Sexual identity of Akh neurons shapes Nucb1-dependent metabolic and reproductive plasticity.

Sex differences in metabolic physiology are widespread across species and are tightly linked to reproductive demands. Here, we identify nucleobindin 1 (Nucb1), a conserved calcium-binding protein, as a female-specific metabolic regulator in the adipokinetic hormone (Akh)-producing cells (APCs) of Drosophila melanogaster. APC-specific nucb1 knockdown increased energy storage and body mass in females but not in males. Cellular sex reversal experiments revealed these female-specific functions of Nucb1 to be dependent on the intrinsic sexual identity of the APCs, rather than the sex of the organis

Immature Neurons in the Postnatal Brain: Markers, Modulation, and Involvement in Normal and Aberrant Plasticity.

Cortical immature neurons (cINs) represent a unique population of prenatally generated, non-dividing neurons that maintain an immature phenotype, characterized by doublecortin (DCX) and polysialylated neural cell adhesion molecule (PSA-NCAM) expression, into adulthood. Unlike canonical adult neurogenesis involving continuous neuron generation from stem cell niches, cINs constitute a distinct form of structural plasticity termed "neurogenesis without division". This review comprehensively examines the molecular markers, morphological diversity, developmental origins, and maturation trajectories

Spared but Not Silent: Ion Channel Plasticity in Uninjured Sensory Neurons in Neuropathic Pain.

Chronic pain is maintained by persistent, maladaptive molecular processes, many of which converge on ion channels. In sensory neurons, these processes include transcriptional reprogramming, altered trafficking, and remodeling of the channel complexes that set membrane excitability. Accumulating evidence indicates that the major ion channel changes driving chronification of neuropathic pain are not restricted to injured sensory neurons but also arise within the adjacent population of spared neurons. Here we review the ion channel alterations that sustain chronic pain states, the mechanisms by w

Sensory-neuron-derived CGRPα controls white adipocyte differentiation and tissue plasticity.

Communication between the brain and adipose tissue is mediated in part by the peripheral nervous system. The sympathetic branch transmits lipolytic and thermogenic signals toward adipose tissue, while sensory nerves relay information to the central nervous system. Importantly, sensory nerve activation also triggers local neuropeptide release. Here, we show that sensory-neuron-derived α-calcitonin gene-related peptide (CGRPα) inhibits white, but not brown, preadipocyte differentiation in a cell-autonomous manner. In vivo, CGRPα expression in mouse subcutaneous adipose tissue during cold exposur

Contributions of cortical neuron firing patterns, synaptic connectivity, and plasticity to task performance.

Neuronal responses during behavior are diverse, ranging from highly reliable 'classical' responses to irregular 'non-classically responsive' firing. While a continuum of response properties is observed across neural systems, little is known about the synaptic origins and contributions of diverse responses to network function, perception, and behavior. To capture the heterogeneous responses measured from auditory cortex of rodents performing a frequency recognition task, we use a novel task-performing spiking recurrent neural network incorporating spike-timing-dependent plasticity. Reliable and

Oral Exposure to Microplastics Affects the Neurochemical Plasticity of Reactive Neurons in the Porcine Jejunum.

Plastics are present in almost every aspect of our lives. Polyethylene terephthalate (PET) is commonly used in the food industry. Microparticles can contaminate food and drinks, posing a threat to consumers. The presented study aims to determine the effect of microparticles of PET on the population of neurons positive for selected neurotransmitters in the enteric nervous system of the jejunum and histological structure. An amount of 15 pigs were divided into three groups (control, receiving 0.1 g, and 1 g/day/animal orally). After 28 days, fragments of the jejunum were collected for immunofluo

Microglia regulate motor neuron plasticity via reciprocal fractalkine and adenosine signaling.

We report an important role for microglia in regulating neuroplasticity within phrenic motor neurons. Brief episodes of low oxygen (acute intermittent hypoxia; AIH) elicit a form of respiratory motor plasticity known as phrenic long-term facilitation (pLTF) that is regulated by the balance of competing serotonin vs adenosine-initiated cellular mechanisms. Serotonin arises from brainstem raphe neurons, but the source of adenosine is unknown. We tested if hypoxic episodes initiate phrenic motor neuron to microglia fractalkine signaling that evokes extracellular adenosine formation using a well-d

Strumpellin/WASHC5 regulates the structural plasticity of cortical neurons involved in gait coordination.

