Broad neuraminidase antibodies confer protection against seasonal and avian influenza viruses.

Neuraminidase (NA) is a critical target for universal influenza vaccines and therapeutic antibodies, yet its antigenic landscape remains incompletely understood. Here we identify two broadly cross-protective monoclonal antibodies, CAV-F6 and CAV-F34, from influenza-infected individuals. These antibodies inhibit NA enzymatic activity across multiple subtypes and confer protection against seasonal influenza in female mouse models. Importantly, the two antibodies also neutralize emerging avian strains, including recent bovine H5N1 and H7N9 strains, both with pandemic potential. Structural studies

Human Immune Response to Influenza Neuraminidase After Vaccination: A Systematic Review.

BACKGROUND: Licensed influenza vaccines primarily target hemagglutinin (HA)-related immunity, but neuraminidase (NA)-based immunity is gaining attention as an independent correlate of protection. Inactivated influenza vaccines contain unspecified quantities of residual NA, with limited understanding of the resulting antibody induction and durability. METHODS: We conducted a systematic literature review across PubMed, Embase, Cochrane Library, ClinicalTrials.gov, and Trialtrove to identify studies reporting NA-related immunity following influenza vaccination. Primary outcomes included pre-exis

Neuraminidase-1 (NEU1): Biological Roles and Therapeutic Relevance in Human Disease.

Neuraminidases catalyze the desialylation of cell-surface glycoconjugates and play crucial roles in the development and function of tissues and organs. In both physiological and pathophysiological contexts, neuraminidases mediate diverse biological activities via the catalytic hydrolysis of terminal neuraminic, or sialic acid residues in glycolipid and glycoprotein substrates. The selective modulation of neuraminidase activity constitutes a promising strategy for treating a broad spectrum of human pathologies, including sialidosis and galactosialidosis, neurodegenerative disorders, cancer, car

Structural basis for Glycan-receptor binding by mumps virus hemagglutinin-neuraminidase.

Mumps virus is one of the main cause of respiratory illnesses in humans, especially children. Among the viral surface glycoproteins, the hemagglutinin - neuraminidase, MuV-HN, plays key roles in virus entry into host cells and infectivity, thus representing an ideal target for the design of novel inhibitors. Here we report the detailed analysis of the molecular recognition of host cell surface sialylated glycans by the viral glycoprotein MuV-HN. By a combined use of NMR, docking, molecular modelling and CORCEMA-ST, the structural features of sialoglycans/MuV-HN complexes were revealed. Evidenc

Neuraminidase A-Exposed Galactose Promotes Streptococcus pneumoniae Biofilm Formation during Colonization.

Streptococcus pneumoniae is an opportunistic pathogen that colonizes the nasopharynx. Herein we show that carbon availability is distinct between the nasopharynx and bloodstream of adult humans: glucose is absent from the nasopharynx, whereas galactose is abundant. We demonstrate that pneumococcal neuraminidase A (NanA), which cleaves terminal sialic acid residues from host glycoproteins, exposed galactose on the surface of septal epithelial cells, thereby increasing its availability during colonization. We observed that S. pneumoniae mutants deficient in NanA and β-galactosidase A (BgaA) fail

Pitfalls in Diagnosing Neuraminidase Deficiency: Psychosomatics and Normal Sialic Acid Excretion.

Neuraminidase deficiency (mucolipidosis I, sialidosis types I and II, cherry-red spot myoclonus syndrome) is a lysosomal storage disorder with an expanding clinical phenotype. Here, we report the striking diagnostic history of late-onset neuraminidase deficiency in two sisters, currently aged 14 (patient 1) and 15 (patient 2).Patient 1 was referred for evaluation of her vision after a traffic accident. During this examination, nummular cataract, macular cherry-red spot, and optic nerve atrophy were seen. Furthermore, tremors were noticed in her arms and legs. This combination suggested a lysos

Pseudovirus-based neuraminidase inhibition assays reveal potential H5N1 drug-resistant mutations.

