Denitrification: microbiology and ecology.

The ability of some aerobic microorganisms to reduce nitrogen oxides, especially nitrate, to gaseous products enables them to grow in oxygen-limiting conditions by using the oxides as electron acceptors. Denitrifying respiration is not as efficient as oxygen respiration so growth is slower. The ability to denitrify is found in a wide variety of unrelated bacteria and even in some fungi, and biochemical mechanisms are now moderately well understood. Major factors regulating denitrification are the availability of the nitrogen oxides, the availability of reductant (mostly organic carbon compound

Marine Colloids Promote the Adaptation of Diatoms to Nitrate Contamination by Directional Electron Transfer.

Nitrate contamination from human activities (e.g., domestic pollution, livestock breeding, and fertilizer application) threatens marine ecosystems and net primary productivity. As the main component of primary productivity, diatoms can adapt to high nitrate environments, but the mechanism is unclear. We found that electron transfer from marine colloids to diatoms enhances nitrogen uptake and assimilation under visible-light irradiation, providing a new pathway for nitrogen adaptation. Under irradiation, marine colloids exhibit semiconductor properties (e.g., the separation of electron-hole pai

Differential modulation of S-nitrosoglutathione reductase and reactive nitrogen species in wild and cultivated tomato genotypes during development and powdery mildew infection.

Nitric oxide plays an important role in the pathogenesis of Pseudoidium neolycopersici, the causative agent of tomato powdery mildew. S-nitrosoglutathione reductase, the key enzyme of S-nitrosothiol homeostasis, was investigated during plant development and following infection in three genotypes of Solanum spp. differing in their resistance to P. neolycopersici. Levels and localization of reactive nitrogen species (RNS) including NO, S-nitrosoglutathione (GSNO) and peroxynitrite were studied together with protein nitration and the activity of nitrate reductase (NR). GSNOR expression profiles a

Molybdenum nutrition of isolates of four Aspergillus species.

The molybdenum requirement for growth and conidial formation by Aspergillus flavus, A. terreus, and A. sulphureus was found to be 0.2 ppb, which was one-fifth that of an A. niger isolate. Molybdenum deficiency depressed growth, conidial formation, dry weight, soluble protein, and the specific activities of nitrate reductase, succinic dehydrogenase, and aconitase in all the isolates of Aspergillus studied, but the specific activities of catalase and peroxidase were depressed only in isolates of A. niger, A. terreus, and A. flavus. Also, molybdenum deficiency stimulated the specific activities o

Metabolome and molecular basis for carbohydrate increase and nitrate reduction in burley tobacco seedlings by glycerol through upregulating carbon and nitrogen metabolism.

Burley tobacco (Nicotiana Tabacum) is a chlorophyll-deficiency mutant. Nitrate is one precursor of tobacco-specific nitrosamines (TSNAs) and is largely accumulated in burley tobacco. To decrease nitrate accumulation in burley tobacco, glycerol, a polyhydric alcohol compound and physiological regulating material, was sprayed and its effects were investigated based on metabolomic technology and molecular biology. The results showed that glucose, glutamine and glutamic acid increased by 2.6, 5.1 and 196, folds, respectively, in tobacco leaves after glycerol application. Nitrate content was signif

Effect and mechanism of cyclodextrins on nitrate reduction and bio-activity by S.oneidensis.MR-1.

Cyclodextrins (CDs) have been widely used due to the excellent solubilization of hydrophobic organics. However, their effect on the biotransformation process of hydrophilic pollutants is unclear. This study first evaluated the effect and mechanism of CDs on nitrate reduction by S.oneidensis.MR-1. The three CDs (α-CD, β-CD and γ-CD) all accelerated nitrate reduction, among which β-CD had the best effect. The nitrate reduction rate was increased by 21.8% with 0.5 mM β-CD. As for the mechanism, β-CD increased the biomass, membrane permeability and EPS of S.oneidensis.MR-1. The nitrate reductase a

Mechanisms impairing blood pressure responses to nitrite and nitrate.

Hypertension is a multifactorial disease associated with impaired nitric oxide (NO) production and bioavailability. In this respect, restoring NO activity by using nitrite and nitrate has been considered a potential therapeutic strategy to treat hypertension. This possibility is justified by the understanding that both nitrite and nitrate may be recycled back to NO and also promote the generation of other bioactive species. This process involves a complex biological circuit known as the enterosalivary cycle of nitrate, where this anion is actively taken up by the salivary glands and converted

Who Reduces Silver? A Critical Review of the Biomolecular Drivers of Fungal-Mediated Silver Nanoparticle Biosynthesis.