Strumpellin/Wiskott-Aldrich syndrome protein and SCAR homologue (WASH) complex subunit 5 (WASHC5) is a core component of the WASH complex, and its mutations confer pathogenicity for hereditary spastic paraplegia (HSP) type SPG8, a rare neurodegenerative gait disorder. WASH complex activates actin-related protein-2/3-mediated actin polymerization and plays a pivotal role in intracellular membrane trafficking in endosomes. In this study, we examined the role of strumpellin in the regulation of structural plasticity of cortical neurons involved in gait coordination. Administration of a lentivirus

Coordinated Regulation of Synaptic Plasticity at Striatopallidal and Striatonigral Neurons Orchestrates Motor Control.

The basal ganglia play a critical role in shaping motor behavior. For this function, the activity of medium spiny neurons (MSNs) of the striatonigral and striatopallidal pathways must be integrated. It remains unclear whether the activity of the two pathways is primarily coordinated by synaptic plasticity mechanisms. Using a model of Parkinson's disease, we determined the circuit and behavioral effects of concurrently regulating cell-type-specific forms of corticostriatal long-term synaptic depression (LTD) by inhibiting small-conductance Ca(2+)-activated K(+) channels (SKs) of the dorsolatera

Trigeminal intersubnuclear neurons: morphometry and input-dependent structural plasticity in adult rats.

Intersubnuclear neurons in the caudal division of the spinal trigeminal nucleus that project to the principal nucleus (Pr5) play an active role in shaping the receptive fields of other neurons, at different levels in the ascending sensory system that processes information originating from the vibrissae. By using retrograde labeling and digital reconstruction, we investigated the morphometry and topology of the dendritic trees of these neurons and the changes induced by long-term experience-dependent plasticity in adult male rats. Primary afferent input was either eliminated by transection of t

Difference in binocularity and ocular dominance plasticity between GABAergic and excitatory cortical neurons.

Neuronal circuits in the cerebral cortex consist mainly of glutamatergic/excitatory and GABAergic/inhibitory neurons. In the visual cortex, the binocular responsiveness of neurons is modified by monocular visual deprivation during the critical period of postnatal development. Although GABAergic neurons are considered to play a key role in the expression of the critical period, it is not known whether their binocular responsiveness and ocular dominance plasticity are different from those of excitatory neurons. Recently, the end of the critical period was found to be not strict so that cortical

Pathway interactions and synaptic plasticity in the dendritic tuft regions of CA1 pyramidal neurons.

Input comparison is thought to occur in many neuronal circuits, including the hippocampus, where functionally important interactions between the Schaffer collateral and perforant pathways have been hypothesized. We investigated this idea using multisite, whole-cell recordings and Ca2+ imaging and found that properly timed, repetitive stimulation of both pathways results in the generation of large plateau potentials in distal dendrites of CA1 pyramidal neurons. These dendritic plateau potentials produce widespread Ca2+ influx, large after-depolarizations, burst firing output, and long-term pote

On neural plasticity, new neurons and the postischemic milieu: an integrated view on experimental rehabilitation.

This review discusses actual and potential contributors to functional improvement after stroke injuries. Topics that will be covered are neuronal re-organization and sprouting, neural stem/progenitor cell activation and neuronal replacement, as well as the neuronal milieu defined by glia, inflammatory cells and blood vessel supply. It is well established that different types of neuronal plasticity ultimately lead to post-stroke recovery. However, an untapped potential which only recently has started to be extensively explored is neuronal replacement through endogenous or exogenous resources. M

Postdecentralization plasticity of voltage-gated Na+ currents in rat glandular sympathetic neurons.