The use of antiviral drugs such as influenza neuraminidase (NA) inhibitors is a critical strategy to prevent and control flu pandemic, but this strategy faces the challenge of emerging drug-resistant strains. For a highly pathogenic avian influenza (HPAI) H5N1 virus, biosafety restrictions have significantly limited the efforts to monitor its drug responses and mechanisms involved. In this study, a rapid and biosafe assay based on NA pseudovirus was developed to study the resistance of HPAI H5N1 virus to NA inhibitor drugs. The H5N1 NA pseudovirus was comprehensively tested using oseltamivir-s

Neuraminidase activity assays for monitoring MDCK cell culture derived influenza virus.

Three assay methods were investigated for monitoring the time-course of neuraminidase (NA) activity of tissue culture derived equine influenza A virus from large-scale microcarrier cultivation and several steps of downstream processing required for the production of inactivated vaccines. Measurements of neuraminidase activity by a thiobarbituric acid (TBA) and a fluorometric method using Amplex Red as a fluorogen (FL-AR) did not correlate with the increase of hemagglutinin (HA) during virus replication. Samples analysed by the TBA method showed unspecific interference from low molecular weight

Parainfluenza virus type 2 haemagglutinin-neuraminidase glycoprotein characterized with monoclonal antibodies.

Thirteen monoclonal antibodies (MAbs) were prepared against human parainfluenza virus type 2 (PIV2). These MAbs reacted with the haemagglutinin-neuraminidase glycoprotein with an Mr of 84K. The MAbs defined one antigenic site which could be divided into five epitopes. A correlation between haemagglutination inhibition (HI) and neutralization activity could be seen although one MAb, which recognized a distinct epitope, showed neutralization and no HI activity to PIV2. The reactivity of the MAbs was tested against Sendai virus, parainfluenza virus type 3, simian virus 5 (SV5), mumps virus, Newca

A neuraminidase-targeted nanobody confers broad protection against influenza B virus.

UNLABELLED: Influenza B virus (IBV) exhibits antigenic diversity and significantly contributes to the annual burden of influenza-related disease, posing substantial impacts on global public health and the economy. In this study, we focused on the neuraminidase (NA) of IBV, immunized an alpaca with tetrameric IBV NA, and identified a series of nanobodies with a common feature of the "DYR" motif in CDR3, exhibiting broad inhibitory activity against diverse NAs of IBV. Among these, the most potent VHH5 broadly inhibits NA from multiple representative strains across ancestral, B/Yamagata/16/88-lik

Population Immunity to Influenza Neuraminidase: From Immune Imprinting to Pandemic Preparedness.

Neuraminidase (NA) is a major surface glycoprotein of influenza viruses that mediates the release of progeny virions from infected cells. Although both hemagglutinin (HA) and NA are targets of protective immune responses, current seasonal influenza vaccines are primarily standardized based on HA content, resulting in variable and often suboptimal induction of NA-specific immunity. Therefore, NA is subject to different immune pressures and exhibits distinct patterns of antigenic evolution. Increasing evidence suggests that neuraminidase-inhibiting (NAI) antibodies contribute to protection again

Functional neuraminidase inhibitor resistance motifs in avian influenza A(H5Nx) viruses.

Neuraminidase inhibitors (NAIs) are antiviral agents recommended worldwide to treat or prevent influenza virus infections in humans. Past influenza virus pandemics seeded by zoonotic infection by avian influenza viruses (AIV) as well as the increasing number of human infections with AIV have shown the importance of having information about resistance to NAIs by avian NAs that could cross the species barrier. In this study we introduced four NAI resistance-associated mutations (N2 numbering) previously found in human infections into the NA of three current AIV subtypes of the H5Nx genotype that

Influenza virus neuraminidase regulates host CD8+ T-cell response in mice.

Influenza A virus (IAV)-specific CD8+ T-cell response was shown to provide protection against pandemic and seasonal influenza infections. However, the response was often relatively weak and the mechanism was unclear. Here, we show that the composition of IAV released from infected cells is regulated by the neuraminidase (NA) activity and the cells infected by NA-defective virus cause intracellular viral protein accumulation and cell death. In addition, after uptake of NA-defective viruses by dendritic cells (DCs), an expression of the major histocompatibility complex class I is induced to acti

Characterization of Influenza B Virus Variants with Reduced Neuraminidase Inhibitor Susceptibility.