Silver nanoparticles (AgNPs) synthesized via fungal-mediated biosynthesis have gained attention as eco-friendly alternatives to chemically produced nanomaterials, with broad biomedical potential. Fungi represent particularly attractive systems because their secretomes contain diverse biomolecules, including enzymes, proteins, polysaccharides, and secondary metabolites, capable of reducing silver ions and stabilizing the resulting nanoparticles. Despite extensive investigation, the molecular mechanisms underlying fungal-mediated AgNP formation remain poorly defined. This review critically exami

Genomic and physiological characterization of the chromate-reducing, aquifer-derived Firmicute Pelosinus sp. strain HCF1.

Pelosinus spp. are fermentative firmicutes that were recently reported to be prominent members of microbial communities at contaminated subsurface sites in multiple locations. Here we report metabolic characteristics and their putative genetic basis in Pelosinus sp. strain HCF1, an isolate that predominated anaerobic, Cr(VI)-reducing columns constructed with aquifer sediment. Strain HCF1 ferments lactate to propionate and acetate (the methylmalonyl-coenzyme A [CoA] pathway was identified in the genome), and its genome encodes two [NiFe]- and four [FeFe]-hydrogenases for H(2) cycling. The reduc

The role of the nitrate respiration element of Thermus thermophilus in the control and activity of the denitrification apparatus.

The nitrate conjugative element (NCE) encodes the ability to respire nitrate in the facultative Thermus thermophilus NAR1 strain. This process is carried out by two heterotetrameric enzymes that catalyse the oxidation of NADH (Nrc) and the reduction of nitrate (Nar), whose expression is activated by the NCE-encoded transcription factors DnrS and DnrT. We report the presence of NCE in other facultative strains of T. thermophilus and analyse its role in subsequent steps of the denitrification pathway. We encountered that nrc mutants of denitrifying strains show a decrease in anaerobic growth rat

Effect of nitrate injection on the bacterial community in a water-oil tank system analyzed by PCR-DGGE.

Sulfide production by sulfate-reducing bacteria (SRB) is a major concern for the petroleum industry since it is toxic and corrosive, and causes plugging due to the formation of insoluble iron sulfides (reservoir souring). In this study, PCR followed by denaturing gradient gel electrophoresis (PCR-DGGE) using two sets of primers based on the 16S rRNA gene and on the aps gene (adenosine-5-phosphosulfate reductase) was used to track changes in the total bacterial and SRB communities, respectively, present in the water-oil tank system on an offshore platform in Brazil in which nitrate treatment wa

Carbon Sequestration and Pollution Reduction Mechanisms of Chlorella sp. for Simulated Flue Gas.

Nitric oxide (NO), a signaling molecule and free radical, regulates microalgal photosynthesis, yet its potential in enhancing carbon fixation alongside pollutant mitigation remains underexplored. This study investigated the effects of NO (0, 50, 100 ppm) in simulated flue gas (15% CO2) on Chlorella sp., examining intracellular/extracellular NO dynamics, antioxidant response, key enzyme activity (nitrate reductase), and growth-photosynthesis coupling. Under optimized aeration (25 mL min-1, 3 h), 50 ppm of NO optimally elevated intracellular (0.62-1.30 nmol L-1) and extracellular (2.56-3.90 nmol

Effect of mixtures of dietary fibres on the enzyme activity of the rat caecal microflora.

The enzyme activity of the caecal microflora from weanling rats was determined after feeding 1 of 3 basal diets (purified fibre-free; purified plus cellulose; and stock), with or without additional dietary fibre (pectin, i-carrageenan or carboxymethylcellulose 5% w/w). The wet weight of caecal contents and total bacterial numbers were similar for the purified fibre-free and purified plus cellulose diets, yet were significantly higher in animals fed the stock diet. Pectin supplementation of the basal diets had no effect of caecal bacterial numbers, but significantly increased total nitrate redu

Dissimilatory Nitrate Reduction to Ammonium (DNRA) and Denitrification Pathways Are Leveraged by Cyclic AMP Receptor Protein (CRP) Paralogues Based on Electron Donor/Acceptor Limitation in Shewanella loihica PV-4.

Under anoxic conditions, many bacteria, including Shewanella loihica strain PV-4, could use nitrate as an electron acceptor for dissimilatory nitrate reduction to ammonium (DNRA) and/or denitrification. Previous and current studies have shown that DNRA is favored under higher ambient carbon-to-nitrogen (C/N) ratios, whereas denitrification is upregulated under lower C/N ratios, which is consistent with our bioenergetics calculations. Interestingly, computational analyses indicate that the common cyclic AMP receptor protein (designated CRP1) and its paralogue CRP2 might both be involved in the

Molecular mechanisms associated with polyphosphate accumulation in Microcystis aeruginosa under nitrate limitation.