The kinetic properties of voltage-gated Na(+) currents in two groups of glandular postganglionic sympathetic neurons were assessed. The first group of neurons remained innervated by preganglionic axons until the day of current recordings, while the second--decentralized 4 weeks prior to recordings. An increase of maximum current amplitude and density was noted in decentralized neurons. Na(+) currents activated and time-dependently inactivated more slowly in decentralized than in control neurons. Furthermore, after decentralization the currents steady-state inactivated at less hyperpolarized po

Timing and plasticity of specification of CaM-Kinase II alpha expression by neocortical neurons.

In this work, the differential expression of a chemical marker, the alpha-isoform of the calcium/calmodulin-dependent protein kinase II (CaM-Kinase II alpha) and the development of the spinal cord projection were used to determine in vivo the embryonic stages at which different aspects of the phenotype of neocortical cells are specified. We first performed a quantitative, immunocytochemical study on the levels of CaM-Kinase II alpha expression in the frontal, parietal and occipital cortical areas of control adult rats. We found that the levels of expression of CaM-Kinase II alpha were larger i

Chemical plasticity of visual cortical neurons in integrative organization of feeding behavior in cats.

The P.K. Anokhin conception of organization of systemic behavior was used to study the discharge activity and chemical plasticity of cortical neurons in a goal-directed behavior. Chemical sensitivity of neurons in the visual cortex of instrumentally conditioned cats was studied during successive stages of food procuring and consuming. The results showed that definite stages of this integrated behavioral act are characterized by differences in the chemical sensitivity of visual neurons to neurotransmitters. The largest number of neurons resistant to neurotransmitters was found among cells that

Calmodulin blockers decrease short-term plasticity of the cholinoreceptors of neurons of the edible snail.

A reversible decrease in the rate and depth of the extinction of the reactions of the cholinoreceptive membrane to repeated iontophoretic applications of acetylcholine to the soma by a number of calmodulin blockers was demonstrated in identified RPa3 and LPa3 neurons in the edible snail using the method of recording transmembrane ionic currents: R 24571 (20-50 mumols/liter), trifluoperazine (50-200 mumols/liter), chlorpromazine (20-60 mumols/liter), and prenylamine lactate (30-400 mumols/liter). The role of Ca2+ channels of the L-type in neurotransmitter plasticity of cultured sympathetic neurons.

We have studied the effects of Ca2+ antagonists and agonists on the development of choline acetyltransferase (ChAT), tyrosine hydroxylase (TOH) and acetylcholinesterase (AChE) in cultures of rat sympathetic neurons maintained for 6-9 days in low K+ (5 mM) or high K+ (35 mM) medium. Previous experiments have shown that high K+ medium increases TOH activity and TOH-mRNA level up to 3.5-fold and depresses the development of AChE, in particular of its asymmetric A12 form. Moreover, high K+ medium inhibits ChAT induction by 90% in muscle-conditioned medium (Raynaud et al., Dev. Biol., 119 (1987) 30

Osmotic stimulation causes structural plasticity of neurone-glia relationships of the oxytocin but not vasopressin secreting neurones in the hypothalamic supraoptic nucleus.

A comparative quantitative analysis was carried out on identified supraoptic neurones of male and female Wistar and Long Evans rats under normal conditions and after chronic osmotic stimulation, and in homozygous Brattleboro rats suffering from diabetes insipidus. The neurones were identified by immunocytochemical or morphological means. Osmotic stimulation resulted in significant increases in the number and extent of direct neuronal appositions and in the number of presynaptic terminals contacting two neurosecretory cells simultaneously ("double" synapses). In the supraoptic nuclei of both se

Sensory plasticity of dorsal horn silent neurons as a critical mechanism for neuropathic pain.

The spinal cord dorsal horn (DH) integrates sensory processing but undergoes critical plasticity following nerve injury, leading to mechanical allodynia, or "touch-evoked pain". It has been proposed that after nerve injury, innocuous sensory neurons gain access to nociceptive-specific (NS) circuits in the DH due to altered spinal inhibitory controls, thereby converting touch into pain. It is however unclear how sensory processing is reorganized under these conditions to generate this symptom. In this study, we used two-photon calcium imaging in mice to show that spinal disinhibition or nerve i