Treatment options for influenza B virus infections are limited to neuraminidase inhibitors (NAIs), which block the neuraminidase (NA) glycoprotein on the virion surface. The development of NAI resistance would therefore result in a loss of antiviral treatment options for influenza B virus infections. This study characterized two contemporary influenza B viruses with known resistance-conferring NA amino acid substitutions, D197N and H273Y, detected during routine surveillance. The D197N and H273Y variants were characterized in vitro by assessing NA enzyme activity and affinity, as well as repli

Computer-based de novo designs of tripeptides as novel neuraminidase inhibitors.

The latest influenza A (H1N1) pandemic attracted worldwide attention and called for the urgent development of novel antiviral drugs. Here, seven tripeptides are designed and explored as neuraminidase (NA) inhibitors on the structural basis of known inhibitors. Their interactions with NA are studied and compared with each other, using flexible docking and molecular dynamics simulations. The various composed tripeptides have respective binding specificities and their interaction energies with NA decrease in the order of FRI > FRV > FRT > FHV > FRS > FRG > YRV (letters corresponding to amino acid

Protective effects of influenza B neuraminidase antibodies against symptomatic influenza virus infection.

BACKGROUND: Influenza vaccines are designed to induce hemagglutinin-inhibiting (HAI) antibodies rather than neuraminidase-inhibiting (NAI) antibodies; therefore, NAI antibodies' protective effects need further investigation. NAI antibodies' protective effects, independent of HAI antibodies, have been demonstrated for influenza A viruses, but not influenza B viruses (IBV). METHODS: Employing H6NB reassortant virus targets, we evaluated NAI antibodies' protective effects against IBV in sera from a prospective influenza vaccine effectiveness study among healthcare personnel (HCP) in Israel. Population immunity to clade 2.3.4.4b H5N1 is dominated by anti-neuraminidase antibodies.

Clade 2.3.4.4b highly pathogenic avian influenza A(H5N1) viruses continue to expand geographically and across mammalian hosts, raising concern about pandemic potential. The degree and specificity of pre-existing immunity in humans are key determinants of this risk. We analyzed hemagglutinin (HA)- and neuraminidase (NA)-specific antibody responses in 300 sera collected from adults in New York City. While HA directed binding antibodies to clade 2.3.4.4b H5 were low and hemagglutination-inhibiting antibodies were absent, we detected widespread binding and functional NA antibodies against N1 neura

Neuraminidase-Associated Vaginal Dysbiosis Correlates with HPV Infection Across Oncogenic Risk Subtypes.

BACKGROUND: Human papillomavirus (HPV) infection is a major cause of cervical cancer, and increasing evidence suggests that cervicovaginal microbial dysbiosis may influence HPV susceptibility. This study evaluated the association between microbial enzymatic markers and HPV infection across different risk subtypes. METHODS: A cross-sectional analysis was conducted in 43,426 women undergoing HPV screening. HPV status and subtype classification (high-, possible-, and low-risk) were determined using standardized genotyping. Clinical and laboratory parameters including age, white blood cells (WBC)

Broadly protective bispecific antibodies that simultaneously target influenza virus hemagglutinin and neuraminidase.

UNLABELLED: Monoclonal antibodies (mAbs) are an attractive therapeutic platform for the prevention and treatment of influenza virus infection. There are two major glycoproteins on the influenza virion surface: hemagglutinin (HA), which is responsible for viral attachment and entry, and neuraminidase (NA), which mediates viral egress by enzymatically cleaving sialic acid to release budding particles from the host cell surface. Broadly neutralizing antibodies (bNAbs) that target the conserved HA central stalk region, such as CR9114, can inhibit both viral entry and egress. More recently, broadly

Protective efficacy of anti-neuraminidase monoclonal antibodies against H7N9 influenza virus infection.

The H7N9 influenza virus has been circulating in China for more than six years. The neuraminidase (NA) has gained great concern for the development of antiviral drugs, therapeutic antibodies, and new vaccines. In this study, we screened seven mouse monoclonal antibodies (mAbs) and compared their protective effects against H7N9 influenza virus. The epitope mapping from escape mutants showed that all the seven mAbs could bind to the head region of the N9 NA close to the enzyme activity sites, and four key sites of N9 NA were reported for the first time. The mAbs D3 and 7H2 could simultaneously i