Bloom-forming cyanobacteria frequently exhibit "luxury phosphorus uptake" and polyphosphate (PolyP) accumulation under nitrate-limited conditions, yet the underlying molecular regulatory mechanisms remain unclear. Our previous study quantitatively demonstrated PolyP accumulation in Microcystis aeruginosa under nitrate limitation. Building on this foundation, the present study investigated the molecular responses of M. aeruginosa to nitrate limitation using integrated transcriptomic and physiological approaches. Nitrate limitation markedly suppressed nitrogen assimilation, as evidenced by reduc

Gallium nitrate in the treatment of lymphoma.

Gallium nitrate is effective and well tolerated for the treatment of cancer-related hypercalcemia. At somewhat higher doses, gallium nitrate also has cytotoxic activity against a variety of cancers. The probable mechanism is inhibition of both ribonucleotide reductase and a protein tyrosine phosphatase. Radioactive gallium ((67)Ga) is concentrated at sites of malignant lymphoma, Hodgkin's disease, and other tumors. Gallium nitrate has substantial single-agent activity in the treatment of patients with advanced lymphoma and has also shown activity when used in combination with other agents. Sig

Integration of Non-Invasive Micro-Test Technology and 15N Tracing Reveals the Impact of Nitrogen Forms at Different Concentrations on Respiratory and Primary Metabolism in Glycyrrhiza uralensis.

Glycyrrhiza uralensis is a highly valued medicinal species worldwide. However, a paradox arises in its cultivation in that high nitrogen fertilization boosts yield at the expense of root quality, a problem linked to nitrogen's regulation of tricarboxylic acid (TCA) cycle-driven respiration. It remains unclear how different nitrogen forms coordinate respiratory and primary metabolism. We examined the regulatory mechanisms of nitrate (NO3-) versus ammonium (NH4+) on these processes in cultivated G. uralensis by supplying seedlings with varying concentrations of K15NO3 or (15NH4)2SO4 in a modifie

Mechanism exploration of divergent partial denitrification performance under tetracycline stress: Insights from functional gene, electron transport and molecular docking.

Nitrates and antibiotics like tetracycline (TC) coexist in wastewater and inhibit nitrite (NO₂--N) accumulation during partial denitrification (PD), restricting anammox coupling. A moving bed biofilm reactor (PD-MBBR) and a sequencing batch reactor (PD-SBR) were compared under TC stress (0-8 mg/L). The PD-MBBR proved more robust, sustaining a high nitrate transformation ratio (NTR) of 95.11% and ∼53% TC removal. Metagenomic sequencing, quantitative polymerase chain reaction (qPCR), and molecular docking revealed this tolerance stemmed from physical shielding and metabolic compensation. Carrier

Leghemoglobin is nitrated in functional legume nodules in a tyrosine residue within the heme cavity by a nitrite/peroxide-dependent mechanism.

Protein tyrosine (Tyr) nitration is a post-translational modification yielding 3-nitrotyrosine (NO2 -Tyr). Formation of NO2 -Tyr is generally considered as a marker of nitro-oxidative stress and is involved in some human pathophysiological disorders, but has been poorly studied in plants. Leghemoglobin (Lb) is an abundant hemeprotein of legume nodules that plays an essential role as an O2 transporter. Liquid chromatography coupled to tandem mass spectrometry was used for a targeted search and quantification of NO2 -Tyr in Lb. For all Lbs examined, Tyr30, located in the distal heme pocket, is t

Enhancement of cold-adapted heterotrophic nitrification and denitrification in Pseudomonas sp. NY1 by cupric ions: Performance and mechanism.

Cupric ions can restrain biological nitrogen removal processes, which comprise nitrite reductase and nitric oxide reductase. Here, Pseudomonas sp. NY1 can efficiently perform heterotrophic nitrification and aerobic denitrification with cupric ions at 15 °C. At optimal culturing conditions, low cupric ion levels accelerated nitrogen degradation, and ammonium and nitrite removal efficiencies increased by 2.33%-4.85% and 6.76%-12.30%, respectively. Moreover, the maximum elimination rates for ammonium and nitrite increased from 9.48 to 10.26 mg/L/h and 6.20 to 6.80 mg/L/h upon adding 0.05 mg/L cup
