Linking Essential Tremor to the Cerebellum: Neurochemical Evidence.

The pathophysiology and the exact anatomy of essential tremor (ET) is not well known. One of the pillars that support the cerebellum as the main anatomical locus in ET is neurochemistry. This review examines the link between neurochemical abnormalities found in ET and cerebellum. The review is based on published data about neurochemical abnormalities described in ET both in human and in animal studies. We try to link those findings with cerebellum. γ-aminobutyric acid (GABA) is the main neurotransmitter involved in the pathophysiology of ET. There are several studies about GABA that clearly po

Functional imaging and neurochemical correlates of stimulant self-administration in primates.

RATIONALE: Recent advances in neuroimaging and in vivo neurochemistry have documented drug-induced functional changes in brain activity under physiologically relevant conditions. These approaches have significant strengths and important limitations that should be considered. OBJECTIVES: The present review describes current in vivo approaches to characterize drug effects as they relate to behavior, and highlights key contributions derived from each approach in the context of stimulant self-administration in primates. METHODS: Techniques relating in vitro neurochemistry to behavioral pharmacol

Elevated primary motor cortex GABA in Parkinson's disease: 7T MR spectroscopy evidence.

INTRODUCTION: Cortical inhibitory-excitatory imbalance may contribute to motor dysfunction in Parkinson's disease (PD), but in vivo primary motor cortex (M1) neurochemistry remains incompletely characterized. METHODS: We used 7T proton MR spectroscopy to quantify γ-aminobutyric acid (GABA) and glutamate (Glu) in M1 in patients with mild-to-moderate PD and healthy controls. The inhibitory/excitatory ratio (I/E ratio, GABA/Glu) was derived as an exploratory measure. After quality control, baseline analyses included 32 patients and 28 controls. A self-paced bilateral handgrip task was conducted

When brain stimulation backfires: the effects of prefrontal cortex stimulation on impulsivity.

Transcranial direct current stimulation (tDCS) can sometimes cause the opposite of its intended effect. These reverse effects may be related in part to individual differences in personality and neurochemistry. Previous studies have demonstrated that dopamine levels can impact the effects of tDCS. In the present study, 124 healthy participants took the UPPS impulsive behavior scale. Participants then underwent a single, randomized anodal or sham tDCS session on the prefrontal cortex. While the effects of tDCS were still active, they performed the Stop Signal Task, a measure of state impulsivity

Neuroinflammation: a contributing factor to the pathogenesis of cancer cachexia.

The clinical journey of cancer patients is frequently complicated by the development of a complex and multifaceted syndrome, the main features of which are reduced appetite, decreased food intake, progressive weight loss, and wasting of muscle mass and adipose tissue, which is not prevented by the provision of calories and proteins. This syndrome, termed Cachexia, is responsible for increased morbidity, reduced survival, and impinged quality of life of cancer patients. The pathogenesis is complex and involves deranged metabolism of peripheral tissues and profound alterations of brain neurochem

Neurochemical anatomy of movement disorders.

Recent advances in our understanding of the neurochemical anatomy of the pyramidal and extrapyramidal motor systems have led to more logical and effective pharmacotherapies for movement disorders. Our knowledge of the neurochemistry of Parkinson's and Huntington's diseases is the most complete. In the future, the same approaches used in these diseases need to be used on the somewhat less common disorders such as progressive supranuclear palsy, dystonia, Tourette's syndrome, and cerebellar ataxias. Postmortem neurochemical analyses of these diseases should be exciting and will improve our overa

Neurochemical and morphological changes associated with human epilepsy.

To date a multitude of studies into the morphology and neurochemistry of human epilepsy have been undertaken with variable, and often inconsistent, results. This review summarises these studies on a range of neurotransmitters, neuromodulators, neuropeptides and their receptors. In addition to this, novel changes in cell viability and sprouting have been identified and are discussed. Whether the alterations observed are a result of the seizures or are a contributory factor is unclear. However, it may be that following an initial insult (such as febrile convulsions, status epilepticus or head in

The Top 5 Neurotransmitters from a Clinical Neurologist's Perspective.

Neurologists are proficient in neuroanatomy and -physiology but their understanding of neurochemistry tends to be mediocre. As a rule, we do not think in biochemical pathways and complex metabolic interactions but rather associate a few neurotransmitters with well-known brain diseases or drugs that we routinely prescribe. Most of us can hardly come up with more than a handful of relevant neurochemicals. From our point of view the most important neurotransmitters are, in alphabetical order, acetylcholine (associated with Alzheimer's disease and myasthenia gravis), dopamine (Parkinson's disease)

Hypothesized neurochemical models for psychiatric syndromes in alcohol and drug dependence.

Exploration of the neurochemistry of psychiatric and substance use disorders in dual diagnosis patients may help explain the greater than chance comorbidity of these disorders and lead to advances in treatment. This paper will focus on the hypothesized neurochemical changes associated with primary substance use disorders which might lead to secondary psychiatric disorders by mimicking the hypothesized neurochemical changes of primary psychiatric disorders. For example, hypothesized serotonergic deficits in alcoholism, endorphin deficits in opioid dependence, and dopamine depletion in cocaine d

Identifying associations and key early life exposure windows for organophosphate esters and adolescent brain morphometry in the HOME study.

BACKGROUND: Organophosphate esters (OPEs) can be used as flame retardants and plasticizers in consumer products. Early life exposure may disrupt brain development by modifying brain structure and neurochemistry. OBJECTIVES: To evaluate the associations between gestational and childhood OPE metabolite urinary concentrations and their mixtures with neurobehavioral outcomes, explore the potential mediation of brain structure and neurochemistry, and identify potential windows of susceptibility. METHODS: We employed quantile-based g-computation models to investigate the mixture associations of OP

Chemical neuroanatomy of the substantia nigra in the ovine brain.

The substantia nigra is an integral component of the basal ganglia circuitry for limbic and motor functions. Dysfunction and degeneration of the basal ganglia are fundamental aspects of neurodegenerative diseases such as Parkinson's disease and Huntington's disease. With the increasing use of sheep to model neurological diseases, it is crucial to understand the anatomy and neurochemistry of these key basal ganglia nuclei in the normal sheep brain and how they compare to the human brain. Therefore, studies of the gross anatomy, cellular morphology, and neurochemical expression patterns within t
