitis and deafness in humans. The non-segmented RNA genome of MuV is encapsidated by the nucleocapsid protein (NP), which forms the ribonucleoprotein (RNP) complex that serves as a template for viral RNA synthesis. To make viral genomic RNA accessible to the viral polymerase, a conformational change within NP occurs. Recently, an atomic model of the NP of MuV was developed w

The role of toll-like receptor 7 signaling in mice infected by severe fever with thrombocytopenia syndrome virus.

Severe fever with thrombocytopenia syndrome virus (SFTSV) is a negative-sense RNA virus that can cause high fever, thrombocytopenia, and multiple-organ dysfunction. Toll-like receptor 7 (TLR7) can recognize single-stranded RNA viruses and induce type I interferons (IFNs). Although type I IFNs play a critical role in SFTSV infection, the role of TLR7 remains incompletely understood. To investigate the role of TLR7 signaling in SFTSV pathogenesis, TLR7-deficient (TLR7-/-) mice and wild-type (WT) mice were inoculated with SFTSV. TLR7-/- mice exhibited severe weight loss and elevated viral RNA lev

Analysis of RNA binding by the dengue virus NS5 RNA capping enzyme.

Flaviviruses are small, capped positive sense RNA viruses that replicate in the cytoplasm of infected cells. Dengue virus and other related flaviviruses have evolved RNA capping enzymes to form the viral RNA cap structure that protects the viral genome and directs efficient viral polyprotein translation. The N-terminal domain of NS5 possesses the methyltransferase and guanylyltransferase activities necessary for forming mature RNA cap structures. The mechanism for flavivirus guanylyltransferase activity is currently unknown, and how the capping enzyme binds its diphosphorylated RNA substrate i

Non-Ebola Filoviruses: Potential Threats to Global Health Security.

Filoviruses are negative-sense single-stranded RNA viruses often associated with severe and highly lethal hemorrhagic fever in humans and nonhuman primates, with case fatality rates as high as 90%. Of the known filoviruses, Ebola virus (EBOV), the prototype of the genus Orthoebolavirus, has been a major public health concern as it frequently causes outbreaks and was associated with an unprecedented outbreak in several Western African countries in 2013-2016, affecting 28,610 people, 11,308 of whom died. Thereafter, filovirus research mostly focused on EBOV, paying less attention to other equall

T7 RNA polymerase-dependent and -independent systems for cDNA-based rescue of Rift Valley fever virus.

Rift Valley fever virus (RVFV) is responsible for large and recurrent outbreaks of acute febrile illness among humans and domesticated animals in Africa. It belongs to the family Bunyaviridae, genus Phlebovirus, and its negative-stranded RNA genome consists of three segments. Here, we report the establishment and characterization of two different systems to rescue the RVFV wild-type strain ZH548. The first system is based on the BHK-21 cell clone BSR-T7/5, which stably expresses T7 RNA polymerase (T7 pol). Rescue of wild-type RVFV was achieved with three T7 pol-driven cDNA plasmids representin
